CD4/CD8 Ratio could be predictor of burden hepatocellular carcinoma in Egyptian chronic hepatitis C after combined sofosbuvir and daclatasvir therapy.

BACKGROUND: During the first years of the use of direct acting Hepatitis C antiviral drugs (DAAS), several studies reported a possible correlation between this new era of treatment and an increased risk of Hepatocellular carcinoma (HCC). Its development could possibly be favored by the changes in the immunological milieu and the different cellular behavior after eradication of HCV infection with them. For this reason, this study aimed to address the immunological effect of DAAS. SUBJECT & METHODS: Prospective paired -sample design, carried out on 90 naïve chronically infected HCV patients bef

Altered peptide ligands trigger perforin- rather than Fas-dependent cell lysis.

CTLs lyse Fas-expressing target cells by the concomitant action of a perforin- and a Fas-dependent mechanism. This study analyzed whether target cells pulsed with T cell antagonists and other altered peptide ligands (APLs) were susceptible selectively to only one of these two mechanisms. In vivo and in vitro activated T cells from transgenic mice expressing a TCR specific for lymphocytic choriomeningitis virus were used as effector cells. To distinguish between perforin- and Fas-dependent cytotoxicity, T cells from normal or perforin-deficient mice were used to lyse peptide-pulsed Fas-positive

Search for biomarkers of asbestos exposure and asbestos-induced cancers in investigations of the immunological effects of asbestos.

The immunological effects of asbestos exposure on various lymphocytes such as the regulatory T cell (Treg), responder CD4+ T helper cell (Tresp), CD8+ cytotoxic T lymphocytes (CTL), and natural killer (NK) cells were investigated. Results show that asbestos exposure impairs antitumor immunity through enhancement of regulatory T cell function and volume, reduction of CXCR3 chemokine receptor in responder CD4+ T helper cells, and impairment of the killing activities of CD8+ cytotoxic T lymphocytes (CTL) and NK cells. These findings were used to explore biological markers associated with asbestos

Interleukin-15 correlates with cytotoxic immune networks in cervical tuberculous lymphadenitis.

INTRODUCTION: Cervical tuberculous lymphadenitis (CTL) represents a localized manifestation of Mycobacterium tuberculosis infection in which immune responses are organized within lymphoid tissue. While cytotoxic lymphocyte responses contribute to antimycobacterial immunity, the cytokine networks coordinating these responses in human lymph node tuberculosis remain incompletely defined. METHODS: We performed integrated immune profiling of patients with CTL (n = 60) and non-tuberculous cervical lymphadenopathy (CNTL; n = 44). Immune-gene expression was quantified in peripheral blood and lymph no

IL-10's contextual role in cytotoxic antitumor immunity: evidence, controversies, and therapeutic translation.

Interleukin-10 (IL-10) is classically defined as a potent anti-inflammatory cytokine that protects tissues by constraining myeloid activation and limiting inflammatory cytokine production. Yet, accumulating evidence indicates that, in defined contexts, IL-10 can also promote antitumor immunity by enhancing the fitness and cytotoxic programs of CD8+ T cells and natural killer (NK) cells. Conversely, in other tumor contexts, IL-10 may reinforce immunosuppressive myeloid and regulatory programs, and the factors that determine these divergent outcomes remain incompletely understood. Although these

Induction of remission results in spontaneous enhancement of anti-tumor cytotoxic T-lymphocyte activity in dogs with B cell lymphoma.

Characterization of the tumor microenvironment, particularly the immune cells that infiltrate tumors, provides important predictive and prognostic information in humans with lymphoma and other types of cancer. Tumor associated T lymphocytes have not been previously described in dogs with lymphoma. Therefore, we investigated the phenotype and function of T cells in the lymph nodes of dogs with B cell Non-Hodgkin's lymphoma (NHL), as well as the function of T cells in circulation of these dogs. We found that CD4+ and CD8+ T lymphocytes were few in number and minimally responsive to mitogenic sti

Antibody dependent cell-mediated cytotoxicity--an additional mechanism in human autoimmune myasthenia gravis.

