Effects of carotid chemoreceptor excitation on medullary expiratory neurons in cats.

A previous report (Lipski et al., 1977, J. Physiol. (London) 269, 797-810) demonstrated an inhibition of the medullary dorsal inspiratory neurons when a phasic chemoreceptor stimulus was applied during expiration. The present study tested the response of expiratory neurons which might mediate this inhibition. Recordings were made in cats anaesthetized with chloralose-urethane from the C5 phrenic rootlet and mainly from the rostral (Bötzinger) and caudal (nucleus retroambigualis, NRA) groups of medullary expiratory neurons. Carotid chemoreceptors were excited by close arterial injections of CO2

Histaminergic mechanisms involved in the centrally mediated effects of ouabain.

The role of central histaminergic mechanisms in cardiac glycoside toxicity was evaluated in chloralose-urethane anesthetized cats. Cardiac rhythm, blood pressure and sympathetic nerve activity were monitored during continuous i.v. infusion of ouabain (1 microgram/kg/min). Histamine administered into the fourth cerebral ventricle decreased significantly the dose of ouabain necessary to produce ventricular arrhythmias and fibrillation. Toxicity potentiation was associated with enhancement of sympathetic efferent activity. Dimaprit, a specific H2 receptor agonist, produced a similar, but slightly

Effect of delta 9-tetrahydrocannabinol on hypothalamic thermosensitive units.

Thermosensitive anterior hypothalamic neurons (pre-optic region) were studied in urethane and chloralose anesthetized cats in an attempt to characterize the hypothermic action of delta 9-THC at the neuronal level. One hundred and seventy-eight single neurons were isolated and subjected to thermal challenge, 66 were found to reproducibly alter firing frequency at a significant level (thermosensitivity (T.S.) greater than 0.75). Twenty-one of these units met the criteria for primary thermodetectors, 34 were heat-sensitive interneurons, and 11 were cold-sensitive interneurons. Administration of d

Responses of abdominal vascular capacitance to stimulation of splachnic nerves.

In chloralose-anesthetized dogs the abdominal circulation was vascularly isolated without opening the abdominal cavity. The region was perfused at constant flow through the aorta and drained at constant pressure from the inferior vena cava. Changes in resistance were calculated from changes in perfusion pressure and changes in capacitance were calculated by integrating changes in venous outflow. Stimulation of both splanchnic nerves at 20 Hz increased resistance by 135% and reduced capacitance by 7.20 ml kg-1. The capacitance responses at 1 and 2 Hz (3.42 and 5.43 ml kg-1) were 48 and 67% of t

Involvement of hindbrain and peripheral prostanoids in gastric motor and cardiovascular responses to delta-9-tetrahydrocannabinol in the rat.

We previously reported that delta-9-tetrahydrocannabinol (delta-9-THC), the primary psychoactive constituent of Cannabis sativa, inhibited gastric motor activity and evoked bradycardia and hypotension upon its parenteral administration in the rat. As prostanoids are important mediators of the actions of cannabinoids, we hypothesized that the inhibitory gastric motor and cardiovascular effects of delta-9-THC could depend on cyclooxygenase (COX) activation in the hindbrain and/or in the periphery. To test this hypothesis, vehicle or delta-9-THC (0.2 mg/kg, i.v.) were administered before and 15-m

Sympathetic rhythms during hyperventilation-induced apnea.

The effect of hyperventilation-induced apnea on the respiratory rhythmicity of sympathetic nerve activity was determined using spectral analysis of sympathetic nerve frequencies. Left phrenic, external intercostal, and inferior cardiac sympathetic nerves were recorded in alpha-chloralose-anesthetized, vagotomized, paralyzed, artificially ventilated cats. The respiratory modulation of sympathetic activity during normoventilation was indicated by spectral peaks of sympathetic activity coinciding with respiratory frequencies determined from the phrenic nerve activity of each cat. The spectral pea

Left ventricular inotropic responses to stimulation of carotid body chemoreceptors in anaesthetized dogs.

1. Dogs were anaesthetized with chloralose and artificially ventilated. The regions of both carotid bifurcations were vascularly isolated and perfused at constant pressure with arterial blood or with venous or hypoxic blood. 2. Inotropic responses were assessed by measuring the maximum rate of change of left ventricular pressure (dP/dtmax) with aortic pressure and heart rate held constant. 3. Stimulation of the chemoreceptors with venous blood from the inferior vena cava resulted in a decrease in dP/dtmax from 510 +/- 27 kPa.s-1 to 418 +/- 21 kPa.s-1 (mean +/- S.E.): a change of 17.9 +/- 1.0%

Comparative haemodynamic effects of tubocurarine and metocurine in the dog.

