Generation of Xenopus Haploid, Triploid, and Hybrid Embryos.

Frog species of the genus Xenopus are widely used for studies of cell and developmental biology, and recent genome sequencing has revealed interesting phylogenetic relationships. Here we describe methods to generate haploid, triploid, and hybrid species starting from eggs and sperm of Xenopus laevis and Xenopus tropicalis that enable investigation of how genome size and content affect physiology at the organismal, cellular, and subcellular levels.

Biological organization of the extraocular muscles.

Extraocular muscle is fundamentally distinct from other skeletal muscles. Here, we review the biological organization of the extraocular muscles with the intent of understanding this novel muscle group in the context of oculomotor system function. The specific objectives of this review are threefold. The first objective is to understand the anatomic arrangement of the extraocular muscles and their compartmental or layered organization in the context of a new concept of orbital mechanics, the active pulley hypothesis. The second objective is to present an integrated view of the morphologic, cel

Hox genes and regional patterning of the vertebrate body plan.

Several decades have passed since the discovery of Hox genes in the fruit fly Drosophila melanogaster. Their unique ability to regulate morphologies along the anteroposterior (AP) axis (Lewis, 1978) earned them well-deserved attention as important regulators of embryonic development. Phenotypes due to loss- and gain-of-function mutations in mouse Hox genes have revealed that the spatio-temporally controlled expression of these genes is critical for the correct morphogenesis of embryonic axial structures. Here, we review recent novel insight into the modalities of Hox protein function in impart

Genes and signals regulating murine trophoblast cell development.

A fundamental step in embryonic development is cell differentiation whereby highly specialised cell types are developed from a single undifferentiated, fertilised egg. One of the earliest lineages to form in the mammalian conceptus is the trophoblast, which contributes exclusively to the extraembryonic structures that form the placenta. Trophoblast giant cells (TGCs) in the rodent placenta form the outermost layer of the extraembryonic compartment, establish direct contact with maternal cells, and produce a number of pregnancy-specific cytokine hormones. Giant cells differentiate from prolifer

Developmental mutant mouse models for external genitalia formation.

Development of external genitalia and perineum is the subject of developmental biology as well as toxicology and teratology researches. Cloaca forms in the lower (caudal) end of endoderm. Such endodermal epithelia and surrounding mesenchyme interact with various signals to form the external genitalia. External genitalia (the anlage termed as genital tubercle: GT) formation shows prominent sexually dimorphic morphogenesis in late embryonic stages, which is an unexplored developmental research field because of many reasons. External genitalia develop adjacent to the cloaca which develops urethra

Genetic study on developmental dysplasia of the hip.

BACKGROUND: Developmental dysplasia of the hip (DDH) is a developmental disorder of the hip joint that results in an abnormal socket of the femoral head, ranging from instability, subluxation and complete dislocation of the hip joint. DDH has a considerable genetic component. DESIGN: We reviewed the recent progress on genetic study of DDH. RESULTS: By linkage and case-control association studies, DDH loci have been identified with several susceptibility genes that involve in bone and joint biology; however, no unequivocal genes with global significance have been detected. The current problem

Childhood exposure to violence and lifelong health: clinical intervention science and stress-biology research join forces.

Many young people who are mistreated by an adult, victimized by bullies, criminally assaulted, or who witness domestic violence react to this violence exposure by developing behavioral, emotional, or learning problems. What is less well known is that adverse experiences like violence exposure can lead to hidden physical alterations inside a child's body, alterations that may have adverse effects on life-long health. We discuss why this is important for the field of developmental psychopathology and for society, and we recommend that stress-biology research and intervention science join forces

The Influence of Pathologies upon Sensory Perception and Sensory Coordination in Children with Developmental Dyslexia and Learning Disorders: A Unified Theory of Developmental Dyslexia.

This case is presented to explain that developmental dyslexia and related autistic spectrum disorders have solely pathological origins. There is a general consensus of opinion which supports the phonological theory. However, this largely ignores the biological basis for all aspects of the brain's development and function, and hence, for its dysfunction. A unified explanation must take into account all salient features including cognitive dysfunction, encephalograph (EEG) frequencies, neural networks, physiological systems, autonomic nervous system and the function of the cerebellum. It must ex

The emergence of pigment cell biology: a personal view.

This is a semi-autobiographical coverage of my research career in pigment cell biology presented in the context of the emergence and growth of the discipline. This anecdotal presentation tells about some historical personages in the field. My undergraduate studies at the University of Rochester are related to my graduate work at the University of Iowa. I tell how my dissertation research was derived from a marriage between my interests in experimental embryology and the new field of comparative endocrinology. Development of cranial placodes: insights from studies in chick.

This review focuses on how research, using chick as a model system, has contributed to our knowledge regarding the development of cranial placodes. This review highlights when and how molecular signaling events regulate early specification of placodal progenitor cells, as well as the development of individual placodes including morphological movements. In addition, we briefly describe various techniques used in chick that are important for studies in cell and developmental biology.

Beyond genomic studies of congenital heart defects through systematic modelling and phenotyping.

