The BHARAT study: a multi-modal, multi-omics investigation of aging signatures in the Indian population.

India is undergoing a rapid demographic transition, with its elderly population projected to exceed 347 million by 2050. Although aging is the primary risk factor for multiple chronic diseases, most biological age (BA) models have been developed for Western populations, with limited applicability to Indian demographics. The BHARAT study (Biomarkers of Healthy Aging, Resilience, Adversity, and Transitions) aims to develop and validate composite signatures of aging in the Indian population by integrating multi-omics, biochemical, clinical, and lifestyle data. The BHARAT study is a multi-center,

An epigenetic clock for human skeletal muscle.

BACKGROUND: Ageing is associated with DNA methylation changes in all human tissues, and epigenetic markers can estimate chronological age based on DNA methylation patterns across tissues. However, the construction of the original pan-tissue epigenetic clock did not include skeletal muscle samples and hence exhibited a strong deviation between DNA methylation and chronological age in this tissue. METHODS: To address this, we developed a more accurate, muscle-specific epigenetic clock based on the genome-wide DNA methylation data of 682 skeletal muscle samples from 12 independent datasets (18-8

Hypomorphic expression of parathyroid Bmal1 disrupts the internal parathyroid circadian clock and increases parathyroid cell proliferation in response to uremia.

The molecular circadian clock is an evolutionary adaptation to anticipate recurring changes in the environment and to coordinate variations in activity, metabolism and hormone secretion. Parathyroid hyperplasia in uremia is a significant clinical challenge. Here, we examined changes in the transcriptome of the murine parathyroid gland over 24 hours and found a rhythmic expression of parathyroid signature genes, such as Casr, Vdr, Fgfr1 and Gcm2. Overall, 1455 genes corresponding to 6.9% of all expressed genes had significant circadian rhythmicity. Biological pathway analysis indicated that the

Epigenetic age acceleration and mortality risk prediction in US adults.

Epigenetic clocks have emerged as novel measures of biological aging and potential predictors of mortality. We examined all-cause, cardiovascular, and cancer mortality prediction by epigenetic age acceleration (EAA) estimated using different epigenetic clocks. Among 2105 participants to the 1999-2002 National Health and Nutrition Examination Survey aged ≥ 50 years old and followed for mortality through 2019, we calculated EAAs from the residuals of nine epigenetic clocks regressed on chronological age. We assessed the association of EAAs and pace of aging with mortality adjusting for covariate

Epigenetic age acceleration is associated with oligodendrocyte proportions in MSA and control brain tissue.

AIMS: Epigenetic clocks are widely applied as surrogates for biological age in different tissues and/or diseases, including several neurodegenerative diseases. Despite white matter (WM) changes often being observed in neurodegenerative diseases, no study has investigated epigenetic ageing in white matter. METHODS: We analysed the performances of two DNA methylation-based clocks, DNAmClockMulti and DNAmClockCortical , in post-mortem WM tissue from multiple subcortical regions and the cerebellum, and in oligodendrocyte-enriched nuclei. We also examined epigenetic ageing in control and multiple

Proteomic organ-specific signatures, ageing traits, disease risks and genetic architecture in an East Asian population.

BACKGROUND: Protein-based ageing clocks can be used to help identify individuals at high risk of death or morbidity. There is a lack of evidence in non-European populations on clocks derived from combining different proteomic platforms and their relationship with age-related traits, diseases and genetic architecture. METHODS: The prospective China Kadoorie Biobank assayed proteins via Olink and SomaScan in ∼4000 individuals with a mean age of 58 years. We used organ-enriched plasma proteins identified from the Genotype-Tissue Expression Project and trained Light Gradient Boosting models on ch

Later-generation epigenetic aging clocks outperform first-generation models in predicting survival in TCGA breast cancer.

BACKGROUND: Epigenetic aging bridges the gap between biological and chronological age by exploiting DNA methylation (DNAm) patterns. Over the past decade, successive DNAm-based clocks have been introduced, beginning with the first-generation Horvath and Hannum models and extending to second-generation PhenoAge and the GrimAge family; complementary measures include DNAm-estimated telomere length and mitotic indices such as epiTOC/pcgtAge. We previously conducted a side-by-side evaluation of these metrics in colorectal cancer using publicly available data from The Cancer Genome Atlas (TCGA) COAD

Epigenetic Age Acceleration as a Modifiable Public Health Target: A Systematic Review and Meta-Analysis of Environmental, Behavioral, and Social Determinants with Development of the MEAB-Index.

Chronological age is a poor indicator of interindividual differences in biological aging. DNA methylation-based epigenetic clocks provide a reliable measure of biological age and epigenetic age acceleration (EAA). Although modifiable behavioral, environmental, and social factors appear to influence EAA, the magnitude, consistency, and potential preventability of these associations have never been systematically quantified. We conducted a systematic review and meta-analysis following PRISMA 2020 guidelines. PubMed/MEDLINE and Scopus were searched from inception to 7 April 2026 for English-langu

Roles for Phosphatase PP4 in Rhythmicity and Compensation in the Neurospora Circadian System.

The circadian clock is a highly conserved timer which allows organisms to anticipate future conditions driven by the 24-hour day on planet Earth. Circadian clocks from fungi to mammals are based on a transcription-translation feedback loop (TTFL) molecular architecture. Progressive phosphorylation of core clock proteins can alter both their activity and stability and is required for all known circadian TTFLs. The mechanism for kinase control of circadian period has been extensively studied; however, the mechanism(s) whereby phosphatases alter period remain less studied. Based on the observatio

Biomarkers for Biological Aging in Postmenopausal Women.

