Barbiturates are widely used as neuroprotective agents during status epilepticus and during surgical procedures that cause cerebral ischemia. The efficacy of this practice is unproved, however, and while barbiturates may counter neuronal excitotoxicity, they can also inhibit mitochondrial ATP production. Since glutamate uptake is energetically costly, mitochondrial inhibition could impair glutamate uptake. To examine this possibility, glutamate uptake was measured in primary rat astrocyte cultures in the presence of several barbiturates. Different barbiturates had differing effects on glutamat

The experimental research carried out on rabbits was aimed at determination of astrocytes reaction during brain inflammation caused by Herpes simplex virus. The study was made with the help of glia marker. Immunocytochemical findings showed that the earliest structural brain responses to the infection were changes in astrocyte glia. This was evident from the swelling and hypertrophy of vessel crus and from increased number of astrocytes around the vessels. Glia complexes (astrocyte agglomeration) were formed in the affected sites. During the exudative reaction the hyperplastic alterations of a

AIM: Electroconvulsive therapy (ECT) is effective for psychiatric disorders. However, its action mechanism remains unclear. We previously reported that transcription factor 7 (TCF7) was increased in patients successfully treated with ECT. TCF7 regulates Wnt pathway, which regulates adult hippocampal neurogenesis. Adult hippocampal neurogenesis is involved in the pathophysiology of psychiatric disorders. Astrocytes play a role in adult hippocampal neurogenesis via neurogenic factors. Of astrocyte-derived neurogenic factors, leukemia inhibitory factor (LIF) and fibroblast growth factor 2 (FGF2)

Heat Shock Protein 60 (HSP60) plays a critical role in maintaining mitochondrial function in astrocytes and has a significant impact on central nervous system (CNS) health. However, how HSP60 regulates the mitochondrial function of astrocytes to inhibit neuroregeneration remains unknown. In this study, we generated astrocyte-specific HSP60 knockout male mice to investigate the consequences of HSP60 deficiency. Firstly, our results confirmed that HSP60 deficiency caused abnormal expression of mitochondrial function-related genes, causing significant mitochondrial dysfunction, which triggered ce

Astrocytes are known to play many important roles in brain function. However, research underscoring the extent to which astrocytes modulate neuronal activity is still underway. Here we review the latest evidence regarding the contribution of astrocytes to neuronal oscillations across the brain, with a specific focus on how astrocytes respond to changes in brain state (e.g., sleep, arousal, stress). We then discuss the general mechanisms by which astrocytes signal to neurons to modulate neuronal activity, ultimately driving changes in behavior, followed by a discussion of how astrocytes contrib

Multiple sclerosis (MS) is a neuroinflammatory multifocal disorder. Optic neuritis is common in MS and leads to visual disability. No current treatments repair this damage. Discerning gene expression changes within specific cell types in optic nerve (ON) may suggest new treatment targets for visual disability in MS. Astrocytes are pivotal regulators of neuroinflammation, playing either detrimental or beneficial roles. Here, we used RiboTag technology to characterize the astrocyte-specific transcriptome in ON in the experimental autoimmune encephalomyelitis (EAE) model of MS. RNA sequencing ana

The retinal tumor provisionally referred to as presumed solitary circumscribed retinal astrocytic proliferation (PSCRAP) is a rare, benign lesion of unknown pathogenesis and histopathology. The authors present a case of PSCRAP that was analyzed with spectral-domain optical coherence tomography (OCT), which demonstrated a demarcation between the mass and the retinal nerve fiber layer. This finding stands in contrast to previous descriptions using time-domain OCT, suggesting that this tumor may originate from mid-retinal layers rather than the nerve fiber layer. In addition, the OCT of PSCRAP is

Cerebral amyloid angiopathy (CAA) is increasingly prevalent, and it is characterized by frequent recurrence and complex etiology. Aberrant ceruloplasmin (Cp) localization at the astrocytic endfeet, coupled with oxidative stress-induced dysregulation of iron regulatory proteins, is a central trigger of the iron dyshomeostasis that drives CAA progression. However, therapeutic strategies that specifically target iron transport regulation in astrocytes remain lacking. Here, we develop lattice-expanded Au/CeO2 with strong antioxidant capacity validated by DFT calculations. Its mesoporous architectu

Astrocytes coordinate neuronal activity with vascular development and metabolic homeostasis during postnatal brain maturation, yet the mechanisms enabling astrocytes to support angiogenesis remain elusive. VPS35, a core component of the retromer complex, regulates transmembrane protein trafficking and is implicated in multiple neurodegenerative diseases; however, its function in astrocytes is unknown. Here, we identify astrocytic VPS35 as a crucial regulator for astrocyte maturation and neurovascular development through its control of iron homeostasis and HIF1α-VEGFa signaling. Conditional del

Familial Alzheimer's disease (FAD) is a progressive neurodegenerative disorder associated with loss of cholinergic neurons, intra- and extracellular accumulation of amyloid beta (iAβ, eAβ), hyperphosphorylated tau, and neuroinflammation i.e., reactive glia. The precise contribution of astrocytes to neurodegeneration in FAD is still incompletely understood. Therefore, there is a need for a reliable and simple in vitro cell culture model of neuroinflammation. The aim of the present investigation was to determine the pathophysiological behavior of astrocyte-like cells (ALCs) derived from wild-typ

