Dihydrofolate Reductase Is a Valid Target for Antifungal Development in the Human Pathogen Candida albicans.

While the folate biosynthetic pathway has provided a rich source of antibacterial, antiprotozoal, and anticancer therapies, it has not yet been exploited to develop uniquely antifungal agents. Although there have been attempts to develop fungal-specific inhibitors of dihydrofolate reductase (DHFR), the protein itself has not been unequivocally validated as essential for fungal growth or virulence. The purpose of this study was to establish dihydrofolate reductase as a valid antifungal target. Using a strain with doxycycline-repressible transcription of DFR1 (PTETO-DFR1 strain), we were able to

Protein polymers proliferate: polymerization of phosphatases and proteases.

UNLABELLED: It has been known for more than half a century that enzymes can polymerize to either activate or repress their catalytic functions. Using electron cryo-microscopy (cryo-EM), we serendipitously found that two commonly used enzymes in molecular microbiology research, alkaline phosphatase and proteinase K, can unexpectedly form filaments. While the physiological significance of fibrillation in alkaline phosphatase and proteinase K is unknown, the atomic models of these polymers show the ability of cryo-EM to discover and determine high-resolution structures of macromolecular assemblie

A plant DNMT3 induces non-CG methylation and affects gene expression in human cells.

CG methylation is the predominant form of DNA methylation across eukaryotes, whereas non-CG methylation in vertebrates is largely restricted to neurons and embryonic stem cells and is mediated by DNMT3 enzymes. In contrast, plant DNMT3 homologs catalyze extensive non-CG methylation across diverse tissues. To test whether a plant DNMT3 can establish non-CG methylation and influence transcription in human cells, we expressed Physcomitrium patens DNMT3b (PpDNMT3) in HEK293 cells, which lack this modification. PpDNMT3 induced robust, genome-wide non-CG methylation in transiently and stably transfe

Placental alkaline phosphatase in nonmalignant human cervix.

At least three loci determine human alkaline phosphatases [orthophosphoric-monoester phosphohydrolase (alkaline optimum), EC 3.1.3.1]: one coding for the placental form of the enzyme; at least one coding for the intestinal forms; and at least one for the liver, bone, and kidney forms. It is generally believed that the locus determining the placental form is, in the normal individual, expressed only in placenta. However, ectopic or aberrant expression of this locus occurs in certain malignancies of other tissues and in cell lines, such as HeLa, derived from malignancies. We have examined by the

Ciprofibrate represses alpha 2u-globulin expression in liver and inhibits d-limonene nephrotoxicity.

Peroxisome proliferators are a class of compounds which induce hepatomegaly and peroxisome proliferation in liver parenchymal cells. One of the earliest known effects of peroxisome proliferators is the rapid transcriptional activation of the genes responsible for the peroxisomal beta-oxidation system in liver. Long term administration of these chemicals to rats and mice results in the development of hepatocellular carcinomas. Here we report that mRNA for alpha 2u-globulin, a rodent male specific protein, is markedly reduced or undetectable by Northern blot analysis of total RNA in the livers o

Immunological characterization of oncofetal alkaline phosphatases from human placenta and HeLa(71) cells.

Human placental alkaline phosphatase (AP) exhibits a genetic polymorphism that is determined by the genotype of the fetus. A similar enzyme is produced by certain neoplasms and by some cells in culture. The relationships between these enzymes were investigated by immunological methods comparing human placental AP to the AP of HeLa(71) cells. Antiserum prepared against placental AP precipitated 90 percent of the enzyme and the catalytic activity was quantitatively recovered in the antigen-antibody precipitate. Enzyme-antibody complexes failed to migrate on starch gel electrophoresis, Antiserum

Identification of Lactobacillus strains from human mother milk and cottage cheese revealed potential probiotic properties with enzymatic activity.

The main attempt of this study is to isolate, determine potential probiotic properties and enzyme production of some lactic acid bacteria (LAB). Among all isolates, two LAB strains isolated from human mother milk and cottage cheese revealed antimicrobial activity against some tested pathogenic strains. Both isolates inhibited all the tested pathogens except Escherichia coli. The two isolates were identified by morphological, biochemical properties and then by 16S rRNA gene sequencing technique as Lactobacillus acidophilus SAM1 and Lactiplantibacillus plantarum SAM2. Potential probiotic charact

Role of HIF1α in FXR-mediated protection and anti-inflammatory macrophage polarization in parenteral nutrition-associated cholestasis.