On accout of its histamine releasing and ganglion blocking properties tubocurarine is known to have significant haemodynamic effects. Methylation of the compound produces metocurine and should decrease both histamine release and ganglionic blockade. The haemodynamic effects of these two compounds were compared in 10 mongrel dogs anaesthetized with chloralose and morphine. Haemodynamic measurements were made 2 min before and 2, 5, 10 and 20 min after administration of the drugs. Each animal received three doses of each drug with a 2-h rest period between doses: tubocurarine 0.35 (muscle twitch

On the mechanism of vestibular disturbances caused by industrial solvents.

The industrial solvents xylene, styrene, trichloroethylene and methylchloroform administered to rabbits caused a positional nystagmus and disturbances in the nystagmus response to rotatory acceleration. The positional nystagmus had a beat direction the opposite to positional alcohol nystagmus, which was in similar experiments elicited by methanol, ethanol and propanol. The three alcohols needed a ten times higher blood concentration to cause a nystagmus than the solvents did.

Ex vivo analysis of lactate and glucose metabolism in the rat brain under different states of depressed activity.

Brain metabolism of glucose and lactate was analyzed by ex vivo NMR spectroscopy in rats presenting different cerebral activities induced after the administration of pentobarbital, alpha-chloralose, or morphine. The animals were infused with a solution of either [1-(13)C]glucose plus lactate or glucose plus [3-(13)C]lactate for 20 min. Brain metabolite contents and enrichments were determined from analyses of brain tissue perchloric acid extracts according to their post-mortem evolution kinetics. When amino acid enrichments were compared, both the brain metabolic activity and the contribution

Properties of postganglionic sympathetic neurons with axons in the right thoracic vagus.

The resting and reflex-evoked activities of single postganglionic sympathetic neurons with axons in the right thoracic vagus were tested in chloralose-anaesthetized cats. The properties of a majority of neurons were found to be similar. Cardiac- and inspiration-related rhythmicities were present in the resting activity of sympathetic neurons. Their resting activity was not affected by hyperventilation which abolished phrenic nerve discharges. Systemic hypoxia (2 min; 8% O2 in N2) increased the activity of the neurons more effectively in the deafferented state than when both sinus nerves remain

The acoustic middle ear muscle reflex in albino rats.

The acoustic middle ear muscle reflex was studied in albino rats anesthetized with chloralose. The best frequency of the reflex and the threshold at this frequency were on average about 3 kHz and 57 dB SPL, respectively. The threshold increased as frequency increased above, and decreased below, the best frequency at a rate of about 20 dB/octave. Above about 12 kHz, the muscular response showed instability and habituation. Thresholds were similar between stapedius and tensor tympani reflexes and between ipsilateral and contralateral reflexes. The middle ear transmission loss due to the reflex w

[The effect of various ventilation conditions on evoked potentials of the visual cortex of the cat in distention of the stomach].

In 10 cats anaesthetized using chloralose and with muscular relaxation by gallamine and continuous artificial ventilation in acute experiments photically evoked potentials (PEP) were recorded from the visual area of the cortex under different intragastric pressures and varying tidal volumes. It was the aim of this study to examine whether an exteroceptive stimulus (light flash) in the visual analyzer, which is influenced by an interoceptive stimulation (gastric pressure), may also be modified by different tidal volumes. Protective effect of lidocaine in acute cerebral ischemia induced by air embolism.

To investigate possible approaches to the prevention and treatment of neural damage induced by air embolism and other forms of acute cerebral ischemia, a model was used in which cerebral air embolism was produced by infusion of air (0.4 ml) into a vertebral artery of chloralose-anesthetized cats. Neurological function was assessed by measuring cortical somatosensory evoked responses in a group of 10 untreated animals and in a group of eight animals pretreated with intravenous lidocaine (5 mg/kg). In the untreated group, the primary somatosensory amplitude was reduced to 28% +/- 9% (mean +/- st

Stimulation sites in periaqueductal gray, nucleus raphe magnus and adjacent regions effective in suppressing oral-facial reflexes.

Electrode penetrations were made in the mesencephalon and caudal brainstem at the levels of the periaqueductal gray matter (PAG) and nucleus raphe magnus (NRM) in chloralose-anaesthetized or decerebrate cats. In a systematic fashion, mesencephalic and brainstem loci at every 1 mm of vertical depth were electrically stimulated in a series of mediolateral or anteroposterior electrode penetrations to determine the lowest stimulation threshold at each locus for suppressing the digastric jaw-opening reflex evoked by tooth pulp or infraorbital nerve stimulation; at some loci, the threshold current r
