A General Model to Explain Repeated Turnovers of Sex Determination in the Salicaceae.

Dioecy, the presence of separate sexes on distinct individuals, has evolved repeatedly in multiple plant lineages. However, the specific mechanisms by which sex systems evolve and their commonalities among plant species remain poorly understood. With both XY and ZW sex systems, the family Salicaceae provides a system to uncover the evolutionary forces driving sex chromosome turnovers. In this study, we performed a genome-wide association study to characterize sex determination in two Populus species, P. euphratica and P. alba. Our results reveal an XY system of sex determination on chromosome

Genome-wide association study reveals novel players in defense hormone crosstalk in Arabidopsis.

Jasmonic acid (JA) regulates plant defenses against necrotrophic pathogens and insect herbivores. Salicylic acid (SA) and abscisic acid (ABA) can antagonize JA-regulated defenses, thereby modulating pathogen or insect resistance. We performed a genome-wide association (GWA) study on natural genetic variation in Arabidopsis thaliana for the effect of SA and ABA on the JA pathway. We treated 349 Arabidopsis accessions with methyl JA (MeJA), or a combination of MeJA and either SA or ABA, after which expression of the JA-responsive marker gene PLANT DEFENSIN1.2 (PDF1.2) was quantified as a readout

Transcriptome and Metabolite Changes during Hydrogen Cyanamide-Induced Floral Bud Break in Sweet Cherry.

Release of bud dormancy in perennial woody plants is a temperature-dependent process and thus flowering in these species is heavily affected by climate change. The lack of cold winters in temperate growing regions often results in reduced flowering and low fruit yields. This is likely to decrease the availability of fruits and nuts of the Prunus spp. in the near future. In order to maintain high yields, it is crucial to gain detailed knowledge on the molecular mechanisms controlling the release of bud dormancy. Here, we studied these mechanisms using sweet cherry (Prunus avium L.), a crop wher

High-frequency shoot regeneration, assessment of genetic fidelity, and histochemical analysis of forskolin production in Coleus forskohlii Briq.

Forskolin, a diterpenoid found in the roots of Coleus forskohlii, has generated significant interest in the medical field due to its various therapeutic uses. This study aimed to establish an effective system for regenerating C. forskohlii plants, ensuring a year-round supply of plant material and forskolin production. We tested different concentrations of cytokinins, either alone or combined with auxin, to see their impact on shoot multiplication and growth. We found that a medium supplemented with 1.5 mg L-1 of meta-topolin (mT) resulted in the highest number of shoots (~ 12.66) and leaves (

Transcriptome-wide modulation by Sargassum vulgare and Acanthophora spicifera extracts results in a prime-triggered plant signalling cascade in tomato and sweet pepper.

Seaweed extracts (SWEs) are becoming integrated into crop production systems due to their multiple beneficial effects including growth promotion and induction of defence mechanisms. However, the comprehensive molecular mechanisms of these effects are yet to be elucidated. The current study investigated the transcriptomic changes induced by SWEs derived from Sargassum vulgare and Acanthophora spicifera on tomato and sweet pepper plants. Tomato and sweet pepper plants were subjected to foliar treatment with alkaline extracts prepared from the above seaweeds. Transcriptome changes in the plants w

Analysis of phytohormones in vermicompost using a novel combinative sample preparation strategy of ultrasound-assisted extraction and solid-phase extraction coupled with liquid chromatography-tandem mass spectrometry.

Vermicompost (VC), a widely used premium organic fertilizer, is the by-product of symbiotic interactions between earthworms and microorganisms living within them. It has been postulated that phytohormones are plausible "magic compounds" in VC that are responsible for making them such good fertilizers. Thus, a novel approach involving ultrasound-assisted extraction (UAE) and solid-phase extraction (SPE) was developed as a fast and efficient sample preparation method to screen for different classes of phytohormones in VC by liquid chromatography-tandem mass spectrometric (LC-MS/MS) analysis. Characterization of the ABC Transporter G Subfamily in Pomegranate and Function Analysis of PgrABCG14.

