Differential gene expression and alternative splicing of survivin following mouse sciatic nerve injury.

STUDY DESIGN: In vivo studies using an axotomy model in adult male Naval Medical Research Institute mice. OBJECTIVE: Survivin, a unique member of the inhibitor of the apoptosis (IAP) protein family, is expressed during embryonal development, but is undetectable in terminally differentiated cells and tissues. Owing to the vital role of survivin in cellular proliferation and apoptotic cell death, and also to the necessity of treatment of the nervous system injuries, we have monitored survivin gene expression as well as its alternative splicing changes at different time points within injured mou

Alternative RNA splicing produces multiple forms of c-Myb with unique transcriptional activities.

The c-Myb transcription factor regulates the proliferation and differentiation of hematopoietic cells, and activated alleles of c-myb induce leukemias and lymphomas in animals. Relatively minor changes in the structure of c-Myb protein change the genes that it regulates and can unleash its latent transforming activities. Here, quantitative assays were used to analyze the alternative splicing of human c-myb transcripts. We identified an array of variant transcripts, expressed in highly regulated, lineage-specific patterns, that were formed through the use of alternate exons 8A, 9A, 9B, 10A, 13A

CIR, a corepressor of CBF1, binds to PAP-1 and effects alternative splicing.

We have reported that PAP-1, a product of a causative gene for autosomal retinitis pigmentosa, plays a role in splicing. In this study, CIR, a protein originally identified as a CBF1-interacting protein and reported to act as a transcriptional corepressor, was identified as a PAP-1 binding protein and its function as a splicing factor was investigated. In addition to a basic lysine and acidic serine-rich (BA) domain and a zinc knuckle-like motif, CIR has an arginine/serine dipeptide repeat (RS) domain in its C terminal region. The RS domain has been reported to be present in the superfamily of

Expression and alternative splicing of mouse Gfra4 suggest roles in endocrine cell development.

Members of the GDNF protein family signal through receptors consisting of a GPI-linked GFRalpha subunit and the transmembrane tyrosine kinase Ret. Here we characterize the mouse Gfra4 and show that it undergoes developmentally regulated alternative splicing in several tissues. The mammalian GFRalpha4 receptor lacks the first Cys-rich domain characteristic of other GFRalpha receptors. Gfra4 is expressed in many tissues, including nervous system, in which intron retention leads to a putative intracellular or secreted GFRalpha4 protein. Efficient splicing occurs only in thyroid, parathyroid, and

Novel mutations of the FANCG gene causing alternative splicing in Japanese Fanconi anemia.

Fanconi anemia (FA), an autosomal recessive disorder characterized by a progressive pancytopenia associated with congenital anomalies and high predisposition to malignancies, is a genetically and clinically heterogeneous disease. At least eight complementation groups (FA-A to FA-H) have been identified. Previously, we studied mutations of the FANCA gene, responsible for FA-A, and found pathogenic mutations in 12 of 15 unclassified Japanese FA patients. Here, we further studied an additional 5 FA patients for sequence alterations of the FANCA gene and found pathogenic mutations in 2 of them. We

Alternative splicing of the human CDC25B tyrosine phosphatase. Possible implications for growth control?

CDC25B2, a protein tyrosine phosphatase closely related to the putative CDC25B oncogene, was identified in a Burkitt lymphoma cDNA library. CDC25B2 differs from CDC25B by a 14 residue insertion and a 41 residue deletion, which are both located in the amino-terminal region of the protein, upstream of the catalytic domain. Examination of the genomic sequence revealed that CDC25B1 (formerly B) and CDC25B2 are splice variants of the same gene. A third variant, CDC25B3, that carries both the 14 and the 41 residue sequences was also identified in the same cDNA library. All three variants were detect

Alternative splicing of dystrophin mRNA complicates carrier determination: report of a DMD family.

Carrier determination is important for genetic counselling in DMD/BMD families. The detection of altered PCR amplified dystrophin mRNA fragments owing to deletions, insertions, or point mutations has increased the possibilities of carrier determination. However, problems may occur because of alternative splicing events. Here we present a family with a DMD patient characterised by a deletion of exons 45 to 54. At the mRNA level we detected a corresponding altered fragment which served for carrier determination. The mother and the sister of the patient showed the same altered dystrophin mRNA fra

The emerging roles of alternative splicing in modulating tumor immune responses and immunotherapies.