Congenital heart defects (CHDs), the most common congenital anomalies, are considered to have a significant genetic component. However, despite considerable efforts to identify pathogenic genes in patients with CHDs, few gene variants have been proven as causal. The complexity of the genetic architecture underlying human CHDs likely contributes to this poor genetic discovery rate. However, several other factors are likely to contribute. For example, the level of patient phenotyping required for clinical care may be insufficient for research studies focused on mechanistic discovery. Although se

Nuclear bodies in the caecum of the rabbit.

The structure and cytochemistry of nuclear bodies in the caecum of the rabbit are described and a developmental pathway from morphologically simple to complex forms is proposed. Simple nuclear bodies are proteinaceous in nature while complex bodies possess a proteinaceous capsule with some associated ribonucleoprotein. The interna of complex bodies possess various individual components consisting either of deoxyribonucleoprotein, ribonucleoprotein, or unassociated proteins. The significance of these nuclear bodies in caecal biology is discussed.

Quantified multidimensional assessment of autism and other pervasive developmental disorders. Application for bioclinical research.

A large number of investigation techniques are used to establish the relationships between the clinical and biological data which are necessary for physiopathological analysis in the field of developmental disorders. It therefore seemed necessary to develop a quantified grouping system, based on developmental assessments, which could allow closer matching between clinical evaluations and biological numerical data. Two hundred and two subjects presenting developmental disorders (autistic disorder, pervasive developmental disorder not otherwise specified and mental retardation) were examined. Fo

Scanning Bessel beam microscopy with a protected and corrective objective for solvent-cleared large samples.

Emerging imaging strategies, such as tissue clearing and advanced light-sheet modalities, are transforming the visualization of cell differentiation, tissue reorganization, and morphogenesis in both model and non-model organisms, thereby driving future discoveries of conserved developmental programs and species-specific innovations. In this study, we present a versatile Bessel light sheet microscopy (BLX) system compatible with both immersion and capped air objectives, optimized for whole-mount three-dimensional imaging of cleared tissues. We demonstrate that autofluorescence-based light-sheet

Generating Diverse Spinal Motor Neuron Subtypes from Human Pluripotent Stem Cells.

Resolving the mechanisms underlying human neuronal diversification remains a major challenge in developmental and applied neurobiology. Motor neurons (MNs) represent a diverse pool of neuronal subtypes exhibiting differential vulnerability in different human neurodegenerative diseases, including amyotrophic lateral sclerosis (ALS) and spinal muscular atrophy (SMA). The ability to predictably manipulate MN subtype lineage restriction from human pluripotent stem cells (PSCs) will form the essential basis to establishing accurate, clinically relevant in vitro disease models. EMT in developmental morphogenesis.

Carcinomas, cancers of epithelial origin, constitute the majority of all cancers. Loss of epithelial characteristics is an early step in carcinoma progression. Malignant transformation and metastasis involve additional loss of cell-cycle control and gain of migratory behaviors. Understanding the relationships among epithelial homeostasis, cell proliferation, and cell migration is therefore fundamental in understanding cancer. Interestingly, these cellular events also occur frequently during animal development, but without leading to tumor formation. Can we learn anything about carcinomas from

Evolutionary changes of developmental mechanisms in the absence of cell lineage alterations during vulva formation in the Diplogastridae (Nematoda).

The origin of novelty is one of the least understood evolutionary phenomena. One approach to study evolutionary novelty comes from developmental biology. During developmental cell fate specification of the nematode Pristionchus pacificus (Diplogastridae), five cell fates can be distinguished within a group of twelve ventral epidermal cells. The differentiation pattern of individual cells includes programmed cell death, cell fusion and vulval differentiation after induction by the gonad. A cell lineage comparison among species of seven different genera of the Diplogastridae indicates that the d

Species Choice and Model Use: Reviving Research on Human Development.

While model organisms have had many historians, this article places studies of humans, and particularly our development, in the politics of species choice. Human embryos, investigated directly rather than via animal surrogates, have gone through cycles of attention and neglect. In the past 60 years they moved from the sidelines to center stage. Research was resuscitated in anatomy, launched in reproductive biomedicine, molecular genetics, and stem-cell science, and made attractive in developmental biology. I explain this surge of interest in terms of rivalry with models and reliance on them. T

The critical role of collagen VI in lung development and chronic lung disease.

Type VI collagen (collagen VI) is an obligate extracellular matrix component found mainly in the basement membrane region of many mammalian tissues and organs, including skeletal muscle and throughout the respiratory system. Collagen VI is probably most recognized in medicine as the genetic cause of a spectrum of muscular dystrophies, including Ullrich Congenital Myopathy and Bethlem Myopathy. Collagen VI is thought to contribute to myopathy, at least in part, by mediating muscle fiber integrity by anchoring myoblasts to the muscle basement membrane. Interestingly, collagen VI myopathies prese

Establishment of a physiological reference for embryonic development of three SPF duck breeds compared to SPF chickens.

Specific pathogen free (SPF) duck and chicken embryos serve as essential biological models bridging fundamental developmental biology and biomedical applications. Although morphological staging has been established for SPF ducks, systematic and standardized reference frameworks for their physiological and biochemical characteristics are still lacking. This study aimed to establish a comprehensive and quantitative reference framework for the embryonic development of three SPF duck breeds (HSM, HBW, and HJD) at different developmental stages, using SPF chickens (BM8 line) as a benchmark.
