The Role of ES&T in Advancing Environmental Toxicology and Chemical Risk Assessment: Past, Present, and Future.

Environmental contamination poses risks to all components of the ecosystem─humans and wildlife─yet toxicological research and regulatory assessment remain largely compartmentalized by discipline and organismal focus. We look back on 60 years of toxicological research published in Environmental Science and Technology (ES&T) and analyze how the field has evolved, what role ES&T has played in this evolution, and suggest a path forward for the future. Chemicals, complex mixtures, and their transformation products act across interconnected biological taxa, including humans, that share conserved mol

Placental transcriptomic signatures of prenatal and preconceptional maternal stress.

Prenatal exposure to maternal psychological stress is associated with increased risk for adverse birth and child health outcomes. Accumulating evidence suggests that preconceptional maternal stress may also be transmitted intergenerationally to negatively impact offspring. However, understanding of mechanisms linking these exposures to offspring outcomes, particularly those related to placenta, is limited. Using RNA sequencing, we identified placental transcriptomic signatures associated with maternal prenatal stressful life events (SLEs) and childhood traumatic events (CTEs) in 1 029 mother-c

Risk assessment for the bladder carcinogenesis of 4-aminobiphenyl in smokers: A source-to-outcome study.

4-aminobiphenyl (4-ABP) is widely distributed in the environment and is implicated in the pathogenesis of cancer. This study aims to clarify the environmental and toxicological science of 4-ABP, which are currently unexplored. By searching articles from PubMed, Web of Science, and MEDLINE, we established an aggregate exposure pathway (AEP) framework that demonstrated the presence of 4-ABP in the cigarette smoke. Compared to nonsmokers, smokers had relatively higher levels of 4-ABP and its adducts in blood, urine, and bladder tissues. Then, an adverse outcome pathway (AOP) was constructed using

Insights into tartrazine-exposure and potential hepatotoxicity.

Tartrazine is a synthetic azo-dye abundantly used in food, pharmaceuticals and cosmetic products. Human exposure to tartrazine occurs primarily through ingestion. Although systemic absorption of tartrazine is limited, the dye undergoes efficient metabolism by intestinal microflora, producing aromatic amine derivatives, some of which exhibit enhanced systemic availability. Experimental data from rodent models and human-based in vitro systems indicate that high dose and prolonged exposure can induce hepatocellular damage. Mechanistic pathways, such as oxidative stress, inflammation, endoplasmic

Mapping Social and Emotional Pathways from Racism to Adverse Health: Applying Racial Trauma Health Theory to the Coronary Artery Risk Development in Young Adults (CARDIA) Study.

BACKGROUND: Racial Trauma Health Theory (RTHT) posits that multiple, intersecting forms of racism—structural, interpersonal, and internalized—across key domains such as housing, healthcare, education, and employment, accumulate causing multiplicative effects on health over time through both direct and indirect social and emotional pathways. OBJECTIVE: Employing path analysis, we translate a theoretical framework into a statistical model to evaluate the significance of key pathways and empirically test the application of data to RTHT. METHODS: The study is a secondary data analysis of the Black

Using network analysis to illuminate the intergenerational transmission of adversity.

UNLABELLED: Objective: The effects of maternal exposure to adverse childhood experiences (ACEs) may be transmitted to subsequent generations through various biopsychosocial mechanisms. However, studies tend to focus on exploring one or two focal pathways with less attention paid to links between different pathways. Using a network approach, this paper explores a range of core prenatal risk factors that may link maternal ACEs to infant preterm birth (PTB) and low birthweight (LBW). Methods: We used data from the Avon Longitudinal Study of Parents and Children (ALSPAC) (n = 8379) to estimate two

Knowledge gaps in understanding the metabolic and clinical effects of excess folates/folic acid: a summary, and perspectives, from an NIH workshop.

Folate, an essential nutrient found naturally in foods in a reduced form, is present in dietary supplements and fortified foods in an oxidized synthetic form (folic acid). There is widespread agreement that maintaining adequate folate status is critical to prevent diseases due to folate inadequacy (e.g., anemia, birth defects, and cancer). However, there are concerns of potential adverse effects of excess folic acid intake and/or elevated folate status, with the original concern focused on exacerbation of clinical effects of vitamin B-12 deficiency and its role in neurocognitive health. Metabolomic and Genomic Insights Into Adverse Events in Paroxysmal Atrial Tachycardia.

Paroxysmal atrial tachycardia (PAT) is a supraventricular arrhythmia associated with increased risk of atrial fibrillation (AF) and adverse cardiovascular events, but mechanisms underlying adverse outcomes remain unclear. We integrated serum metabolomics (LC-MS/MS), pathway enrichment, transcriptomic profiling, and in vitro validation in a PAT-onset cohort (five patients and five matched controls) and a PAT-events cohort (10 with and 10 without adverse events). Differential metabolites and pathways were identified, and GSEA focused on ABC transporters and alanine, aspartate, and glutamate meta

Dephospho-Uncarboxylated Matrix Gla-Protein Is Associated With Adverse Outcomes in Heart Failure.

BACKGROUND: MGP (matrix Gla-protein), a known inhibitor of vascular calcification, becomes biologically active by vitamin K-dependent carboxylation. Circulating levels of dpucMGP (dephospho-uncarboxylated matrix Gla-protein), the inactive form of MGP, have been associated with large artery stiffening and reduced skeletal muscle mass in heart failure (HF). Whether dpucMGP is related to adverse outcomes in patients with HF is unknown. METHODS: In this cohort study, we measured plasma dpucMGP among 2247 PHFS (Penn HF Study) participants. We examined the relationship between dpucMGP and ≈5000 oth

Impact of adverse pregnancy outcomes on brain vascular health and cognition.

A State of the Art lecture titled "Impact of Adverse Pregnancy Outcomes on Brain Vascular Health and Cognition" was presented at the International Society on Thrombosis and Haemostasis Congress in 2023. Adverse pregnancy outcomes, encompassing conditions such as gestational hypertension, eclampsia, preeclampsia, preterm birth, fetal growth restriction, stillbirth, and gestational diabetes, may form part of an underrecognized pathway from early adulthood reproductive health factors to later-life vascular cognitive impairment and dementia in women. Adverse pregnancy outcomes are caused by dysreg

The dual adverse effects of TGF-beta secretion on tumor progression.

When a cancer escapes the growth-inhibitory effects of TGF-beta secreted by cancer cells themselves or by cells in the local stroma, a further adverse outcome for the host is the associated TGF-beta-induced suppression of anticancer T cell immunity. In addition to the previously described dampening of T cell activation and proliferation, TGF-beta markedly and directly suppresses the transcription of genes encoding multiple key proteins of the "cytotoxic program" of CD8+ CTL, such as perforin and granzymes, cytotoxins that act through the granule exocytosis pathway. The findings described below
