Selective turn-on fluorescent sensing of endogenous glutamate using N-CQDs/Al nanocomposites in zebrafish CNS.

L-Glutamate is the primary excitatory neurotransmitter in the central nervous system, and its dysregulation is associated with numerous neurological disorders, underscoring the importance of sensitive, real-time detection in living organisms. In this study, we created a biocompatible fluorescent nanosensor using a nitrogen-doped carbon quantum dot aluminium ion composite (N-CQDs/Al) to selectively detect L-glutamate through a straightforward fluorescence "off-on" process. The N-CQDs and the complex were thoroughly analysed with DLS, TEM, FT-IR, EDX, UV-Vis, and fluorescence spectroscopy, confi

A quantitative identification fluorescent probe for highly selective detection of Cu2+ in food and environmental samples.

The potential accumulation of copper ions (Cu2+) in the food chain poses significant risks, making it imperative to develop efficient tools for real-time monitoring of Cu2+ levels in food, plants, and biological systems. In this study, two novel fluorescent probes, RC-1 and RC-2, were synthesized using triphenylamine derivatives through a condensation reaction for the detection of Cu2+. Among the two probes, probe RC-2, which contains 4-aminobenzamide, exhibited excellent selectivity, ultra-fast response (<30 s), a low detection limit of 26.8 nM, high water solubility, and ease of synthesis. Development of an "Off-On" Fluorescence Sensing System Based on Nitrogen-Doped Carbon Quantum Dot/Fe³⁺ for Sensitive Detection of Cysteine.

Cysteine, one of the twenty proteinogenic amino acids, is distinguished by its unique sulfhydryl (–SH) functional group, which endows it with distinctive biochemical properties. In this work, nitrogen-doped carbon quantum dots (N-CQDs) were synthesized through a microwave-assisted strategy, with citric acid serving as the carbon source and urea as the nitrogen source. The as-prepared N-CQDs were employed for the detection of cysteine in liquid samples via an “off-on” fluorescence signaling mechanism. The fluorescence of N-CQDs was significantly quenched by Fe³⁺, whereas the subsequent addition

A single fluorescent probe for simultaneous detection of Zn2+, Cd2+, and Pb2+ in industrial wastewater with application in bio-imaging and molecular logic gates.

Detecting multiple metal ions, especially those from the same periodic group, is quite challenging with a single fluorophore-based sensing method because of their similar chemical properties. We introduce a unique fluorescent probe that can identify Zn2+, Cd2+, and Pb2+ simultaneously from their mixture, even in the presence of other metal ions, with the first two being in the same group of the periodic table. The design of the probe (L1) is based on the selective binding of Zn2+ or Cd2+, which triggers the fluorescence response of the phenolic aldehyde chromophore moiety, changing it from the

A Review on Pyridine Based Colorimetric and Fluorometric Chemosensor for Detection of Hg2+ ion.

Pyridine, N-containing heterocyclic organic compound, displays strong coordination capabilities with various metal ions. It can be synthesized through various methods, such as Friedlander synthesis, heterocumulene synthesis, cross-coupling reactions, the Radziszewski reaction, Bonnemann cyclization, as well as cobalt-catalyzed synthesis. Experimental and spectroscopic analyses have demonstrated a strong binding affinity between pyridine and several heavy metal ions, including Pb2+, Hg2+, and Cd2+ ions. The escalating environmental pollution caused by the disposal of heavy metal ions in rivers,

Simple Way to Fabricate Chiral Fluorescent Molecules and Chirality-Dependent Biomolecule Recognition and Cell Imaging.

Employing chiral fluorescent molecules to enable a visual communication with bio-objects (enzymes, signaling molecules, and living cells, etc.) is meaningful but extremely challenging, and the fabrication of the chirality concept into the molecule's architecture is a great obstacle for scientists. In this study, two chiral compounds (TPA-PyrT@CSAD and TPA-PyrT@CSAL) were obtained using an anion replacement strategy, which shares the same fluorescent skeleton of TPA-PyrT and the chiral anion of camphor sulfonic salt (CAS) with opposite configurations as D or L. First, the chirality-dependent re

A Dual-Modal NIR-FLIM Probe for Hypothalamic Norepinephrine Imaging in Neurogenic Hypertension.

