Flagella are a positive predictor for virulence in Legionella.

Pathogenesis of Legionnaires>> disease is strictly related to the ability of the legionellae to infect phagocytic cells, yet surface markers of virulence in Legionella isolates are currently unknown. Rabbit antibodies raised against purified flagella of Legionella pneumophila serogroup 1 recognized a total of 24 of 30 laboratory-maintained isolates of L. pneumophila serogroups 1-15 and 16 of 24 other Legionella species tested by rapid immunoblot and indirect immunofluorescence assay. All isolates possessing flagella detectable with these anti-flagella antibodies, regardless of species, were ca

Ectoparasite virulence is linked to mode of transmission.

Theory suggests that the evolution of parasite virulence is linked to the dynamics of parasite transmission. All else being equal, parasites transmitted vertically from parents to offspring should be less virulent than parasites capable of horizontal transmission to unrelated hosts. This is because the fitness of vertically transmitted parasites is tightly linked to the reproductive success of the host, whereas the fitness of horizontally transmitted parasites is relatively independent of host reproduction. The virulence-transmission relation has seldom been tested because of difficulties inhe

The lipopolysaccharide of Shigella bacteria as a virulence factor.

The virulence factors of the lipopolysaccharide of Shigella species bacteria include the endotoxic activities of the lipid A component of the molecule and the ability of the polysaccharide chain--the core and the O-antigenic polysaccharide--to provide the bacterium with resistance to host defense mechanisms such as opsonization, phagocytosis, and intracellular killing. Structural features of the lipopolysaccharides of four Shigella species-S. dysenteriae, S. flexneri, S. boydii, and S. sonnei--are described.

Virulence and lienotoxicity of Bordetella bronchiseptica in mice.

Whole-cell suspensions (WCSs) and cell-free sonicated extracts (SEs) of seven Bordetella bronchiseptica strains were studied for lethality and lienotoxicity in mice. Lethality was assessed after intravenous and intracerebral inoculation, and lienotoxicity by splenic atrophy after intravenous inoculation. The strains represented phase I isolates with or without cytotoxin production, their phase III subcultures and a phase IV variant. The lethality and lienotoxicity of the SEs were in close positive correlation with cytotoxin production. The WCSs of all phase I strains were lethal, irrespective

Plaque assay for virulent Legionella pneumophila.

Methods of assessing virulence of Legionella pneumophila, the etiologic agent of Legionnaires disease, include the infection of guinea pigs, fertile chicken eggs, and mammalian and protozoan cell cultures. Guinea pig assays, in particular, are expensive, laborious, or unsuitable for routine screening of Legionella isolates. We have developed a virulence assay that requires the enumeration of viruslike plaques which are the result of virulent L. pneumophila infecting mouse L929 cells. Each plaque is the consequence of the initial infection of an L cell with a single bacterium. A nonvirulent mut

Identification and enumeration of virulent Listeria strains.

Recent outbreaks and a continuous increase in cases of listeriosis underscore the need for rapid, sensitive and reliable techniques to detect Listeria. Of the species of Listeria, only L. monocytogenes has been found to be associated with human infections. One factor which definitely contributes to its pathogenicity is the presence of hemolysins, although L. ivanovii and L. seeligeri also elaborate hemolysins. Based upon cloned hemolysin genes, we have developed DNA probes specifically for detecting L. monocytogenes. The technique combines growth of bacterial colonies on selective agar plates

The central metabolism regulator EIIAGlc switches Salmonella from growth arrest to acute virulence through activation of virulence factor secretion.

The ability of Salmonella to cause disease depends on metabolic activities and virulence factors. Here, we show that a key metabolic protein, EIIAGlc, is absolutely essential for acute infection, but not for Salmonella survival, in a mouse typhoid fever model. Surprisingly, phosphorylation-dependent EIIAGlc functions, including carbohydrate transport and activation of adenylate cyclase for global regulation, do not explain this virulence phenotype. Instead, biochemical studies, in vitro secretion and translocation assays, and in vivo genetic epistasis experiments suggest that EIIAGlc binds to

Assessment of hypermucoviscosity as a virulence factor for experimental Klebsiella pneumoniae infections: comparative virulence analysis with hypermucoviscosity-negative strain.

BACKGROUND: Klebsiella pneumoniae displaying the hypermucoviscosity (HV) phenotype are considered more virulent than HV-negative strains. Nevertheless, the emergence of tissue-abscesses-associated HV-negative isolates motivated us to re-evaluate the role of HV-phenotype. RESULTS: Instead of genetically manipulating the HV-phenotype of K. pneumoniae, we selected two clinically isolated K1 strains, 1112 (HV-positive) and 1084 (HV-negative), to avoid possible interference from defects in the capsule. These well-encapsulated strains with similar genetic backgrounds were used for comparative analy

The role of cytochrome oxidases in bacterial virulence.

