Effects of calcium antagonists on KCl-evoked calcium uptake by rat cortical synaptosomes.

A series of calcium antagonists were used to study their blocking effect on high potassium-induced calcium uptake into rat cortical synaptosomes; these antagonists were classified into five groups: dihydropyridine group (i.e. nifedipine and nitrendipine), benzothiazepine group (i.e. diltiazem), phenylalkylamine group (i.e. verapamil and D600), phenothiazine group (i.e. trifluoperazine) and diphenylpiperazine group (i.e. flunarizine and cinnarizine). Voltage-dependent 45Ca2+-uptake into this fraction was measured after 20 sec KCl-induced depolarization. The ID30 values of the above-mentioned an

Dynorphin reduces calcium-dependent action potential duration by decreasing voltage-dependent calcium conductance.

The opioid peptide dynorphin decreased somatic calcium-dependent action potential duration in a portion of mouse dorsal root ganglion (DRG) neurons without altering resting membrane potential or conductance. Dynorphin action was antagonized by naloxone. Responses of DRG neurons to dynorphin differed from responses to the opioid peptides leucine-enkephalin, which binds preferentially to delta-opiate receptors, and morphiceptin, which binds preferentially to mu-opiate receptors. Firstly, many DRG neurons responded to dynorphin but not to leucine-enkephalin or morphiceptin. Secondly, dynorphin re

Treatment of early-onset neonatal hypocalcemia. Effects on serum calcium and ionized calcium.

For the treatment of early-onset neonatal hypocalcemia, 50 neonates received either a high dose (group H), a low dose (group L), or no parenteral calcium (Ca) (group CON). Groups H and L received parenteral Ca either as a continuous infusion (group C) or by intermittent injection (group I). After the first 24 hours of treatment, groups H, C, and I had higher serum Ca concentrations than did group CON. After the entire 72-hour study period, group H had a lower incidence of hypocalcemia (serum Ca level < 7.0 mg/dL) when compared with all other patients. Although a relatively high Ca intake stati

Effects of diet calcium and 1,25-dihydroxyvitamin D3 on colon calcium active transport.

Unidirectional fluxes of calcium were studied in the absence of electrochemical gradients across rat descending colon segments in vitro. Dietary calcium restriction and 1,25-dihydroxyvitamin D3 [1,25-(OH)2D3]enhanced absorption by increasing the mucosal-to-serosal fluxes, whereas secretory (serosal-to-mucosal) fluxes were unchanged. Low-calcium diet also stimulated calcium uptake by everted gut sac segments of ascending as well as descending colon, whereas transverse colon was unresponsive. These results show that the colon is a target organ for 1,25-(OH)2D3 and demonstrate the participation o

Calcium antagonists and islet function. IX. Is extracellular calcium required for insulin release?

The physiological relevance of extracellular Ca2+ and Mg2+ to the process of glucose-induced insulin release is examined in both the isolated perfused rat pancreas and rat isolated pancreatic islets. When no Ca2+ is added to the perfusion or incubation media, both the initial and late phases of insulin release are severely impaired. The effect of Ca2+ deprivation is an immediate and immediately reversible phenomenon, although the length of the period of Ca2+ deprivation prior to stimulation with glucose modulates the severity of the impairment in the secretory response. When Mg2+ is omitted fr

Bacterial calcium transport: energy-dependent calcium uptake by membrane vesicles from Bacillus megaterium.

Membrane vesicles capable of energy-dependent calcium uptake have been prepared from Bacillus megaterium cells in log-phase growth or when undergoing sporulation. The uptake is dependent on the calcium concentration and appears saturable in vesicles from cells in log-phase growth. Both ascorbate and phenazine methosulfate are needed as a source of electrons for the energy-dependent increase in calcium uptake. Addition of 8 mM sodium cyanide inhibited the energy-dependent uptake. If this calcium uptake mechanism is a component of the sporulation-specific calcium accumulation process, the latter

The Alkaline Trap: Severe Calcium-Alkali Syndrome Secondary to Chronic Calcium Carbonate Ingestion.

