Grey matter (GM) alterations may contribute to cognitive decline in individuals with white matter hyperintensities (WMH) but no consensus has yet emerged. Here, we investigated cortical thickness and grey matter volume in 23 WMH patients with mild cognitive impairment (WMH-MCI), 43 WMH patients without cognitive impairment, and 55 healthy controls. Both WMH groups showed GM atrophy in the bilateral thalamus, fronto-insular cortices, and several parietal-temporal regions, and the WMH-MCI group showed more extensive and severe GM atrophy. The GM atrophy in the thalamus and fronto-insular cortice

This study was conducted to examine the neuropsychological effects of white matter and subcortical gray matter pathology. Nineteen patients with multiple sclerosis (MS), 16 with Huntington's disease (HD), and 17 normal controls (NC) participated. Participants completed the California Verbal Learning Test (CVLT), Rotary Pursuit (RP) and Mirror Tracing (MT) tasks, and the Symbol Digit Modalities Test (SDMT). The principal findings pertain to a dissociation in procedural memory: on RP, the HD group demonstrated impaired sequence learning compared to the MS group, which performed similarly to the

BACKGROUND AND OBJECTIVES: Many reversible brain MRI abnormalities have been described, among these the most frequently reported are cortical hyperintensities on FLAIR/T2 occurring during seizures. Much less attention has been given to those situations where White Matter goes Dark: subcortical white matter hypointensity on T2/FLAIR. Our aim is to identify the medical condition "Dark White Matter" (DWM) is more frequently associated with. This is the first systematic review on DWM.
METHODS: PubMed was searched in August 2023. Included studies were those reporting Diffuse Subcortical White Matt

BACKGROUND AND PURPOSE: Neuropsychiatric symptoms (NPS) are frequent in dementia and mild cognitive impairment (MCI), yet their reported associations with white matter hyperintensities (WMHs) remain inconsistent. We examined item-level associations between WMHs and NPS across Alzheimer's disease dementia (ADD), dementia with Lewy bodies (DLB), and amnestic MCI (aMCI).
METHODS: This multicenter retrospective study involved 523 patients: 276 with ADD, 67 with DLB, and 180 with aMCI. Three-dimensional T1-weighted and T2-fluid-attenuated inversion recovery magnetic resonance images were processed

BACKGROUND AND OBJECTIVES: White matter hyperintensities (WMH) of presumed vascular origin are a neuroimaging hallmark of cerebral small vessel disease (CSVD). Their spatial heterogeneity may reflect different clinical phenotypes. Most prior studies relied on principal component analysis to characterise such heterogeneity, which has limited ability to stratify individuals into discrete and interpretable WMH subtypes. We therefore propose a data-driven framework to identify WMH spatial subtypes, characterise their demographic and clinical profiles, and investigate their predictive value for fut

Posttraumatic stress symptoms (PTSS) are commonly experienced in people with chronic pain. Reduced white matter microstructural integrity in the uncinate fasciculus and the cingulum has separately been reported in chronic pain and posttraumatic stress disorder (PTSD) studies. However, the relationship between chronic pain, PTSS and white matter integrity remains unclear. This study aims to disentangle the relationship between PTSS severity and white matter microstructural integrity common across different chronic pain conditions. Thirty-six subjects with chronic pain and 20 without chronic pai

Mathematics is a complex skill requiring the coordination of distributed gray matter brain regions connected by white matter tracts. Diffusion tensor imaging (DTI) studies have revealed a network of white matter tracts that support math processing, but the specific microstructural features driving this relationship remain unclear. Other magnetic resonance imaging (MRI) methods-neurite orientation dispersion and density imaging (NODDI), inhomogeneous magnetization transfer (ihMT), multicomponent driven-equilibrium single-pulse observation of T1 and T2 (mcDESPOT), and g-ratio imaging-can probe m

Obsessive-compulsive disorder (OCD) emerging during adolescence coincides with a critical period of white matter maturation, yet the structural, functional, and molecular characteristics of white matter abnormalities remain incompletely understood. We acquired multimodal magnetic resonance imaging data from 40 adolescents with OCD and 40 healthy controls and quantified fractional anisotropy (FA) and fractional amplitude of low-frequency fluctuations (fALFF) along white matter tracts using Automated Fiber Quantification. Group comparisons, clinical correlation analyses, support vector machine c

Immune cells, especially regulatory T (Treg) cells, contribute to strengthening white matter integrity, yet little is known about the underlying mechanism. To examine this, we established a transient middle cerebral artery occlusion male mouse model. We found that physical exercise elevated brain Treg cells, thereby enhancing neurological recovery, reducing neuroinflammation, promoting myelin debr

INTRODUCTION: In our previous study, we have identified white matter subnetworks linked to phonological processing deficits (e.g., subnetworks centered at the left middle temporal gyrus) in dyslexic children from alphabetic languages using a data-driven hub-based white matter network analysis approach. Yet, white matter subnetworks associated with phonological processing skills in individuals from nonalphabetic languages (e.g., Chinese) have never been studied. This study aims to identify hub-related white matter networks associated with phonological processing skills in Chinese readers.

