Decellularization of the human urethra for tissue engineering applications.

Recently, several scaffolds have been introduced for urethral tissue engineering. However, acellular human urethral scaffold harvested from deceased donors may provide significant advantages compared to synthetic, composite, or other biological scaffolds. This study aims to develop the protocol for decellularization of the human urethra that preserves substantial extracellular matrix (ECM) components, which are essential for subsequent recellularization mimicking the natural environment of the native ECM. A total of 12 human urethras were harvested from deceased donors. An equal part of every

Rapid Preparation Method for Preparing Tracheal Decellularized Scaffolds: Vacuum Assistance and Optimization of DNase I.

Decellularized scaffolds are an effective way for tracheal tissue engineering to perform alternative treatments. However, clinically used decellularized tracheal scaffolds have a long preparation cycle. The purpose of this study is to improve the efficiency of decellularization by vacuum assistance and optimizing the concentration of DNase I in the decellularization process and to quickly obtain tracheal decellularized scaffolds. The trachea of New Zealand white rabbits was decellularized with 2, 4, 6, and 8 KU/mL DNase I under vacuum. The performance of the decellularized tracheal scaffold wa

Decellularization techniques of human foreskin for tissue engineering application.

The rapid development of tissue engineering (TE) and regenerative medicine brings an acute need for biocompatible and bioactive biological scaffolds to regenerate or restore damaged tissue. Great attention is focused on the decellularization of tissues or even whole organs, and the subsequent colonization of such decellularized extracellular matrices by recipient cells. The foreskin is an integral, normal part of the external genitalia that forms the anatomical covering of the glans penis and the urinary meatus of all human and non-human primates. It is mucocutaneous tissue that marks the boun

A Method for Fabricating Long Decellularized Scaffolds from Skeletal Muscle.

Skeletal muscle tissue engineering (SMTE) is a rapidly evolving field with applications spanning regenerative medicine, disease modeling, drug screening, and biohybrid robotics. Effective SMTE requires scaffolds that reproduce the anisotropic architecture and mechanical properties of native muscle while supporting macroscale tissue formation. Decellularized tissues are strong candidates; however, existing approaches face key limitations. Whole-muscle decellularization requires complex perfusion systems and often fails to fully clear the tissue core. In contrast, minced-tissue processing comple

Biointegration of a partially decellularized tracheal scaffold in a porcine model - preliminary results.

Some pediatric tracheal pathologies remain therapeutic dead ends for which current palliative strategies are fraught with serious complications. With the aim of a tracheal replacement, our team has previously developed and patented a clinical grade partially decellularized trachea (PDT) from porcine tracheas. The aim of this work was to study and compare the biointegration mechanisms of this PDT in vivo, in a pig cervical muscle, with or without immunosuppressant. The secondary objective was to evaluate the optimal maturation time of the PDT in this heterotopic position. In total, 11 female La

Challenges in Fabrication of Tissue-Engineered Cartilage with Correct Cellular Colonization and Extracellular Matrix Assembly.

A correct articular cartilage ultrastructure regarding its structural components and cellularity is important for appropriate performance of tissue-engineered articular cartilage. Various scaffold-based, as well as scaffold-free, culture models have been under development to manufacture functional cartilage tissue. Even decellularized tissues have been considered as a potential choice for cellular seeding and tissue fabrication. Pore size, interconnectivity, and functionalization of the scaffold architecture can be varied. Increased mechanical function requires a dense scaffold, which also eas

Recellularized rat testis scaffolds with embryoid bodies cells: a promising approach for tissue engineering.

Tissue engineering is gaining use to investigate the application of its techniques for infertility treatment. The use of pluripotent embryonic cells for in vitro production of viable spermatozoa in testicular scaffolds is a promising strategy that could solve male infertility. Due to cell-extracellular matrix (ECM) interactions, here we aim to investigate the differentiation of embryoid bodies (EBs) in cultured into decellularized rat testis scaffolds. Decellularized testis (P = 0.019) with a low concentration of gDNA (30.58 mg/ng tissue) was obtained by sodium dodecyl sulfate perfusion. The s

Histopathologic findings in a novel decellularized pulmonary homograft: an autopsy study.

Innovative valved conduits for reconstruction of the right ventricular outflow tract are desirable to overcome the risk of valve deterioration of conventional homografts. This study describes the histopathology of a novel decellularized pulmonary homograft (SynerGraft) implanted in the right ventricular outflow tract of a 60-year-old man 5 weeks before death. The histomorphology of this decellularized homograft showed integrity of its extracellular matrix and a gradual cellular infiltrate consisting predominantly of macrophages, resembling an early nonspecific inflammatory phase of recellulari

Fabrication and Characterization of Bioengineered Sandwich Model Scaffold for Urethral Stricture Reconstruction.

