Old disease, new targets. Part-II, haematological malignancies.

In order to understand the physiological role of the caudate nucleus, we combine here our laboratory data on cats with reports of patients with selective damage to this nucleus. Cats with bilateral removal of the caudate nuclei showed a stereotyped behavior consisting of persistently approaching and then following a person, another cat, or any object, and attempting to contact the target. Simultaneously, the animals exhibited a friendly disposition and persistent docility together with purring and forelimbs treading/kneading. The magnitude and duration of this behavior was proportional to the

Acupuncture analgesia in a new rat model of ankle sprain pain.

The lack of suitable experimental animal models for persistent pain showing clear acupuncture analgesia, has been the major stumbling block in the investigation of the physiological mechanisms of acupuncture analgesia. The present study developed a new rat model of ankle sprain pain and the effect of electroacupuncture (EA) on this model was examined. A common source of persistent pain in humans is the lateral ankle sprain. To model this condition, the rat's right ankle was bent repeatedly, overextending lateral ligaments, for 4 min under halothane anesthesia. The rat subsequently showed swell

Fos induction following systemic kainic acid: early expression in hippocampus and later widespread expression correlated with seizure.

We determined the distribution of Fos protein expression in a model of generalised epilepsy caused by excessive neuronal excitation. Fos immunoreactivity was mapped in forebrain in unrestrained rats, previously prepared with an indwelling venous catheter, after the intravenous administration of kainic acid (10 mg/kg). We determined cerebral activation following various periods of exposure to kainic acid by using intravenous administration of pentobarbitone to prevent further activation. Within a few minutes, kainic acid caused episodes of staring, sniffing, wet dog shakes, nodding and chewing.

Distinct Motor Cortex Somatotopy in Experimental Alzheimer's Disease.

Alzheimer's Disease (AD) and related dementias (AD/RDs) impact cortical motor and sensory biology whereas the precise changes to these systems, and their clinical relevance, remain under debate. We hypothesized that cortical representations of complex and simple movements are differently altered during disease progression in 5XFAD mice, a well-established model of AD. Motor cortex somatotopy was determined in 5XFAD and Wild-Type Control (WT Control) mice at 6 and 12 months (mos.) of age using long-duration intracortical microstimulation (LD-ICMS) to systematically identify cortical sites evoki

Intramuscular Antibiotics Modify Infectious Outcomes in Contaminated, Ischemic Limbs.

INTRODUCTION: Tourniquets are sometimes necessary for hemorrhage control in the setting of traumatic limb injuries but may increase the risk of ischemic complications, including infection and impaired wound healing. Antibiotics may help to reduce infection rates in combat-associated injuries; however, intravenous access is not always readily available, particularly in austere environments. This study evaluates the effect of ertapenem administered intramuscularly at the tourniquet site on infection rates in a 24-hour porcine contaminated wound model. MATERIALS AND METHODS: Yorkshire pigs (n =

Texture analysis of T1- and T2-weighted images identifies myofiber atrophy and grip strength decline in streptozotocin-induced type 1 diabetic sarcopenia rats.

BACKGROUND: Type 1 diabetes mellitus is associated with accelerated skeletal muscle aging and sarcopenia, a condition characterized by muscle mass and function loss. Early and noninvasive evaluation of muscle microstructural damage is critical for managing sarcopenia in diabetes. This study evaluated the potential of MRI texture analysis as a noninvasive imaging tool to assess myofiber size and grip strength alterations in a rat model of diabetic sarcopenia. MATERIALS AND METHODS: Twenty SD rats were randomly and evenly allocated to the control (CON) and diabetic sarcopenia (DS) groups. Blood

Atrazine exposure induces abnormal swimming behavior of tadpoles under light and/or dark stimuli: A comprehensive multi-omics insights from eyes and brain.

