Immunohistological detection of pentosidine in the epiglottis:A contribution to molecular age estimation.

Pentosidin (Pen) accumulates in various tissues with increasing age, among others in the epiglottis. So far, all published Pen concentrations in the epiglottis are based on the analysis of total tissue samples via HPLC. The morphological structures and proteins in which Pen accumulates remain unknown. Such insights would be of great importance for optimizing Pen based methods for age estimation, especially to overcome the problem of sample inhomogeneity in older individuals as source of error in age estimation based on Pen.Against this background, pentosidine was detected immunohistochemically

Cervical chondrocutaneous branchial remnants: A report of 29 cases and review of the literature.

OBJECTIVE: Cervical chondrocutaneous branchial remnants (CCBRs) are rare masses located in the anterior region of the neck. Though the basic characteristics of these rare masses were first described by Atlan in 1997, a critical amount of information about these masses remains unknown. This study aimed to further clarify the characteristics of these rare masses. METHODS: We retrospectively reviewed the clinical records of patients with CCBRs in our facility during a 32-year period ranging from 1988 to 2019. We then compared our clinical records with other case reports. RESULTS: There were 29

Trauma Management of the Auricle.

Smaller injuries of the auricle, such as lacerations without tissue loss, have more or less standardized treatment protocols that require thorough wound closure of each affected layer. Even extended lacerations of larger parts of the ear quite often heal with only minor irregularities. New in vivo diagnostic tools have aided the understanding of this outstanding "skin flap behavior." At the other end of the trauma severity spectrum are partial or complete amputations of the ear. Here, the debate has become more intense over the last decade. There were numerous reports of successful microvascul

3D-printed bionic tracheal stents based on tunable triblock thermoplastic copolyester elastomers for tracheal stenosis.

Poly(ε-caprolactone) (PCL) has attracted significant attention in biomedical applications, including drug delivery, tissue engineering, and biodegradable implants. However, its inherently poor elasticity restricts its applicability in mechanically dynamic tissues and organs. Here, 4-methyl-ε-caprolactone (MCL), a biobased monomer structurally similar to ε-caprolactone (CL), was incorporated to produce poly(ε-caprolactone)-b-poly(4-methyl-ε-caprolactone)-b-poly(ε-caprolactone) (PCL-PMCL-PCL) triblock polymers via sequential ring-opening polymerization. The resulting triblock polymers exhibited

VEGF-neutralized platelet-rich plasma with adipose-derived stromal vascular fraction enhanced osteochondral repair in a point-of-care goat model.

BACKGROUND/OBJECTIVE: Focal articular cartilage defects do not heal spontaneously. Platelet-rich plasma (PRP) is as an autologous source of growth factors that has been reported to promote tissue healing. However, PRP contains angiogenic factors such as vascular endothelial growth factor (VEGF) that are known to inhibit chondrogenesis and promote osteoarthritis progression. Mesenchymal stem cells (MSCs) have also shown promise in promoting focal cartilage repair, but MSC therapies typically require cell isolation and expansion before implantation in a second surgery. Adipose-derived stromal va

Softening Substrates Promote Chondrocytes Phenotype via RhoA/ROCK Pathway.

Due to its evascular, aneural, and alymphatic conditions, articular cartilage shows extremely poor regenerative ability. Thus, directing chondrocyte toward a desired location and function by utilizing the mechanical cues of biomaterials is a promising approach for effective tissue regeneration. However, chondrocytes cultured on Petri dish will lose their typical phenotype which may lead to compromised results. Therefore, we fabricated polydimethylsiloxane (PDMS) materials with various stiffness as culture substrates. Cell morphology and focal adhesion of chondrocytes displayed significant chan

Intra-Articular Injections of Mesenchymal Stem Cell Exosomes and Hyaluronic Acid Improve Structural and Mechanical Properties of Repaired Cartilage in a Rabbit Model.

PURPOSE: To compare the efficacy of mesenchymal stem cell (MSC) exosomes with hyaluronic acid (HA) against HA alone for functional cartilage regeneration in a rabbit osteochondral defect model. METHODS: Critical-size osteochondral defects (4.5-mm diameter and 1.5-mm depth) were created on the trochlear grooves in the knees of 18 rabbits and were randomly allocated to 2 treatment groups: (1) exosomes and HA combination and (2) HA alone. Three 1-mL injections of either exosomes and HA or HA alone were administered intra-articularly immediately after surgery and thereafter at 7 and 14 days after

Cricoarytenoid Articulation in Elderly Japanese With Special Reference to Morphology of the Synovial Tissue.

OBJECTIVE: To clarify composite fibers and cells in the synovial tissues of the cricoarytenoid joint (CA joint). METHODS: Routine histology and immunohistrochemistry using sagittal or nearly sagittal sections obtained from 18 elderly cadaveric specimens. RESULTS: The CA joint capsule was thin and contained few elastic fibers. A limited supportive ligament, namely, a thickened fascia of the posterior cricoarytenoid muscles, was sometimes evident on the lateral aspect of the CA joint. However, even in the weaker medial aspect of the joint, no marked destruction of the synovial tissues was foun

Digital Volume Correlation for large deformations of soft tissues: Pipeline and proof of concept for the application to breast ex vivo deformations.