Urethral stricture remains a challenging clinical condition, particularly in patients with long or complex defects where suitable donor tissue is limited. Tissue engineering offers a promising alternative through the development of biomimetic scaffolds capable of restoring urethral structure and function. In this study, we fabricated a sandwich nanofibrous scaffold based on polyglycolic acid (PGA) and decellularized urethral extracellular matrix (uECM) using a sandwich design (uECM/PGA/uECM) and compared it with PGA and mix (PGA-uECM) scaffolds. Successful decellularization was confirmed by hi

Optimization of swim bladder decellularization with sodium dodecyl sulfate, triton X-100, and aminosulfabetaine-16 for use as a biomaterial.

Swim bladders are biomaterials with numerous potential applications, but effective decellularization is essential to maximize retention of the extracellular matrix (ECM) and minimize immunogenicity. However, systematic optimization of detergent-based decellularization protocols for swim bladder remains limited, particularly with respect to balancing efficient nuclear removal with preservation of ECM, mechanics, and anti-calcification properties. We performed a systematic evaluation of the efficacy of three detergents-sodium dodecyl sulfate (SDS), Triton X-100 (TX-100), and aminosulfobetaine-16

Recellularization of scaffolds derived from precision-cut kidney slices.

The global rise in chronic kidney disease necessitates innovative solutions for end-stage renal disease that can help to overcome the limitations of the only available treatment options, transplantation and dialysis. Tissue engineering presents a promising alternative, leveraging decellularized scaffolds to retain the extracellular matrix (ECM). However, optimizing methods for decellularization and recellularization remains a challenge. Here we present novel work which builds on our previous study where we investigated several decellularization protocols. In this study we analyzed the suitabil

Peripheral Nerve Decellularisation Protocol for Allogeneic Transplantation: From Tissue Procurement to Banking.

Peripheral nerve injuries affect over one million individuals annually worldwide due to various causes such as trauma, metabolic disorders, and autoimmune diseases. While autologous nerve grafting remains the gold standard for treating large-gap nerve injuries, its limitations, including limited tissue availability, donor site morbidity, infection risk, and suboptimal functional recovery, have spurred interest in alternative approaches. Among these, allogeneic nerve grafting has emerged as a promising option, offering structural and functional advantages due to the native architecture of donor

Regional and cell specific bioactivity of injectable extracellular matrix biomaterials in myocardial infarction.

Myocardial infarction (MI) remains a global health concern. To mitigate subacute and chronic MI pathophysiology, we previously investigated a pro-reparative decellularized extracellular matrix hydrogel. Despite increasing interest in biomaterial scaffolds, single cell and spatially resolved transcriptomics have not been used to probe their therapeutic activity in the heart. Here, we utilize spatial transcriptomics and single nucleus RNA sequencing to delineate the regional and cell-specific bioactivity of extracellular matrix biomaterials. Extracellular matrix hydrogel subacute treatment in fe

Decellularized Pancreatic Tail as Matrix for Pancreatic Islet Transplantation into the Greater Omentum in Rats.

Infusing pancreatic islets into the portal vein currently represents the preferred approach for islet transplantation, despite considerable loss of islet mass almost immediately after implantation. Therefore, approaches that obviate direct intravascular placement are urgently needed. A promising candidate for extrahepatic placement is the omentum. We aimed to develop an extracellular matrix skeleton from the native pancreas that could provide a microenvironment for islet survival in an omental flap. To that end, we compared different decellularization approaches, including perfusion through th

Reconstruction of pulmonary venous conduit with CorMatrix in lung transplant.

Anomalous pulmonary vein anatomy is infrequently encountered during reconstructive lung surgery, especially lung transplantation. Complications of pulmonary venous anastomosis carry high morbidity and mortality. We report a case of anomalous pulmonary vein reconstruction with a decellularized porcine small intestinal submucosa-derived extracellular matrix during bilateral lung transplantation in an 18-year-old woman with cystic fibrosis.

TGF-β1-induced deposition of provisional extracellular matrix by tracheal basal cells promotes epithelial-to-mesenchymal transition in a c-Jun NH2-terminal kinase-1-dependent manner.

Epithelial cells have been suggested as potential drivers of lung fibrosis, although the epithelial-dependent pathways that promote fibrogenesis remain unknown. Extracellular matrix is increasingly recognized as an environment that can drive cellular responses in various pulmonary diseases. In this study, we demonstrate that transforming growth factor-β1 (TGF-β1)-stimulated mouse tracheal basal (MTB) cells produce provisional matrix proteins in vitro, which initiate mesenchymal changes in subsequently freshly plated MTB cells via Rho kinase- and c-Jun NH2-terminal kinase (JNK1)-dependent proce