Bacterial physiology plays an important role in virulence. Being a pathogen is energetically expensive, requiring the production of specialized virulence factors and other proteins that enable survival within the host. The most efficient way for bacteria to generate the energy required for virulence is by electron-coupled oxidative phosphorylation, with oxygen as the terminal acceptor, also called aerobic respiration. Cytochrome oxidases are critical components of the electron transport chain used in aerobic respiration. Studies on the role of cytochrome oxidases during infection in clinically

Effects of fisetin on virulence of Actinobacillus Pleuropneumoniae.

Porcine infectious pleuropneumonia (PCP), caused by Actinobacillus pleuropneumoniae (APP), is a highly infectious respiratory disease of pigs, resulting in significant economic losses to the industry. Apx toxins are cytolytic virulence factors produced by APP. The natural flavonoid fisetin has a variety of biological activities, including antioxidant and anti-inflammatory properties. This study aimed to investigate the protective effects of fisetin against APP. Fisetin was found to effectively inhibit the hemolytic activity of both APP culture supernatants and the purified toxins, and blood ag

The respiratory route of transmission of virulent polioviruses.

AIMS: The route of transmission of wild and circulating vaccine-derived polioviruses remains controversial, between respiratory and faecal-oral, and we aim to identify the most plausible one to settle the controversy. METHODS: We explored available epidemiological clues and evidence in support of either route in order to arrive at an evidence-based conclusion. RESULTS: Historically the original concept was respiratory transmission based on epidemiological features of age distribution, which was later revised to faecal-oral as the rationale for popularising the live attenuated oral polio vacc

On the determination of Toxoplasma gondii virulence in mice.

Toxoplasma gondii is one of the most successful pathogens on earth, capable of infecting an extremely broad range of mammals and birds and causing potentially fatal disease in humans. The house mouse (Mus musculus) has been used as the primary laboratory animal model for determining the virulence of T. gondii strains. Epidemiological evidence also suggests a potential association between virulence in mice and disease severity in human toxoplasmosis. However, many factors can affect virulence measurements, including route of infection, life stage of the parasite, number of passages of the paras

Role of dupA in virulence of Helicobacter pylori.

Helicobacter pylori (H. pylori) is a gastric human pathogen associated with acute and chronic gastritis, 70% of all gastric ulcers, 85% of all duodenal ulcers, and both forms of stomach cancer, mucosal-associated lymphoid tissue (MALT) lymphoma and adenocarcinoma. Recently, attention has focused on possible relationship between presence of certain virulence factor and H. pylori-associated diseases. Some contradictory data between this bacterium and related disorders has been observed since not all the colonized individuals develop to severe disease. The reported diseases plausibility related t

The Functions of Effector Proteins in Yersinia Virulence.

Yersinia species are bacterial pathogens that can cause plague and intestinal diseases after invading into human cells through the Three Secretion System (TTSS). The effect of pathogenesis is mediated by Yersinia outer proteins (Yop) and manifested as down-regulation of the cytokine genes expression by inhibiting nuclear factor-κ-gene binding (NF-κB) and mitogen-activated protein kinase (MAPK) pathways. In addition, its pathogenesis can also manipulate the disorder of host innate immune system and cell death such as apoptosis, pyroptosis, and autophagy. Among the Yersinia effector proteins, Yo

Importance of adhesins in virulence of Paracoccidioides spp.

Members of the Paracoccidioides genus are the etiologic agents of paracoccidioidomycosis (PCM). This genus is composed of two species: Paracoccidioides brasiliensis and Paracoccidioides lutzii. The correct molecular taxonomic classification of these fungi has created new opportunities for studying and understanding their relationships with their hosts. Paracoccidioides spp. have features that permit their growth under adverse conditions, enable them to adhere to and invade host tissues and may contribute to disease development. Cell wall proteins called adhesins facilitate adhesion and are cap

Poxviruses and the evolution of host range and virulence.

Poxviruses as a group can infect a large number of animals. However, at the level of individual viruses, even closely related poxviruses display highly diverse host ranges and virulence. For example, variola virus, the causative agent of smallpox, is human-specific and highly virulent only to humans, whereas related cowpox viruses naturally infect a broad spectrum of animals and only cause relatively mild disease in humans. The successful replication of poxviruses depends on their effective manipulation of the host antiviral responses, at the cellular-, tissue- and species-specific levels, whi

Role of CTR4 in the Virulence of Cryptococcus neoformans.

UNLABELLED: While research has identified an important contribution for metals, such as iron, in microbial pathogenesis, the roles of other transition metals, such as copper, remain mostly unknown. Recent evidence points to a requirement for copper homeostasis in the virulence of Cryptococcus neoformans based on a role for a CUF1 copper regulatory factor in mouse models and in a human patient cohort. C. neoformans is an important fungal pathogen that results in an estimated 600,000 AIDS-related deaths yearly. In the present studies, we found that a C. neoformans mutant lacking the CUF1-depende
