Relationship between endothelial dependent vasodilation and size of abdominal aortic aneurysms.

BACKGROUND: Flow-mediated dilation of the brachial artery (FMDB) indirectly reflects the action of nitric oxide liberated by the endothelium. In patients with abdominal aortic aneurysms (AAA), the changes in nitric oxide metabolism in close association with inflammation, appear to play a leading role in the aetiopathology of this disease, although it is still not clear. The objective was to study the correlation and behavior of FMDB relative to the aneurysm diameter (AD). To evaluate the relationship between C-reactive protein (CRP) and the FMDB in these patients. To study the correlation and

Impaired endothelium-dependent flow-mediated vasodilation in hypertensive subjects with hyperaldosteronism.

BACKGROUND: Recent studies suggest that aldosterone may impair endothelium-dependent vascular function through suppression of nitric oxide formation. Assessments of forearm blood flow or arterial compliance suggest a similar effect in humans. The present study was designed to determine whether chronic aldosterone excess in subjects with resistant hypertension impairs endothelium-dependent vascular reactivity as indexed by direct assessment of brachial artery flow-mediated dilation (FMD). METHODS AND RESULTS: Consecutive subjects (n=80) with resistant hypertension were prospectively evaluated

Immediate exercise hyperemia: contributions of the muscle pump vs. rapid vasodilation.

A striking characteristic of the blood flow adaptation at exercise onset is the immediate and substantial increase in the first few (0-5 s) seconds of exercise. The purpose of this mini-review is to put into context the present evidence regarding mechanisms responsible for this phase of exercise hyperemia. One potential mechanism that has received much attention is the mechanical effect of muscle contraction (the muscle pump). The rapid vasodilatory mechanism(s) is another possible mechanism that has recently been shown to exist. This review will provide the reader with 1) an understanding of

Flavanol-rich cocoa induces nitric-oxide-dependent vasodilation in healthy humans.

BACKGROUND: Consumption of flavonoid-rich beverages, including tea and red wine, has been associated with a reduction in coronary events, but the physiological mechanism remains obscure. Cocoa can contain extraordinary concentrations of flavanols, a flavonoid subclass shown to activate nitric oxide synthase in vitro. OBJECTIVE: To test the hypothesis that flavanol-rich cocoa induces nitric-oxide-dependent vasodilation in humans. DESIGN: The study prospectively assessed the effects of Flavanol-rich cocoa, using both time and beverage controls. Participants were blinded to intervention; the en

Impaired vasodilation and nitric oxide synthase activity in glucocorticoid-induced hypertension.

Synthetic glucocorticoids are among the most widely prescribed medications by physicians. Although they have a vast array of beneficial effects such as immunosuppression and anti-inflammation, excess glucocorticoids can lead to iatrogenic Cushing's syndrome, which includes hypertension and cardiovascular disease. The exact mechanism by which glucocorticoids elevate blood pressure is not completely understood, but it appears to be a complex pathology that involves increased responsiveness to vasoconstrictors and decreased vasodilator production. Nitric oxide is a vasodilator that plays a key ro

Endothelium-independent and -dependent vasodilation in alkalotic and acidotic piglet lungs.

Although significant pulmonary hypertension can occur in patients treated with either hypocapnic alkalosis or "permissive" hypercapnic acidosis, the effects of sustained alkalosis or acidosis on subsequent vasodilator responses have not been established. This study measured the effects of 60-100 min of sustained alkalosis or acidosis on endothelium-independent and -dependent vasodilation with inhaled nitric oxide (iNO) and acetylcholine (ACh) in isolated lungs from 1-week-old piglets. After stabilization, lungs were divided into control (pH 7.40, PaCO(2) 40 torr, n = 5), alkalotic (pH 7.60, Pa

Vascular potassium channels mediate oxygen-induced pulmonary vasodilation in fetal lambs.

The pulmonary vascular resistance decreases at birth secondary to release of endothelium-derived nitric oxide (EDNO). EDNO release is a calcium-dependent process, and endothelial potassium (K+) channels regulate intracellular calcium flux. We investigated the hypothesis that potassium channels mediate oxygen-induced pulmonary vasodilation and EDNO release in fetal lambs. We instrumented 18 near-term fetal lambs at 122-126 days of gestation to measure pulmonary pressures, flow, and resistance. We studied hemodynamic effects of (1) 100% oxygen; (2) pinacidil, an ATP-sensitive K+ (KATP) channel a

Endothelial function in Marfan syndrome: selective impairment of flow-mediated vasodilation.

BACKGROUND: The cardiovascular complications of Marfan syndrome arise due to alterations in the structural and functional properties of fibrillin, a constituent of vascular connective tissues. Fibrillin-containing microfibrils are closely associated with arterial endothelial cells, indicating a possible functional role for fibrillin in the endothelium. Plasma concentrations of endothelial cell products are elevated in Marfan subjects, which indirectly indicates endothelial dysfunction. This study directly assessed flow- and agonist-mediated endothelium-dependent brachial artery reactivity in M

Nitric oxide and cutaneous active vasodilation during heat stress in humans.

Whether nitric oxide (NO) is involved in cutaneous active vasodilation during hyperthermia in humans is unclear. We tested for a role of NO in this process during heat stress (water-perfused suits) in seven healthy subjects. Two forearm sites were instrumented with intradermal microdialysis probes. One site was perfused with the NO synthase inhibitor NG-nitro-L-arginine methyl ester (L-NAME) dissolved in Ringer solution to abolish NO production. The other site was perfused with Ringer solution only. At those sites, skin blood flow (laser-Doppler flowmetry) and sweat rate were simultaneously an

Angiotensin II potentiates vasodilation of rat aorta by cAMP elevating agonists.

