Using Selective Agar Containing Ciprofloxacin and Tetracycline Reveals Resistant Oral Microbiota in Healthy and Periodontitis Patients.

The oral cavity may act as a reservoir for antibiotic resistance. This study aimed to directly isolate and identify phenotypically resistant bacteria from the oral biofilm of healthy individuals and patients with periodontitis, using tetracycline, and ciprofloxacin containing selective agar. Furthermore, resistance of selected bacteria towards ampicillin was also evaluated. Plaque samples were collected from 12 patients (six healthy, six with periodontitis). Bacteria were cultured on selective agar containg defined antibiotic concentration and non-selective media under aerobic and anaerobic co

Anthocyanin-Incorporated Chromogenic Agar for Rapid, Selective Detection of Streptococcus pneumoniae via Hydrogen Peroxide-Mediated Oxidation.

The rapid and cost-effective identification of Streptococcus pneumoniae (S. pneumoniae) is crucial for the prompt treatment of pneumonia and meningitis. However, conventional culture methods are time-consuming and molecular techniques are prohibitively expensive in resource-limited settings. To address this challenge, we developed a novel anthocyanin chromogenic agar as a diagnostic tool for the selective and rapid detection of S. pneumoniae. Unlike traditional pH-based indicators, this method exploits the unique oxidative capability of S. pneumoniae, which releases excessive amounts of hydrog

Agar-collagen fluid gels as a dysphagia food: Rheological characterization vs fiberoptic endoscopic evaluation of swallowing.

The completely different structuring approach of the fluid gels makes them rheologically-tunable systems with a high underexplored potential for application in dysphagia diets. The aim of this study was to characterize the swallowing behavior of high-protein collagen (20 %w/w) fluid gels structured with various agar concentrations (0.5 to 2.5 %w/w) using different analytical methods. The oral performance during different phases of dynamic swallowing processes was characterized by rheological methods (shear yield stress, shear-dependent viscosity, strain and frequency-dependent viscoelasticity,

Pulsed power treatment demonstrates complete inactivation of anisakid larvae in agar penetration and rabbit infection tests.

Anisakiasis is a foodborne zoonosis caused by ingesting raw seafood containing anisakid larvae. Its global incidence has increased with rising seafood consumption, highlighting the need for effective prevention methods. While freezing at -20 °C for 24 h is a standard preventive measure, it compromises seafood quality. Pulsed power (PP) technology, delivering high-voltage microsecond pulses, has emerged as a promising non-thermal alternative. Here, we evaluated PP inactivation efficacy using agar penetration and rabbit infection tests. Larvae treated with 10 pulses of 13 kV for 10 µs failed to

Case Report: An unexpected diagnosis of cutaneous Mycobacterium marinum infection based on potato dextrose agar culture.

Non-tuberculous mycobacteria (NTM) are slender, non-motile, acid-fast bacilli widely distributed in diverse environmental reservoirs across the globe. In recent years, the incidence of NTM-related diseases has risen significantly, leading to their recognition as major emerging human pathogens. Cutaneous NTM infections typically arise following traumatic injury, surgical intervention, or cosmetic procedures. Due to their heterogeneous clinical presentations, these infections often pose diagnostic challenges. Löwenstein-Jensen medium and Middlebrook medium are widely employed in mycobacterial mi

Agar Plate Culture: An Alternative Sensitive Routine Laboratory Detection Method for Strongyloides stercoralis and Hookworm Parasites.

BACKGROUND: Human infection with Strongyloides stercoralis and hookworm parasites is usually under reported due to less sensitive diagnostic methods. Agar plate culture (APC) is the most sensitive technique for parasites having larval stage. However, using APC in routine diagnosis is uncommon. This study aimed to determine the detection rate and sensitivity of APC in comparison with formal ether concentration technique (FECT) and spontaneous tube sedimentation techniques (STSTs) for S. stercoralis and hookworm larvae. METHODS: Stool samples collected from 844 schoolchildren in Amhara Regional

Extreme testing of undiluted e-cigarette aerosol in vitro using an Ames air-agar-interface technique.

There is a growing consensus that e-cigarettes hold the potential for reducing the harm associated with cigarette smoking. Recently published studies have reported in vitro testing of e-cigarettes, demonstrating reduced toxicological and biological effects. Few studies however have reported the use of e-cigarettes under extreme testing conditions. To assess the full mutagenic potential of a commercially available electronic-cigarette (Vype ePen), this study investigated the delivery of aerosol under extreme conditions, using a scaled-down 35 mm plate Ames bacterial reverse mutagenicity assay.

