Parasitism of a US traveler by a nymphal Amblyomma tapirellum Dunn, 1933 (Ixodida: Ixodidae) and review of exotic tick interceptions on humans in the United States.

A resident of Monmouth County, New Jersey, United States removed an engorged nymphal tick after returning from travel to Costa Rica. The tick was identified by cox1 barcoding as Amblyomma tapirellum  Dunn, 1933, a Central American species whose immature stages are undescribed. This species is associated with wet, tropical forests, and most host records come from Baird's tapirs (Tapirus bairdii), though feeding on other mammalian orders and on humans has been observed. To date, no human pathogens have been detected in A. tapirellum, although very few specimens have been tested. The A. tapirellu

Enduring Challenge of Invasive Ticks: Introduction of Amblyomma oblongoguttatum (Acari: Ixodidae) into the United States on a Human Traveler Returning from Central America.

Introduction of exotic tick vectors of bacteria, protozoa, viruses, and filarial parasites into the United States has accelerated in recent years, primarily because of globalization, increased frequency of travel, and a rise in legal and illegal animal trades. We herein report introduction of a live specimen of Amblyomma oblongoguttatum on a human into the United States from Central America, and we review 4 previous similar incidents. This tick species occurs widely in the neotropics, from western and southern Mexico, southwards through Central America, to the northern half of South America. I

The Transcriptome of the Salivary Glands of Amblyomma aureolatum Reveals the Antimicrobial Peptide Microplusin as an Important Factor for the Tick Protection Against Rickettsia rickettsii Infection.

The salivary glands (SG) of ixodid ticks play a pivotal role in blood feeding, producing both the cement and the saliva. The cement is an adhesive substance that helps the attachment of the tick to the host skin, while the saliva contains a rich mixture of antihemostatic, anti-inflammatory, and immunomodulatory substances that allow ticks to properly acquire the blood meal. The tick saliva is also a vehicle used by several pathogens to be transmitted to the vertebrate host, including various bacterial species from the genus Rickettsia. Rickettsia rickettsii is a tick-borne obligate intracellul

Characterization of reptile-associated Borrelia sp. in the vector tick, Amblyomma geoemydae, and its association with Lyme disease and relapsing fever Borrelia spp.

The genus Borrelia is arthropod-borne infectious agents in vertebrates, and is classified into Lyme disease (LD) Borrelia spp. and Relapsing fever (RF) Borrelia spp. In addition to these Borrelia groups, we recently reported reptile-associated (REP) Borrelia spp. from reptiles and from hard-bodied ticks, which probably transmitted the REP Borrelia spp. In this study, we investigated the presence of REP Borrelia sp. in moulted ticks, and found that trans-stadial transmission of REP Borrelia sp. occurred in the midgut, while it was observed that REP Borrelia sp. entered the salivary gland during

Ablation of immunity to Amblyomma americanum by anti-basophil serum: cooperation between basophils and eosinophils in expression of immunity to ectoparasites (ticks) in guinea pigs.

Basophils infiltrate the skin and other tissues as part of T lymphocyte and/or antibody-mediated immune responses to certain protein antigens, viruses, tumors, and parasites. Although basophils may comprise a significant fraction of leukocytes in these reactions, their precise role has been poorly understood. Guinea pigs expressing acquired immunity to the tick Amblyomma americanum develop basophil- and eosinophil-rich cutaneous inflammatory responses at tick feeding sites, and tick rejection is associated with extensive local basophil degranulation. We report that a specific anti-basophil ser

Molecular discovery of a unique Litomosoides lineage in the Gulf coast tick Amblyomma maculatum: First evidence of potential tick infection in North America.

Tick-transmitted filarioids are increasingly recognized globally, yet their diversity and transmission dynamics in North American ticks remain poorly characterized. We investigated the prevalence and genetic diversity of filarioids in Amblyomma maculatum (Gulf Coast tick), a vector of multiple pathogens in the United States. We screened 383 samples comprising 471 individual ticks collected from small mammals and vegetation between 2012 and 2016 from across Texas, using PCR assays targeting filarioid mitochondrial 12S rDNA and cox1 genes. The overall prevalence of filarioid DNA detection was 6.

