Enhanced screening via a pure DNA-encoded peptide library enabled by an Fmoc modification.

Peptide-based molecules have widespread therapeutic applications due to their potent binding affinity and relative metabolic safety. Peptide therapeutics are developed using library screening that samples a diverse chemical space within primary sequences and secondary structural conformations. DNA-encoded peptide libraries (PDELs) are ideal for the development of peptide-based therapeutics as novel building blocks, and diverse chemical modifications are easily incorporated. However, current PDEL construction is limited by precipitation-based purification, which constrains libraries to three or

In Situ Synthesis of Multifunctional 3D Polymeric Artificial Organelles in Living Cells.

Artificial organelles have been developed to restore dysfunction or to introduce new-to-nature functions to the host cells, although limitations, such as instability, still exist. Here, we develop a new strategy to fabricate structurally stable polymeric "artificial organelles" by in situ crosslinking of multibranched monomers intracellularly using bioorthogonal click chemistry. The fabricated artificial organelles bearing bioorthogonal reactive centers allow for functional molecule conjugation and cargo release, enabling effective cell modification and manipulation. This demonstration opens u

Radical 1-Fluorosulfonyl-2-alkynylation of Unactivated Alkenes.

Sulfonyl fluorides have found widespread use in chemical biology and drug discovery. The development of synthetic methods for the introduction of the sulfonyl fluoride moiety is therefore of importance. Herein, a transition-metal-free radical 1,2-difunctionalization of unactivated alkenes via FSO2 -radical addition with subsequent vicinal alkynylation to access β-alkynyl-fluorosulfonylalkanes is presented. Alkynyl sulfonyl fluorides are introduced as highly valuable bifunctional radical trapping reagents that also serve as FSO2 -radical precursors. The β-alkynyl-fluorosulfonylalkanes obtained

Harnessing a single molecule for dual bioorthogonal regulation of RNA function and m6A methylation.

This study presents a multifunctional RNA regulation strategy that enables RNA molecules to undergo both bioorthogonal ligation and cleavage reactions within the same system. Using guide RNA (gRNA) as an example, we demonstrate on-demand inactivation via click chemistry and subsequent reactivation by light exposure. Applied to CRISPR-mediated, site-specific RNA methylation, this technology enables continuous, multistep programmable control, overcoming the one-way limitation of traditional methods. Results show that gRNA can stably tolerate both azide and photodegradable groups, facilitating ef

In vivo stem cell tracking with imageable nanoparticles that bind bioorthogonal chemical receptors on the stem cell surface.

It is urgently necessary to develop reliable non-invasive stem cell imaging technology for tracking the in vivo fate of transplanted stem cells in living subjects. Herein, we developed a simple and well controlled stem cell imaging method through a combination of metabolic glycoengineering and bioorthogonal copper-free click chemistry. Firstly, the exogenous chemical receptors containing azide (-N3) groups were generated on the surfaces of stem cells through metabolic glycoengineering using metabolic precursor, tetra-acetylated N-azidoacetyl-d-mannosamine(Ac4ManNAz). Next, bicyclo[6.1.0]nonyne

Convergent Synthesis of a Bifunctional GPI as a Universal Platform to Access Diverse GPI and GPI-Anchored Protein Probes.

A bifunctional glycosylphosphatidylinositol (GPI) anchor containing an azide and a biotin to facilitate additional functionalization via click chemistry and avidin conjugation, respectively, was synthesized by a convergent strategy using an orthogonal protection tactic. It is a general platform to rapidly access diverse GPI and GPI-anchored protein (GPI-AP) probes. Spin-labeled GPI and GPI-AP derived from this platform were used in a related study to show that GPI governs the membrane orientation of GPI-AP, validating GPI as a unique protein anchor.

Clickable tryptophan modification for late-stage diversification of native peptides.

As the least abundant residue in proteins, tryptophan widely exists in peptide drugs and bioactive natural products and contributes to drug-target interactions in multiple ways. We report here a clickable tryptophan modification for late-stage diversification of native peptides, via catalyst-free C2-sulfenylation with 8-quinoline thiosulfonate reagents in trifluoroacetic acid (TFA). A wide range of groups including trifluoromethylthio (SCF3), difluoromethylthio (SCF2H), (ethoxycarbonyl)difluoromethylthio (SCF2CO2Et), alkylthio, and arylthio were readily incorporated. The rapid reaction kinetic

Preparation of Polyphenylene Ring Derivative Dyes with Wide Wave Absorption Properties and Their Performance Study.

Some conjugated benzene ring molecules were prepared using the Sonogashira reaction, and the molecules were post-functionally modified using click chemistry. The optical and electrical band gaps were measured using UV-VIS absorption spectroscopy and the three-electrode method, and the results of both were verified against each other to prove the accuracy of the characterization. In addition, the optical performances of the material were studied by z-scan; almost all materials exhibited good nonlinear optical properties and interconversion between saturable and anti-saturable absorption due to

Engineered endothelium-mimicking antithrombotic surfaces via combination of nitric oxide-generation with fibrinolysis strategies.

Thrombosis associated with implants can severely impact therapeutic outcomes and lead to increased morbidity and mortality. Thus, developing blood-contacting materials with superior anticoagulant properties is essential to prevent and mitigate device-related thrombosis. Herein, we propose a novel single-molecule multi-functional strategy for creating blood-compatible surfaces. The synthesized azide-modified Cu-DOTA-(Lys)3 molecule, which possesses both NO release and fibrinolysis functions, was immobilized on material surfaces via click chemistry. Due to the specificity, rapidity, and complete

A versatile cholera toxin conjugate for neuronal targeting and tracing.