Multiple sclerosis (MS) is an inflammatory neurodegenerative disease of the central nervous system (CNS) with heterogeneous pathophysiology. In its progressive course oligodendrocyte and neuroaxonal damage is sustained by compartmentalized inflammation due to glial dysregulation. Siponimod (BAF312), a modulator of two sphingosine-1-phosphate (S1P) receptors (S1P1 and S1P5) is the first oral treatment specifically approved for active secondary progressive MS. To address potential direct effects of BAF312 on glial function and glia-neuron interaction, we set up a series of in vitro functional as

BACKGROUND: We previously demonstrated that dopamine (DA) overload may be a key mechanism behind development of minimal hepatic encephalopathy (MHE) in rats. It has been shown that low-grade cerebral oedema and oxidative stress play important roles in the pathogenesis of MHE. In the current study, DA-triggered oxidative injury in cerebral cortex was studied.
METHODS: An MHE rat model was used. DA was injected intracerebroventricularly (i.c.v.) into rats and added to primary cortical astrocytes (PCAs). Immunoblotting, immunoprecipitation and immunostaining were conducted after DA injection.

Intracerebral hemorrhage (ICH) is a particularly severe form of stroke, and reactive astrogliosis is a common response following injury to the central nervous system (CNS). Mesenchymal stem cells (MSCs) are reported to promote neurogenesis and alleviate the late side effects in injured brain regions. Gap junctions (Gjs) are abundant in the brain, where the richest connexin (Cx) is Cx43, most prominently expressed in astrocytes. Nuclear factor erythroid 2-related factor 2 (Nrf2) is an essential transcription factor regulating antioxidant reactions. Here, we aimed to explore whether bone marrow

Genetic loss of the mitochondrial control enzyme PINK1 leads to Parkinson's disease, characterized by dopaminergic neuron degeneration and neuroinflammation, yet its role in glia remains poorly understood. To address this gap, we investigated how the function of astrocytes and their ability to support neurons is influenced by PINK1 deficiency. For the first time, we demonstrate that human astrocytes exhibit robust PINK1 activity. Next, the first bulk transcriptomic study of human PINK1 mutant astrocytes was performed followed by biochemical validation at the protein level, uncovering homeostat

Alzheimer's disease (AD), the most prevalent form of dementia, is neuropathologically defined by the accumulation of extracellular amyloid-beta (Aβ) plaques and intracellular neurofibrillary tangles of hyperphosphorylated tau. Although traditionally viewed as a neuron-centric disorder, increasing evidence underscores the pivotal role of glial cells-particularly microglia and astrocytes-in AD pathogenesis. Once regarded as passive support cells, glia are now recognized as active participants in neuroinflammation, synaptic dysfunction, and disease progression. Microglia, the resident immune cell

Alzheimer's disease (AD) is a progressive neurodegenerative disorder marked by extracellular amyloid beta (Aβ) plaques, intracellular neurofibrillary tangles (NFTs), astrogliosis, loss of neurons, and cognitive decline. In this narrative review, we explore astrocytes' dual role in the healthy brain and the brain with AD, reflecting on the available human studies and animal models. Astrocytes are multifunctional regulators of brain homeostasis and neuronal activity within the central nervous system (CNS). These highly plastic cells undergo morphological and functional changes in response to the

OBJECTIVE: Adenosine is a key metabolic and neuroregulatory factor in the brain, and an adenosine-rich immunosuppressive microenvironment is formed post-stroke, making the adenosine pathway a crucial therapeutic target for improving stroke immunotherapy efficacy. This study aims to address the knowledge gaps hindering adenosine therapy translation, summarize the integrated network of extracellular and intracellular adenosine metabolism, and highlight the dynamic changes of adenosine metabolism in astrocytes and the potential of purine-converting enzymes as therapeutic targets for cerebral isch

Astrocytes are morphologically complex and functionally diverse glial cells that play central roles in neural circuits and disease. Although transcriptomic and physiological analyses have advanced understanding of astrocyte function, the intricacies of the relationship between gene expression and protein levels remains poorly understood. Proteins, rather than transcripts, execute the molecular processes that define astrocyte function, often within specialized subcellular compartments whose molecular composition has remained largely unresolved. Recent advances with genetically encoded proximity

Statins are commonly used cholesterol-lowering drugs, but their effects on astrocyte oxidative metabolism are poorly understood. To investigate this, rat astrocytes were exposed to 200 nM atorvastatin or simvastatin for 6 days and then assessed for changes in coenzyme Q (CoQ) homeostasis, mitochondrial function, and energy metabolism. Both statins comparably decreased cellular CoQ9 and CoQ10 levels (~35%), with greater losses of their reduced antioxidant forms (60-75%). Lower intracellular and mitochondrial levels of reactive oxygen species (ROS) were accompanied by the upregulation of nuclear
