Effects of fibrinogen in vivo on dentine permeability in the dog.

Dentine permeability measurements were made each hour for 6 h in tooth cavities prepared in control dogs or dogs which had been pretreated with a purified fraction of snake venom to deplete their plasma of fibrinogen. The control dogs exhibited a progressive fall in dentine permeability (approximately 50 per cent reduction in 6 h); the permeability in the fibrinogen-depleted dogs only fell 15 per cent in 6 h. The results implicate plasma proteins, particularly high molecular-weight proteins such as fibrinogen, as agents which reduce dentine permeability in vivo.

Pulpal response to the application of phosphoric acid to dentin.

Experimental cavities were prepared in forty-six clinically intact premolars of young individuals to evaluate the biologic effects of 50 per cent phosphoric acid etchant containing 7 percent zinc oxide by weight. The experimental periods ranged from 30 minutes to 150 days. The remaining dentin thickness varied from 1.8 to 3.5 mm. The teeth treated with the acid showed no more than a moderate pulpal response at the varying time periods. All of the acid-treated teeth demonstrated pulpal imflammation in the absence of clinical pain. Acid etchants should not be used on exposed or unprotected denti

Effect of Different Irrigating Solutions on the Microhardness of Dentin.

BACKGROUND: The selection of irrigating solutions during root canal therapy is essential for maintaining dentin structure and ensuring effective disinfection. However, various irrigants can alter the microhardness of dentin, which might have an impact on the treated tooth's mechanical characteristics and long-term viability. MATERIALS AND METHODS: To produce uniform dentin discs from the central portion of the root, forty human premolars were sectioned. Based on the irrigating solution employed, the specimens were split into four groups (n = 10): Group 1 received 2.5% sodium hypochlorite (NaO

Interventions for treating cavitated or dentine carious lesions.

BACKGROUND: Traditionally, cavitated carious lesions and those extending into dentine have been treated by 'complete' removal of carious tissue, i.e. non-selective removal and conventional restoration (CR). Alternative strategies for managing cavitated or dentine carious lesions remove less or none of the carious tissue and include selective carious tissue removal (or selective excavation (SE)), stepwise carious tissue removal (SW), sealing carious lesions using sealant materials, sealing using preformed metal crowns (Hall Technique, HT), and non-restorative cavity control (NRCC). OBJECTIVES:

Effect of proanthocyanidin on ultrastructure and mineralization of dentine collagen.

OBJECTIVE: Proanthocyanidin (PA) is a natural collagen cross-linker that has been used in dentine matrix biomodification for reparative and preventive therapies. This study evaluated the ultrastructure of collagen after its interaction with PA. Furthermore, the mineralization of PA-biomodified collagen matrix was observed. METHODS: Ten freshly extracted sound human molars were sectioned into 0.5mm×1.7mm×7mm beams for ultrastructural evaluation of PA and dentine matrix under Field Emission Scanning Electron Microscopy (FESEM) and Transmission Electron Microscopy (TEM). Specimens for TEM were c

Stress and its role in the dentin hypersensitivity in rats.

INTRODUCTION: The perception of pain varies individually. Chronic stress leads to analgesia. The use of animal chronic mild stress model to mimic human condition was previously developed and now applied in the evaluation of pain perception in rats with dentin hypersensitivity (DH). AIMS: Using DH model induced by dentin erosion (DE) mediated by acidic solution, the present study aimed the evaluation of the interaction of chronic stress and pain induced by DH in rats with DE. METHODS: DH was induced by ad libitum 30-day intake of acidic solution. Stress was induced by the New York subway mode

The role of toothpaste in the aetiology and treatment of dentine hypersensitivity.

Dentine hypersensitivity (DH) is a common, painful dental condition with a multi-factorial aetiology. The hydrodynamic mechanism theory to explain dentine sensitivity also appears to fit DH: lesions exhibiting large numbers of open dentinal tubules at the surface and patent to the pulp. By definition, DH can only occur when dentine becomes exposed (lesion localisation) and tubules opened (lesion initiation), thus permitting increased fluid flow in tubules on stimulation. Erosion, particularly from dietary acids appears to play a dominant role in both processes. Toothbrushing with most toothpas

Inflammation-regeneration interplay in the dentine-pulp complex.

OBJECTIVES: Dental tissue disease and trauma provides an excellent model for the interaction between tissue defence and regenerative processes and has application to many of the body's other tissues. Following dental tissue infection, characterised by caries, the molecular and cellular mediators of the immune/inflammatory processes clearly impact on the dental tissues' natural regenerative responses. This review of the literature was performed to better understand how these two processes interact and identify whether cross-talk may provide novel areas for future research and subsequent transla

Dentinal dysplasia type I: report of a case.

