Bovine babesiosis is the most important protozoan disease transmitted by ticks. In Plasmodium falciparum, another Apicomplexa protozoan, the interaction of rhoptry neck protein 2 (RON2) with apical membrane antigen-1 (AMA-1) has been described to have a key role in the invasion process. To date, RON2 has not been described in Babesia bigemina, the causal agent of bovine babesiosis in the Americas. In this work, we found a ron2 gene in the B. bigemina genome. RON2 encodes a protein that is 1351 amino acids long, has an identity of 64% (98% coverage) with RON2 of B. bovis and contains the CLAG d

The apicoplast is an essential organelle found in Apicomplexa, a large phylum of intracellular eukaryotic pathogens. The apicoplast produces metabolites that are utilized for membrane biogenesis and energy production. A majority of apicoplast-resident proteins are encoded by the nuclear genome and are trafficked to the apicoplast and are referred to as nuclear-encoded and apicoplast-trafficked (NEAT) proteins. In this study, we characterized a NEAT protein named TgBipA, which is a homolog of the highly conserved prokaryotic translational GTPase BipA. BipA is essential for bacterial survival in

Gregarines (Apicomplexa: Gregarinasina) are widespread protist symbionts of invertebrates, occupying roles across the symbiotic spectrum from mutualism to parasitism. Despite their ecological importance, they remain far less studied than other apicomplexans, leaving many aspects of their diversity, host specificity, and evolutionary history unresolved. This is particularly true for freshwater taxa for which only a handful of small subunit (SSU) rDNA sequences from species colonising freshwater hosts are available in public databases. In this study, we screened ten freshwater invertebrates (Art

The chrompodellids are a collection of photosynthetic "chromerids" and heterotrophic "colpodellids," which are together the sister lineage to the apicomplexans,1 a group of parasites containing many human disease-causing agents and species of ecological and economic importance. These lineages all descend from a photosynthetic ancestor and have been a model to understand the evolutionary transition from free-living alga to obligate parasite. Only a handful of species of chrompodellids are described-most are free-living predators, while two are photosynthetic and two are parasitic.1,2,3,4,5 The

Phosphoinositides are the phosphorylated derivatives of the structural membrane phospholipid phosphatidylinositol. Single or combined phosphorylation at the 3, 4 and 5 positions of the inositol ring gives rise to the seven different species of phosphoinositides. All are quantitatively minor components of cellular membranes but have been shown to have important functions in multiple cellular processes. Here we describe our current knowledge of phosphoinositide metabolism and functions in apicomplexan parasites, mainly focusing on Toxoplasma gondii and Plasmodium spp. Even though our understandi

The developmental stages of Theileria parva within the intestine of ticks were studied by electron microscopy. On the 2nd-4th day post repletionem (p.r.) the intestine of the ticks contained numerous stages, which were very similar to microgamonts and microgametes of haemosporidia. From these results it seemed likely that gamogony of Theileria parva takes place in the intestine of ticks, too. The microgamont-like stages were spear-head-like; they measured about 10.5 mum in length with a maximum diameter of about 2.1 mum in their middle region. These parasites, which were limited by a unit memb

Toxoplasma gondii is a significant zoonotic pathogen that infects nearly all warm-blooded animals. Available drugs for clinical treatment of toxoplasmosis are limited, creating an urgent need for development of new compounds. Toltrazuril (TOL) is a highly effective and broad-spectrum drug against parasites in Apicomplexa. In the hosts, TOL is metabolized to toltrazuril sulfone (TOLSO2) and toltrazuril sulfoxide (TOLSO), which also show potency against Apicomplexa protozoa. However, the knowledge about the effects of these metabolites on T. gondii remains unknown. The present study demonstrated

Apicomplexa is a diverse protistan phylum composed almost exclusively of metazoan-infecting parasites, including the causative agents of malaria, cryptosporidiosis, and toxoplasmosis. A single apicomplexan genus, Nephromyces, was described in 2010 as a mutualist partner to its tunicate host. Here we present genomic and transcriptomic data from the parasitic sister species to this mutualist, Cardiosporidium cionae, and its associated bacterial endosymbiont. Cardiosporidium cionae and Nephromyces both infect tunicate hosts, localize to similar organs within these hosts, and maintain bacterial en