Biological aging describes the cumulative decline in physiologic function that can differ from chronological age. Menopause marks a reproductive transition that may influence biological aging through loss of estrogen, changes in body composition, chronic inflammation, and epigenetic regulation. These pathways may contribute to heterogeneity in cardiovascular, skeletal, cognitive, and functional outcomes among postmenopausal women. Researchers use composite indices and molecular measures, including phenotypic age and DNA methylation or epigenetic clocks, to quantify these processes. However, av

The mediating role of DNA methylation clocks in associations of race, ethnicity, education, income, and occupation with mortality: findings from NHANES 1999-2002.

Race, ethnicity, education, income, and occupation are important social factors affecting health and longevity. DNA methylation aging biomarkers may mediate the effects of these social factors on mortality, but so far, no studies have investigated the extent of this mediation systematically in a nationally represents samples of the U.S. population. This study aims to provide systematic evidence on the mediating proportion of DNA methylation clocks between these social factors and mortality using the US National Health and Nutrition Examination Survey (NHANES) 1999-2002 (N=2,402) linked with Na

Variations in Innate Immune Cell Subtypes Correlate with Epigenetic Clocks, Inflammaging and Health Outcomes.

Epigenetic clocks in blood have shown promise as tools to quantify biological age, displaying robust associations with morbidity and all-cause mortality. Whilst the effect of cell-type heterogeneity on epigenetic clock estimates has been explored, such studies have been limited to studying heterogeneity within the adaptive immune system. Much less is known about whether heterogeneity within the innate immune system can impact epigenetic clock estimates and their associations with health outcomes. Here, we apply a high-resolution DNAm reference panel of 19 immune cell-types, including young and

The Angiotensin-melatonin axis.

Accumulating evidence indicates that various biological and neuroendocrine circadian rhythms may be disrupted in cardiovascular and metabolic disorders. These circadian alterations may contribute to the progression of disease. Our studies direct to an important role of angiotensin II and melatonin in the modulation of circadian rhythms. The brain renin-angiotensin system (RAS) may modulate melatonin synthesis, a hormone with well-established roles in regulating circadian rhythms. Angiotensin production in the central nervous system may not only influence hypertension but also appears to affect

Circadian clock control of breast cancer hallmarks: molecular mechanisms and therapeutic implications.

Circadian rhythms regulate physiological functions and critical biological processes in almost all living forms. The circadian system enables organisms to anticipate and adapt to environmental fluctuations, thereby optimizing physiological responses. Proper synchronization between the master pacemaker in the brain and peripheral clocks throughout the organism is essential for maintaining homeostasis and promoting health. Conversely, circadian misalignment is increasingly recognized as a contributing factor in the pathogenesis of various diseases. Emerging evidence implicates circadian disrupti

Broad epigenetic shifts in the aging Drosophila retina contribute to its altered rhythmic transcriptome.

Alterations in biological rhythms are a common feature of aging, and disruption of circadian rhythms can exacerbate age-associated pathologies. The retina is critical for detecting light for both vision and for transmitting time-of-day information to the brain, synchronizing rhythms throughout the body. Disruption of circadian rhythms by manipulating the molecular clock leads to premature retinal degeneration in flies and mice, and gene expression rhythms are disrupted in models of age-associated ocular disease. Despite this, it is unknown how or why the gene expression rhythms of the retina c

Phase independent resetting in relaxation and bursting oscillators.

Relaxation oscillators that depend on one slow variable, such as the Fitzhugh-Nagumo oscillator, reset in a phase-dependent manner. A complete oscillation can be divided into two parts, the "plateau" and "trough", and a prematurely induced plateau or trough is significantly shorter than normal. The class of square-wave bursting oscillators can be viewed as relaxation oscillators with rapid spikes during the plateau, and reset similarly when modeled with one slow variable. However, it has been reported that a physiological bursting oscillator, the membrane potential of the pancreatic beta-cell,

Epigenetic ageing and the risk of Parkinson's disease.

BACKGROUND: Estimators of biological age, such as epigenetic clocks, are promising biomarkers for neurological disorders where the risk significantly increases with age, such as Parkinson's disease (PD). The purpose of this study was to prospectively investigate whether epigenetic age acceleration can predict PD risk, age at PD onset and time to phenoconversion. METHODS: We conducted a prospective, nested case-control study within the Nurses' Health Study, including participants who provided two blood samples before being diagnosed with PD. DNA methylation profiles were obtained from 75 indiv

Daily expression of clock genes in central and peripheral tissues of tree sparrow (Passer montanus).

Almost all organisms live in a fluctuating environment. To achieve synchrony with the fluctuating environment, organisms have evolved with time-tracking mechanism commonly known as biological clocks. This circadian clock machinery has been identified in almost all cells of vertebrates and categorized as central and peripheral clocks. In birds, three independent circadian clocks reside within the nervous tissues in the hypothalamus, pineal and retina, which interact with each other and produce circadian time at a functional level. There is limited knowledge available of the molecular clockwork,

Aging Biomarkers in Osteoporosis: A Systematic Review of Molecular Mechanisms, Biological Aging, and Population Evidence.

While chronological age is an important risk factor for osteoporosis, biological aging measures may better capture nuances in systemic and bone-tissue aging among same-age individuals. This review aims to summarize the associations between DNA methylation/telomere dysfunction (mainly telomere shortening) and bone health. We searched for human epidemiological studies that examined the associations between blood-based epigenetic aging clocks or leukocyte telomere length (LTL) and osteoporosis-related outcomes on PubMed and Embase.
