Amine-containing pharmaceuticals are the most often detected pharmaceuticals in wastewater and ambient aquatic environments. They can usually be degraded by manganese oxide (MnO2), which is a common natural oxidant in soils. Surfactants often coexist with pharmaceuticals in wastewater. Some amine-containing pharmaceuticals, such as diclofenac (DIC), are acidic and are thus ionic compounds in neutral conditions. These compounds, therefore, have similar properties to surfactants. Surfactants, thus, may influence the adsorption and degradation processes of DIC by MnO2. The effect of the type of s

Homozygotes for homocystinuria due to cystathionine synthase (CS) deficiency accumulate homocysteine and methionine in their blood and tissues. High-dose pyridoxin, folic acid, vitamin B12, or betaine are therapeutical options to lower the elevated homocysteine concentration. These compounds stimulate the transsulfuration or remethylation of homocysteine. Despite such treatment, elevated blood homocysteine concentrations may persist in many homocystinurics. Therefore, it is warranted to study alternative regimen to reduce the blood homocysteine concentration in homocystinurics. Apart from ente

We evaluated the effect of sialadenectomy on hexokinase activity and on rates of lactate formation and of [U-14C]glucose decarboxylation in 3 cellular fractions of the small intestine epithelium from male adult mice. The surgery was carried out under ether anesthesia and a sham-operated group was used as control. Three cell fractions were obtained by shaking the inverted small intestine: 1) tip of the villus, 2) villus and 3) villus and crypt cells. Five days after sialadenectomy, hexokinase activity was reduced in fractions 1 (3.53 +/- 0.65 vs 1.98 +/- 0.25 nmol min-1 mg protein-1, expressed

Gut endocrine cells possess the capacity to take up and decarboxylate biogenic amine precursors. Decarboxylation is mediated by aromatic L-amino acid decarboxylase (AADC) which is encoded by mRNAs differing in their 5' untranslated regions (UTR) depending on the usage of alternative first exons, 1a and 1b, each with its own acceptor site (-13 and -8 bases relative to the translation start site, respectively). We describe here a novel splice variant of exon 1a-AADC mRNA in rat antral mucosa. Both exon 1a and 1b mRNAs were expressed in rat antral mucosa, but the 1a form was spliced into the acce

A method is described for the synthesis of α-aryl and allylic sulfones and sulfonamides through CuBr/oxalamide-catalyzed coupling at 40 °C. This transformation involves coupling with (hetero)aryl or vinyl bromides and 2-substituted sulfonylacetates (or amide analogues), proceeding via spontaneous hydrolysis and decarboxylation. The conditions are compatible with various functional groups and heterocycles, allowing for the diverse syntheses of the target compounds.

A female infant with an intermediate variant of maple syrup urine disease is described. The patient had psychomotor retardation and high plasma levels of branched chain aminoacids. Leucine decarboxylation rate in leukocytes was diminished. Cranial computed tomography showed decreased density in the cerebral white matter. After starting dietary treatment the infant resumed her psychomotor development and the abnormal images previously seen on computed tomography disappeared.

Picolinic acid (PA), a typical C-2-carboxylated pyridine derivative, is a metabolite of l-tryptophan and many other aromatic compounds in mammalian and microbial cells. Microorganisms can degrade and utilize PA for growth. However, the precise mechanism of PA metabolism remains unknown. Alcaligenes faecalis strain JQ135 utilizes PA as its carbon and nitrogen source for growth. In this study, we screened a 6-hydroxypicolinic acid (6HPA) degradation-deficient mutant through random transposon mutagenesis. The mutant hydroxylated 6HPA into an intermediate, identified as 3,6-dihydroxypicolinic acid

The effects of increased cardiac work, pyruvate and insulin on the state of pyruvate dehydrogenase (PDH) activation and rate of pyruvate decarboxylation was studied in the isolated perfused rat heart. At low levels of cardiac work, 61% of PDH was present in the active form when glucose was the only substrate provided. The actual rate of pyruvate decarboxylation was only 5% of the available capacity calculated from the percent of active PDH. Under this condition, the rate of pyruvate decarboxylation was restricted by the slow rate of pyruvate production from glycolysis. Increasing cardiac work

Friedel-Crafts alkylation is a cornerstone transformation for arene functionalization; however, strongly electron-deficient nitroarenes fail to undergo alkylation under conventional conditions. This long-standing limitation is notable given the prevalence of nitroarenes in pharmaceuticals and agrochemicals and their central role as synthetic building blocks. Here we report a photocatalytic strategy that overcomes this challenge, enabling direct C-H alkylation of nitroarenes using readily available carboxylic acids as alkyl radical precursors. Under mild conditions, alkyl radicals generated thr