Intracellular pathogens need to develop sophisticated mechanisms to survive and thrive in the hostile environment within host cells. Unicellular, eukaryotic parasites from the Apicomplexa phylum have become masters of manipulating their host cells, exploiting signaling, and metabolic pathways to hijack host gene expression to their own advantage. These intracellular parasites have developed a wide range of strategies that affect transcriptional machineries and epigenetic events in the host cell nucleus. In recent years, many laboratories have risen to the challenge of studying the epigenetics

The glideosome is an actomyosin-based machinery that powers motility in Apicomplexa and participates in host cell invasion and egress from infected cells. The central component of the glideosome, myosin A (MyoA), is a motor recruited at the pellicle by the acylated gliding-associated protein GAP45. In Toxoplasma gondii, GAP45 also contributes to the cohesion of the pellicle, composed of the inner membrane complex (IMC) and the plasma membrane, during motor traction. GAP70 was previously identified as a paralog of GAP45 that is tailored to recruit MyoA at the apical cap in the coccidian subgrou

Members of the genus Cryptosporidium in the phylum Apicomplexa were long thought to be closely related to the coccidia. However, despite strong morphological similarities to these organisms, Cryptosporidium has notable differences with them and similarities with the gregarine protozoa. On the basis of phylogenetic analysis of molecular data, some authors place Cryptosporidium at the basis of the phylum Apicomplexa, others consider species of this genus to be phylogenetically too distant from the coccidia and do not include them in this group of protozoa, and others think that Cryptosporidium i

Toxoplasma gondii is a unicellular organism of the Apicomplexa that occurs worldwide and is therefore a close relative of the malaria pathogen Plasmodium. As T. gondii infests every warm-blooded vertebrate species as an intermediate host and has a very high prevalence worldwide, toxoplasmosis is one of the most important international foodborne diseases. Potential vaccines (human as well as veterinary) play a crucial role in controlling this disease.

Malaria is caused by protozoan parasites, Plasmodium spp., that belong to the phylum Apicomplexa. The life cycle of these parasites depends on two different hosts; the definitive host, or vector, is a mosquito, and the intermediate host is a vertebrate, such as human. Malaria parasites use a unique form of substrate-dependent motility for host cell invasion and egress, which is dependent on an actomyosin motor complex called the glideosome. Apicomplexa have a small set of actin regulators, which are poorly conserved compared to their equivalents in higher eukaryotes. Actin-depolymerizing facto

Toxoplasma gondii, an apicomplexan parasite and agent of the human disease toxoplasmosis, possesses a non-photosynthetic relic plastid, named the apicoplast. Thought to be evolved from a red algal plastid, the apicoplast houses major metabolic pathways, such as heme, isoprenoid and lipid synthesis, crucial for parasite survival, and thus considered attractive drug targets. However, despite similarities with plant chloroplast lipid synthesis pathways, the apicoplast lacks canonical plant/chloroplast lipid transporters and so metabolite import/export is at present, poorly characterised. Here we

Apicomplexans are a diverse phylum of single-celled protists that are all obligate animal symbionts. The most well-studied members are severe pathogens of humans and livestock, such as the causative agents of malaria, cryptosporidiosis, toxoplasmosis, and coccidiosis, but comparatively little is known about their invertebrate-infecting counterparts, which are evolutionarily and ecologically important, but often overlooked. One of these groups is the Cephaloidophoroidea gregarine superfamily which is one of the most often-detected apicomplexan groups in molecular surveys of marine environments.

BACKGROUND: Colpodella species are classified within the domain Eukaryota, specifically under the order Colpodellida, family Colpodellaceae, and genus Colpodella, which are close relative of the phylum Apicomplexa. These organisms are unicellular, predatory flagellates. In recent years, their frequent detection in animal tissues, vector insect samples, and particularly in human has garnered significant attention as an emerging zoonotic threat. This review is to scope the biological characteristics and epidemiological features of Colpodella species.

Parasites belonging to the Apicomplexa phylum are among the most successful pathogens known in nature. They can infect a wide range of hosts, often remain undetected by the immune system, and cause acute and chronic illness. In this phylum, we can find parasites of human and veterinary health relevance, such as Toxoplasma, Plasmodium, Cryptosporidium, and Eimeria. There are still many unknowns about the biology of these pathogens due to the ethical and practical issues of performing research in their natural hosts. Animal models are often difficult or nonexistent, and as a result, there are ap