The anal pore route is efficient to infect Amblyomma spp. ticks with Rickettsia rickettsii and allows the assessment of the role played by infection control targets.

Adult Amblyomma sculptum and Amblyomma aureolatum ticks are partially refractory to Rickettsia rickettsii when fed on infected hosts, hindering the functional characterization of potentially protective targets in the bacterial acquisition. In the current study, we used the anal pore route to infect adult A. sculptum and A. aureolatum ticks with R. rickettsii and to assess the effects of the knockdown of microplusin in infection control. The anal pore route was efficient to infect both species, resulting in a prevalence of around 100% of infected ticks. Higher loads of R. rickettsii were detect

Rhipicephalus microplus and Amblyomma sculptum (Ixodidae) infestation of Nellore cattle (Bos taurus indicus) in a farm of the Brazilian Cerrado: seasonality and infestation patterns.

We herein describe zebuine cattle tick infestation in a farm in southeast Brazil with an examination accurate enough to detect tick immatures and species other than R. microplus. Cattle were inspected monthly for ticks from May 2015 to May 2017 and 7604 ticks were collected along 276 bovine inspections. Rhipicephalus microplus was the dominant species (7197 specimens, 94.5% from the total), but Amblyomma sculptum was also collected (407/5.5%). Horse tick infestations were evaluated for comparison purposes of A. sculptum infestations of a primary host sharing pastures with bovines. Ticks were c

Population dynamics of immature Amblyomma maculatum (Acari: Ixodidae) and other ectoparasites on meadowlarks and northern bobwhite quail resident to the coastal prairie of Texas.

A survey of ectoparasites from 219 meadowlarks conducted during 2 consecutive fall-winter periods in a coastal prairie found immature Amblyomma maculatum Koch to be the most abundant parasite. Peak larval infestations occurred in December with 80-100% of collected birds infested and with a monthly mean of up to 34 larvae per bird. Peak nymphal infestations occurred in February or March with 95-100% of birds infested and with a monthly mean of up to 11 nymphs per bird. Seasonal dynamics of these stages offered possible insight into the persistence of A. maculatum in an area long infested with t

Rickettsia parkeri and Candidatus Midichloria sp. in Amblyomma triste ticks from protected areas of Buenos Aires Province (La Plata River Hydrographic Basin, Argentina).

Ticks are important vectors of bacteria belonging to the order Rickettsiales, several of which are causes zoonotic diseases. Amblyomma triste is the main vector of Rickettsia parkeri, the etiological agent responsible for most cases of human spotted fever rickettsiosis in Argentina. However, information on the circulation of rickettsial agents in protected areas of the La Plata River Hydrographic Basin, a region that includes highly urbanized environments and frequent contact between wildlife, domestic animals and humans, remains limited. The aim of this study was to investigate the presence o

Molecular Evidence of Novel Spotted Fever Group Rickettsia Species in Amblyomma albolimbatum Ticks from the Shingleback Skink (Tiliqua rugosa) in Southern Western Australia.

Tick-borne infectious diseases caused by obligate intracellular bacteria of the genus Rickettsia are a growing global problem to human and animal health. Surveillance of these pathogens at the wildlife interface is critical to informing public health strategies to limit their impact. In Australia, reptile-associated ticks such as Bothriocroton hydrosauri are the reservoirs for Rickettsia honei, the causative agent of Flinders Island spotted fever. In an effort to gain further insight into the potential for reptile-associated ticks to act as reservoirs for rickettsial infection, Rickettsia-spec

First Record of the Tortoise Tick, Amblyomma geoemydae (Cantor, 1847) (Acari: Ixodidae) Parasitizing a Tree Shrew, Tupaia glis (Scandentia: Tupaiidae) in West Malaysia.