In recent years, the development of neonatal brain networks has become a research focus, with traditional studies primarily emphasizing gray matter (GM) functional networks. This study systematically explores the developmental characteristics of white matter (WM) functional networks in neonates. Utilizing data from the third release of the Developing Human Connectome Project (dHCP), we analyzed resting-state functional magnetic resonance imaging (rs-fMRI) data from 730 full-term and 157 preterm neonates. We successfully identified ten large-scale WM functional networks and validated their corr

This study aimed to develop an automatic segmentation method for brainstem fiber bundles. We utilized the brainstem as a seed region for probabilistic tractography based on multishell, multitissue constrained spherical deconvolution in 40 subjects from the Human Connectome Project (HCP). All tractography data were registered into a common space to construct a brainstem fiber cluster atlas. A total of 100 fiber clusters were identified and annotated. Cortical parcellation-based fiber selection was then performed to extract fibers within the annotated clusters that projected to their correspondi

Hallucinations are a prominent transdiagnostic psychiatric symptom but are also prevalent in individuals who do not require clinical care. Moreover, persistent psychosis-like experience in otherwise healthy individuals may be related to an increased risk to transition to a psychotic disorder. This suggests a common etiology across clinical and non-clinical individuals along a multidimensional psychosis continuum that may be detectable in structural variations of the brain. The current diffusion tensor imaging study assessed 50 healthy individuals (35 females) to identify possible differences i

White matter signals in resting state blood oxygen level dependent functional magnetic resonance (BOLD-fMRI) have been largely discounted, yet there is growing evidence that these signals are indicative of brain activity. Understanding how these white matter signals capture function can provide insight into brain physiology. Moreover, functional signals could potentially be used as early markers for neurological changes, such as in Alzheimer's Disease. To investigate white matter brain networks, we leveraged the OASIS-3 dataset to extract white matter signals from resting state BOLD-FMRI data

Damage or microstructural alterations of the white matter can cause dysfunction of the intrinsic neural networks in a condition termed as white matter disease (WMD). Frequently detected on brain computed tomography and magnetic resonance imaging scans, WMD is commonly presented in inflammatory demyelinating diseases like multiple sclerosis (MS) and vascular diseases such as cerebral small vessel disease (CSVD). Prevention of MS and CSVD progression requires early treatments with drastically different medications and approaches, as such, early and accurate diagnosis of WMD, derived from vascula

Familial cerebral amyloid disorders are characterized by the accumulation of fibrillar protein aggregates, which deposit in the parenchyma as plaques and in the vasculature as cerebral amyloid angiopathy (CAA). Amyloid β (Aβ) is the most common of these amyloid proteins, accumulating in familial and sporadic forms of Alzheimer's disease and CAA. However, there are also a number of rare, hereditary, non-Aβ cerebral amyloidosis. The clinical manifestations of these familial cerebral amyloid disorders are diverse, including cognitive or neuropsychiatric presentations, intracerebral hemorrhage, se

Previous studies have shown that the cholinergic nucleus basalis of Meynert and its white matter projections are affected in Alzheimer's disease dementia and mild cognitive impairment. However, it is still unknown whether these alterations can be found in individuals with subjective cognitive decline, and whether they are more pronounced than changes found in conventional brain volumetric measurements. To address these questions, we investigated microstructural alterations of two major cholinergic pathways in individuals along the Alzheimer's disease continuum using an in vivo model of the hum

BACKGROUND: White matter hyperintensities are an important marker of cerebral small vessel disease. This disease burden is commonly described as hyperintense areas in the cerebral white matter, as seen on T2-weighted fluid attenuated inversion recovery magnetic resonance imaging data. Studies have demonstrated associations with various cognitive impairments, neurological diseases, and neuropathologies, as well as clinical and risk factors, such as age, sex, and hypertension. Due to their heterogeneous appearance in location and size, studies have started to investigate spatial distributions an

Diffusion MRI has provided insight into the widespread structural connectivity changes that characterize epilepsies. Although syndrome-specific white matter abnormalities have been demonstrated, studies to date have predominantly relied on statistical comparisons between patient and control groups. For diffusion MRI techniques to be of clinical value, they should be able to detect white matter microstructural changes in individual patients. In this study, we apply an individualized approach to a technique known as fixel-based analysis, to examine fibre-tract-specific abnormalities in individua
