Ataxia-telangiectasia mutated (ATM) deficiency decreases reprogramming efficiency and leads to genomic instability in iPS cells.

During cell division, one of the major features of somatic cell reprogramming by defined factors, cells are potentially exposed to DNA damage. Inactivation of the tumor suppressor gene p53 raised reprogramming efficiency but resulted in an increased number of abnormal chromosomes in established iPS cells. Ataxia-telangiectasia mutated (ATM), which is critical in the cellular response to DNA double-strand breaks, may also play an important role during reprogramming. To clarify the function of ATM in somatic cell reprogramming, we investigated reprogramming in ATM-deficient (ATM-KO) tail-tip fib

BRD2 impedes iPSC reprogramming by regulating lipid biosynthesis and the matrisome via its ET tail.

Induction of human pluripotent stem cells (hiPSCs) from somatic cells encounters significant barriers that remain poorly understood. Lipids are fundamental cellular molecules with many essential roles. However, the roles of lipids in reprogramming are unknown. Here, we show that BRD2 is a barrier to hiPSC reprogramming by suppressing lipid genes and maintaining the somatic transcriptional program, particularly the somatic matrisome program. Strikingly, the acetylation epigenetic reader BRD2 unorthodoxly suppresses the expression of lipid-related genes in the reprogramming cells. The two rate-l

Induced Neurons for Disease Modeling and Repair: A Focus on Non-fibroblastic Cell Sources in Direct Reprogramming.

Direct cellular reprogramming exhibits distinct advantages over reprogramming from an induced pluripotent stem cell intermediate. These include a reduced risk of tumorigenesis and the likely preservation of epigenetic data. In vitro direct reprogramming approaches primarily aim to model the pathophysiological development of neurological disease and identify therapeutic targets, while in vivo direct reprogramming aims to develop treatments for various neurological disorders, including cerebral injury and cancer. In both approaches, there is progress toward developing increased control of subtyp

Enhanced generation of human induced pluripotent stem cells by ectopic expression of Connexin 45.

Somatic cells can be successfully reprogrammed into pluripotent stem cells by the ectopic expression of defined transcriptional factors. However, improved efficiency and better understanding the molecular mechanism underlying reprogramming are still required. In the present study, a scrape loading/dye transfer assay showed that human induced pluripotent stem cells (hiPSCs) contained functional gap junctions partially contributed by Connexin 45 (CX45). We then found CX45 was expressed in human embryonic stem cells (hESCs) and human dermal fibroblasts (hDFs) derived hiPSCs. Then we showed that C

Epigenetic Modifications in the Retinal Pigment Epithelium of the Eye During RPE-Related Regeneration or Retinal Diseases in Vertebrates.

The retinal pigment epithelium (RPE) is a cellular source of retinal regeneration in lower vertebrates and a cellular source of retinal diseases in mammals, including humans. Both processes are based on a genetic program for the conversion of RPE cells into cells of other phenotypes: neural in the first case and mesenchymal in the second. RPE reprogramming in the neural direction is realized in tailed amphibians and bird embryos in vivo, but in higher vertebrates and humans, this process is realized in vitro. Epigenetic regulation determines the phenotypic plasticity of RPE cells, i.e., their

A circRNA platform for safe partial reprogramming and rejuvenation of mesenchymal stem cells without pluripotent conversion.

BACKGROUND: Mesenchymal stem cells (MSCs) are widely used in cell therapies; however, replicative senescence during in vitro expansion limits their potency and consistency. Partial reprogramming has emerged as a promising strategy to rejuvenate aged cells, yet concerns regarding unintended pluripotent conversion have hindered its translational application. METHODS: Here we developed a circular RNA (circRNA)-based platform enabling safe partial reprogramming and rejuvenation of MSCs. We generated a lipid nanoparticle (LNP)-delivered circRNA co-expressing OCT4, SOX2, and KLF4 (circ-OSK) to achi

The SynMuvA lin-15A licenses natural transdifferentiation by antagonizing identity safeguarding mechanisms.