We previously observed an effect of the Ca++-mobilizing peptide, angiotensin II (ANG II), to potentiate agonist-stimulated adenosine 3',5'-cyclic monophosphate (cAMP) formation in rat cultured aortic smooth muscle cells. Consequently, it was postulated that the relaxant effects of dilator agents that act through cAMP formation would be enhanced in the presence of ANG II. To test this idea, we examined the influence of ANG II on agonist-induced relaxation of rat isolated aortic rings. Angiotensin II (0.1 microM) evoked a transient increase in the tone of KCI (30 mM)-precontracted aortae that re

Cerebral vasodilation after the thermocoagulation of the trigeminal ganglion in humans.

The resulting changes in the regional cerebral blood flow of 18 patients suffering from idiopathic trigeminal neuralgia and treated by selective thermocoagulation of the trigeminal ganglion were measured by xenon-133 emission tomography. One hour after thermal stimulation, there was an asymmetric increase (P < 0.05) in cerebral blood flow, with a 14.7% mean increase in the ipsilateral cerebral hemisphere (P < 0.001) and a 12.7% mean increase in the contralateral side (P < 0.01). The increase in regional cerebral blood flow was not uniform but was most marked in the ipsilateral middle cerebral

Antagonists of EDRF attenuate acetylcholine-induced vasodilation in isolated hamster lungs.

To evaluate the role of endothelium-dependent relaxing factor (EDRF) in acetylcholine- (ACh) induced vasodilation in the intact pulmonary circulation, we examined the effects of atropine and three EDRF antagonists that have been shown to be effective in vitro: nitro-L-arginine (NOARG), hemoglobin (Hb), and methylene blue (MB). We studied ACh-induced dilation after preconstriction with angiotensin II and prostaglandin F2 alpha (PGF2 alpha) in hamster lungs perfused with Krebs solution containing Ficoll (4 g/dl) and indomethacin (10 microM). Selective hemoglobin inhibition of endothelium-dependent vasodilation of rabbit basilar artery.

The effect of hemoglobin on endothelium-dependent vasodilation of the isolated rabbit basilar artery was examined using an isometric tension recording method. Acetylcholine (ACh) (10(-7) - (10(-4) M) evoked a dose-dependent vasodilation of isolated rabbit basilar artery previously contracted by 10(-6) M serotonin. This vasodilating action disappeared after removal of the endothelium. The ACh-induced vasodilation of rabbit basilar artery is thought to be strictly endothelium-dependent. Hemoglobin (10(-7) - 10(-5) M) inhibited this ACh-induced endothelium-dependent vasodilation conditional upon

Vasomotor effects of facial nerve stimulation: noncholinergic vasodilation in the eye.

The effect of facial nerve stimulation on ocular blood flow was studied in rabbits. The intracranial part of the facial nerve was stimulated electrically and the regional blood flow was measured with labelled microspheres. Effects on the intraocular pressure were determined in a separate series of experiments. Stimulation increased choroidal blood flow by about 200%. The blood flow of the iris and the ciliary processes also increased. The blood flow of the eyelids and the nictitating membrane increased by approximately 1000%, and the blood flow of Harder's gland increased by about 200%. The bl

Ganglion cells in arterioles of skeletal muscle: role in sympathetic vasodilation.

Ganglion cells were found in arterioles of gracilis muscles of dogs 2 wk after complete extrinsic denervation. We tested the possibility that they function in active sympathetic vasodilation (SVD) induced in isolated gracilis muscles by hypothalamic stimulation. To this end various drugs were injected into the gracilis perfusate. (Drugs did not reach the systemic circulation, and exerted their effects within the gracilis itself). C6 had no effect. Submaximal doses of atropine or l-hyoscyamine delayed and slowed SVD; higher doses blocked completely. d-Hyoscyamine did not change SVD or acetylcho

Singlet oxygen controls vasodilation through the activation of calcium signalling in astrocytes.

The central nervous system is highly active and one of the fastest consumers of oxygen. Thus, the brain requires immediate and effective oxygenation by arterial blood supply. Singlet oxygen is a non-radical form of oxygen which can be produced in cells by non-enzymatic and enzymatic means. Generation of non-toxic doses of singlet oxygen in a primary co-culture of neurons and astrocytes by 1267 nm laser induces calcium signalling in astrocytes via the induction of lipid peroxidation, activation of phospholipase C and production of IP3. [Ca2+]i rise in astrocytes triggers fusion of vesicular com

Chloroquine induces endothelium-dependent nitric oxide-mediated vasodilation in isolated rat aorta.

Toxic doses of chloroquine induce vasodilation, which contributes to hypotension. In this study, we aimed to investigate the involvement of endothelial nitric oxide in the vasodilatory effect of chloroquine on isolated rat aortas and elucidate the underlying mechanisms. We evaluated the effects of endothelial denudation and several inhibitors, including the nitric oxide synthase inhibitor NW-nitro-L-arginine methyl ester (L-NAME), the non-specific guanylate cyclase (GC) inhibitor methylene blue, the nitric oxide-sensitive GC inhibitor 1H-[1,2,4]oxadiazolo[4,3-a]quinoxalin-1-one (ODQ), and the

Quantification and interpretation of nitric oxide-dependent cutaneous vasodilation during local heating.

Human cutaneous microdialysis approaches for assessing nitric oxide (NO)-dependent blood flow include local heating (LH) of the skin until a plateau is reached, followed by infusion of a NO synthase inhibitor such as NG-nitro-l-arginine methyl ester (l-NAME); however, varied methods of quantifying and expressing NO-dependent vasodilation can obfuscate data interpretation and reproducibility. We retrospectively assessed NO-dependent vasodilation during LH to 39°C or 42°C, calculated as the 1) absolute contribution of the NO-dependent component (along with baseline and the non-NO-dependent compo