Calcium-alkali syndrome is an increasingly recognised cause of parathyroid hormone (PTH)-independent hypercalcaemia resulting from excessive calcium and absorbable alkali ingestion, commonly through calcium-containing antacids or supplements. Its clinical presentation may mimic neurological or malignant disease, potentially leading to extensive investigation and delayed diagnosis. A 64-year-old woman presented following a fall with paraparesis and lower back pain. Investigations demonstrated severe hypercalcaemia (adjusted calcium 4.83 mmol/L), suppressed PTH of 1.4 pmol/L, metabolic alkalosis

Calcium Channel Blockers Modulate Calcium Homeostasis and Signaling Gene Expression in Sophora tonkinensis.

BACKGROUND: Calcium signaling plays a critical role in plant growth, development, and stress adaptation. However, the tissue-specific mechanisms governing calcium homeostasis in medicinal plants adapted to high-calcium karst environments remain poorly understood. This study investigated the functional divergence and potential molecular basis of calcium homeostasis in leaves and root tips of Sophora tonkinensis under pharmacological perturbation. METHODS: Treatments included lanthanum chloride (LaCl3) (a plasma membrane Ca2+ channel blocker), sodium orthovanadate (Na3VO4) (a Ca2+-ATPase inhibi

Total Versus Ionized Calcium: Valid Index of Calcium Status in Critically Ill Patients.

CONTEXT.—: Accurate evaluation of calcium status is essential in critically ill patients. However, reliance on total calcium (tCa) and albumin-corrected calcium often leads to misclassification, influencing clinical decisions. OBJECTIVE.—: To compare various calcium measurement methods with ionized calcium (iCa), the gold standard, and evaluate their diagnostic and prognostic relevance in intensive care unit (ICU) patients. DESIGN.—: A cross-sectional study was conducted for 1 year in 145 ICU patients at a tertiary care center. Measured parameters included tCa, iCa, albumin, total protein, a

Calcium-binding protein TgpCaBP regulates calcium storage of the zoonotic parasite Toxoplasma gondii.

Toxoplasma gondii, the causative parasite of toxoplasmosis, is an apicomplexan parasite that infects warm-blooded mammals. The ability of the calcium-binding proteins (CBPs) to transport large amounts of Ca2+ appears to be critical for the biological activity of T. gondii. However, the functions of some members of the CBP family have not yet been deciphered. Here, we characterized a putative CBP of T. gondii, TgpCaBP (TGME49_229480), which is composed of four EF-hand motifs with Ca2+-binding capability. TgpCaBP was localized in the cytosol and ER of T. gondii, and parasites lacking the TgpCaBP

Calcium Channel Blocker Overdose Treated with Calcium Resulting in Pancreatitis: A Case Report.

Calcium channel blockers (CCBs) are a mainstay for the treatment of hypertension. Here we report a case of a male who after intentionally ingesting amlodipine presented with overdose symptomology. His QTc (corrected QT) was 525 ms (millisecond) on admission, he was treated with calcium intravenous infusion and subsequently his QTc narrowed to 393 ms, but he also developed iatrogenic pancreatitis. His serum calcium levels were not checked during the infusion. He was treated with supportive care, which led to the normalization of serum calcium levels and a favorable outcome. Further studies are

[Body weight and bone/calcium metabolism. Bone and calcium metabolism in anorexia nervosa.].

Osteoporosis is one of chief complications of anorexia nervosa. Their calcium intake decreases and 84%are lack of vitamin D. The abnormal bone metabolism in severely emaciated patients with anorexia nervosa involves both a reduction in bone formation and an increase in bone resorption. The annual change in lumbar bone mineral density(BMD)is significantly correlated with body mass index(BMI)at the entry. The critical BMI for a positive increase in BMD was 16.4±0.3 kg/m2. Since 30%of patients are lack of vitamin K, their bone quality deteriorates. The risk factors of a decrease in lumbar vertebr

Calcium-sensing receptor (CaSR) mutations and disorders of calcium, electrolyte and water metabolism.