The mechanisms that restrict or enable latent cellular plasticity have attracted growing interest over the past decade, with important implications for cancer and regenerative therapies. However, the diversity of both pro- and anti-plasticity mechanisms remains incompletely understood. Here, we identify the THAP domain gene lin-15A as a novel factor involved in the natural rectal-to-neuronal Y-to-PDA transdifferentiation in Caenorhabditis elegans. Our genetic analyses show that lin-15A acts in parallel to previously described plasticity factors. Null mutant suggests that, rather than driving t

Bone marrow mesenchymal stromal cells metabolic reprogramming in systemic lupus erythematosus: remodeling of bone marrow microenvironment and regulation of immune cell fate.

Systemic Lupus Erythematosus (SLE) is a chronic immune-mediated inflammatory disease characterized by dysregulated immune tolerance, abnormal secretion of autoantibodies, and multi-organ damage. Among them, mutations in genetic susceptibility genes, abnormal epigenetic modifications, excessive oxidative stress, abnormal accumulation of inflammatory factors, and intestinal flora disorders are all key specific factors that lead to immune dysfunction, abnormal production of autoantibodies, and multi-organ damage in patients with SLE. The bone marrow microenvironment, as a key niche for immune cel

In Vivo Reprogramming of Tissue-Derived Extracellular Vesicles for Treating Chronic Tissue Injury Through Metabolic Engineering.

Extracellular vesicles (EVs) have emerged as promising therapeutics for regenerative medicine, but the efficacy of current exogenous EV-based therapies for treating chronic tissue injury is still unsatisfactory. Exercise can affect skeletal muscle EV secretion and that this process regulates the systemic health-promoting role of exercise, suggesting that fine-tuning of endogenous tissue EV secretion may provide a new therapeutic avenue. Here, this work reports that in vivo reprogramming of EV secretion via metabolic engineering is a promising strategy for treating chronic diseases. Briefly, ex

Electromagnetized MXenes Enhance the Efficient Direct Reprogramming of Dopamine Neurons for Parkinson's Disease Therapy.

MXenes, a two-dimensional transition metal carbide and nitride, have shown significant potential in various biological applications. In particular, the distinct properties of MXenes─including their functionalizable surface, biocompatibility, and conductive characteristics, make them highly promising materials for advancing biomedical technologies. Here, we report that MXene, under specific electromagnetic field (EMF) conditions, effectively promotes the direct lineage reprogramming of induced dopaminergic (iDA) neurons both in vitro and in vivo. Remarkably, we found that electromagnetized MXen

Essential and dual effects of Notch activity on a natural transdifferentiation event.

Cell identity can be reprogrammed, naturally or experimentally, albeit with low frequency. Why some cells, but not their neighbours, undergo a cell identity conversion remains unclear. We find that Notch signalling plays a key role to promote natural transdifferentiation in C. elegans hermaphrodites. Endogenous Notch signalling endows a cell with the competence to transdifferentiate by promoting plasticity factors expression (hlh-16/Olig and sem-4/Sall). Strikingly, ectopic Notch can trigger additional transdifferentiation in vivo. However, Notch signalling can both promote and block transdiff

Direct Reprogramming Improves Cardiac Function and Reverses Fibrosis in Chronic Myocardial Infarction.

BACKGROUND: Because adult cardiomyocytes have little regenerative capacity, resident cardiac fibroblasts (CFs) synthesize extracellular matrix after myocardial infarction (MI) to form fibrosis, leading to cardiac dysfunction and heart failure. Therapies that can regenerate the myocardium and reverse fibrosis in chronic MI are lacking. The overexpression of cardiac transcription factors, including Mef2c/Gata4/Tbx5/Hand2 (MGTH), can directly reprogram CFs into induced cardiomyocytes (iCMs) and improve cardiac function under acute MI. However, the ability of in vivo cardiac reprogramming to repai

IFI16 Impacts Metabolic Reprogramming during Human Cytomegalovirus Infection.

