Multiomics Dissection Reveals Natural Killer Cell-CD8+ T-cell Cooperation in Shaping an Immunostimulatory Tumor Microenvironment.

UNLABELLED: Immune evasion remains a major barrier to effective immunotherapy in cancer. Coordinated activation of different immune cell types shapes immune responses within the tumor microenvironment (TME). In this study, we uncovered a spatiotemporal synergy between cytotoxic T lymphocytes (CTL) and natural killer (NK) cells in tumors with low T-cell infiltration. An integrative multiomics approach combining spatial transcriptomics, single-cell RNA sequencing, and high-dimensional flow cytometry demonstrated that vaccination, capable of activating both innate and adaptive immunity, rapidly r

TLR2-induced CD8+ T-cell deactivation shapes dendritic cell differentiation in the bone marrow during sepsis.

Sepsis is associated with profound immune dysregulation that increases the risk for life-threatening secondary infections: Dendritic cells (DCs) undergo functional reprogramming due to yet unknown changes during differentiation in the bone marrow (BM). In parallel, lymphopenia and exhaustion of T lymphocytes interfere with antigen-specific adaptive immunity. We hypothesized that there exists a link between T cells and the modulation of DC differentiation in the BM during murine polymicrobial sepsis. Sepsis was induced by cecal ligation and puncture (CLP), a model for human bacterial sepsis. At

Three-Dimensional Shapes and Cell Deformability of Rat Red Blood Cells during and after Asphyxial Cardiac Arrest.

Changes in microcirculation are believed to perform an important role after cardiac arrest. In particular, rheological changes in red blood cells (RBCs) have been observed during and after ischemic-reperfusion injury. Employing three-dimensional laser interferometric microscopy, we investigated three-dimensional shapes and deformability of RBCs during and after asphyxial cardiac arrest in rats at the individual cell level. Rat cardiac arrest was induced by asphyxia. Five rats were maintained for 7 min of no-flow time, and then, cardiopulmonary resuscitation (CPR) was started. Blood samples wer

Cancer cell genetics shaping of the tumor microenvironment reveals myeloid cell-centric exploitable vulnerabilities in hepatocellular carcinoma.

Myeloid cells are abundant and plastic immune cell subsets in the liver, to which pro-tumorigenic, inflammatory and immunosuppressive roles have been assigned in the course of tumorigenesis. Yet several aspects underlying their dynamic alterations in hepatocellular carcinoma (HCC) progression remain elusive, including the impact of distinct genetic mutations in shaping a cancer-permissive tumor microenvironment (TME). Here, in newly generated, clinically-relevant somatic female HCC mouse models, we identify cancer genetics' specific and stage-dependent alterations of the liver TME associated w

Autopsy case of a patient with rapidly progressive combined small-cell lung carcinoma with spindle-shaped cell tumor.

A 69-year-old Japanese man visited our hospital because of worsening shortness of breath. His chest computed tomography (CT) showed a giant left lung mass with a massive left pleural effusion. He could not be treated with chemotherapy and eventually died from a rapidly progressive tumor. He was diagnosed with combined small cell lung carcinoma (C-SCLC) with spindle-shaped cell tumor at autopsy. C-SCLC is characterized by pathologically concurrent SCLC and adenocarcinoma or squamous cell carcinoma, or rarely, spindle-shaped cell tumor. The clinical course of C-SCLC with spindle-shaped cell tumo

Leydig cell tumors of the testis with unusual features: adipose differentiation, calcification with ossification, and spindle-shaped tumor cells.

We report 19 Leydig cell tumors (LCTs) of the testis with adipose differentiation (n = 12) and/or spindle cell growth (n = 8) in patients 28-70 years of age; three tumors with adipose differentiation showed psammomatous calcifications, two of which also had foci of ossification. In eight tumors fat-like cells apparently derived from lipid accumulation within neoplastic Leydig cells and appeared as focal to prominent clusters in a background of vacuolated, neoplastic Leydig cells. The fat-like cells were usually immunoreactive for Leydig cell markers (inhibin-alpha, calretinin, and melan-A) but

Shaping of Drosophila Neural Cell Lineages Through Coordination of Cell Proliferation and Cell Fate by the BTB-ZF Transcription Factor Tramtrack-69.