Alternative splicing is a fundamental mechanism that expands transcriptomic and proteomic diversity and contributes to multiple biological processes, including immune regulation. Increasing evidence shows that dysregulated alternative splicing influences tumor immunogenicity, the immune landscape of the tumor microenvironment, and responses to cancer immunotherapy. Alternative splicing can alter the expression and peptide repertoire of tumor antigens, modulate major histocompatibility complex-mediated antigen presentation, and generate immunomodulatory isoforms that promote immune evasion. In

Characterization of aberrant alternative splicing landscape in patients with metastatic renal cell carcinoma.

PURPOSE: Aberrant alternative splicing (AS) events have been implicated in cancer progression; however, their role in metastatic renal cell carcinoma (mRCC) remains underexplored. This study aims to identify AS events associated with clinical benefits from immune checkpoint inhibitors and targeted therapies in mRCC. MATERIALS AND METHODS: We conducted a retrospective analysis on 101 patients with mRCC who received systemic therapy and underwent RNA sequencing. Patients were divided into subgroups based on ICIs (alone or in combination) and targeted therapies. Responders and non-responders wer

Metformin improves RAN protein pathology, alternative splicing, and behavioral phenotypes in SCA8 mice.

Spinocerebellar ataxia type 8 (SCA8) is a member of a group of dominantly inherited, debilitating neurological diseases caused by CAG•CTG expansions for which there are no effective treatments. RAN translation, which was discovered in SCA8, has previously been shown to occur across CAG and CUG expansion transcripts, making treatments for SCA8 potentially relevant to a broad group of diseases, including SCA1, SCA2, SCA3, SCA6, SCA7, SCA12, Huntington's disease, and myotonic dystrophy type 1. In addition, CUG and CAG expansion transcripts have been reported to cause RNA gain-of-function effects.

Alternative splicing in breast cancer drug resistance: Mechanisms, therapeutic targeting, and clinical translation.

Alternative splicing (AS) is a pivotal but underexplored driver of therapeutic resistance in breast cancer. This review provides an integrative synthesis of how AS remodels molecular networks to undermine chemotherapy, endocrine therapy, targeted therapy, and immunotherapy. We highlight specific AS events, such as NCOR2 splice variants in chemoresistance and human epidermal growth factor receptor 2 (HER2) isoforms in anti-HER2 therapy failure, that reshape drug response, survival outcomes, and tumor adaptation. Core mechanisms include splicing factor dysregulation (e.g., SRSF1, SF3A2), stress-

CCDC174 deficiency impaired human fertility by affecting the alternative splicing of maternal mRNAs.

Precise regulation of alternative splicing (AS) of maternal mRNAs is crucial for maintaining mRNA homeostasis and for acquiring oocyte competence. However, the regulatory factors and mechanisms of AS regulating oocyte competence and human fertility remain largely unknown. Here, we identified biallelic variants in CCDC174 that cause human oocyte competence defects and female infertility. Oocyte-specific knockout of Ccdc174 resulted in oocyte maturation arrest and female infertility in mice, and transcriptomic and proteomic analyses indicated that deletion of Ccdc174 disrupted mRNA and protein h

Transcriptomic characterization of the aberrant alternative splicing in skeletal muscles of sarcopenia patients.

Sarcopenia represents a substantial health risk for the expanding elderly population globally, characterized by a progressive decline in skeletal muscle mass and strength as people age. To investigate the potential role of the alternative splicing change in sarcopenia pathogenesis, we conducted a re-analysis of previously published RNA-seq data from muscle biopsies of sarcopenia patients, age-matched healthy controls, and individuals exhibiting partial symptoms (either low muscle mass or reduced muscle function). Our results revealed 2260 differential alternative splicing events (DASEs) in 168

HSP90 inhibition partially rescues alternative splicing dysregulation in cell models of myotonic dystrophy.

