Circadian rhythms orchestrate a wide array of behavioral and physiological functions, coordinating cellular and organismal processes on an approximately 24-h cycle through an intrinsic timekeeping system. Among the many processes subject to this temporal regulation, mitochondrial function has emerged as a critical and dynamic target of circadian control. Mitochondria, far from being static organelles, undergo continuous morphological remodeling through cycles of fusion and fission, collectively termed mitochondrial dynamics, that are essential for maintaining metabolic homeostasis, energy prod

The occurrence of headache and its change after treatment with melatonin 5 mg were studied in 30 patients with delayed sleep phase syndrome. The medication was taken 5 hours before the endogenous nocturnal plasma melatonin concentration had reached 10 pg/mL. Three women (aged 14, 14, and 23 years) suffered from chronic tension-type headache. Their headache disappeared within 2 weeks after the start of treatment with melatonin. One 54-year-old man suffered from disabling migraine attacks without aura, twice a week. After starting melatonin treatment, only three migraine attacks were reported in

An altered autonomic balance is considered to be a pathogenetic factor in cluster headache syndrome, although there is varying data on sympathetic and/or parasympathetic activation during attacks and/or attack-free intervals. The aim of the present study was to evaluate the day/night pattern of heart rate during the active and remission phases of cluster headache. In addition, the relationship between heart rate changes and the site of pain was investigated to determine if an autonomic imbalance was related to the lateralization of pain. Thirty-nine patients (34 with primary episodic cluster h

INTRODUCTION: The sleep-wake cycle and circadian rhythm disturbances are common in Alzheimer's disease (AD). However, it is not known if exercise has any benefit for the sleep disorders in AD.
METHODS: We conducted a 2-month voluntary wheel running (VWR) exercise (Ex) in an animal model of AD (APPSWE/PS1dE9 mice). We assessed behavioral circadian rhythm, sleep structure, circadian clock genes, cognitive function, and neurodegeneration in the suprachiasmatic nucleus (SCN), the hippocampus, and the cortex.
RESULTS: After VWR exercise in the AD mice, the rapid eye movement sleep was increased b

Sleep and circadian rhythms play critical roles in pain regulation, yet the influence of chronotype remains underexplored. This systematic review and meta-analysis synthesized evidence across 43 studies (cross-sectional, cohort, and Mendelian randomization designs) investigating associations between chronotype and pain outcomes. Participants ranged from small clinical cohorts to large-scale biobanks and represented diverse pain conditions, including fibromyalgia, temporomandibular disorders, migraine, musculoskeletal pain, and rheumatoid arthritis. Qualitative synthesis indicated that evening

Many physiological functions in humans and other mammals exhibit circadian rhythms with an approximate 24-hour period, enabling efficient adaptation to periodic environmental changes driven by the Earth's rotation. Clock genes generate circadian rhythms at the cellular level through transcription-translation feedback loops and contribute to the maintenance of physiological homeostasis. Accordingly, dysfunction of clock genes or chronic disruption of circadian rhythms has been implicated in increased disease risk and exacerbation of pathological conditions, and accumulating evidence indicates t

The global obesity epidemic has increased the burden of metabolic diseases. Dietary interventions are key to managing obesity and improving metabolic health. Intermittent fasting (IF) has emerged as a promising chrononutritional intervention that may modulate metabolic metabolic homeostasis through timed eating-fasting cycles. This review examines four main IF patterns:time-restricted feeding (TRF), alternate day fasting (ADF), the 5:2 fasting, and the 4:3 fasting, and evaluates their effects on body weight, glucose homeostasis, insulin sensitivity, and lipid profiles. Furthermore, it delineat

BACKGROUND: A system of photosensitive retinal ganglion cells provides 'non-visual' information on the circadian sequences of light to the suprachiasmatic nucleus (SCN), which, as the 'master clock', synchronizes the chronobiological mechanisms of all the biological clocks. Damage to SCN structure alters circadian behavioral and hormonal rhythms and interferes with a regular sleep-wake pattern. Several studies have shown that, in aging and in Alzheimer's disease (AD), circadian rhythms change their synchronization with the environment and behavior loses sync with light.
OBJECTIVE: The current