Myasthenia gravis (MG) is an autoimmune disease characterised by serum anti-acetylcholine receptor (AChR) antibodies and several pathogenic mechanisms for the action of these antibodies have been elucidated. In this study, we have analysed a possible additional mechanism for these antibodies, namely antibody-dependent cell-mediated cytotoxicity (ADCC). Using as target cells a cell line expressing AChR, we could show an increased ADCC mediated by sera from MG patients. Sera with AChR antibodies induced a higher cytotoxicity than sera from patients without these antibodies or healthy individuals

Crucial role of marginal zone macrophages and marginal zone metallophils in the clearance of lymphocytic choriomeningitis virus infection.

Macrophages play a key role in the immune defense against pathogens. They control early invasion by antigen-unspecific phagocytosis of pathogens and act as professional antigen-presenting cells to induce antigen-specific T cell responses. To investigate the involvement of particular subsets of the splenic macrophages in an antiviral immune response, we selectively depleted mice of splenic marginal zone macrophages (MZM) and marginal zone metallophils (MM) using the clodronate liposome depletion technique. MZM- and MM-depleted mice were not able to control an infection with lymphocytic choriome

Morphologic analysis of the interactions between lymphocytic choriomeningitis virus-specific cloned cytotoxic T cells and virus infected targets.

The interactions between lymphocytic choriomeningitis virus-specific cloned cytotoxic T lymphocytes (CTL) and virus infected targets have been examined by electron microscopy. CTLs, which were readily differentiated from target cells by the presence of cytoplasmic granular inclusions, made intimate contact with infected cells. Some CTLs contacted infected cells via numerous interdigitating processes; others were observed thrusting finger-like protrusions deep into the target cell; some were seen with their plasma membranes lying closely opposed to that of the infected cell. The majority (55%)

Cytolysis of Junin infected target cells by immune guinea pig spleen cells.

Spleen cells from guinea pigs infected with an attenuated strain of Junin virus (the causative agent of Argentine hemorrhagic fever) specifically lysed virus-infected syngeneic target cells in vitro. This activity was detected as early as 6 days after infection, reached a maximum on days 10-13, and persisted at lower levels, at least through day 30. Monoclonal antibody to guinea pig T cells had no effect on the activity. After B or T cell enrichment techniques, the cytolysis was found with the B cell fraction. Aggregated IgG blocked the cytolysis. These characteristics suggested lytic activity

An altered peptide ligand antagonizes antigen-specific T cells of patients with human T lymphotropic virus type I-associated neurological disease.

Human T lymphotropic virus type I (HTLV-I)-associated myelopathy/tropical spastic paraparesis (HAM/TSP) is an inflammatory neurologic disease associated with HTLV-I infection, in which chronically activated, HTLV-I-specific CD8+ CTL have been suggested to be immunopathogenic. In HLA-A2 HAM/TSP patients, CD8+ HTLV-I-specific CTLs recognize an immunodominant peptide of the HTLV-I Tax protein, Tax11-19. We examined the functional outcome on activation of both cloned peripheral blood and cerebrospinal spinal fluid-derived CTL and bulk PBMC from HAM/TSP patients by altered peptide ligands (APL) der

Defective cytotoxicity of T lymphocytes in myelodysplastic syndrome.

OBJECTIVE: The capacity of mononuclear blood cells to form autoreactive cytotoxic T lymphocytes was investigated in order to elucidate the mechanism of successful immunosuppressive therapy in some myelodysplastic syndrome (MDS) patients (autoreactivity studies). The failure in autoreactivity studies raised the question of alloreactive cytotoxic T lymphocyte formation in MDS (alloreactivity studies). MATERIALS AND METHODS: Sixteen MDS patients and relevant controls were examined. Autoreactive lymphocytes directed against autologous bone marrow mononuclear cells and alloreactive lymphocytes dir

Characterization of a cytotoxic factor induced in mouse peritoneal fluid by OK-432.

