The global dissemination of carbapenem-resistant Acinetobacter baumannii (CRAB) is primarily driven by the expansion of two major international clones, IC1 and IC2, which have achieved widespread geographic distribution. Their success is further facilitated by the acquisition and dissemination of resistance determinants in healthcare settings. However, the evolutionary dynamics and genomic features of dominant clones in endemic settings such as Thailand remain insufficiently characterized. In this study, comparative genomic analyses were conducted on 650 A. baumannii genomes, including 38 newl

Carbapenem-resistant Acinetobacter baumannii (CRAB) is a major global public health threat. The World Health Organization (WHO) has classified CRAB as a critical priority pathogen. This threat is mainly attributed to its multidrug resistance, efficient nosocomial transmission, and limited therapeutic options. Timely and accurate detection is essential for early diagnosis, targeted antimicrobial therapy, and effective infection control. This review summarizes recent advances in CRAB detection technologies, including conventional phenotypic methods, nucleic acid amplification-based assays, genom

Nosocomial infections are a major health problem worldwide. The increasing time patients spend in hospitals has led to an increase in mortality. Acinetobacter baumannii is an opportunistic pathogen that is a significant factor in nosocomial infections. The main aim of the current study was to investigate biofilm and pathogenicity-related genes among multidrug-resistant A. baumannii strains (n = 54) obtained from patients with respiratory infections. ERIC-PCR was also used to determine their molecular typing correlation. Disk diffusion and MIC methods were used to test antibiotic susceptibility

The escalating problem of bacterial drug resistance poses a severe threat to global public health, with Acinetobacter baumannii (A. baumannii) exhibiting particularly high resistance rates that are closely linked to its Quorum Sensing (QS) system. This narrative review synthesizes current knowledge on the A. baumannii QS system, focusing on its essential role in mediating antimicrobial resistance, as well as the latest progress in developing QS inhibitors. Key findings indicate that the A. baumannii QS system enhances bacterial survival by promoting biofilm formation, and regulating the expres

Acinetobacter calcoaceticus-baumannii (ACB) complex comprises of both environmental and pathogenic bacterial species. Pathogenic species are responsible for nosocomial infections, which are increasingly difficult to treat due to growing resistance to antibiotics. In addition to nosocomial settings, ACB complex has also been found in other environmental niches, including animals, food, soil, and water, where human activity has contributed to the development of antibiotic resistance. For instance, improper disposal of solid waste and release of wastewater effluent carrying antibiotic-resistant p

Antibiotic-resistant infections are a pressing clinical challenge. Plasmids are known to accelerate the emergence of resistance by facilitating horizontal gene transfer of antibiotic resistance genes between bacteria. We explore this question in Acinetobacter baumannii, a globally emerging nosocomial pathogen responsible for a wide range of infections with a worrying accumulation of resistance, particularly involving plasmids. In this species, plasmids of the Rep_3 family harbor antibiotic resistance genes within variable regions flanked by potential site-specific recombination sites recognize

Multidrug-resistant (MDR) Acinetobacter baumannii is a major clinical threat with limited treatment options, as current therapies rely on polymyxins such as colistin. Targeting the lipopolysaccharide (LPS) biosynthetic pathway offers a new target for new antibiotic discovery, yet most efforts have focused on enzyme-based assays that do not reflect cell level physiology. Here, we developed a cell-based screening strategy that links colistin resistance to inhibition of lipooligosaccharide (LOS) biogenesis in A. baumannii. Using colistin, we established a phenotypic platform in which compounds th

Acinetobacter baumannii is a globally disseminated Gram-negative bacterium that causes several types of serious nosocomial infections, the most worrisome being ventilator-associated pneumonia and bacteremia related to using venous catheters. Due to its great ability to form biofilms, combined with its survival for prolonged periods on abiotic surfaces and its potential to acquire and control the genes that determine antibiotic resistance, A. baumannii is at the top of the World Health Organization's priority list of pathogens in urgent need of new therapies. In this sense, this review aimed to