Carboxyl radicals are reactive intermediates, which potentially offer opportunities to develop unique transformations in radical chemistry, yet their straightforward generation from carboxylic acids has largely relied on the use of environmentally less benign methods, and their application has been mainly limited to decarboxylative functionalizations. Here, we present a photoactivated ketone-catalyzed direct generation of carboxyl radicals via hydrogen atom transfer (HAT) from the O-H bond of carboxylic acids. An integrated approach combining theoretical predictions and experimental validation

DMSO is well-known as a solvent but here we have explored DMSO as a sole reagent for dehydrogenation of tetrahydrocarbazoles, oxidation of alcohols to carbonyl compounds, decarboxylation of keto-acids to aldehydes, and C-C/C-N bond formations via cyclization, self-coupling, the Povarov reaction and Friedlander synthesis via a radical pathway. This methodology involves simple, metal-free and catalyst-free conditions.

A practical and unified strategy has been described for the preparation of mono- and difluoromethylated phenanthridine derivatives using a visible-light-promoted alkylation and decarboxylation sequence from biphenyl isocyanides with ethyl bromofluoroacetate (EBFA) or ethyl bromodifluoroacetate (EBDFA). These reactions could be carried out at room temperature in good to excellent chemical yields. Both stepwise and one-pot procedures have been developed, which makes this strategy more attractive.

In this paper, Pd-catalyzed [4+2] decarboxylative cycloaddition of 4-vinylbenzodioxinones with barbiturate-derived alkenes has been developed, leading to various spirobarbiturate-chromane derivatives in high yields with excellent diastereo- and enantioselectivities. The scale-up reaction and further derivation of the product were demonstrated. A plausible reaction mechanism was also proposed.

A mild, metal- and additive-free, inexpensive electrochemical decarboxylation of arylalkanecarboxylic acids, including all the most important nonsteroidal anti-inflammatory drugs based on such a framework, is discussed. An efficient, nonaqueous, and essentially no-chromatography workup procedure enhances the attractiveness of the disclosed solution. This methodology can facilitate scaffold diversification by functionalization of NSAIDs or by providing easy access to building blocks from a simple arylalkanecarboxylic acid feedstock.

A dithionite-involved multicomponent reaction of redox-active esters and alkenes/alkynes is comprehensively achieved for the construction of alkyl and alkenyl tertiary sulfones. The industrial feedstock sodium dithionite is employed as a sulfur dioxide surrogate and a single-electron reductant to initiate the decarboxylation of redox-active esters. Mechanistic studies further indicated that the transformation underwent a proton trapping process, which was different from the traditional radical trapping pathway.

Orotidine-5'-monophosphate decarboxylase (ODCase) has evolved to catalyze a decarboxylation reaction, most probably via a carbanion species at the C6 position of orotidine-5'-monophosphate. We reveal an unusual biochemical pathway of conversion of 6-cyano-uridine-5'-monophosphate by ODCase to barbiturate-5'-monophosphate via perhaps an electrophilic center at the C6 position, leading to inhibition. This potential of ODCase is very useful in the design of novel inhibitors.

BACKGROUND: This study investigates the safety evaluation of enterocin-producing 11 E. mundtii and two E. faecium strains previously isolated from small livestock colostrums. Enterococcus species do not possess Generally Recognized as Safe (GRAS) status. Hence, it is critical to scrutinize enterococci's antibiotic resistance, virulence characteristics, and biogenic amine production capabilities in order to assess their safety before using them as starter or adjunct cultures.
RESULTS: Enterococcus strains showed susceptibility to medically significant antibiotics. Multiple-drug resistance (MDR

Maple syrup urine disease (MSUD) is an inherited metabolic disorder due to decreased decarboxylation of branched chain keto acids triggering an accumulation of leucine, isoleucine, and valine. We describe two infants with biochemically confirmed MSUD in whom computed tomography (CT) revealed cerebral edema. In one of these cases repeat CT 40 days after institution of appropriate therapy revealed that the edema had disappeared and the ventricles had enlarged.

This paper describes the preparation of 1,2,3-triazole derivatives of salicylic acid from the methyl 4-azidosalicylate, by 1,3-dipolar cycloaddition reactions to beta-ketoesters, beta-ketoamides, beta-diketones and to symmetrical or asymmetrical alkynes. Other derivatives were obtained by hydrolysis and decarboxylation reactions. Some compounds were evaluated for antiinflammatory activity by an in vitro test based upon the inhibition of the cyclooxygenase enzyme, but they generally showed a weak activity.

Values of various alpha-keto acids in whole blood were determined from cases of cerebrocortical necrosis (CCN), a thiamine deficiency disease, and were compared with those from normal animals. Thiamine pyrophosphate is an essential cofactor in the decarboxylation of many alpha-keto acids and this was reflected in elevated values not only of pyruvic acid but of glyoxylic, alpha-keto glutaric, phenyl pyruvic and hydroxyphenyl pyruvic acids in the CCN cases.