Atrazine, a widely used pesticide, can damage organs and affect the respond ability of aquatic animals to environmental stimuli. To explore the amphibian response to light and/or dark stimuli following pesticide exposure, a comparative analysis was conducted on the swimming behavior of Pelophylax nigromaculatus tadpoles (Gs 8 - Gs 36, from fertilised egg to forelimb appearance) following a 60-day exposure period to atrazine. Additionally, an examination of ocular structures, eye metabolism, and brain transcription was undertaken across the treatment groups. This comprehensive approach aimed to

Investigating the pharmacodynamic mechanism of Erxian decoction to alleviate sarcopenia generated by ovarian aging through the ER-mediated estrogen signaling pathway.

ETHNOPHARMACOLOGICAL RELEVANCE: Erxian decoction (EXD) is widely used in the treatment of various gynecological conditions, including menopausal syndrome, ovarian insufficiency, and premature ovarian failure (POF). As society ages, the prevalence of postmenopausal osteoporosis (PMOP) rises annually, accompanied by a progressive increase in the incidence of sarcopenia, a skeletal muscle disorder associated with osteoporosis. Recent studies have revealed that the diverse pharmacological agents and active compounds in EXD have been utilized in clinical and experimental contexts for sarcopenia; ne

Constraint-induced movement therapy combined with mesenchymal stem cell transplantation promotes myelination and functional recovery by inhibiting PRKCD/MEK/ERK pathway in hemiplegic cerebral palsy rats.

BACKGROUND: The core problem of hemiplegic cerebral palsy (HCP) is upper limb motor deficits with high rates of disability. Given the shared goals of stem cell therapy and rehabilitation, this study investigated the synergistic effects of constraint-induced movement therapy (CIMT) and human umbilical cord-derived mesenchymal stem cells (hUC-MSCs) transplantation in promoting motor recovery and elucidates the underlying mechanisms in HCP. METHODS: The rats were allocated to a control group and HCP groups receiving different interventions (CIMT, hUC-MSCs or combination treatment), namely, the C

3D biological scaffold delivers Bergenin to reduce neuroinflammation in rats with cerebral hemorrhage.

BACKGROUND: Intracerebral hemorrhage (ICH) is a severe form of stroke characterized by high incidence and mortality rates. Currently, there is a significant lack of effective treatments aimed at improving clinical outcomes. Our research team has developed a three-dimensional (3D) biological scaffold that incorporates Bergenin, allowing for the sustained release of the compound. METHODS: This 3D biological scaffold was fabricated using a combination of photoinitiator, GEMA, silk fibroin, and decellularized brain matrix (dECM) to encapsulate Bergenin through advanced 3D bioprinting techniques.

How might we build limbs in vitro informed by the modular aspects and tissue-dependency in limb development?

Building limb morphogenesis in vitro would substantially open up avenues for research and applications of appendage development. Recently, advances in stem cell engineering to differentiate desired cell types and produce multicellular structures in vitro have enabled the derivation of limb-like tissues from pluripotent stem cells. However, in vitro recapitulation of limb morphogenesis is yet to be achieved. To formulate a method of building limbs in vitro, it is critically important to understand developmental mechanisms, especially the modularity and the dependency of limb development on the

P2Y6 Receptor Activation Aggravates NLRP3-dependent Microglial Pyroptosis via Downregulation of the PI3K/AKT Pathway in a Mouse Model of Intracerebral Hemorrhage.

Pro-inflammatory signals generated after intracerebral hemorrhage (ICH) trigger a form of regulated cell death known as pyroptosis in microglia. White matter injury (WMI) refers to the condition where the white matter area of the brain suffers from mechanical, ischemic, metabolic, or inflammatory damage. Although the p2Y purinoceptor 6 (P2Y6R) plays a significant role in the control of inflammatory reactions in central nervous system diseases, its roles in the development of microglial pyroptosis and WMI following ICH remain unclear. In this study, we sought to clarify the role of P2Y6R in mic

Growth hormone secretagogues modulate inflammation and fibrosis in mdx mouse model of Duchenne muscular dystrophy.

