The blood meal origins of 222 tsetse flies (213 Glossina palpalis palpalis, 7 Glossina pallicera pallicera, one Glossina nigrofusca and one Glossina caliginea) caught in 2008 in two Human African trypanosomiasis foci (Bipindi and Campo) of south Cameroon were investigated. 88.7% of tsetse flies blood meals were identified using the heteroduplex method and the origin of the remaining blood meals (11.3%) was identified by sequencing the cytochrome B gene. Most of the meals were from humans (45.9%) and pigs (37.4%), 16.7% from wild animals. Interestingly, new tsetse fly hosts including turtle (Tr

BACKGROUND: Tsetse flies (Glossina spp.) are the definitive biological vectors of African trypanosomes in humans and animals. Controlling this vector is the most promising method of preventing trypanosome transmission. This requires a comprehensive understanding of tsetse biology and host preference to inform targeted design and management strategies, such as the use of olfaction and visual cues in tsetse traps. No current review exists on host preference and blood meal analyses of tsetse flies.
METHODS: This review presents a meta-analysis of tsetse fly blood meal sources and the methodologi

We have previously shown that the slender form of Trypanosoma (T.) brucei is able to infect teneral tsetse flies, develop to the first fly form, which is the procyclic form, and complete the life cycle in the insect vector (Schuster et al., 2021). Further, analysis of the transmission index (TI; defined as the number of salivary gland infections relative to the number of midgut infections) revealed a higher TI for slender as compared to stumpy forms under laboratory conditions, which included the addition of N-acetylglucosamine (NAG) to the infective bloodmeal. Here, we show that slender trypa

Arthropod vectors have multiple physical and immunological barriers that impede the development and transmission of parasites to new vertebrate hosts. These barriers include the peritrophic matrix (PM), a chitinous barrier that separates the blood bolus from the midgut epithelia and modulates vector-pathogens interactions. In tsetse flies, a sleeve-like PM is continuously produced by the cardia organ located at the fore- and midgut junction. African trypanosomes, Trypanosoma brucei, must bypass the PM twice; first to colonize the midgut and secondly to reach the salivary glands (SG), to comple

Human African Trypanosomiasis (HAT) and Nagana in cattle, commonly called sleeping sickness, is caused by trypanosome protozoa transmitted by bites of infected tsetse flies. We present a deterministic model for the transmission of HAT caused by Trypanosoma brucei gambiense between human hosts, cattle hosts and tsetse flies. The model takes into account the growth of the tsetse fly, from its larval stage to the adult stage. Disease in the tsetse fly population is modeled by three compartments, and both the human and cattle populations are modeled by four compartments incorporating the two stage

BACKGROUND: Tsetse flies (Glossina) transmit Trypanosoma brucei gambiense which causes Gambian human African trypanosomiasis (gHAT) in Central and West Africa. Several countries use Tiny Targets, comprising insecticide-treated panels of material which attract and kill tsetse, as part of their national programmes to eliminate gHAT. We studied how the scale and arrangement of target deployment affected the efficacy of control.
METHODOLOGY AND PRINCIPAL FINDINGS: Between 2012 and 2016, Tiny Targets were deployed biannually along the larger rivers of Arua, Maracha, Koboko and Yumbe districts in N

Sterile Insect Technique (SIT) has proven effective to reduce tsetse population density in large infected areas where animal African trypanosomosis (AAT) and human African trypanosomiasis (HAT) elimination was difficult to achieve. However, the decrease in mass production of insectary-reared tsetse and the limited but incomplete knowledge on symbiont-trypanosome interaction over time, impede large-scale use of SIT. We investigated the spatiotemporal changes in symbiont prevalence and symbiont-trypanosome interactions in wild tsetse of Sora-Mboum AAT focus in northern Cameroon, collected in 201

The unicellular parasite Trypanosoma brucei is transmitted between mammals by tsetse flies. Following the discovery that flagellar phosphodiesterase PDEB1 is required for trypanosomes to move in response to signals in vitro (social motility), we investigated its role in tsetse flies. Here we show that PDEB1 knockout parasites exhibit subtle changes in movement, reminiscent of bacterial chemotaxis mutants. Infecting flies with the knockout, followed by live confocal microscopy of fluorescent parasites within dual-labelled insect tissues, shows that PDEB1 is important for traversal of the peritr

Between March 2004 and February 2005, the monthly incidence of trypanosome infections was measured in cattle from nine sentinel herds in the Adamaoua province of Cameroon. Three herds of 20 cattle each were kept on the plateau which has been cleared from tsetse flies about 10 years ago, three other herds were grazing in the tsetse infested valley whereas the last three were herded in the buffer zone. The cross-sectional study showed that the initial trypanosomosis prevalence was 1.8, 5.2 and 2.0% on the plateau, in the buffer zone and the valley, respectively. During the longitudinal study, th

BACKGROUND: Trypanosomoses are parasitic diseases caused by Trypanosoma protozoa transmitted by tsetse flies (Glossina spp.) to humans and animals. These diseases cause major health and economic disruptions in sub-Saharan Africa. Despite the development and wide implementation of control strategies, the disease burden remains high and complementary tools are needed. Ivermectin is an endectocide toxic to arthropods, including Glossina. The aim of this study was to test the efficacy of different doses of ivermectin administered to cattle on the survival and fecundity of Glossina palpalis gambien

