Insilico engineering of transaminase variants for enhanced biocatalytic conversion of an ACE inhibitor precursor.

Angiotensin-converting enzyme (ACE) inhibitors are widely prescribed for cardiovascular disorders, yet their conventional chemical synthesis involves multiple steps, high energy consumption, and poor stereoselectivity. In this work, we present a fully insilico framework for the computational design and prediction of transaminase (TA) variants capable of catalyzing the asymmetric amination of ethyl 2-oxo-4-phenylbutanoate (OPBE) to yield L-homophenylalanine ethyl ester (L-HPE), a key chiral intermediate in ACE inhibitor synthesis. A homology model of the Silicibacter transaminase (62.7 % identi

The Dynamics of Transaminase and Alkaline Phosphatase Activities in the "Mother-Placenta-Fetus" Functional System.

Background: The timing of delivery depends on the condition of the fetus and the mother's body, which is reflected in both the incretion of enzymes in the pregnant woman's body and their use by the developing fetus in the anabolic process. The aim: This work was aimed to analyze the activities of transaminases (aspartate aminotransferase (AST) and alanine aminotransferase (ALT)) and alkaline phosphatase (AlPh) in liquid media (blood serum, amniotic fluid, umbilical cord blood, and placental homogenate) in pregnant women in each trimester of pregnancy and in the postpartum period, considering t

Design and Fabrication of a Versatile Microreactor to Study Transaminase-Based Heterogeneous Biocatalysts in Flow.

Microfluidics is a very attractive discipline for implementing more versatile high-throughput screening methods to improve enzymes. However, state-of-the-art microfluidics set-ups are mainly devoted to screening enzymes in solution, while screening of enzyme immobilization protocols using microfluidics is scarce. In this work, a microreactor device is designed, fabricated and applied to test different heterogeneous biocatalysts with transaminase activity. This microsystem set-up can be operated in two different modes: packed-bed (PBR) and fluidized (FBR) microreactors without exchanging the sa

Crystallization-Assisted Asymmetric Synthesis of Enantiopure Amines Using Membrane-Immobilized Transaminase.

The production of active pharmaceutical ingredients (APIs) requires enantiopure chiral amines, for which greener synthesis processes are needed. Transaminases (TAs) are enzymes that catalyze the enantioselective production of chiral amines from prochiral ketones through transamination under mild conditions. Yet, industrial applications of biocatalytic transamination remain currently hindered by the limited stability of soluble enzymes and by the unfavorable thermodynamic equilibrium of targeted asymmetric reactions. Enzyme immobilization can be applied to address stability, recoverability, and

To PLP or Not to PLP: Stereodivergent Transaminase-Catalyzed Reactions Directed by Kinetic and Thermodynamic Control.

Selective catalysis is a key objective in organic synthesis, and reactions with differing kinetic and thermodynamic products present the opportunity for divergent reaction outcomes with a single catalyst. We report a biocatalytic method in which a single transaminase can form either cis or trans cyclohexylamines in high diastereoselectivity. With the model substrate 4-methylcyclohexanone, E. coli cells expressing WT-Vf-ATA form either diastereomer of the amine product in >10:1 dr and >70% conversion, depending on reaction conditions, stereodivergence that also extends to a range of substrates.

[Semi-rational evolution of ω-transaminase from Aspergillus terreus for enhancing the thermostability].

ω-transaminase (ω-TA) is a natural biocatalyst that has good application potential in the synthesis of chiral amines. However, the poor stability and low activity of ω-TA in the process of catalyzing unnatural substrates greatly hampers its application. To overcome these shortcomings, the thermostability of (R)-ω-TA (AtTA) from Aspergillus terreus was engineered by combining molecular dynamics simulation assisted computer-aided design with random and combinatorial mutation. An optimal mutant AtTA-E104D/A246V/R266Q (M3) with synchronously enhanced thermostability and activity was obtained. Comp

Enzymatic synthesis of L-norephedrine by coupling recombinant pyruvate decarboxylase and ω-transaminase.

L-Norephedrine, a natural plant alkaloid, possesses similar activity as ephedrine and can be used as a vicinal amino alcohol for the asymmetric synthesis of a variety of optically pure compounds, including pharmaceuticals, fine chemicals, and agrochemicals. Because of the existence of two asymmetric centers, efficient synthesis of L-norephedrine has been challenging. In the present study, an R-selective pyruvate decarboxylase from Saccharomyces cerevisiae and an S-selective ω-transaminase from Vibrio fluvialis JS17 were coupled to develop a sequential process for the stereoselective biosynthes

Serum transaminase level changes in dengue fever and its correlation with disease severity.

It was observed that liver enzymes are elevated in dengue fever. In this study our aims were to determine the changes in serum transaminases in dengue fever, dengue hemorrhagic fever (DHF) and dengue shock syndrome (DSS) and to find out the relation of transaminase level changes with the disease severity. This cross sectional, prospective hospital based observational study was carried out in the department of Gastrointestinal Hepatobiliary and Pancreatic diseases and Internal Medicine department of BIRDEM Hospital, Dhaka. Patients are classified into 3 groups depending on clinical & laboratory

Correction of venlafaxine- and duloxetine-induced transaminase elevations with desvenlafaxine in a patient with Gilbert's syndrome.

Recent reviews have questioned whether the serotonin-norepinephrine reuptake inhibitor (SNRI) desvenlafaxine succinate offers any practical clinical advantages over existing SNRIs. The following case is one instance where it appears that this SNRI offers unique safety and benefit. Presented is a case report of a patient with Gilbert's syndrome, longstanding social phobia, and more recent depressive disorder not otherwise specified, who was found to have elevated liver transaminases when prescribed both duloxetine and venlafaxine. The patient subsequently responded to desvenlafaxine but without

Measuring human brain GABA in vivo: effects of GABA-transaminase inhibition with vigabatrin.

Gamma-aminobutyric acid (GABA) plays a pivotal role in suppressing the origin and spread of seizure activity. Low occipital lobe GABA was associated with poor seizure control in patients with complex partial seizures. Vigabatrin irreversibly inhibits GABA-transaminase, raising brain and cerebrospinal fluid (CSF) GABA concentrations. The effect of vigabatrin on occipital lobe GABA concentrations was measured by in vivo nuclear magnetic-resonance spectroscopy. Using a single oral dose of vigabatrin, the rate of GABA synthesis in human brain was estimated at 17% of the Krebs cycle rate. As the da

Isozyme patterns of branched-chain amino acid transaminase in human tissues and tumors.

The isozymes (enzymes I and III) of branched-chain amino acid transaminase (EC 2.6.1.42) from various human tissues were separated by DEAE-cellulose column chromatography. Their distributions were found to be considerably different from those in rat tissues reported before. In rats, all tissues examined contained enzyme I and in addition enzyme II was found in the liver and enzyme III in the brain, ovary, and placenta. In humans, however, enzyme II was not found in any tissues examined including the liver, and enzyme III was found in many other tissues besides brain, ovary, and placenta. All n