Being able to reposition tumors from prone imaging to supine surgery stances is key for bypassing current invasive marking used for conservative breast surgery. This study aims to demonstrate the feasibility of using Digital Volume Correlation (DVC) to measure the deformation of a female quarter thorax between two different body positioning when subjected to gravity. A segmented multipart mesh (bones, cartilage and tissue) was constructed and a three-step FE-based DVC procedure with heterogeneous elastic regularization was implemented. With the proposed framework, the large displacement field

Soft Hydrogel Environments that Facilitate Cell Spreading and Aggregation Preferentially Support Chondrogenesis of Adult Stem Cells.

Mesenchymal stem/stromal cells (MSCs) represent a promising cell type for treating damaged synovial joints. The therapeutic potential of MSCs will be facilitated by the engineering of biomaterial environments capable of directing their fate. Here the interplay between matrix elasticity and cell morphology in regulating the chondrogenic differentiation of MSCs when seeded onto or encapsulated within hydrogels made of interpenetrating networks (IPN) of alginate and collagen type I is explored. This IPN system enables the independent control of substrate stiffness (in 2D and in 3D) and cell morph

Micromechanical characterisation of osteoarthritic subchondral bone by micropillar compression.

Osteoarthritis (OA) is a multifaceted joint disease which poses significant socioeconomic burdens and remains a significant clinical challenge. Evidence suggests that structural and mechanical changes in subchondral bone influence the pathogenesis and development of OA, leading to diminished bone quality and cartilage degeneration. While changes in microstructure and tissue scale elastic properties are well reported, the tissue yield response of subchondral bone in OA and their correlation with compositional changes have not been investigated. Here, we performed quasistatic micropillar compres

An unusual cause of conductive hearing loss: bilateral complete meatal obstruction following otoplasty.

There are many causes of conductive hearing loss. Otoplasty is a commonly performed cosmetic surgery to correction for prominent ear. We discussed an unusual cause of conductive hearing loss, bilateral complete meatal obstruction following otoplasty, in this article. This complication was probably due to unsuitable placement of the Furnas sutures. In the literature, as a cause of conductive hearing loss, unilateral complete meatal obstruction has been described rarely, but bilateral complete obstruction has not been defined. Hearing loss recovered completely after revision surgery. Correct dia

A stiffness-tunable 3D alginate-collagen-hyaluronic acid model reveals the matrix-dependent chondroprotection of quercetin in osteoarthritis.

Current clinical management of osteoarthritis (OA) follows a stepwise, symptom-oriented approach that provides analgesia but exhibits limited disease-modifying efficacy. Quercetin (QUE), a natural flavonoid, possesses broad anti-inflammatory and antioxidant properties. However, its chondroprotective capacity against early chondrocyte degenerative injury triggered by inflammatory cytokine and abnormal matrix stiffness, alongside the relevant molecular mechanisms, remain incompletely defined. Existing in vitro OA chondrocyte models, predominantly based on 2D cultures with interleukin-1β (IL-1β)

Structure-function of cartilage in osteoarthritis: An ex-vivo correlation analysis between its structural, viscoelastic and frictional properties.

Healthy articular cartilage is characterized by extremely low friction and high compressive stiffness. This dual-functionality is tailored by its biphasic structure, whereby a fluid phase interacts with the extracellular matrix. Osteoarthritis (OA) causes structural changes, thereby altering the biomechanical and frictional properties. How the structural and functional properties of human cartilage are associated with OA remain unknown. To address this, we identified relationships between structural parameters, viscoelastic and frictional properties of degenerated human cartilage through corre

Mechanical behavior of bovine nasal cartilage under static and dynamic loading.

Abnormal mechanical loading may trigger cartilage degeneration associated with osteoarthritis. Tissue response to load has been the subject of several in vitro studies. However, simple stimuli were often applied, not fully mimicking the complex in vivo conditions. Therefore, a rolling/plowing explant test system (RPETS) was developed to replicate the combined in vivo loading patterns. In this work we investigated the mechanical behavior of bovine nasal septum (BNS) cartilage, selected as tissue approximation for experiments with RPETS, under static and dynamic loading. Biphasic material proper

Failure in articular cartilage: Finite element predictions of stress, strain, and pressure under micro-indentation induced fracture.

Articular cartilage is found at the distal end of long bones and is responsible for assisting in joint articulation. While articular cartilage has remarkable resistance to failure, once initially damaged, degeneration is nearly irreversible. Thus, understanding damage initiation is important. There are a few proposed mechanisms for articular cartilage failure initiation: (A) a single collagen fibril stress-based regime; (B) a rate-dependent regime captured by brittle failure at slow displacement rates (SDR) and ductile failure at fast displacement rates (FDR); and (C) a rate-dependent regime w