Cellular lipid metabolism plays a pivotal role in human cytomegalovirus (HCMV) infection, as increased lipogenesis in HCMV-infected cells favors the envelopment of newly synthesized viral particles. As all cells are equipped with restriction factors (RFs) able to exert a protective effect against invading pathogens, we asked whether a similar defense mechanism would also be in place to preserve the metabolic compartment from HCMV infection. Here, we show that gamma interferon (IFN-γ)-inducible protein 16 (IFI16), an RF able to block HCMV DNA synthesis, can also counteract HCMV-mediated metabol

Induced pluripotent stem cells from urine of Duchenne muscular dystrophy patients.

BACKGROUND: The most common muscular dystrophy, Duchenne muscular dystrophy (DMD), is a lethal, X-linked disorder with no widespread cure. Worldwide, in vitro studies involving new, mutation-specific cures and regenerative therapies are employing disease-specific patient-specific cells. However, these may not be completely relevant for Pakistani children because of the human genome diversities and geographic variation in mutation type and frequency. Therefore, this study aimed to generate DMD induced pluripotent stem cells (iPSCs) from the urine of Pakistani children with DMD, to serve as a pr

Dedifferentiated melanomas: Morpho-phenotypic profile, genetic reprogramming and clinical implications.

Phenotypic plasticity of malignant melanoma is a well-known phenomenon. Several translational studies and small case series have reported this clinical and biological entity, particularly in metastatic melanoma, showing frequent aberrant expression of non-melanocytic differentiation markers of different lineages, posing remarkable challenges due to several alternative differential diagnoses including undifferentiated carcinoma and sarcomas. When melanoma loses its typical morpho-phenotype by routinely used diagnostic immunohistochemical markers, it is defined as "dedifferentiated melanoma". Hi

Generation of induced pluripotent stem cell line (XDCMHi001-A) from an Ankylosing spondylitis patient with JAK2 mutation.

Heredity is the major factor contributing to the susceptibility to ankylosing spondylitis(AS). Janus kinase 2 (JAK2) has been associated with AS. Urine-derived cells from an AS patient with JAK2 mutation were used to generate induced pluripotent stem cells (iPSCs) with five episomal iPSC reprogramming vectors (pCXLE-hOCT3/4-shp53-F, pCXLE-hSK, pCXLE-hUL, pCXLE-EGFP and pCXWB-EBNA1). The iPSCs were pluripotent and will be valuable for research on the role and mechanism of JAK2 in the pathogenesis of AS.

NAD+ inhibits the metabolic reprogramming of RPE cells in early AMD by upregulating mitophagy.

Age-related macular degeneration (AMD) is a leading cause of blindness and is becoming a global crisis, with the number of affected people expected to reach 288 million by 2040 worldwide. Retinal pigment epithelium (RPE) performs a number of highly diverse functions that are essential to maintaining the normal health and function of the retina. Alterations to retinal metabolism and remodeling are an early feature of AMD. The pathology of AMD is associated with mitochondrial dysfunction. Mitophagy is vital to promote a metabolic shift towards glycolysis that is required for cell differentiation

A multi-omics analysis reveals metabolic reprogramming in THP-1 cells upon treatment with the contact allergen DNCB.

Dendritic cell (DC) activation by contact allergens is one of the key steps in the development of allergic contact dermatitis (ACD). Recent evidence suggests that metabolic reprogramming is a prerequisite for the activation of DCs, macrophages and monocytes. Therefore, we used an integrated approach by combining proteomics and metabolomics to investigate the metabolism of human THP-1 cells in response to the strong contact allergen, 2,4-dinitrochlorobenzene (DNCB). Cells were treated with 5, 10 and 20μM DNCB for 4, 8, and 24h, respectively. Using a targeted metabolomics approach, we quantified

Subretinal fibrosis in neovascular age-related macular degeneration: From clinical features to fibrotic mechanisms and therapeutic opportunities.

Neovascular age-related macular degeneration (nAMD) is a leading cause of irreversible central vision loss worldwide. Although intravitreal anti-vascular endothelial growth factor therapy has substantially improved short-term outcomes, long-term vision is frequently limited by the development of subretinal fibrosis. This progressive scarring process occurs in a substantial proportion of patients and represents a major determinant of permanent visual impairment during long-term follow-up. Subretinal fibrosis arises from a complex interaction among structural injury, inflammatory signaling, and