Parenteral nutrition-associated cholestasis (PNAC) arises from suppressed FXR signaling and macrophage activation driven by intestinal LPS and PN-derived phytosterols. We investigated whether tissue hypoxia and HIF1α signaling play a role in FXR-mediated protection in PNAC. PNAC mouse intrahepatic mononuclear cells (IHMCs) displayed evidence of hypoxia and upregulation of HIF1α and hypoxia-responsive genes, also demonstrated in livers from human infants with PNAC. Myeloid-specific Hif1a knock-out reduced liver injury, decreased macrophage infiltration, restored FXR signaling, and promoted anti

Regulation of a catabolic enzyme whose substrate is always available.

Certain hydrogenases are capable of converting the trace amounts of molecular hydrogen constantly present in Earth's atmosphere to supply cells with a little energy during periods of starvation. Recently, Kropp and colleagues investigated the regulation of one of these "high-affinity" hydrogenases in response to different growth conditions for the ubiquitous soil bacterium Mycobacterium smegmatis (A. Kropp, J. D. Archer, M. Jespersen, T. D. Watts, et al., mSystems 11:e01678-25, 2026, https://doi.org/10.1128/msystems.01678-25). A mutation in the gylR gene, whose product acts as a positive trans

Improving erectile function in diabetic male mice by rescuing depalmitoylated FBP1 to reduce cavernosal lactate.

The high prevalence of diabetes mellitus-induced erectile dysfunction (DMED) in males and the limited efficacy of existing therapies necessitate deeper mechanistic insights. We identify pathological lactate accumulation in the corpus cavernosum as a major driver of DMED in male mice, promoting structural degeneration and smooth muscle cell loss. Mechanistically, transcriptional repression and functional impairment of fructose-1,6-bisphosphatase 1 (Fbp1), a rate-limiting gluconeogenic enzyme, underlie this lactate accumulation. Epigenetic profiling revealed suppression of Fbp1 transcription ass

MicroRNAs regulate renal tubule maturation through modulation of Pkd1.

MicroRNAs (miRNAs) contribute to the regulation of early kidney development, but their role during later stages of renal tubule maturation is not well understood. Here, we found that ablation of the miRNA-processing enzyme Dicer from maturing renal tubules produces tubular and glomerular cysts in mice. Inactivation of Dicer is associated with downregulation of miR-200, a kidney-enriched miRNA family, and upregulation of the polycystic kidney disease gene Pkd1. Inhibition of miR-200 in cultured renal epithelial cells disrupted tubulogenesis and led to upregulation of Pkd1. Using bioinformatic a

Podocyte-specific loss of functional microRNAs leads to rapid glomerular and tubular injury.

MicroRNAs (miRNAs) are in a class of endogenous, small, noncoding RNAs that exert their effects through posttranscriptional repression of specific target mRNAs. Although miRNAs have been implicated in the regulation of diverse biologic processes, little is known about miRNA function in the kidney. Here, mice lacking functional miRNAs in the developing podocyte were generated through podocyte-specific knockout of Dicer, an enzyme required for the production of mature miRNAs (Nphs2-Cre; Dicer(flx/flx)). Podocyte-specific loss of miRNAs resulted in significant proteinuria by 2 wk after birth, rap

Decreased IRS-2 and increased SREBP-1c lead to mixed insulin resistance and sensitivity in livers of lipodystrophic and ob/ob mice.

In mice with too little fat (lipodystrophy) or too much fat (ob/ob), leptin deficiency leads to hyperglycemia, hyperinsulinemia, and insulin resistance. In both disorders, the liver overproduces glucose as a result of resistance to the normal action of insulin in repressing mRNAs for gluconeogenic enzymes. Here we show that chronic hyperinsulinemia downregulates the mRNA for IRS-2, an essential component of the insulin-signaling pathway in liver, thereby producing insulin resistance. Despite IRS-2 deficiency, insulin continues to stimulate production of SREBP-1c, a transcription factor that ac

Multiple RNA- and DNA-binding proteins exhibit direct transfer of polynucleotides with implications for target-site search.

