Human granulocytic ehrlichiosis is an emerging infectious disease in the United States and Europe, and PCR methods have been shown to be effective for the diagnosis of acute infections. Numerous PCR assays and primer sets have been reported in the literature. The analytical sensitivities (limits of detection) of 13 published PCR primer sets were compared using DNA extracted from serial dilutions of Anaplasma phagocytophilum-infected HL-60 cells. The specificity of the assays that were able to detect <or=2.5 infected cells was tested by the use of template DNA extracted from Ehrlichia chaffeens

The present study aimed to evaluate under dairy farm conditions the predisposing factors, impact on milk production and productivity, and the role of Rhipicephalus microplus in the epidemiology of tick fever agents in Holstein calves grazing in a tropical region. A total of 4292 pure female Holsteins were evaluated at a commercial farm. Until April 2020, calves had contact with R. microplus for between 3 and 24 months, while after April 2020, no animal had further contact with ticks. Three times a week the rectal temperature (RT) of all animals was determined, and blood samples were collected

Blood smears from a total of 282 wild Bovidae from Ngamiland, Botswana were examined for the presence of blood parasites. The following species were sampled: 190 African buffalo, 23 impala, 10 blue wildebeest, 18 tsessebe, 1 eland, 13 lechwe, 16 kudu and 11 sable antelope. In addition, blood from 36 of the above antelope and from a further 48 buffalo was inoculated into rodents to test for the presence of trypanosomes. An anaplasm morphologically indistinguishable from Anaplasma marginale Theiler, 1910 was found in 28,4% of buffalo. The incidence of detectable cases of A. marginale infection i

The southern cattle fever tick (SCFT) Rhipicephalus (Boophilus) microplus, is considered the most important ectoparasite of livestock in the world because of high financial losses associated with direct feeding and transmission of the hemoparasites Babesia bovis, B. bigemina, and Anaplasma marginale. Unfortunately, SCFT in many parts of the world have evolved resistance to all market-available pesticides thus driving development of new control technologies. Vaccination against ticks using the tick gut protein Bm86 has been shown to be effective against acaricide-resistant ticks. This technique

Samples from 17 free-ranging hunter-killed grey brocket deer (Mazama gouazoubira) in the Gran Chaco, Bolivia, were collected during June-August 1999. All 17 deer appeared to be in good condition at the time of death. Gross necropsies were performed, serum was collected for serologic evaluation of selected infectious disease agents, and feces and ectoparasites were collected for evaluation of internal and external parasites. Serologic tests were positive for antibodies against bovine respiratory syncytial virus and four Leptospira interrogans serovars, with questionable results for epizootic he

Diseases transmitted by arthropod vectors such as mosquitoes, ticks and sand flies greatly impact human and animal health, and therefore, their control is important for the eradication of vectorborne diseases (VBD). Vaccination is an environmentally friendly alternative for vector control that allows control of several VBD by targeting their common vector. Recent results have suggested that subolesin (SUB) and its orthologue in insects, akirin (AKR) are good candidate antigens for the control of arthropod vector infestations and pathogen infection. SUB was discovered as a tick-protective antig

Bovine babesiosis is a tick-transmitted hemoprotozoan disease that is mainly caused by Babesia bovis and/or Babesia bigemina and is characterized by significant morbidity and mortality worldwide. This disease is widespread in most parts of China. However, it is difficult to rapidly discriminate between the B. bovis and B. bigemina species. To detect and distinguish these species, a loop-mediated isothermal amplification (LAMP) platform that targets specific sequences of the internal transcribed spacer (ITS) genes was developed. Specificity testing revealed that there was no cross-reaction with

Rhipicephalus microplus is a vector of cattle tick fever, a disease caused by the protozoans Babesia bovisand B. bigemina, and also anaplasmosis, produced by the Rickettsiales Anaplasma marginale. These tick-borne pathogens cause considerable losses to Brazilian livestock breeders and represent an obstacle to the expanded use of taurine breeds due to their higher sensitivity to ticks and hemoparasites compared to zebu breeds. Differences in the susceptibility to hemoparasites were also verified within breeds, suggesting that may be possible to select a most resistant phenotype. Therefore, repe

Immunization with the Ixodes scapularis protein, subolesin, has previously been shown to protect hosts against tick infestation and to decrease acquisition of Anaplsma marginale and Babesia bigemina. Here we report the efficacy of subolesin, a conserved tick protein that can act as a regulator of gene expression, expressed from vaccinia virus for use as an orally delivered reservoir - targeted vaccine for prevention of tick infestation and acquisition/transmission of Borrelia burgdorferi to its tick and mouse hosts. We cloned subolesin into vaccinia virus and showed that it is expressed from m

Ticks are blood-feeding arthropods and transmit a variety of medically important viral, bacterial, protozoan pathogens to animals and humans. Ticks also harbor a diverse community of microbes linked to their biological processes, such as hematophagy, and hence affect vector competence. The interactions between bacterial and/or protozoan pathogens and the tick microbiome is a black-box, and therefore we tested the hypothesis that the presence of a protozoan or bacterial pathogen will alter the microbial composition within a tick. Hence, this study was designed to define the microbial compositio

