Visceral adipose tissue-derived serine protease inhibitor augments acetylcholine-induced relaxation via the inhibition of acetylcholine esterase activity in rat isolated mesenteric artery.

AIM: Visceral adipose tissue-derived serine protease inhibitor (vaspin) is an adipocytokine with insulin-sensitizing activity originally identified in visceral adipose tissues of obesity-related type II diabetic rats. We previously showed that vaspin inhibits vascular cell migration and apoptosis as well as inflammatory responses, which are crucial for the development of hypertension. However, little is known about the effects of vaspin on vascular reactivity. The aim of this study was thus to explore the effects of vaspin on contraction and relaxation of isolated blood vessel. METHODS: After

Acetylcholine-related bowel dysmotility in homozygous mutant NCX/HOX11L.1-deficient (NCX-/-) mice-evidence that acetylcholine is implicated in causing intestinal neuronal dysplasia.

BACKGROUND/PURPOSE: Homozygous mutant Ncx/Hox11L.1-deficient (Ncx-/-) mice develop mega-ileo-ceco-colon (mega-ICC) with a caliber change in the proximal colon. The authors investigated the mechanism of intestinal dysmotility in these mice. METHODS: Five-week-old Ncx-/- mice with mega ICC were compared with age-matched BDF1 control mice. Jejunum, ileum, and colon were excised from all mice and 1.0-cm-long strips of each organ, each with a resting tension of 0.5g, were suspended in an organ bath filled with Tyrode's solution at 37 degrees C and bubbled with a mixture of 95% oxygen and 5% carbon

Desensitization of membrane-bound Torpedo acetylcholine receptor by amine noncompetitive antagonists and aliphatic alcohols: studies of [3H]acetylcholine binding and 22Na+ ion fluxes.

Measurements of the kinetics of binding of [3H]acetylcholine ([3H]AcCh) to membrane-bound nicotinic AcCh receptors from Torpedo electric tissue have been used to characterize the effects of a series of amine and alcohol noncompetitive antagonists on receptor conformational equilibria. The receptor exists in multiple, interconvertible conformations distinguished by agonist binding affinity. In the absence of cholinergic ligands, certain aromatic amines including proadifen, dimethisoquin, and lidocaine, as well as propanol and butanol, produce a dose-dependent increase in the fraction of recepto

Nicotinic acetylcholine receptors and their role in ghrelin-producing cells.

Gastric orexigenic peptide ghrelin has been implicated in tobacco addiction, but there are conflicting results regarding the effects of acute smoke on the plasma ghrelin levels. In this study, we screened the effects of tobacco-related chemicals, especially focusing on the effects of nicotine on ghrelin regulation at cellular level. Nicotine alone modestly stimulated ghrelin secretion, and enhanced the stimulatory effects of isoproterenol and oxytocin on ghrelin secretion from ghrelin-producing MGN3-1 cells evaluated by a newly developed ghrelin-HiBiT assay system. MGN3-1 cells expressed high

Multidimensional roles of acetylcholine receptors in neurobiology and their therapeutic significance.

Dysfunction of cholinergic signaling has emerged as a convergent mechanism across neurological and neuromuscular disorders, linking disturbances in synaptic, immune, and metabolic regulation. This review summarizes recent structural, functional, and imaging advances, demonstrating that nicotinic and muscarinic acetylcholine receptors play central roles in neural plasticity, neurotransmission, and neuroimmune homeostasis. Studies in structural biology and molecular pharmacology have clarified receptor architectures and conformational transitions, which enhance our understanding of subtype-speci

[Molecular mechanism of verbascoside in promoting acetylcholine release of neurotransmitter].

The molecular mechanism of verbascoside(OC1) in promoting acetylcholine(ACh) release in the pathogenesis of Alzheimer's disease(AD) was studied. Adrenal pheochromocytoma cells(PC12) of rats induced by β-amyloid protein(1-42)(Aβ_(1-42)) were used as AD models in vitro and were divided into control group, model group(Aβ_(1-42) 10 μmol·L~(-1)), OC1 treatment group(2 and 10 μg·mL~(-1)). The effect of OC1 on phosphorylated proteins in AD models was analyzed by whole protein phosphorylation quantitative omics, and the selectivity of OC1 for calcium channel subtypes was virtually screened in combinat

[Congenital myasthenic syndromes with kinetic abnormalities of the acetylcholine receptor].

Congenital myasthenic syndromes (CMS) are genetically and phenotypically very heterogeneous conditions resulting in a defect in the neuromuscular transmission. Post-synaptic forms are the most frequent CMSs, and acetyl choline receptor (low expressor) deficiency is the most commonly involved pathophysiological mechanism. CMS with kinetic abnormalities of the acetylcholine receptor (AChr) are much rarer and can give rise to potentially life-threatening phenotypes. Among them, two types have been described: the slow channel syndrome (SCS) and the fast channel syndrome (FCS). Diagnosis and therap

Chemotaxis of the Human Pathogen Pseudomonas aeruginosa to the Neurotransmitter Acetylcholine.

Acetylcholine is a central biological signal molecule present in all kingdoms of life. In humans, acetylcholine is the primary neurotransmitter of the peripheral nervous system; it mediates signal transmission at neuromuscular junctions. Here, we show that the opportunistic human pathogen Pseudomonas aeruginosa exhibits chemoattraction toward acetylcholine over a concentration range of 1 μM to 100 mM. The maximal magnitude of the response was superior to that of many other P. aeruginosa chemoeffectors. We demonstrate that this chemoattraction is mediated by the PctD (PA4633) chemoreceptor. Usi

Fast desensitization of acetylcholine receptors induced by a spider toxin.