A case of dentinal dysplasia type I is presented. This rare hereditary disturbance of dentine is characterized by short-rooted teeth with sharp conical apical constrictions, aberrant growth of dentine in the pulp chamber leading to reduced pulp space in permanent teeth and total pulpal obliteration in the primary dentition. Clinical, radiographic and histopathological material from a 7-year-old boy, showing the typical features of this disorder in which teeth are prematurely lost through periapical abscesses, cysts or spontaneous exfoliation, is described. Effect of bisphosphonates and gallium on dentin resorption in vitro.

Replacement resorption may follow the replantation of an avulsed tooth. Currently there is no effective treatment for replacement resorption. The purpose of this study was to investigate the effect of bisphosphonates and gallium nitrate, which have been shown to reduce bone resorption, on cells which resorb dentin. Osteoclast-like cells were obtained by culturing cells from prenatal chick tibeas. These cells were seeded onto slices of human dentin which had been soaked in either saline (control), or solutions of 10(-5) M 1-hydroxyethylidene-1, 1-bisphosphonic acid (EHBP), 10(-6) M dichlorometh

[Morphological and functional properties of odontoclasts on dentine resorption].

Tooth root resorption in tooth movement is the principal unsolved problem in orthodontics. In order to study this mechanism, odontoclasts in bovine deciduous tooth root-resorbing tissues (RRT) were cultured and characterized for their biological properties in comparison with those of osteoclasts. Multinucleated cells isolated from the RRT had tartrate-resistant acid phosphatase activity and responded to calcitonin and also formed the resorption lacunae on the dentine. These results are consistent with the notion that odontoclasts resemble osteoclasts with respect to enzyme-cytochemical and mor

Relationship between age and deposit of peritubular dentine.

Peritubular dentine is a mineralised deposit formed centripetally in the dentine tubules with advancing age, so that the tubular diameter is smaller in teeth from older persons. The aim of the present study was to investigate the relationship between age in humans and the amount of peritubular dentine and the extent of the consequent obliteration of the tubules, and to find out whether this relationship was strong enough to be used as a parameter for age estimation. Fifty mandibular central and lateral human incisors were ground on the lingual aspect of the root and examined in a scanning elec

Effect of wettability of adhesive resins on bonding to dentin.

The purpose of this study was to investigate on human dentin the relationships between, on the one hand, the wettability of five adhesive compounds, characterized by different solubility parameters delta, and on the other, the thickness of the resin impregnated dentin layer (RIL) and the shear bond strength (SBS). Cut dentin surfaces were treated with conditioners of pH 1.25, 3.6 or 7.4. Shear bond strength was measured on 20 specimens each. RIL was determined by scanning electron microscope on specimens fractured perpendicularly to the bonding interface. When resins with delta 32, 30 or 39 (M

Traumatic root resorption in dentine-immunized mice.

Traumatic root resorption in a mouse model has been shown to coincide with a decline in naturally occurring serum antibody levels to dentin. It has been proposed that root resorption may be dentin antibody mediated. The purpose of this study was to examine the traumatic root resorption response in mice after hyperimmunization with a crude tooth extract (dentin). The hypothesis of this study was that elevated dentin antibody titers would positively correlate with root resorption. Mice were immunized with mouse dentin and controls were sham immunized. All mice were boosted 4 weeks later with or

Odontoblast-mediated regulation of the progression of dentinal caries.

Osteoblasts and odontoblasts are both derived from the same mesenchymal cell line. Our aim was to investigate whether the processes of bone destruction and dentinal caries are biologically similar. The working hypothesis was that after the initiation of caries in the enamel, its rate of progression in the dentine is regulated by cell-mediated factors. Experimental caries was induced in the rat with a high sucrose diet combined with Streptococcus sobrinus infection. Both destruction of dentine and its apposition in the pulp under the carious lesions were measured after vital staining with tetra

In vivo morphogenetic activity of dentine matrix proteins.

Matrix proteins (EDTA-soluble and collagenase-released) from rabbit dentine have been demonstrated to show in vivo morphogenetic activity in exposed pulps of ferret canine teeth. In young adult ferrets, the reparative dentine resembled normal primary tubular dentine with odontoblast-like cells lining its formative surface. The reparative response in older animals was seen as osteo-dentine deposition. In both cases, the reparative response was characterized by its intensity within a comparatively short time period and also by the low grade or complete absence of any localized inflammatory respo
