Cannabis sativa is known for its therapeutic benefit in various diseases including pain relief by targeting cannabinoid receptors. The primary component of cannabis, Δ9-tetrahydrocannabinol (THC), and other agonists engage the orthosteric site of CB1, activating both Gi and β-arrestin signaling pathways. The activation of diverse pathways could result in on-target side effects and cannabis addiction, which may hinder therapeutic potential. A significant challenge in pharmacology is the design of a ligand that can modulate specific signaling of CB1. By leveraging insights from the structure-fun

Virus-like particles (VLPs) are highly suited platforms for protein-based vaccines. In the present work, we adapted a previously designed non-infectious adenovirus-inspired 60-mer dodecahedric VLP (ADDomer) to display a multimeric array of large antigens through a SpyTag/SpyCatcher system. To validate the platform as a potential COVID-19 vaccine approach, we decorated the newly designed VLP with the glycosylated receptor binding domain (RBD) of SARS-CoV-2. Cryoelectron microscopy structure revealed that up to 60 copies of this antigenic domain could be bound on a single ADDomer particle, with

The emergence of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) lineages that are more transmissible and resistant to currently approved antibody therapies poses a considerable challenge to the clinical treatment of coronavirus disease (COVID-19). Therefore, the need for ongoing discovery efforts to identify broadly reactive monoclonal antibodies to SARS-CoV-2 is of utmost importance. Here, we report a panel of SARS-CoV-2 antibodies isolated using the linking B cell receptor to antigen specificity through sequencing (LIBRA-seq) technology from an individual who recovered from COV

The dynamin-related GTPase OPA1 is mutated in autosomal dominant optic atrophy (DOA) (Kjer type), an inherited neuropathy of the retinal ganglion cells. OPA1 is essential for the fusion of the inner mitochondrial membranes, but its mechanism of action remains poorly understood. Here we show that OPA1 has a low basal rate of GTP hydrolysis that is dramatically enhanced by association with liposomes containing negative phospholipids such as cardiolipin. Lipid association triggers assembly of OPA1 into higher order oligomers. In addition, we find that OPA1 can promote the protrusion of lipid tubu

Intestinal fluids dictate the intraluminal environment, and therefore, they substantially affect the absorption of orally taken drugs. The characterization of human intestinal fluids (HIF) and the design of simulated intestinal fluids (SIF) mainly focus on composition, not necessarily taking into account the ultrastructure of HIF. Colloidal structures in HIF and SIF can enhance the solubilizing capacity for lipophilic drugs while decreasing the bioaccessible fraction. As such, colloids present in HIF play a crucial role and require an in-depth characterization. Therefore, the present study pur

The ancient pyruvate dehydrogenase complex (PDHc) performs the "link reaction" of cellular respiration-a discovery from the 1930s that was central in the award of the 1953 Nobel Prize in Physiology and Medicine to Krebs and Lipmann. Fast forward to 2024, PDHc emerges with roles in Alzheimer's, cancer, and neurodegeneration, as well as in obesity and aging processes. Due to these recent reports, structural analysis of PDHc, a 10-megadalton enzymatic complex, comes into focus-only now this analysis begins to unveil an enormous and challenging molecular complexity. Cutting-edge techniques and met

Revealing the 3D conformational variability of biomolecules is crucial for understanding their function, while cryo-EM reconstruction of rare states remains difficult due to data imbalance and structural detail loss in existing generative models. We present a dual-stage pipeline consisting of a generative stage and a validation stage. In the generative stage, we employ a 3D dual-frequency variational autoencoder (3DDF-VAE) that separately models low- and high-frequency components of protein density maps to enhance global coherence and local structural detail. In the validation stage, a pose-co

Bacterial microcompartments (BMCs) are pseudo-organelles that sequester metabolic enzymes, intermediates, and/or gases within the bacterial cytosol. One model BMC is the carboxysome (CB). CBs facilitate rubisco-driven fixation of CO2, increasing efficiency and maximizing the phosphoglycerate output in CB-containing bacteria. The α-CBs are of particular interest due to their small size and relative simplicity, making them ideal targets for bioengineering applications. These CBs were the first BMC observed and have been a long-studied model; however, they are challenging to study in native syste

Enzymes that reside in the membrane are responsible for a myriad of vital tasks such as assembly of and changes to the membrane itself, and modification of proteins that reside therein or are secreted through the various cellular compartments. Understanding how these enzymes work at a molecular level is most often dependent on high resolution structural information. While the rates of success for the determination of structures of integral membrane proteins in general, enzymes included, have improved considerably-most notably because of recent advances in single particle cryo-electron microsco

Microtubule nucleation is a crucial process in cell biology that describes the formation of a new microtubule from free tubulin. As one of the key steps in establishing organized microtubule networks, microtubule nucleation is fundamental to the roles of the microtubule cytoskeleton in mediating intracellular transport, mitotic spindle formation, and cellular morphology. At the core of this process is the γ-tubulin ring complex (γTuRC), a multi-protein complex that serves as a template for microtubule nucleation, thus ensuring spatial and temporal control of microtubule growth. Understanding t