ATP-binding cassette subfamily G (ABCG) proteins play important roles in plant growth and development by transporting metabolites across cell membranes. To date, the genetic characteristics and potential functions of pomegranate ABCG proteins (PgrABCGs) have remained largely unknown. In this study, we found that 47 PgrABCGs were divided into five groups according to a phylogenetic analysis; groups I, II, III, and IV members are half-size proteins, and group V members are full-size proteins. PgrABCG14, PgrABCG21, and PgrABCG47 were highly expressed in the inner seed coat but had very low expres

Nitrate transiently rewires transcription in developing xylem of Populus wood to promote cell expansion via CRF4.

Nitrate functions both as a nutrient and a signaling molecule, but how nitrate is perceived and translated into developmental outputs within woody tissues is unknown. Here, we show that developing xylem in hybrid aspen (Populus tremula × P. tremuloides) mounts a rapid, cell-type-specific transcriptional response to nitrate that promotes xylem cell expansion. To isolate local nitrate signaling in wood-forming tissues, we delivered nitrate directly into the xylem sap and profiled transcriptomic responses from 2 to 48 h after treatment. Bulk RNA sequencing revealed a transient response peaking wi

Evolutionary origin of a new gene drives root growth in one desert poplar.

Populus euphratica develops an extensive root system that supports survival in arid environments. We identified PeuPLDζ4, a new gene arising from tandem duplication in P. euphratica, and evaluated its effects on root growth and drought-related performance. Phylogenetic and promoter-reporter analyses, enzyme assays, and subcellular localization characterized PeuPLDζ4 divergence. Its effects were evaluated in transgenic Arabidopsis thaliana, P. alba var. pyramidalis, wheat, and rice. Phosphatidic acid (PA) and hormone measurements, reporter imaging, transcriptomics, protein-lipid overlay, dual-l

Protective capacity of Rutin against oxidative damage induced by saline stress in the roots of the model organism Allium cepa.

Salt stress limits plant growth by impairing physiological and biochemical processes, especially in roots, which are the main absorption organs. This study aimed to evaluate the phytoprotective potential of rutin against NaCl-induced salt stress in Allium cepa, focusing on morphological, biochemical, and molecular responses. Seeds were treated with 512 µg/mL and 1024 µg/mL rutin, both with 50 mM NaCl, and compared to a control group (NaCl only). Germination rate, root development, oxidative stress markers (TBARS, non-protein thiols, protein thiols), and elemental analysis of iron accumulation

Unveiling exogenous potential of phytohormones as sustainable arsenals against plant pathogens: molecular signaling and crosstalk insights.

Plants frequently confront pathogens that disrupt physiological and molecular functions, ultimately reducing agricultural yields. To counter these challenges, plants activate sophisticated defense mechanisms to recognize stress signals while optimizing growth. Phytohormones signaling pathways and their crosstalk are central to regulating plant growth, development and defense. Numerous proteins associated with phytohormone signaling pathways have been identified, including receptors for several vital hormones. Previous studies indicate that defense phytohormones, like salicylic acid (SA), jasmo

Role of methylglyoxal and glyoxalase in the regulation of plant response to heavy metal stress.

Methylglyoxal and glyoxalase function a significant role in plant response to heavy metal stress. We update and discuss the most recent developments of methylglyoxal and glyoxalase in regulating plant response to heavy metal stress. Methylglyoxal (MG), a by-product of several metabolic processes, is created by both enzymatic and non-enzymatic mechanisms. It plays an important role in plant growth and development, signal transduction, and response to heavy metal stress (HMS). Changes in MG content and glyoxalase (GLY) activity under HMS imply that they may be potential biomarkers of plant stres

The alleviation of Cr(Ⅵ) stress on simultaneous nitrification and denitrification process of Acinetobacter haemolyticus RH19.