A cytotoxic factor (peritoneal cytotoxic factor, PCF) was strongly induced by the injection of LPS into the peritoneal fluids of mice which had been previously primed with OK-432. In order to clarify characteristics of PCF, physicochemical and immunological studies were conducted. When incubated with LPS, the macrophages from mice primed with OK-432 induced PCF whereas the lymphocytes did not. These results indicate that PCF is different from lymphotoxin. PCF appears to be quite similar to tumor necrosis factor (TNF) in the serum for the following reasons: The two factors are similar in the mo

Synovial fibroblasts as target cells for staphylococcal enterotoxin-induced T-cell cytotoxicity.

Rheumatoid arthritis (RA) is a chronic autoimmune disease of unknown aetiology. Recently, superantigens have been implied in the pathogenesis of RA. Superantigens activate a large fraction of T cells leading to the production of cytokines and proliferation. In addition, superantigens direct cellular cytotoxicity towards major histocompatibility complex (MHC) class II-expressing cells. There is now increasing evidence that cytotoxic T cells may be involved in the pathogenesis of RA. In the inflamed synovia class II-positive synovial fibroblasts (SFC) are found. In the present study it was teste

CD3-negative lymphokine-activated cytotoxic cells express the CD3 epsilon gene.

The expression of genes encoding different polypeptide chains of the TCR-CD3 complex was analyzed in a panel of cloned MHC-unrestricted cytotoxic cells. The clones were derived from CD3+ and CD3- human PBL. After expansion in rIL-2, all clones were able to lyse the NK-sensitive target cell line K562. In contrast, lysis of fresh tumor cells was achieved almost exclusively by CD3- clones. To test whether a known TCR-CD3 complex may be involved in MHC-unrestricted cytotoxicity, total RNA from nine CD3+ and 11 CD3- clones was isolated and hybridized with DNA probes for the TCR alpha-, beta-, and g

Effector functions and specificities of normal murine T cells stimulated by syngeneic blasts.

Large numbers of syngeneically stimulated T cells are easily obtained by stimulation of splenic T cell populations with irradiated lipopolysaccharide (LPS) blasts. Thus, proliferative T cell responses were obtained after stimulation with allogeneic LPS blasts, concanavalin A (Con A) blasts, normal spleen cells or peritoneal macrophages while only LPS and Con A blasts were competent syngeneic stimulators. The cytolytic activity of T cells stimulated with allogeneic LPS blasts was H-2 specific, while that of T cells stimulated with syngeneic LPS blasts was often completely nonspecific, and highe

Immunosuppressive therapy in collagen vascular diseases.

Over the past ten to fifteen years cytotoxic drugs have gradually been added to the therapeutic armamentarium of physicians other than oncologists, particularly dermatologists. They are being used to treat nonmalignant cutaneous diseases such as pemphigus vulgaris, bullous pemphigoid, and psoriasis. The rationale for their use in these diseases is conjectural, yet those dermatologists who use them feel strongly not only that cytotoxic drugs are effective, but that when used properly, they are relatively safe. The immunosuppressive activity of cytotoxic drugs has been accepted and widely acclai

Deciphering the syntax of cytotoxic lymphocyte degranulation.

In the killer lymphocyte, the targeted delivery of perforin- and granzyme-containing cytotoxic granules to the immunological synapse is crucial for the eradication of pathogen-infected or transformed targets. This process is achieved through a tightly controlled and highly efficient granule exocytosis pathway. Mutations in the granule trafficking proteins Munc13-4, syntaxin 11, Munc18-2 or Rab27 leads to a fatal lapse of immune surveillance and can be manifested as haemophagocytic lymphohistiocytosis in humans. Elucidation of the role of these proteins in exocytic trafficking is pivotal for ou

Water avoidance stress exacerbates orthotopic pancreatic cancer growth by suppressing cytotoxic T lymphocyte infiltration.

BACKGROUND/OBJECTIVES: The incidence of pancreatic cancer is increasing, and it has a poor prognosis. Also, it is often refractory to standard chemotherapy. Psychological stress influences the development of some malignant neoplasms; however, its immunological mechanism is unclear. In this study, we investigated whether water avoidance stress (WAS) promotes mouse orthotopic pancreatic cancer growth and the mechanism through which WAS influences anticancer immunological responses. METHODS: Mouse pancreatic adenocarcinoma cells were orthotopically inoculated into the pancreas of mice. After 2 w