Myotonic dystrophy type 1 (DM1) and type 2 (DM2) are multisystemic diseases caused by the expression of toxic expansion RNAs that sequester muscleblind-like (MBNL) proteins, resulting in extensive alternative splicing dysregulation. Given that there are no current disease-modifying treatments for DM, we sought to identify compounds that rescue the underlying splicing dysregulation. A medium throughput splicing screen utilizing DM1 patient-derived fibroblasts was developed and used to screen 1584 compounds from the NIH NCI Diversity Set VI, leading to the identification of macbecin II, an HSP90

Alternative splicing of the CaV1.2 calcium channel and its role in cardiovascular diseases.

The L-type calcium channel CaV1.2 serves as a core trigger for excitation-contraction and excitation-transcription coupling in cardiovascular cells. Its abnormalities are closely linked to multiple cardiovascular diseases, including hypertension, arrhythmias, and cardiac hypertrophy. Alternative splicing (AS) generates CaV1.2 channel isoforms with distinct electrophysiological properties and drug sensitivity, finely tuning channel functions. This review systematically summarizes AS regulation and pathological significance of CaV1.2 in the cardiovascular system. Three key splicing events, mutua

Modulating alternative splicing of MECP2 is a potential therapeutic strategy for Rett syndrome.

Rett syndrome (RTT) is a neurological disorder caused by loss-of-function mutations in methyl-CpG-binding protein 2 (MECP2), which encodes a transcriptional regulator essential for maintenance of normal neuronal function. The current US Food and Drug Administration-approved treatment for RTT, trofinetide, mildly alleviates some symptoms. In contrast, reintroducing MeCP2 or increasing its amount through transgenesis in mouse RTT models improves most neurological phenotypes and enhances survival. Here, we devised a therapeutic strategy to moderately increase MeCP2 protein by modulating the alter

Alternative RNA Splicing in Cancer: Molecular Mechanisms, Functional Consequences, Biomarkers and Therapeutic Opportunities.

Alternative pre-mRNA splicing is a central layer of gene regulation that enables a limited number of genes to generate a far larger and more context-dependent transcriptome and proteome. In cancer, splicing is disrupted by mutations in cis-regulatory sequences, recurrent lesions in spliceosome components, altered abundance or activity of RNA-binding proteins, and changes in transcription, chromatin, RNA modification, metabolism and stress signalling. These alterations are not merely by-products of malignant transformation. They can create oncogenic protein isoforms, eliminate tumour-suppressiv

HNRNPC lactylation promotes pancreatic cancer progression through mediating the alternative splicing of PAK6.

Pancreatic cancer (PC) is a highly malignant and lethal tumor in gastrointestinal tract. Lactate accumulation is a classical feature of metabolic reprogramming in cancers. Lactate-derived lysine lactylation (Kla) is identified as a new type of post-translational modifications (PTMs), which is confirmed to be involved in multiple biological processes. However, the cancer-specific regulation of protein Kla in PC requires further elucidation. Here, we report a range of dysregulated Kla sites specifically related to RNA splicing in human pancreatic cancer, leading to the observation that the Kla a

NAD+ reverses Alzheimer's neurological deficits via regulating differential alternative RNA splicing of EVA1C.

Dysfunctional alternative splicing events (ASEs) in RNA are markers of aging and Alzheimer's disease (AD). As a key neuronal resilience metabolite, the oxidized nicotinamide adenine dinucleotide (NAD+) slows down AD progression in preclinical studies with several clinical trials ongoing. However, the underlying molecular mechanisms around how NAD+ enhances neuronal resilience, especially whether it has any effect on ASEs, have remained elusive. This study shows that NAD+ augmentation corrects the ASEs of many genes via a key protein, EVA1C (epithelial V-like antigen 1 homolog C), which is invo

Alternative Splicing of Serum Response Factor Reveals Isoform-Specific Remodeling in Cardiac Diseases.

Background: Alternative splicing is an important mechanism of transcriptomic and proteomic diversity and is progressively involved in cardiovascular disease (CVD) pathogenesis. Serum response factor (SRF), a critical transcription factor in cardiac development and function, may itself undergo splicing regulation, potentially altering its function in disease states. Objective: The objective of this study is to identify SRF-associated alternative splicing events in cardiac pathological conditions and examine regulatory interactions with splicing factors using RNA-seq data. Methods: Three human h