The extracellular calcium-sensing receptor (CaSR) is a family C G-protein-coupled receptor (GPCR) that is expressed at multiple sites, including the parathyroids and kidneys. The human CASR gene, located on chromosome 3q21.1, encodes a 1078 amino acid protein. More than 230 different disease-causing mutations of the CaSR have been reported. Loss-of-function mutations lead to three hypercalcemic disorders, which are familial hypocalciuric hypercalcemia (FHH), neonatal severe hyperparathyroidism and primary hyperparathyroidism. Gain-of-function mutations, on the other hand, result in the hypocal

Calcium signaling of thyrocytes is modulated by TSH through calcium binding protein expression.

TSH is an important stimulus to maintain thyroid epithelial differentiation. Impairment of TSH signal transduction can cause thyroid pathologies such as hot nodules, goiter and hyperthyroidism. In a gene expression study in Fischer rat thyroid cells (FRTL-5) using cDNA microarrays we found a TSH-dependent regulation of several calcium binding proteins, S100A4, S100A6 and annexin A6. Expression of these genes in FRTL-5 and regulation by TSH was confirmed with LightCycler qPCR and Western blotting. The differential expression of S100A4 was confirmed for cultured primary human thyrocytes. Calcium

Calcium hydroxide root canal sealers: evaluation of pH, calcium ion concentration and conductivity.

The calcium hydroxide ionization of four root canal sealers (Sealapex, CRCS, Sealer 26, and Apexit) was studied by measuring conductivity and pH and by conducting atomic absorption spectrophotometry. Samples 6 mm in diameter and 15 mm long were prepared from these sealers. After setting and 48 h storage in a desiccator, five samples of each material were placed in 50 mL distilled water and analysed after 0,1,2,4,6 and 24 h and 5, 15 and 30 days. The results showed that Sealapex was the root canal sealer showing the highest pH, ionic calcium and total calcium values (P < 0.05) throughout the ex

Calcium activated neutral protease & calcium ATPase in glucose-6-phosphate dehydrogenase deficiency hemizygotes.

To study the mechanism of haemolysis in G6PD deficient erythrocytes, studies were undertaken in G6PD deficiency and in normal erythrocytes artificially loaded with calcium. Significantly increased concentrations of calcium, calcium activated neutral protease (CANP) and calcium ATPase were found in patients of G6PD deficiency. However, the membrane bound calcium, the total glycoprotein and sulphydryl groups of membrane were observed to be decreased. Similar results were also observed in the normal erythrocytes when loaded with calcium. These results point to the role of the proteolytic process

Calcium binding by bile acids: in vitro studies using a calcium ion electrode.

In this study, we compared in vitro calcium binding by the taurine and glycine conjugates of the major bile acids in human bile: cholic (CA), chenodeoxycholic (CDCA) and deoxycholic (DCA) acids, together with the cholelitholytic bile acids ursodeoxycholic (UDCA) and ursocholic (UCA) acids. At physiological total calcium (CaTOT) (1-15 mM) and bile acid (BA) (10-50 mM) concentrations, all the bile acids caused concentration-dependent falls in [Ca2+], suggesting calcium binding. Except for glycine-conjugated CDCA, all the other calcium-bile acid complexes were soluble in 150 mM NaCl. The calcium

Comparison between the fractional isotopic calcium absorption and an oral calcium tolerance test.

In 27 subjects with several disorders of calcium metabolism, the fractional intestinal absorption of 47CaCl2 was rather poorly correlated with the urinary output of calcium or with the maximal increase of serum calcium after an oral calcium load. Conversely, a good correlation was observed with the product of these parameters. We propose that this product be used as an estimate of intestinal calcium absorption when a radioisotopic method is not available.

Deoxynivalenol induced intestinal barrier injury, mitochondrial dysfunction and calcium overload by inositol 1,4,5-triphosphate receptors (IP3Rs)-mitochondrial calcium uniporter (MCU) calcium axis.

Deoxynivalenol (DON) contamination is widespread in crops and could easily cause intestinal injury, which brings hazards to animals. Mitochondria are considered as an important target of DON, nevertheless, the mechanism is still unclear. Mitochondria-associated endoplasmic reticulum (ER) membranes (MAMs) have gained arousing interest and are recognized as critical signaling hubs that control calcium signaling transduction between ER and mitochondria. This study aims to investigate the effects of DON on intestinal barrier, mitochondria, MAMs and inositol 1,4,5-triphosphate receptors (IP3Rs)-mit
