Achieving the Reverse Intersystem Crossing in Chalcone Based Donor-Acceptor System through Down-Conversion of Triplet Exciton.

In recent years, understanding the mechanism of thermally activated delayed fluorescence (TADF) has become the primary choice for designing high-efficiency, low-cost, metal-free organic light emitting diodes (OLEDs). Herein, we propose a strategically designed chalcone based donor-acceptor system, where intensification of delayed fluorescence with decrease in temperature (300 K to 100 K) is observed; the theoretical investigations of electronic states and orbital characters uncovered a new cold rISC pathway in donor-acceptor system, where rISC occurs through the down-conversation of higher tri

Beneficial biological effects of Flavokawain A, a chalcone constituent from kava, on surgically induced endometriosis rat model.

ETHNOPHARMACOLOGICAL RELEVANCE: Shrub kava has long been grown and utilized, primarily in the South Pacific region, for ceremonial, religious, and social occasions. It has been used as a pain reliever and muscle relaxant in medicinal practices from the eighteenth century. Interestingly, relatively low incidence of lung cancer may attribute to the high consumption of kava products in this region. AIM OF THE STUDY: Kava extracts were used to produce the kava chalcones Flavokawain A, B and C, which have a variety of bioactivities. In the present study, we show that Flavokawain A has positive eff

Optimization and antifungal activity of quinoline derivatives linked to chalcone moiety combined with FLC against Candida albicans.

In present work, a series of quinoline derivatives linked to chalcone moiety have been prepared, and their in vitro and in vivo antifungal activities against C. albicans have been evaluated. The results indicated that quinoline combined with fluconazole (FLC) showed good inhibitory activity against C. albicans. Especially, compound PK-10 combined with FLC displayed the best antifungal activity against 14 FLC-resistant C. albicans strains with almost no cytotoxicity. Preliminary mechanistic studies proved that PK-10 combined with FLC could inhibit the hyphae formation of C. albicans, induce the

2',4',6'-Tris(methoxymethoxy) chalcone induces apoptosis by enhancing Fas-ligand in activated hepatic stellate cells.

Suppression of hepatic stellate cell (HSC) activation and proliferation, and induction of apoptosis in activated HSCs have been proposed as therapeutic strategies for the treatment and prevention of the hepatic fibrosis. We previously showed that 2',4',6'-tris(methoxymethoxy) chalcone (TMMC), a synthesized chalcone derivative, inhibits platelet-derived growth factor-induced HSC proliferation at 5-20 μM. Here, we showed that TMMC induces apoptosis in activated HSCs at higher concentrations (30-50 μM), but is not cytotoxic to primary hepatocytes. Moreover, TMMC induces hyperacetylation of histon

Design, synthesis, and biological evaluation of novel ebselen-chalcone hybrids as multifunctional neuroprotective agents for ischemic stroke.

Ischemic stroke is a multifactorial neurological disorder characterized by oxidative stress, neuroinflammation, and mitochondrial dysfunction. Currently, effective neuroprotective agents for treating ischemic stroke remain limited. To integrate the glutathione peroxidase (GPx)-mimicking activity of ebselen with the antioxidant and anti-inflammatory properties of chalcones, a total of 67 ebselen-chalcone hybrids were rationally designed and synthesized. Biological evaluation identified B21 as an initial lead compound with improved neuroprotective activity and reduced cytotoxicity compared to eb

Discovery of novel indole-chalcone derivatives as potent hexokinase 2 inhibitors for triple negative breast cancer therapy.

Accelerated glucose metabolism is one of the important characteristics of cancer cells. Hexokinase 2 (HK2) is the rate-limiting enzyme that catalyzes the first step reaction of the glycolytic pathway and it is always found overexpressed in various tumor cells. In tumor cells, the overexpression of HK2 not only promotes aerobic glycolysis to provid energy and metabolite support for the rapid proliferation of tumor cells, but also inhibits apoptosis by binding to the outer mitochondrial membrane protein VDAC. It is recognized as important target of tumor metabolic reprogramming and it owns signi

A novel chalcone analog inhibits cancer stemness in CD44+ and CD133+ populations via suppression of ERK signaling.

Cancer stem-like cells (CSC) that express cluster of differentiation (CD)44 or CD133 contribute to tumor aggressiveness, therapeutic resistance, and disease recurrence across multiple malignancies. Consequently, current therapeutic modalities frequently fail to achieve durable disease control; hence, novel therapeutic strategies are urgently required. Therapeutic strategies that disrupt CSC maintenance may therefore contribute to more durable cancer control. Suitably, chalcone-based analogs have emerged as promising anticancer candidates. Therefore, this study aimed to investigate the effects

Nutrient deprivation enhances the cytotoxic effects of a sulfonamide-chalcone hybrid in PC-3 prostate cancer cells.

Hybrid molecules integrating different pharmacophoric motifs have emerged as a promising strategy in anticancer drug discovery. In this study, we report for the first time the biological effects of a sulfonamide-chalcone hybrid in the PC-3 prostate cancer cell line under standard culture conditions and nutrient deprivation. The molecular design of SSC185 (µ-(2,5-dichloro-N-{4-[(3E)-4-(3-nitrophenyl)buta-1,3-dien-2-yl]phenyl}benzenesulfonamide) incorporates an α,β-unsaturated system conjugated to the chalcone scaffold together with a sulfonamide moiety, structural features frequently associated

Prenylated chalcone analog-mediated Inhibition of Toxoplasma gondii growth in human trophoblast cell line and villous explants.

Congenital toxoplasmosis is a significant public health issue caused by the transplacental passage of Toxoplasma gondii to the embryo/fetus. The standard treatment involves a combination of sulfadiazine and pyrimethamine, drugs often associated with adverse effects and high toxicity. The current study aimed to investigate the potential of prenylated chalcones (C2, C4 and C9) in controlling T. gondii infection in human trophoblast cells (BeWo) and human placental explants. As results, non-cytotoxic doses of C2, C4 and C9 impaired parasite invasion and subsequent intracellular proliferation in B

Polyvinyl alcohol-based electrospun matrix as a delivery system for nanoemulsion containing chalcone against Leishmania (Leishmania) amazonensis.

Cutaneous leishmaniasis is a worldwide public health problem. Conventional therapies, in addition to the high cost, have many adverse effects and cases of parasite's resistance. Chalcones are secondary metabolites precursors in the flavonoid pathway and can be obtained naturally, but with low yield from plant raw material. Thus, the use of synthetic chalcones has been a promising strategy for the development of molecules with leishmanicidal activity. Thus, this work aimed to develop a controlled release system of two synthetic chalcone (trans-chalcones and 3'-(trifluormethyl)-chalcone) using p