Nicotinic acetylcholine receptors (nAChRs) are members of the "cys-loop" ligand-gated ion channel superfamily that play important roles in both the peripheral and central system. At the neuromuscular junction, the endplate current is induced by ACh binding and nAChR activation, and then, the current declines to a small steady state, even though ACh is still bound to the receptors. The kinetics of nAChRs with high affinity for ACh but no measurable ion conductance is called desensitization. This adopted desensitization of nAChR channel currents might be an important mechanism for protecting cel

Role of the α7 Nicotinic Acetylcholine Receptor in the Pathophysiology of Atherosclerosis.

Atherosclerosis constitutes a major risk factor for cardiovascular diseases, the leading cause of morbidity and mortality worldwide. This slowly progressing, chronic inflammatory disorder of large- and medium-sized arteries involves complex recruitment of immune cells, lipid accumulation, and vascular structural remodeling. The α7 nicotinic acetylcholine receptor (α7nAChR) is expressed in several cell types involved in the genesis and progression of atherosclerosis, including macrophages, dendritic cells, T and B cells, vascular endothelial and smooth muscle cells (VSMCs). Recently, the α7nACh

Direct and indirect action modes of acetylcholine in cholinergic urticaria.

Cholinergic urticaria (CholU) manifests small, itchy and/or painful wheals occurring upon perspiration and mechanically involving acetylcholine (Ach). Although a considerable number of studies have been conducted, the pathomechanisms underlying perspiration-associated release of histamine remain to be elucidated. We have proposed that CholU can be categorized into two major subtypes: Ach-indirectly induced, sweat allergic type and Ach-directly induced, depressed sweating type. In the former type, Ach evokes perspiration, and some sweat antigen(s) leaking from the sweat ducts to the dermis may

α9-containing nicotinic acetylcholine receptors and the modulation of pain.

UNLABELLED: Neuropathic pain is a complex and debilitating syndrome for which there are few effective pharmacological treatments. Opioid-based medications are initially effective for acute pain, but tolerance to their analgesic effects quickly develops, and long-term use often leads to physical dependence and addiction. Furthermore, neuropathic pain is generally resistant to non-steroidal anti-inflammatory drugs. Other classes of medications including antidepressants, antiepileptics and voltage-gated calcium channel inhibitors are only partially effective in most patients, may be associated wi

Role of antibodies to neuronal α7-acetylcholine receptors in myasthenia.

The role of antibodies to a fragment of neuronal acetylcholine receptor was studied by EIA in patients with myasthenia. Antibody levels were significantly higher in patients with generalized myasthenia. Enzyme immunoassay of antibodies by the reaction with acetylcholine receptor fragment can serve as an additional method for studies of autoimmune myasthenia pathogenesis.

Motion sickness may be caused by a neurohumoral action of acetylcholine.

The mechanism by which motion stimulation results in the autonomic responses, known as motion sickness, has remained somewhat of an enigma. Neural connections between the vestibular nuclei and the autonomic and emetic centers in the brainstem have been described, but these appear to be relatively sparse. Thus, new or additional mechanisms seem warranted to account for this curious relationship. A new hypothesis is herein presented which posits that acetylcholine (ACh) acts as a neurohumeral agent to bring about the symptoms associated with motion sickness. Motion stimulation will activate prim

Regulation of self-incompatibility by acetylcholine and cAMP in Lilium longiflorum.

Elongation of pollen tubes in pistils of Lilium longiflorum cv. Hinomoto after self-incompatible pollination was here found to be promoted by acetylcholine (ACh) and other choline derivatives, such as acetylthiocholine, l-alpha-phosphatidylcholine and chlorocholinechloride [CCC; (2-chloroethyl) trimethyl ammonium chloride]. Moreover, the elongation was promoted by neostigmine, a potent inhibitor of acetylcholinesterase (AChE; acetylcholine-decomposing enzyme) (EC 3.1.1.7.) and activities of this and choline acetyltransferase (ChAT; acetylcholine-forming enzyme) (EC 2.3.1.6.) in pistils were as

Efficacy of naltrexone on acetylcholine-induced alloknesis in atopic eczema.

Atopic eczema (AE) is a chronically pruritic inflammatory skin disease. Although the mediators and exact mechanisms eliciting and sustaining pruritus are not completely known, AE patients in clinical trials have been shown to benefit under treatment with morphine antagonists. Naltrexone (NAL) is a relatively pure morphine antagonist that blocks the effects of opioids twice as much as naloxone. NAL exhibits minimal pharmacological activity and displaces endorphines at mu- and kappa-receptors without its own intrinsic activity. NAL's excellent oral bioavailability and linear increases in the are

Neuronal nicotinic acetylcholine receptors: from the gene to the disease.

The neuronal nicotinic acetylcholine receptors are excitatory ligand-gated channels. Widely expressed throughout the peripheral and central nervous system, their properties depend upon their subunit composition. Furthermore, genetic studies have revealed a high degree of variation at the genomic level and alternative splicing of the mRNAs coding for these integral membrane proteins. In particular, genes coding for alpha4 and alpha7 subunits harbour a high degree of polymorphisms. Although well characterised at their molecular and functional level, the role of these receptors in the central ner

Day-night rhythm of acetylcholine in the rat pineal gland.

Using high-performance-liquid-chromatography (HPLC) measurement of acetylcholine, choline acetyltransferase (ChAT) enzyme assay and anti-ChAT immunohistochemistry, we have investigated the expression of the cholinergic system in pineal glands of male rats. Glands procured during the day period (1200 h) contained significant amounts of acetylcholine (0.5 pmol/gland). A similar content was found in pineal glands after a 48 h culture period, i.e. when the intrapineal nerve fibres have degenerated. This strongly indicates that the pinealocytes are the cells which contain acetylcholine. To confirm
