The age-related effects on orthodontic tooth movement and the surrounding periodontal environment.

Orthodontic treatment in adults is often related to longer treatment time as well as higher periodontal risks compared to adolescents. The aim of this review is to explore the influence of age-related chages on orthodontic tooth movement (OTM) from macro and micro perspectives. Adults tend to show slower tooth movement speed compared to adolescence, especially during the early phase. Under orthodontic forces, the biological responses of the periodontal ligament (PDL) and alveolar bone is different between adult and adolescents. The adult PDL shows extended disorganization time, increased cell

Temporal and spatial patterning of axial myotome fibers in Xenopus laevis.

Somites give rise to the vertebral column and segmented musculature of adult vertebrates. The cell movements that position cells within somites along the anteroposterior and dorsoventral axes are not well understood. Using a fate mapping approach, we show that at the onset of Xenopus laevis gastrulation, mesoderm cells undergo distinct cell movements to form myotome fibers positioned in discrete locations within somites and along the anteroposterior axis. We show that the distribution of presomitic cells along the anteroposterior axis is influenced by convergent and extension movements of the

Heartburn: cardiac potassium channels involved in parietal cell acid secretion.

Hydrochloric acid is produced in parietal cells of the gastric glands by an H(+)/K(+)-ATPase. This proton pump couples the outwards movement of H(+) to the inwards movement of K(+) thus requiring the presence of luminal K(+) to operate. To maintain the activity of the pump, K(+) recirculates over the apical membrane via conductive pathways, the molecular nature of which has been identified in the past few years. This review gives a short overview about the recent advances in the understanding of the role of K(+) channels in the process of parietal cell H(+) secretion and focuses on the identif

Endocardial transcription factor HAND2 orchestrates hypoxic and TGF-β signaling to modulate coronary artery formation.

The endocardium is a major source of coronary angiogenesis and arterialization, through coordinated cell fate transition and migration. However, the transcriptional regulatory network synchronizing cell fate determination and movement remains unclear. Here, we identified transcription factor HAND2 as a key candidate for coronary vascular formation. Endocardial deletion of Hand2 in mice disrupted arterial-venous networks and stunted coronary arteries, paralleling a ventricular noncompaction phenotype. Moreover, deletion of Hand2 produced excessive tip cells with defective movement. RNA-seq anal

Whole-Mount Dissection of the Vestibular Complex and Quantification of Vestibular Ganglion Neurons in Mice.

The vestibular system is crucial for our sense of balance, for detecting head movements, and for regulating gaze and posture. There are five vestibular organs located inside the temporal bone, including two otolith organs (utricle and saccule), which detect linear movement, and three cristae (lateral/horizontal, anterior, and posterior), which detect rotational movement. Each organ contains a sensory epithelium with hair cells that detect head movements and supporting cells that provide structural support, release growth factors and cytokines, and phagocytose dying hair cells. The neurons that

Sequelae of experimental tympanic and inferior canal wall perforations: the double meaning of epithelial migration.

The term "epithelial migration" has been used to describe both the normal surface movement of the tympanic epithelium and the movement of epithelial basal cells in repair processes. In an attempt to distinguish between these two processes and to determine their role in the repair of a wounded tympanic membrane, 20 guinea pig tympanic membranes were perforated inferiorly and tattooed with ink through an external incision. Sequential histology of whole temporal bones at intervals from the time of injury up to three weeks showed evidence of movement of the superficial layers of epidermis which wa

Tisssue responses in corticotomy- and osteotomy-assisted tooth movements in rats: histology and immunostaining.

INTRODUCTION: The purpose of this histologic study was to examine underlying cellular responses to corticotomy- and osteotomy-assisted tooth movements. METHODS: Thirty-six rats were divided into 5 groups: corticotomy-assisted tooth movement (CO + TM), sham corticotomy without tooth movement (CO alone), osteotomy-assisted tooth movement (OS + TM), sham osteotomy without tooth movement (OS alone), and unassisted tooth movement (TM alone). Standard orthodontic springs were activated to produce mesial tooth movement. The rats were killed at 3, 21, and 60 days after activation for osteoclast and b

The spatial landscape of glial pathology and T cell response in Parkinson's disease substantia nigra.

Parkinson's Disease (PD) is an incurable neurodegenerative disease that causes movement disorders. Neurons in PD aggregate α-synuclein and are depleted from the substantia nigra (SN), which is a movement control hub. The presence of α-synuclein-reactive T cells in PD patient blood suggests a role for adaptive immunity in the pathogenesis of PD. However, the characteristics of this response within the brain are not well understood. Here, we employed single-nucleus RNAseq, spatial transcriptomics, and T cell receptor (TCR) sequencing to analyze T cell and glial cell states in post-mortem PD brai

Motility voltage sensor of the outer hair cell resides within the lateral plasma membrane.

The outer hair cell (OHC) from the organ of Corti is believed to be responsible for the mammal's exquisite sense of hearing. A membrane-based motile response of this cell underlies the initial processing of acoustic energy. The voltage-dependent capacitance of the OHC, possibly reflecting charge movement of the motility voltage sensor, was measured in cells during intracellular dialysis of trypsin under whole cell voltage clamp. Within 10 min after dialysis, light and electron microscopic examination revealed that the subplasmalemmal structures, including the cytoskeletal framework and subsurf

Pathogenic LRRK2 mutations cause loss of primary cilia and Neurturin in striatal parvalbumin interneurons.