BACKGROUND: Tumor necrosis factor-alpha (TNFα) is an important therapeutic target for treating a range of inflammatory and autoimmune disorders. The TNFα trimer functions by interacting with its receptors (TNFRs) on the immune cell surface and triggers downstream intracellular signaling. (-)-Epigallocatechin-3-gallate (EGCG) is the primary bioactive polyphenol compound in green tea. Our previous study found that EGCG can inhibit TNFα activity; however, the underlying molecular mechanism remains unclear.
PURPOSE: In this study, we investigated the interaction between EGCG and TNFα to elucidate

Transient receptor potential melastatin 1 (TRPM1) is a membrane protein essential for vision in dim light, and mutations in TRPM1 cause complete congenital stationary night blindness. Although TRPM1 shares sequence similarity to other TRPM ion channels such as TRPM3, whether it independently functions as an ion channel remains controversial. This controversy is largely caused by TRPM1's challenging biochemical behaviors that prevent detailed molecular characterization. In this work, we isolate TRPM1 and determine its structures using cryogenic electron microscopy (cryo-EM). The structures reve

Developing an effective malaria vaccine is challenging because Plasmodium parasites use complex strategies to invade host cells. The blood stage, where parasites replicate inside red blood cells, causes disease symptoms and relies on large multiprotein surface complexes that are difficult to target. Understanding their structure is essential for rational vaccine design. Cryogenic electron microscopy (cryoEM) enables atomic-level analysis of dynamic and membrane-associated complexes without crystallization, revealing key processes such as protein trafficking, nutrient uptake, and host cell remo

A diverse subset of cyanobacteria can transiently modify their photosynthetic machinery during far-red light photoacclimation to drive photosynthesis with less energetic photons (700 nm-800 nm). To achieve this, all the main light-driven components of the photosynthetic apparatus, including their allophycocyanin antenna, are replaced with red-shifted paralogues. Recent studies based on the structure of an incomplete complex provided some insights into the tuning of the far-red phycobiliproteins. Here, we solved the structure of the intact bicylindrical allophycocyanin complex from the cyanobac

Distinct tau amyloid assemblies underlie diverse tauopathies but defy rapid classification. Cell and animal experiments indicate tau functions as a prion, as different strains propagated in cells cause unique, transmissible neuropathology after inoculation. Strain amplification requires compatibility of the monomer and amyloid template. We used cryo-electron microscopy to study one cell-based yellow fluorescent protein (YFP)-tagged strain, resolving its amyloid nature. We then used sequential alanine (Ala) substitution (scan) within tau repeat domain (RD) to measure incorporation to preexistin

In response to various intracellular stress or damage-associated signals, inflammasomes can be activated and trigger a pyroptotic cell death process through the sequential assembly of structurally compatible and interacting filamentous oligomers involving the pyrin domains (PYD) of important inflammasome components. The PYD-containing interferon-inducible protein 16 (IFI16) has been suggested as a viral DNA sensor that can induce inflammasome formation, but it also has other inflammasome-independent functions, including interferon production. Here, the cryo-EM structure of the filament assembl

Distinguishing signal from background in cryogenic electron microscopy (cryo-EM) micrographs is a critical processing step but remains challenging owing to the inherently low signal-to-noise ratio (SNR), contaminants, variable ice thickness, and densely packed particles of heterogeneous sizes. Recent image-segmentation methods provide pixel-level precision and thus offer several advantages over traditional object-detection approaches: segmented-blob mass can be computed to suppress false-positive particles, particle centering can be improved by leveraging the full brightness profile, and irreg

Single-particle cryo-electron microscopy (cryo-EM) has become a very powerful technique in the field of membrane protein structural biology. Historically, protein structure determination requires homogenous and pure samples, and sample heterogeneity often hampered the progress of drug design and development, especially those targeting membrane proteins and their complexes. With the rapid development of the instrumentation, software and methodologies of cryo-EM, it is now feasible to obtain high-resolution cryo-EM structural information of membrane proteins, from both pure/homogeneous and impur

Primary malignant tumors of small bowel constitute only about 1-2% of all gastrointestinal neoplasms. Although neuroendocrine tumors (NETs) are relatively rare, they still represent the second most common malignancy of the small bowel (after adenocarcinoma). Clinical manifestations include abdominal pain, bowel obstruction, diarrhea, weight loss and bleeding. The differential diagnosis of obscure gastrointestinal bleeding can sometimes be challenging for endoscopy as well as for radiology methods. We present the case of an 80-year-old man suffering from severe hypochromic anemia. Routine endos

Matriglycan is a linear glycan (xylose-β1,3-glucuronate)n, which binds proteins in the extracellular matrix that contain laminin-globular domains and Lassa Fever Virus. It is indispensable for neuromuscular function. Matriglycan of insufficient length can cause muscular dystrophy with abnormal brain and eye development. LARGE1 (Like-acetylglucosaminyltransferase-1) uniquely synthesizes matriglycan on dystroglycan. The mechanism of matriglycan synthesis is not obvious from cryo-EM reconstructions of LARGE1. However, by reconstituting activity in vitro on recombinant prodystroglycan we show that