We previously demonstrated that the polycomb repressive complex 2 chromatin-modifying enzyme can directly transfer between RNA and DNA without a free-enzyme intermediate state. Simulations suggested that such a direct transfer mechanism may be generally necessary for RNA to recruit proteins to chromatin, but the prevalence of direct transfer capability is unknown. Herein, we used fluorescence polarization assays and observed direct transfer for several well-characterized nucleic acid-binding proteins: three-prime repair exonuclease 1, heterogeneous nuclear ribonucleoprotein U, Fem-3-binding fa

Epigenetic Heterogeneity of B-Cell Lymphoma: Chromatin Modifiers.

We systematically studied the expression of more than fifty histone and DNA (de)methylating enzymes in lymphoma and healthy controls. As a main result, we found that the expression levels of nearly all enzymes become markedly disturbed in lymphoma, suggesting deregulation of large parts of the epigenetic machinery. We discuss the effect of DNA promoter methylation and of transcriptional activity in the context of mutated epigenetic modifiers such as EZH2 and MLL2. As another mechanism, we studied the coupling between the energy metabolism and epigenetics via metabolites that act as cofactors o

A time-course study of long term over-expression of ARR19 in mice.

A leucine-rich protein, ARR19 (androgen receptor corepressor-19 kDa), is highly expressed in male reproductive organs and moderately in others. Previously, we have reported that ARR19 is differentially expressed in adult Leydig cells during the testis development and inhibits steroidogenesis by reducing the expression of steroidogenic enzymes. Whereas in prostate, ARR19 represses the transcriptional activity of AR (androgen receptor), it is important for male sexual differentiation and maturation in prostate and epididymis, through the recruitment of HDAC4. In this study we show that long term

Regulation of sulfate assimilation in Physcomitrella patens: mosses are different!

Sulfur is an essential nutrient, taken up as sulfate from soil, reduced and incorporated into bioorganic compounds in plant cells. The pathway of sulfate assimilation is highly regulated in a demand-driven manner in seed plants. To test the evolutionary conservation of the regulatory mechanisms, we analyzed regulation of the pathway in the model for basal plants, the moss Physcomitrella patens. While in Arabidopsis the key enzyme of sulfate assimilation, adenosine 5'-phosphosulfate reductase (APR), is feedback repressed by thiols and induced by reduced levels of glutathione, in P. patens such

Distinct pulmonary and hepatic forms of flavin-containing monooxygenase in sheep.

1. Flavin-containing monooxygenase (FMO) in pulmonary and hepatic microsomes from sheep was analyzed by western blotting by probing with antibodies raised against FMO purified from rabbit lung and pig liver. 2. Pulmonary microsomes from sheep contain a single major protein which cross-reacts with the antibody to rabbit lung FMO, but no band can be observed when probed with the antibody to the pig liver enzyme. Likewise, sheep liver microsomes contain a protein which cross-reacts with the antibody to pig liver FMO, but no significant staining is observed following incubation with antibody to th

N-6-Methyladenosine in Vasoactive microRNAs during Hypoxia; A Novel Role for METTL4.

N-6-methyladenosine (m6A) is the most prevalent post-transcriptional RNA modification in eukaryotic cells. The modification is reversible and can be dynamically regulated by writer and eraser enzymes. Alteration in the levels of these enzymes can lead to changes in mRNA stability, alternative splicing or microRNA processing, depending on the m6A-binding proteins. Dynamic regulation of mRNA m6A methylation after ischemia and hypoxia influences mRNA stability, alternative splicing and translation, contributing to heart failure. In this study, we studied vasoactive microRNA m6A methylation in fib

Ovine arylalkylamine N-acetyltransferase in the pineal and pituitary glands: differences in function and regulation.

The enzyme arylalkylamine N-acetyltransferase (AANAT; EC 2.3.1.87) has been conventionally linked with the biosynthesis of melatonin within the pineal gland and retina. This study establishes that AANAT messenger RNA (mRNA) and functional enzyme occurs within the pars tuberalis (PT) and to a lesser degree within the pars distalis (PD) of the sheep pituitary gland; expression in these tissues is approximately 1/15th (PT) and 1/300th (PD) of that in the ovine pineal gland. AANAT mRNA in the PT appears to be expressed in the same cells as the Mel1a receptor. No evidence was obtained to indicate t