Parkinson's disease-associated, activating mutations in the LRRK2 kinase block primary cilium formation in cell culture and in specific cell types in the brain. In the striatum that is important for movement control, about half of astrocytes and cholinergic interneurons, but not the predominant medium spiny neurons, lose their primary cilia. Here, we show that mouse and human striatal parvalbumin interneurons that are inhibitory regulators of movement also lose primary cilia. Without cilia, these neurons are not able to respond to Sonic hedgehog signals that normally induce the expression of P

Decoupling of motor cortex to movement in Parkinson's dyskinesia rescued by sub-anaesthetic ketamine.

Gamma-band and single-unit neural activity in primary motor cortex are involved in the control of movement. This activity is disrupted in Parkinson's disease (PD) and levodopa-induced dyskinesia (LID), a debilitating consequence of dopamine replacement therapy for PD. Physiological features of LID include pathological narrowband gamma oscillations, finely tuned gamma and altered primary motor cortex firing activity. Given that most studies characterize LID through visual scoring, little is known about the relationships between ongoing dyskinetic movements, gamma and neuronal activity at fast (

Nuclear migration in mammalian brain development.

During development of the mammalian brain, neural stem cells divide and give rise to adult stem cells, glia and neurons, which migrate to their final locations. Nuclear migration is an important feature of neural stem cell (radial glia progenitor) proliferation and subsequent postmitotic neuronal migration. Defects in nuclear migration contribute to severe neurodevelopmental disorders such as microcephaly and lissencephaly. In this review, we address the cellular and molecular mechanisms responsible for nuclear migration during the radial glia cell cycle and postmitotic neuronal migration, wit

Occlusion-related cue used for analysis of motion in the primate visual cortex.

Although cells in ventral part of the medial superior temporal area (MST) do not respond to movements of a wide textured field, many of them start to respond when a stationary object is placed in front of the moving field. The effective direction in this stimulus configuration is opposite to the preferred direction of individual cells for movement of an object. Here, we examined what stimulus cues were detected by the cells in response to movements of a background pattern behind an object. The responses disappeared when the border of the object was blurred. Because blurring the border degraded

Modes of mechanical guidance of adhesion-independent cell migration.

Adhesion-independent migration is a prominent mode of cell motility in confined environments, yet the physical principles that guide such movement remain incompletely understood. We present a phase-field model for simulating the motility of deformable, non-adherent cells driven by contractile surface instabilities of the cell cortex. This model couples surface and bulk hydrodynamics, accommodates large shape deformations and incorporates a diffusible contraction-generating molecule (myosin) that drives cortical flows. These capabilities enable a systematic exploration of how mechanical cues di

An isoform of 14-3-3 protein regulates transbilayer lipid movement at the plasma membrane.

Exogenous phosphatidylserine (PS) is cytotoxic to Chinese hamster ovary (CHO) cells with a PS-resistant CHO cell mutant exhibiting impaired transbilayer movement of a fluorescent PS analog at the plasma membrane. Here, we demonstrate that both mRNA and protein levels of 14-3-3 zeta are markedly reduced in this mutant, while knockdown of 14-3-3 zeta in wild-type CHO cells confers partial resistance to exogenous PS. Consistently, these knockdown cells display defective transbilayer movement of 1-palmitoyl-2-{6-[(7-nitro-1,3-benz-2-oxadiazol-4-yl) amino] hexanoyl}-sn-glycero-3-phosphoserine (C6-N

Hippocampal conjunctive and complementary CA1 populations relate sensory events to movement.

Hippocampal CA1 neurons respond to sensory stimuli during enforced immobility, movement, and their transitions in a new conveyor belt task. Head-fixed mice were exposed to light flashes or air streams while at rest, spontaneously moving, or running a fixed distance. Two-photon calcium imaging of CA1 neurons revealed that 62% of 3341 imaged cells were active during one or more of 20 sensorimotor events. Of these active cells, 17% were active for any given sensorimotor event, with a higher proportion during locomotion. The study found two types of cells: Conjunctive cells that were active across

Directional specificity for head rotation in the pigeons posterior cerebellum.

Neurons in the posterior cerebellum, including the vestibulocerebellum, which respond to rotation in the plane of the horizontal semicircular canal exhibit considerably greater plane specificity than is known for primary afferents. This is shown by a response amplitude decline with tilt out of the canal plane which exceeds the decline predicted to a cosine function. Of 96 horizontal canal cells, 41 also responded to yaw movements about neck joints with only three of these cells exhibiting responses to roll movements as well. Other neurons responding to head rotation in roll very commonly respo

4D imaging analysis of the aging mouse neural stem cell niche reveals a dramatic loss of progenitor cell dynamism regulated by the RHO-ROCK pathway.

In the adult ventricular-subventricular zone (V-SVZ), neural stem cells (NSCs) give rise to transit-amplifying progenitor (TAP) cells. These progenitors reside in different subniche locations, implying that cell movement must accompany lineage progression, but the dynamic behaviors of adult NSCs and TAPs remain largely unexplored. Here, we performed live time-lapse imaging with computer-based image analysis of young and aged 3D V-SVZ wholemounts from transgenic mice with fluorescently distinguished NSCs and TAP cells. Young V-SVZ progenitors are highly dynamic, with regular process outgrowth a

Purkinje cells of the cerebellar dorsal vermis: simple-spike activity during pursuit and passive whole-body rotation.

To track a slowly moving object during whole body rotation, smooth-pursuit and vestibularly induced eye movements must interact to maintain the accuracy of eye movements in space (i.e., gaze), and gaze movement signals must eventually be converted into eye movement signals in the orbit. To understand the role played by the cerebellar vermis in pursuit-vestibular interactions, in particular whether the output of the vermis codes gaze-velocity or eye-velocity, we examined simple-spike activity of 58 Purkinje (P-) cells in lobules VI-VII of head-stabilized Japanese monkeys that were trained to el