Tracing of neurons plays an essential role in elucidating neural networks in the brain and spinal cord. Cholera toxin B subunit (CTB) is already widely used as a tracer although its use is limited by the need for immunohistochemical detection. A new construct incorporating non-canonical azido amino acids (azido-CTB) offers a novel way to expand the range and flexibility of this neuronal tracer. Azido-CTB can be detected rapidly in vivo following intramuscular tongue injection by 'click' chemistry, eliminating the need for antibodies. Bioorthogonal chemistry conjugates: ADCs and beyond.

Bioorthogonal click chemistry has played a transformative role in developing advanced hybrid conjugates. These molecular constructs are formed by chemically linking a targeting component to a functional moiety through a suitable linker. Therapeutic conjugates are designed to deliver a biologically active agent to a specific target to treat, prevent, or modify a disease process. By directing the payload to a defined site of action, these conjugates can improve efficacy while reducing off-target effects and systemic toxicity. Among these, antibody-drug conjugates stand out as the most clinically

Click-code-seq reveals strand biases of DNA oxidation and depurination in human genome.

DNA modifications drive aging, neurodegeneration, carcinogenesis and chemotherapy drug action. Accurate mapping of diverse DNA modifications with single-nucleotide precision in complex genomes remains challenging. We upgraded click-code-seq, a click-chemistry-aided DNA-modification mapping strategy, to enable its first application for sequencing oxidation and depurination in the human genome. We developed a companion fluorescence assay, click-fluoro-quant, to rapidly quantify common DNA modifications and novel adaptors to minimize false positives and assess modification frequency. We uncovered

Cysteine-Directed Bioconjugation of a Platinum(II)-Acridine Anticancer Agent.

Classical maleimide Michael addition chemistry in conjunction with copper-free click chemistry was investigated as a synthetic strategy to attach cytotoxic platinum-acridine hybrid agents to carrier proteins. The structural integrity and selectivity of the model payloads, which were validated in human serum albumin (HSA) using mass spectrometric analysis and heteronuclear 2D 1H-15N HSQC NMR experiments, may have broad utility for the targeted delivery of highly cytotoxic platinum acridines and other nonclassical platinum containing anticancer agents.

Fast and Bioorthogonal Release of Isocyanates in Living Cells from Iminosydnones and Cycloalkynes.

Bioorthogonal click-and-release reactions are powerful tools for chemical biology, allowing, for example, the selective release of drugs in biological media, including inside animals. Here, we developed two new families of iminosydnone mesoionic reactants that allow a bioorthogonal release of electrophilic species under physiological conditions. Their synthesis and reactivities as dipoles in cycloaddition reactions with strained alkynes have been studied in detail. Whereas the impact of the pH on the reaction kinetics was demonstrated experimentally, theoretical calculations suggest that the n

Click-Activated Supramolecular Targeting Chimeras for Spatiotemporally Resolved Protein Degradation In Vivo.

Targeted protein degradation (TPD) holds enormous therapeutic potential, yet spatiotemporally controlled, tissue-selective protein degradation remains challenging. Herein, we report Click-activated Supramolecular TArgeting Chimeras (ClSupTAC), a multimodular platform leveraging bioorthogonal chemistry to realize spatially and temporally resolved TPD in vivo. ClSupTAC follows a two-step "click-to-activate" paradigm: self-assembled, functionally dormant tissue-targeting supramolecular nanoparticles presenting protein-binding ligands and trans-cyclooctene (TCO) moieties are locally activated upon

Safirinium Fluorescent "Click" Molecular Probes: Synthesis, CuAAC Reactions, and Microscopic Imaging.

Fluorescent labeling utilizing Cu(I)-catalyzed azide-alkyne cycloaddition reactions (CuAAC) is among the leading applications of the "click" chemistry strategy. Fluorescent probes for this approach can be constructed by linking an azide or alkyne group to a fluorophore, such as the recently developed Safirinium derivatives. These compounds are water-soluble, highly fluorescent heterocycles based on 1,2,4-triazolium, with significant potential for various labeling applications, although they have not yet been converted to azide or alkyne probes. Herein, we report the synthesis of Safirinium-bas

High Yield Branched Puromycin Linker Design Enables Efficient cDNA Display and Chemical Modification of Peptides.

Peptide chemical modification is a valuable technique for improving peptide stability and bioactivity, particularly in drug discovery applications. Here, we report the development of a novel linker junction strategy using strain-promoted azide-alkyne cycloaddition (SPAAC) that enables the efficient formation of branched puromycin linkers with an average yield of 97%. This approach represents an improvement over traditional Michael-Addition methods, which typically yield ~15%-20%. The high yield of the SPAAC reaction and the near absence of by-products make the linkage by click reaction easy to

Tagging lipoproteins in mycobacteria via metabolic incorporation of modified fatty acids.

Lipoproteins are lipid-modified proteins and critical components of the mycobacterial cell envelope with diverse functions related to virulence and survival. In Mycobacterium tuberculosis (Mtb), the causative agent of tuberculosis, lipoproteins account for ∼3 % of the Mtb proteome, yet their precise functions remain poorly characterized. Prior experimental efforts have addressed the validation and preliminary functional annotation of specific lipoproteins. Whereas these studies elucidated aspects of lipoprotein localization and involvement in host-pathogen interactions, the methodologies emplo