Cattle fever ticks, Rhipicephalus microplus and R. annulatus have been eradicated from the United States and inspectors from the U.S. Department of Agriculture (USDA), Animal Plant Health Inspection Service (APHIS), Cattle Fever Tick Eradication Program (CFTEP) monitor the quarantine zone along the Texas border to prevent the introduction of livestock carrying cattle fever ticks from Mexico. Stray livestock apprehended by CFTEP in the zone are checked for ticks and tested for infectious disease-causing pathogens but are not evaluated for evidence of infection with tick-borne pathogens. We test

A cross-sectional study of serum antibody responses of cattle to tick-borne pathogens (Theileria parva, Theileria mutans,Anaplasma marginale, Babesia bigemina and Babesia bovis) was conducted on smallholder dairy farms in Tanga and Iringa Regions of Tanzania. Seroprevalence was highest for T. parva (48% in Iringa and 23% in Tanga) and B. bigemina (43% in Iringa and 27% in Tanga) and lowest for B. bovis (12% in Iringa and 6% in Tanga). We use spatial and non-spatial models, fitted using classical and Bayesian methods, to explore risk factors associated with seroprevalence. These include both fi

BACKGROUND: Ticks are transmitting a wide range of bacterial pathogens that cause substantial morbidity and mortality in domestic animals. The full pathogen burden transmitted by tick vectors is incompletely studied in many geographical areas, and extensive studies are required to fully understand the diversity and distribution of pathogens transmitted by ticks.
RESULTS: We sampled 824 ticks of 11 species collected in 19 counties in Romania. Ticks were collected mainly from dogs, but also from other domestic and wild animals, and were subjected to molecular screening for pathogens. Rickettsia

The global expansion of South American camelid farming contrasts with the limited knowledge of the pathogens that can infect them. This study investigated the presence of multiple infectious agents, including Anaplasmataceae, Bartonella sp., hemotropic Mycoplasma, piroplasmids, trypanosomatids, and gastrointestinal parasites, in 10 surviving llamas sampled following an outbreak of clinical anemia. Ehrlichia sp. was molecularly detected in 1/10 animal, hemotropic Mycoplasma sp. in 4/10, and 7/10 were infected with apicomplexans, while parasitological stool analysis showed that 6/9 were infected

The development of sensitive surveillance technologies using PCR-based detection of microbial DNA, such as the reverse line blot assay, can facilitate the gathering of epidemiological information on tick-borne diseases, which continue to hamper the productivity of livestock in many parts of Africa and elsewhere. We have employed a reverse line blot assay to detect the prevalence of tick-borne parasites in an intensively studied cohort of indigenous calves in western Kenya. The calves were recruited close to birth and monitored for the presence of infectious disease for up to 51 weeks. The fina

When considering an elk (Cervus elaphus) restoration program, wildlife managers must evaluate the positive and negative elements of translocation. We prepared this protocol to give an overview of health considerations associated with translocation of elk, with an emphasis on movement of free-ranging elk from western North America to the southeastern USA. We evaluated infectious agents and ectoparasites reported in elk from two perspectives. First, we made a qualitative estimate of the ability of the agent to be introduced and to become established. This was done using a selected set of epidemi

BACKGROUND: Ticks are important ectoparasites for both humans and animals and can also transmit a wide range of different viral, bacterial, and parasitic pathogens, which are commonly known as "tick-borne pathogens" (TBPs). In Zambia (Southern Africa), a number of studies have been conducted on TBPs, but information on their distribution and genetic variation is still incomplete.
METHODS: Between 2022 and 2023, 588 ticks were collected in 3 provinces from the environment/vegetation via flagging as well as from cattle and wild host species (buffaloes and tortoises). After tick species identifi

BACKGROUND: Small ruminant playing significant economic development and cultural roles for smallholders and reared in different agroecological systems in Ethiopia. However, ticks and tick-borne haemopathogens are major threats to the health of small ruminants leading to loss of production and productivity in Ethiopia especially in north-western Ethiopia, due to climate and agroecological system favourable to the tick survival in the area.
OBJECTIVES: The objectives of this study were to determine the prevalence of Ixodid tick infestation and tick-borne haemopathogens, to identify the existing

A survey to determine the prevalence of tick-borne diseases (TBDs) and ticks infesting cattle was conducted in the communal areas of the north-eastern region of the Eastern Cape Province (ECP) between January 2019 and October 2019. Tick counts, packed cell volume (PCV), body condition scores (BCS), and serological test for TBDs were seasonally conducted in communally grazed cattle (n = 240) in Joe Gqabi district in two local municipalities (Elundini and Senqu). A standard indirect fluorescent antibody test (IFAT) was used to determine antibodies to Babesia bigemina, Babesia bovis, and Ehrlichi