Cell diversity in multicellular organisms relies on coordination between cell proliferation and the acquisition of cell identity. The equilibrium between these two processes is essential to assure the correct number of determined cells at a given time at a given place. Using genetic approaches and correlative microscopy, we show that Tramtrack-69 (Ttk69, a Broad-complex, Tramtrack and Bric-à-brac - Zinc Finger (BTB-ZF) transcription factor ortholog of the human promyelocytic leukemia zinc finger factor) plays an essential role in controlling this balance. In the Drosophila bristle cell lineage

Osteocyte-lacuna shape and canaliculi architecture dictate fluid flow around osteocyte, and strain of cell and bone matrix: implications for cell mechanobiology and bone fragility.

Osteocytes, the most abundant cells in bone, play a critical role in maintaining bone quality by sensing mechanical loads and orchestrating bone modeling and remodeling. These cells are housed in lacunae and connected by a complex network of canaliculi, through which interstitial fluid flows in response to mechanical loading. Osteocyte-lacuna shape can vary from elongated in healthy lamellar bones subjected to directional loading to more spherical shapes, often seen in flat bones, or in aging and diseases. Additionally, canaliculi can be star-shaped or run perpendicular to the lacunar major ax

Epithelial-Mesenchymal Transition Shapes the Lipotoxic Response of Colon Cancer Cells to Palmitic Acid.

Saturated fatty acids such as palmitic acid (PA) can induce lipotoxic stress, whereas monounsaturated fatty acids like oleic acid (OA) often promote adaptive responses through lipid droplets (LDs) formation. Here, we reveal that epithelial-mesenchymal transition (EMT) profoundly influences the lipotoxic response of colorectal cancer cells. Using the epithelial-like HCT15 and mesenchymal-like HCT116 cell lines, we combined proteomic, metabolic, and imaging analyses to elucidate how EMT status determines lipid storage capacity and resistance to PA-induced toxicity. A basal proteomic profiling hi

CNS1-dependent regulatory T cells shape recovery from acute lung injury.

Regulatory T cells (Tregs) play a crucial role in mediating recovery from acute lung injury (ALI). However, the complex roles of functionally heterogeneous Treg subsets in the lung during the resolution of acute inflammation remain unclear. To investigate the role of peripherally induced Tregs, we utilized mice lacking conserved noncoding sequence 1 (CNS1) of the Foxp3 locus, a genetic deletion that impairs peripheral Treg induction. We found that CNS1-deficient mice exhibit greater mortality and delayed resolution during ALI. Tregs induced via CNS1 modulated antiviral and proinflammatory immu

Langerhans cell control of early-life dermal lymphatic development shapes adult immunity.

The communication between skin and draining lymph nodes is crucial for immune responses to skin insults. Lymphatic vessels play key roles in this communication by transporting antigens from skin to lymph nodes, and mechanisms that compromise dermal lymphatic function can disrupt skin immunity. Here, we show that Langerhans cells (LCs), epidermis-derived antigen-presenting cells, promote dermal lymphatic expansion and phenotype acquisition during early life. This function was mediated by PlGF (placental growth factor) and VEGF-C (vascular endothelial growth factor C), and LCs selectively shaped

Type I interferons modulate autophagy to shape gemcitabine response in pancreatic cancer cells.

Pancreatic ductal adenocarcinoma (PDAC) is an aggressive malignancy characterized by poor prognosis and limited response to gemcitabine, the standard first-line chemotherapy. One major contributor to chemoresistance is autophagy, a process frequently upregulated in PDAC. In this study, we examined the ability of type I interferons (IFNα2b and IFNβ1a) to modulate autophagy and disturb tumor cell resistance to gemcitabine. PDAC cells were treated with increasing concentrations of IFNα2b or IFNβ1a, and cell proliferation was assessed by [3H]-thymidine incorporation. Apoptosis was evaluated by TUN

Tissue factor and regenerative medicine: shaping cell and extracellular vesicle-based therapeutic approaches.