Previous studies indicate that the gaseous messenger carbon monoxide (CO) is released from the eye into the ophthalmic venous blood depending on the intensity of sunlight. This study was designed to determine whether the increased concentration of CO in ophthalmic venous blood affects the synthesis of melatonin and therefore, whether CO released from the eye under normal lighting conditions can be a carrier of light intensity information. Thirty six mature male wild boar and pig crossbreeds (n = 36) were studied. We measured the difference in the scotophase melatonin pathway response in terms

Ion channels are the gatekeepers to neuronal excitability. Retinal neurons of vertebrates and invertebrates, neurons of the suprachiasmatic nucleus (SCN) of vertebrates, and pinealocytes of non-mammalian vertebrates display daily rhythms in their activities. The interlocking transcription-translation feedback loops with specific post-translational modulations within individual cells form the molecular clock, the basic mechanism that maintains the autonomic approximately 24-h rhythm. The molecular clock regulates downstream output signaling pathways that further modulate activities of various i

Osler disease is an autosomal dominant disorder of the fibrovascular tissue characterized by arteriovenous malformations with multi-systemic haemorrhages. Recurrent epistaxis is the predominant symptom in more than 90% of patients. Recent studies showed circadian and seasonal patterns in the onset of nosebleeds, similar to acute cardiovascular events, such as myocardial infarction and stroke. The aim of this study was to determine whether such patterns would also apply to the onset of epistaxis in patients with Osler disease. In all, 110 patients with Osler disease who were under treatment for

An increase in the circadian amplitude (A) of blood pressure (BP) had been reported to precede a rise in the circadian BP average (MESOR, M), as pre-hypertension in the stroke-prone Okamoto rat. In humans, children with a positive family history of high BP and/or related cardiovascular disease had, on average, a larger BP-A than children with a negative family history, and an elevated BP-A was associated with intermediate values of the left ventricular mass index (LVMI), whereas an elevation in BP-M was only observed for larger LVMI values. Against this background, with 24-hour ambulatory moni

BACKGROUND: Circadian rhythm (CR) disruption is a major risk factor for metabolic dysfunction in skeletal muscle and liver. Although endurance training (ETR) is known to improve metabolic health, it remains unclear whether exercise timing and training duration influences metabolic adaptations under CR disruption.
METHODS: Twenty-four male Sprague-Dawley rats were randomly assigned to four groups: regular sleep cycle sedentary (RSC), irregular sleep cycle sedentary (ISC), irregular sleep cycle with late-stage ETR (ISE), and irregular sleep cycle with prolonged ETR (IEE). CR disruption was indu

SUNA is a trigeminal autonomic cephalalgia (TAC) characterized by short unilateral attacks centered on the ophthalmic trigeminal distribution, and accompanied by at least one of a number of cranial autonomic symptoms that can include lacrimation, redness of the ipsilateral eye, nasal congestion, rhinorrhea, and eyelid edema. It exists in episodic and chronic form. We have described an atypical case of episodic SUNA with an exclusive seasonal pattern as previously reported in other trigeminal autonomic cephalalgia, commonly known as TACs.

Neurological disorders represent a disproportionate fraction of pediatric intensive care unit (PICU) admissions, accounting for approximately one quarter of all critically ill children. These conditions-including traumatic brain injury, hypoxic-ischemic encephalopathy, stroke, infectious and inflammatory disorders, epilepsy, and disorders of consciousness-are frequently accompanied by profound disruption of circadian rhythms. The circadian timekeeper is a fundamental biological system that coordinates physiology across scales, from molecular and cellular processes to systemic functions such as

BACKGROUND: Parkinson's disease (PD) is a chronic neurodegenerative disorder marked by motor symptoms and nonmotor complications, notably cognitive impairment, which severely impairs patients' quality of life. While circadian disruption (CD) correlates with cognitive decline in PD, it remains unclear whether CD is merely secondary to motor symptoms or directly contributes to cognitive dysfunction. The objective of this study was to investigate whether chronic CD exacerbates cognitive decline in PD.
BASIC METHODS: Male mice were subjected to the 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MP