The tick genus Amblyomma Koch, 1844 (Acari: Ixodidae) has received little attention in Malaysia; therefore, its associated hosts and distribution records are poorly known. In this study, we collected six Amblyomma sp. individuals (two larvae and four adults) that infested a common treeshrew, Tupaia glis (Diard, 1820) (Scandentia: Tupaiidae) caught in a recreational area in Sungai Lembing, Pahang (West Malaysia). The adult female ticks were morphologically identified according to taxonomic keys prior to molecular identification using cytochrome oxidase subunit I (COI) and 16S rDNA genes. The ti

Tick-Host Range Adaptation: Changes in Protein Profiles in Unfed Adult Ixodes scapularis and Amblyomma americanum Saliva Stimulated to Feed on Different Hosts.

Understanding the molecular basis of how ticks adapt to feed on different animal hosts is central to understanding tick and tick-borne disease (TBD) epidemiology. There is evidence that ticks differentially express specific sets of genes when stimulated to start feeding. This study was initiated to investigate if ticks such as Ixodes scapularis and Amblyomma americanum that are adapted to feed on multiple hosts utilized the same sets of proteins to prepare for feeding. We exposed I. scapularis and A. americanum to feeding stimuli of different hosts (rabbit, human, and dog) by keeping unfed adu

Ehrlichia and spotted fever group Rickettsiae surveillance in Amblyomma americanum in Virginia through use of a novel six-plex real-time PCR assay.

The population of the lone star tick Amblyomma americanum has expanded in North America over the last several decades. It is known to be an aggressive and nondiscriminatory biter and is by far the most common human-biting tick encountered in Virginia. Few studies of human pathogen prevalence in ticks have been conducted in our state since the mid-twentieth century. We developed a six-plex real-time PCR assay to detect three Ehrlichia species (E. chaffeensis, E. ewingii, and Panola Mountain Ehrlichia) and three spotted fever group Rickettsiae (SFGR; R. amblyommii, R. parkeri, and R. rickettsii)

New insights into the systematics of the afrotropical Amblyomma marmoreum complex (Acari: Ixodidae) and the genome of a novel Rickettsia africae strain using morphological and metagenomic approaches.

The Amblyomma marmoreum complex includes afrotropical species, such as Amblyomma sparsum, a three-host tick that parasitizes reptiles, birds, and mammals, and is a recognized vector of Ehrlichia ruminantium. However, the lack of morphological, genetic and ecological data on A. sparsum has caused considerable confusion in its identification. In this study, we used microscopy and metagenomic approaches to analyze A. sparsum ticks collected from a puff adder snake (Bitis arietans) in southwest Senegal (an endemic rickettsioses area) in order to supplement previous morphological descriptions, prov

The Asian turtle tick Amblyomma geoemydae Cantor, 1847 (Acari: Ixodidae) in the Philippines: first confirmed local host and locality with a complete host index.

Amblyomma geoemydae is an oriental tick species primarily associated with turtles and tortoises but also sometimes reported from other vertebrates, including humans. Although A. geoemydae was first reported from the Philippines more than 70 years ago, it has not been reported since, and no local distributional or host data have been published. Herein, we present the first definitive locality report and host record for A. geoemydae in the Philippines. A complete checklist of the known host species from which A. geoemydae has been collected is also presented.

Ultrastructure features of the midgut of the female adult Amblyomma cajennense ticks Fabricius, 1787 (Acari: Ixodidae) in several feeding stages and subjected to three infestations.

The digestive tract of Amblyomma cajennense, as well as other species of ticks, is divided into the anterior, medium and posterior intestines and is the organ responsible for digestion of blood ingested during the meal on the host. The anterior and posterior regions are derived from the ectoderm and the medium one from the endoderm. In the present ultrastructural study, we analyzed the midgut of females in a semi- and fully engorged stages, on the rabbit host, which was subjected to three infestations, where were analyzed the changes that the epithelial cells went through.