Regenerative medicine, characterized by breakthroughs in stem cell technology, cell-free therapies, and tissue engineering, holds immense promise for treating various diseases. However, along with the encouraging advancements in translational and clinical research, emerging evidence has demonstrated a potential risk of developing instant blood-mediated inflammatory reactions (IBMIRs) and venous thromboembolism (VTE) with the intravascular delivery of cellular products that do not naturally come into contact with blood. Tissue factor (TF), an initiator of the extrinsic coagulation cascade, play

Evolution of compound eye cell types shapes visual behaviors across Heliconius butterflies.

The evolution of visual systems is tightly linked to the diversification of visually guided behaviors. Using Heliconius butterflies, renowned for their rapid diversification of wing color patterns and corresponding visual mate preferences, we investigate the evolution of compound eye cell types across a complete speciation continuum. By integrating population genomics, chromatin conformation capture, and single-nucleus multi-omics, we uncover multi-modal mechanisms driving visual system evolution. Within a population polymorphic for mate preference, we identify senseless-2 as a glia-expressing

Matrix stiffness shapes early transcriptional trajectories during hepatic stellate cell activation.

BACKGROUND: Matrix stiffness increases during fibrosis and drives hepatic stellate cell (HSC) activation through YAP-dependent mechanotransduction. Although recent studies have revealed HSC heterogeneity beyond classical quiescent and fully activated states, how physiological-range stiffness influences activation trajectories underlying this heterogeneity remains unclear. This study aimed to determine how substrate stiffness shapes the transcriptomic profiles and biases activation trajectories. METHODS: We performed time-resolved single-cell RNA sequencing of primary mouse HSCs cultured on st

Derivation of Mouse Dome-Shaped Trophoblast Stem Cells With Low Heterogeneity From Blastocysts.

Trophoblast stem cells (TSCs) have emerged as a valuable model for investigating placentation and related disorders. Despite their ability to be maintained over multiple passages without losing stemness, conventional TSCs are characterised by notorious heterogeneity and a spontaneous differentiation tendency. In this study, we established a type of mouse TSCs with dome-shaped colonies (dTSCs). The dTSCs expressed canonical markers of conventional TSCs and exhibited relatively low levels of heterogeneity and spontaneous differentiation when cultured in conventional trophoblast stem cell medium.

CellMet: Extracting 3D shape and topology metrics from confluent cells within tissues.

During development and tissue repair, cells reshape and reconfigure to ensure organs take specific shapes. This process is inherently three-dimensional (3D). Yet, in part due to limitations in imaging and data analysis, cell shape analysis within tissues has largely been studied in two-dimensions (2D), e.g., the Drosophila wing disc. With recent advances in imaging and machine learning, there has been significant progress in our understanding of 3D cell and tissue shape in vivo. However, even after gaining 3D segmentation of cells, it remains challenging to extract cell shape metrics beyond vo

Targeting NF-kappaB-inducing kinase shapes B-cell homeostasis in myasthenia gravis.

BACKGROUND: B cell immune dysregulation plays a critical role in myasthenia gravis (MG). However, targeted B-cell therapy such as rituximab may result in long-term peripheral B cell clearance and allow for the survival of plasma cells, contributing to frequent infections and relapses. Therefore, we aimed to identify potential novel therapeutic targets that preserve part of B cell function while inhibiting antibody-secreting cells (ASCs). METHODS: The transcriptome of sorted CD19+B cells obtained from MG patients in active and remission state was performed by RNA sequencing. The hallmark gene

Particle and Cell Separation in Deterministic Lateral Displacement Arrays with Inverse L-Shaped Pillars.

Deterministic lateral displacement (DLD) has emerged as a powerful microfluidic technique for label-free particle separation with high resolution. Although recent innovations in pillar geometry have broadened its biomedical applications, the fundamental mechanisms dictating flow behavior and separation efficiency remain not fully understood. In this study, we conducted dissipative particle dynamics simulations to systematically investigate the separation of rigid spherical particles and red blood cells (RBCs) in DLD arrays with inverse L-shaped pillars. The simulations established a predictive