Bioremediation of Cr(Ⅵ) and ammonia is considered as a promising and cost-effective alternative to chemical and physical methods. However, Cr(Ⅵ) could inhibit nitrogen removal by inhibiting intra-/extracellular electron (IET/EET) transfer or nitrifying and denitrifying enzymes activity due to its higher solubility. In this study, we isolated a simultaneous nitrification and denitrification (SND) microorganism Acinetobacter haemolyticus RH19, capable of outcompeting oxygen to take nitrogen oxides/ammonia as electron acceptors, and studied a combined accelerant (cysteine, biotin and cytokinin) t

Comparative transcriptome analysis of flower bud transition and functional characterization of EjAGL17 involved in regulating floral initiation in loquat.

Floral initiation plays a critical role for reproductive success in plants, especially fruit trees. However, little information is known on the mechanism of the initiation in loquat (Eriobotrya japonica Lindl.). Here, we used transcriptomic, expression and functional analysis to investigate the candidate genes in floral initiation in loquat. Comparative transcriptome analysis showed differentially expressed genes (DEGs) were mainly enriched in the metabolic pathways of plant hormone signal transduction. The DEGs were mainly involved in the gibberellin, auxin, cytokinin, abscisic acid, salicyli

Early steps of adventitious rooting: morphology, hormonal profiling and carbohydrate turnover in carnation stem cuttings.

The rooting of stem cuttings is a common vegetative propagation practice in many ornamental species. A detailed analysis of the morphological changes occurring in the basal region of cultivated carnation cuttings during the early stages of adventitious rooting was carried out and the physiological modifications induced by exogenous auxin application were studied. To this end, the endogenous concentrations of five major classes of plant hormones [auxin, cytokinin (CK), abscisic acid, salicylic acid (SA) and jasmonic acid] and the ethylene precursor 1-aminocyclopropane-1-carboxylic acid were ana

Differential effects of yeast derivatives on pepper productivity, stress resilience, and soil microbial communities.

The excessive application of chemical fertilizers is a prevalent issue in modern agriculture, as it increases production costs, undermines economic returns, and degrades soil health. In this study, the effects of two compositionally distinct yeast derivatives, namely Yeast Glycoside Type I (YD1) and Water-Soluble Yeast Glycoside (YD2), on the growth, nutrient use efficiency, low-temperature (LT) stress tolerance, and rhizosphere microbial communities of pepper (Capsicum annuum L.) were investigated. The yeast derivatives (YD) were characterized using scanning electron microscopy (SEM) and bioc

Development of an efficient in vitro micropropagation and biochemical profiling of Salvia halophila, an endemic Turkish sage.

BACKGROUND: Salvia halophila Hedge is an endemic sage species confined to the salt steppe ecosystems of Central Anatolia (Türkiye), particularly around the Lake Tuz Basin, and is classified as Endangered (EN) by the IUCN. Its narrow distribution, low seed germination rates, and habitat pressures threaten its survival and restrict the sustainable exploitation of its valuable phytochemicals, such as rosmarinic acid and other phenolics. In vitro micropropagation represents a strategic approach for producing genetically uniform and phytochemically rich plant material, thereby supporting both ex si

Molecular regulation by H2S of antioxidant and glucose metabolism in cold-sensitive Capsicum.

BACKGROUND: Cold is an important environmental limiting factor affecting plant yield and quality. Capsicum (chili pepper), a tropical and subtropical vegetable crop, is extremely sensitive to cold. Although H2S is an important signaling regulator in the responses of plant growth and development to abiotic stress, few studies have examined its effects on cold-sensitive capsicum varieties. Through biotechnology methods to enhance the cold resistance of peppers, to provide some reference for pepper breeding, investigated molecular regulation by H2S of responses to cold stress in cold-sensitive ca

Harnessing chickpea bacterial endophytes for improved plant health and fitness.

Endophytic bacteria live asymptomatically inside the tissues of host plants without inflicting any damage. Endophytes can confer several beneficial traits to plants, which can contribute to their growth, development, and overall health. They have been found to stimulate plant growth by enhancing nutrient uptake and availability. They can produce plant growth-promoting substances such as auxins, cytokinins, and gibberellins, which regulate various aspects of plant growth and development. Endophytes can also improve root system architecture, leading to increased nutrient and water absorption. So