Comparison of nine agar media for the recovery of shigatoxin-producing Escherichia coli (STEC) from the food.

Pathogenic Shiga toxin-producing Escherichia coli (STEC) are a major public health concern and a key issue for the agri-food sector. However, their isolation remains challenging because STEC lack distinctive biochemical characteristics that clearly differentiate them from other Escherichia coli. This study evaluated the performance of nine commonly used agar media for STEC isolation using three types of samples: pure cultures, experimentally spiked food matrices, and naturally contaminated foods. Two isolation approaches were compared: direct streaking and immuno-separation followed by plating

Unveiling the impact of additives on structural integrity, thermal and color stability of C-phycocyanin - Agar hydrocolloid.

C-Phycocyanin and sugar (C-PC/S) blended agar hydrocolloid was prepared and its rheological, thermo-functional and morphological properties were examined based on the fluorescence excitation-emission matrix profile. Sucrose (40%, w/v) determined as a superior preservative, maintaining the native conformation of C-PC effectively. C-PC/S exhibited enhanced structural integrity with high storage modulus (G') and 86.4% swelling index. FT-IR demonstrated strong intramolecular bonding. TGA revealed that the presence of sucrose prolonged the devolatilization peak up to 325 °C, with a degradation rate

Comparative investigations into the growth details of Histoplasma capsulatum var farciminosum on four different agar media.

Epizootic equine lymphangitis (EEL) is a chronic fungal disease that affects equids. The causative agent is a dimorphic fungus called Histoplasma capsulatum var farciminosum. Histoplasmacapsulatum var farciminosum field strain 7 (D 2878/2023) isolated from the eye socket of an EEL Ethiopian horse was sub-cultured on four different solid media and incubated at 26°C and 37°C for 6 weeks. Details of growth morphology were recorded and shown in images during 6 weeks of incubation. Histoplasmacapsulatum var farciminosum grew best at 26°C on all four agars, but only on sheep blood agar at 37°C as sm

Validation of Selective Agars for Detection and Quantification of Escherichia coli Strains Resistant to Critically Important Antimicrobials.

Success in the global fight against antimicrobial resistance (AMR) is likely to improve if surveillance can be performed on an epidemiological scale. An approach based on agars with incorporated antimicrobials has enormous potential to achieve this. However, there is a need to identify the combinations of selective agars and key antimicrobials yielding the most accurate counts of susceptible and resistant organisms. A series of experiments involving 1,202 plates identified the best candidate combinations from six commercially available agars and five antimicrobials, using 18 Escherichia coli s

An optimized agar plate culture improves diagnostic efficiency for Strongyloides stercoralis infection in an endemic community.

We aimed to compare the efficacy of modified agar plate fecal culture (mAPC) and standard agar plate culture (sAPC) for diagnosis of Strongyloides stercoralis infection in a community at Khon Kaen Province, Thailand. Fecal samples were collected from participants individually (n = 1076) and were tested using these two methods. Modified APC and sAPC detected 129 (11.99%) and 91 (8.46%) infected individuals, respectively. Thirty-eight participants were negative according to sAPC, but positive for mAPC. Moreover, in the participants who were positive for both methods, the number of worm developme

Isolation of Previously Uncultured Slow-Growing Bacteria by Using a Simple Modification in the Preparation of Agar Media.

Most microorganisms living in the environment have yet to be cultured, owing at least in part to their slow and poor propagation properties and susceptibility to oxidative stress. Our previous studies demonstrated that a simple modification in the preparation of agar media, i.e., autoclaving the phosphate and agar separately (termed "PS" medium), can greatly improve the culturability of microorganisms by mitigating oxidative stress compared with the use of "PT" medium (autoclaving the phosphate and agar together). Here, we attempted to isolate phylogenetically novel bacteria by combining PS me

Single-Center Evaluation of an Agar-Based Screening for Azole Resistance in Aspergillus fumigatus by Using VIPcheck.

Antifungal susceptibility testing is an essential tool for guiding therapy, although EUCAST and CLSI reference methods are often available only in specialized centers. We studied the performance of an agar-based screening method for the detection of azole resistance in Aspergillus fumigatus cultures. The VIPcheck consists of four wells containing voriconazole, itraconazole, posaconazole, or a growth control. Ninety-six A. fumigatus isolates were used. Thirty-three isolates harbored a known resistance mechanism: TR34/L98H (11 isolates), TR46/Y121F/T289A (6 isolates), TR53 (2 isolates), and 14 i