INTRODUCTION: Growth hormone secretagogues (GHSs) exert multiple actions, being able to activate GHS-receptor 1a, control inflammation and metabolism, to enhance GH/insulin-like growth factor-1 (IGF-1)-mediated myogenesis, and to inhibit angiotensin-converting enzyme. These mechanisms are of interest for potentially targeting multiple steps of pathogenic cascade in Duchenne muscular dystrophy (DMD). METHODS: Here, we aimed to provide preclinical evidence for potential benefits of GHSs in DMD, via a multidisciplinary in vivo and ex vivo comparison in mdx mice, of two ad hoc synthesized compoun

Combination of epidural electrical stimulation with ex vivo triple gene therapy for spinal cord injury: a proof of principle study.

Despite emerging contemporary biotechnological methods such as gene- and stem cell-based therapy, there are no clinically established therapeutic strategies for neural regeneration after spinal cord injury. Our previous studies have demonstrated that transplantation of genetically engineered human umbilical cord blood mononuclear cells producing three recombinant therapeutic molecules, including vascular endothelial growth factor (VEGF), glial cell-line derived neurotrophic factor (GDNF), and neural cell adhesion molecule (NCAM) can improve morpho-functional recovery of injured spinal cord in

Animal models used to study direct peripheral nerve repair: a systematic review.

OBJECTIVE: Peripheral nerve repair is required after traumatic injury. This common condition represents a major public health problem worldwide. Recovery after nerve repair depends on several factors, including the severity of the injury, the nerve involved, and the surgeon's technical skills. Despite the precise microsurgical repair of nerve lesions, adequate functional recovery is not always achieved and, therefore, the regeneration process and surgical techniques are still being studied. Pre-clinical animal models are essential for this research and, for this reason, the focus of the presen

Effects of PDE5 inhibition on dystrophic muscle following an acute bout of downhill running and endurance training.

Lack of sarcolemma-localized neuronal nitric oxide synthase mu (nNOSμ) contributes to muscle damage and fatigue in dystrophic muscle. In this study, we examined the effects of compensating for lack of nNOSμ with a phosphodiesterase type 5 (PDE5) inhibitor in mdx mice following downhill running and endurance training. Dystrophic mice (mdx) were treated with sildenafil citrate and compared with untreated mdx and wild-type mice after an acute bout of downhill running and during a progressive low-intensity treadmill running program (5 days/wk, 4 wk). Ryanodine channel complex stabilizer compound S48168/ARM210 as a disease modifier in dystrophin-deficient mdx mice: proof-of-concept study and independent validation of efficacy.

Muscle fibers lacking dystrophin undergo a long-term alteration of Ca2+ homeostasis, partially caused by a leaky Ca2+ release ryanodine (RyR) channel. S48168/ARM210, an RyR calcium release channel stabilizer (a Rycal compound), is expected to enhance the rebinding of calstabin to the RyR channel complex and possibly alleviate the pathologic Ca2+ leakage in dystrophin-deficient skeletal and cardiac muscle. This study systematically investigated the effect of S48168/ARM210 on the phenotype of mdx mice by means of a first proof-of-concept, short (4 wk), phase 1 treatment, followed by a 12-wk trea

Characterization of Rett Syndrome-like phenotypes in Mecp2-knockout rats.

BACKGROUND: Rett Syndrome (RTT) is a neurodevelopmental disease caused by the disruption of the MECP2 gene. Several mouse models of RTT have been developed with Mecp2 disruptions. Although the mouse models are widely used in RTT research, results obtained need to be validated in other species. Therefore, we performed these studies to characterize phenotypes of a novel Mecp2 (-/Y) rat model and compared them with the Mecp2 (tm1.1Bird) mouse model of RTT. METHODS: RTT-like phenotypes were systematically studied and compared between Mecp2 (-/Y) rats and Mecp2 (-/Y) mice. In-cage conditions of th