In order to verify whether chronic trypanosomal infections can affect the transmissibility of Trypanosoma congolense by tsetse flies, batches of Glossina morsitans morsitans were fed on mice infected with the same level of parasitemia (10(8.1)trypanosomes/ml of blood) of two cloned low virulent T. congolense strains during the acute and the chronic phases of infection. Results showed that the proportions of procyclic infections in flies that were fed during the acute phase (32.6% and 45.4% for isolates 1 and 2, respectively) were significantly higher (chi(2)=4.7, P<0.05 and chi(2)=23.7, P<0.00

An epidemic of sleeping sickness, which started in 1976 in a focus within the county of Luuka in Central Busoga, has spread to cover the three districts of Busoga and large parts of the neighbouring districts of Tororo and Mukono. Forty-three isolates of the subgenus Trypanozoon from Busoga and Tororo (27 from man, 9 from cows, 2 from pigs and 5 from tsetse flies) were compared by thin-layer starch-gel electrophoresis for seven enzymes. Thirty zymodemes were identified; 17 of them were found circulating in the human population. The zymodemes seen previously in Busoga were still circulating tog

Tsetse-transmitted trypanosomosis remains a major constraint to the development of agriculture, particularly to that of livestock production in sub-Saharan Africa. It is estimated that 10 million km2 of Africa are tsetse infested, exposing some 50 million people and 60 million cattle to the risk of trypanosomosis. The epidemiology of the disease is complex and is greatly influenced by management and farming practices. The different control strategies are reviewed and their comparative advantages assessed. It is concluded that eradication of tsetse flies, while desirable, is rarely achieved. It

The interactions of host, vector and parasite in bovine trypanosomiasis transmission cycles in southwest Nigeria are not yet well understood. Trypanosoma (Trypanosomatida: Trypanosomatidae) species infection prevalences and bloodmeal sources were determined in transmitting vectors of the genera Glossina (Diptera: Glossinidae), Tabanus (Diptera: Tabanidae) and Stomoxys (Diptera: Muscidae) collected using Nzi traps in cattle settlements in southwest Nigeria. Sequenced cytochrome B mitochondrial DNA segments obtained from vector digestive tracts identified bloodmeal sources from eight host specie

BACKGROUND: As the reality of eliminating human African trypanosomiasis (HAT) by 2020 draws closer, the need to detect and identify the remaining areas of transmission increases. Here, we have explored the feasibility of using commercially available LAMP kits, designed to detect the Trypanozoon group of trypanosomes, as a xenomonitoring tool to screen tsetse flies for trypanosomes to be used in future epidemiological surveys.
METHODS AND FINDINGS: The DNA extraction method was simplified and worked with the LAMP kits to detect a single positive fly when pooled with 19 negative flies, and the

BACKGROUND: African trypanosomes are parasites mainly transmitted by tsetse flies. They cause trypanosomiasis in humans (HAT) and animals (AAT). In Chad, HAT/AAT are endemic. This study investigates the diversity and distribution of trypanosomes in Mandoul, an isolated area where a tsetse control campaign is ongoing, and Maro, an area bordering the Central African Republic (CAR) where the control had not started.
METHODS: 717 human and 540 cattle blood samples were collected, and 177 tsetse flies were caught. Trypanosomal DNA was detected using PCR targeting internal transcribed spacer 1 (ITS

African trypanosomiasis (AT) is a vector-borne disease caused by protozoan parasites of the genus Trypanosoma, primarily transmitted through the bite of infected tsetse flies in the genus Glossina. The disease is a major constraint to livestock production, causing reduced productivity, anemia, and mortality in affected animals. A cross-sectional study was conducted from November to December 2024 to assess the spatial prevalence of AAT in cattle as well as tsetse fly abundance in Busia County, Kenya. A total of 762 cattle randomly selected from seven villages in Busia County were screened for A

In Burkina Faso, we assessed the efficacy of treating cattle with a footbath containing aqueous formulations of pyrethroids to control two tsetse-fly species, Glossina tachinoides Westwood, 1850 (Diptera, Glossinidae) and Glossina palpalis gambiensis Vanderplank 1949. Legs were the most targeted parts of the body for tsetse-fly blood meals: 81% (95% CI: 73, 89) for G. tachinoides and 88% (81, 95) for G. palpalis. The in-stable efficacy of footbath treatments was compared with manual full spraying with a 0.005% alphacypermethrin (Dominex, FMC, Philadelphia, USA) formulation (250mL versus 2L). T

The tsetse fly Glossina is the vector of the protozoan Trypanosoma brucei spp., which causes Human and Animal African Trypanosomiasis in sub-Saharan African countries. To supplement their unbalanced vertebrate bloodmeal diet, flies permanently harbor the obligate bacterium Wigglesworthia glossinidia, which resides in bacteriocytes in the midgut bacteriome organ as well as in milk gland organ. Tsetse flies also harbor the secondary facultative endosymbionts (S-symbiont) Sodalis glossinidius that infects various tissues and Wolbachia that infects germ cells. Tsetse flies display viviparous repro

Palpalis group tsetse flies are the major vectors of human African trypanosomiasis, and visually-attractive targets and traps are important tools for their control. Considerable efforts are underway to optimise these visual baits, and one factor that has been investigated is coloration. Analyses of the link between visual bait coloration and tsetse fly catches have used methods which poorly replicate sensory processing in the fly visual system, but doing so would allow the visual information driving tsetse attraction to these baits to be more fully understood, and the reflectance spectra of ca