In the current era of antibiotic resistance, researchers are exploring alternative ways to treat bacterial infections that are resistant to multiple drugs. Acinetobacter baumannii (A. baumannii) is a bacterium that is commonly encountered in clinical settings and is known to be resistant to several drugs. Due to the increase in drug-resistant infections caused by this bacteria, there is an urgent need to investigate alternative treatment options such as phage therapy and combination therapy. Despite the success of phages in some cases, there are some limitations in their clinical application t

PURPOSE: Colistin is used as a last resort antibiotic against infections caused by multidrug-resistant gram-negative bacteria, especially carbapenem-resistant bacteria. However, colistin-resistance in clinical isolates is becoming more prevalent. Cinnamaldehyde and baicalin, which are the major active constituents of Cinnamomum and Scutellaria, have been reported to exhibit antibacterial properties. The aim of this study was to evaluate the capacity of cinnamaldehyde and baicalin to enhance the antibiotic activity of colistin in Enterobacterales and Acinetobacter baumannii strains.

Nosocomial infections represent one of the biggest health problems nowadays. Acinetobacter baumannii is known as an opportunistic pathogen in humans, affecting people with compromised immune systems, and is becoming increasingly important as a hospital-derived infection. It is known that in recent years, more and more bacteria have become multidrug-resistant (MDR) and, for this reason, the development of new drugs is a priority. However, these products must not affect the human body, and therefore, cytotoxicity studies are mandatory. In this context, antimicrobial peptides with potential antib

Carbapenem-resistant Acinetobacter baumannii (CRAB) is resistant to almost all antibiotics. Eravacycline, a newer treatment option, has the potential to treat CRAB infections, however, the mechanism by which CRAB isolates develop resistance to eravacycline has yet to be clarified. This study sought to investigate the features and mechanisms of eravacycline heteroresistance among CRAB clinical isolates. A total of 287 isolates were collected in China from 2020 to 2022. The minimum inhibitory concentration (MIC) of eravacycline and other clinically available agents against A. baumannii were dete

Cerebrospinal fluid shunts are the primary treatment for hydrocephalus. However, prolonged external ventricular drain (EVD) use can lead to central nervous system (CNS) infections such as ventriculitis. In the ICU setting, nosocomial infections with gram-negative, multi-drug resistant (MDR) organisms such as Acinetobacter baumannii (AB) prevail, leading to poor outcomes. AB infections are notably challenging due to their genetic drug resistance. Colistin has been reintroduced for use against gram-negative MDR pathogens but has limitations in CNS penetration when administered intravenously. The

BACKGROUND: Acinetobacter baumannii is a Gram-negative, non-fermenting coccobacillus of the Moraxellaceae family. It is an opportunistic pathogen responsible for several hospital-acquired infections (HAIs) associated with skin and tissue infections at surgical sites, catheter-associated urinary tract infections, and central line catheters. Multidrug-resistant (MDR) A. baumannii has caused hospital outbreaks that are difficult to eradicate and represent one of the leading producers of HAIs. MDR-A. baumannii presents a broad range of resistance to different antimicrobials, including carbapenems.

Acinetobacter baumannii (A. baumannii) has become a notorious pathogen causing nosocomial and community-acquired infections, especially ventilator-associated pneumonia. This opportunistic pathogen is found to possess powerful genomic plasticity and numerous virulence factors that facilitate its success in the infectious process. Although the interactions between A. baumannii and the pulmonary epitheliums have been extensively studied, a complete and specific description of its overall pathogenic process is lacking. In this review, we summarize the current knowledge of the antibiotic resistance

Several infections, such as pneumonia, urinary tract infections (UTIs), as well as bloodstream, skin, and soft tissue infections, are caused by Acinetobacter baumannii, a nosocomial pathogen and Gram-negative coccobacillus. Due to its resistance to a variety of medications, multidrug therapy, and occasionally pan therapies, this bacterium is a huge public health concern. Drug resistance is a big worry not only in A. baumannii, but it is also a major challenge in many other diseases. Antibiotic resistance, biofilm development, and genetic alterations are all linked to variables like the efflux