Hypothalamic norepinephrine (NE) dysregulation drives hypertensive pathogenesis, yet lacks precise in vivo quantitative monitoring tools. We engineered CyQs probes integrating near-infrared (NIR) intensity and fluorescence lifetime imaging (FLIM) for spatiotemporal NE tracking. CyQ-2 employs a hemicyanine scaffold conjugated to an S-phenyl carbonate moiety, enabling 16-fold fluorescence turn-on, 0.21 μM detection limit, and 0.8 ns lifetime extension upon NE-specific activation via nucleophilic cyclization. CyQ-2 mapped endogenous NE dynamics in PC12 cells and the K+-evoked neurotransmitter rel

Near-infrared phenothiazine-fused rhodol with large Stokes shift for fluorogenic imaging of butyrylcholinesterase in vivo.

Butyrylcholinesterase (BChE) is a serine hydrolase that plays diverse roles in physiological and pathological processes, and imaging BChE activity is of great importance for disease diagnosis and drug development. Here we develop a new activatable near-infrared (NIR) fluorescent probe, DR760, for high contrast imaging of BChE activities in vivo. A new NIR fluorophore with a large Stokes shift is engineered by fusing phenothiazine with rhodol. We further develop a fluorogenic probe for BChE by caging the hydroxyl group with a cyclopropyl ester moiety. It is shown that BChE-mediated release of t

Harnessing a MMP-sensitive fluorescent probe for in-situ and reversible monitoring MMP alteration in dual color based on aggregation/monomer mechanism.

BACKGROUND: Mitochondria are highly dynamic organelles which exhibit multiple functional states. The mitochondrial membrane potential (MMP) plays a vital role in sustaining mitochondrial functions and serves as an indicator of cellular health and viability. So, monitoring of MMP variations in real time is vital for gaining insights into mitochondria-associated biological events. The direct dual-color visualization of MMP alteration could decrease errors resulting from non-uniform staining and instrumentation inconsistencies. Nevertheless, achieving in-situ and direct detection of dynamic MMP c

An Ultraresponsive Green Biosensor for Robust in Vivo Imaging of Synaptic Zinc Dynamics.

Despite its crucial role in neurotransmission and neural processing, direct in vivo measurement of Zn2+ transients has been hindered by the limited responsiveness and stability of available fluorescent indicators. Here, we report hpGRISZ (high-performance green indicator for synaptic Zn2+), an ultraresponsive, thermostable, turn-on green fluorescent biosensor engineered for high brightness, large dynamic range, and affinity tuned for detecting extracellular synaptic Zn2+ release. We comprehensively characterized hpGRISZ in vitro, in mammalian cells, and in vivo. A membrane-anchored version of

A novel borondifluoro indolenine-functionalized red-shift ratiometric fluorescent probe for detection of hypochlorous acid in drug-induced liver injury.

The diagnosis of drug-induced liver injury (DILI) remains a significant clinical challenge due to the lack of reliable tools for visualizing related biomarkers in vivo. Hypochlorous acid (HClO), a key indicator of oxidative stress, has been demonstrated to be closely linked to the onset and progression of DILI. Herein, to address the limitations of conventional HClO/ClO- detection, we developed a D-π-A architecture probe BFI-Naph through rational molecular engineering of a borondifluoro indolenine (BFI) core and a naphthol group. The probe exhibits a rapid (∼30 s), sensitive (LOD = 120 nM), la

Dual-channel fluorescent probe for in-situ differential detection of HClO and H2S in crops and food.

Hypochlorous acid (HClO) and Hydrogen sulfide (H2S) are vital redox regulators in crops, yet their imbalance can trigger oxidative stress. Simultaneous detection of both is challenging with conventional single-target probes. To address this, we engineered a dual-functional probe, ZZ, based on a coumarin-quinolinium structure. It selectively identifies HClO and H2S via distinct fluorescence channels: blue emission for HClO and near-infrared (NIR) for H2S. Additionally, it shows a visible colour change from yellow to colorless or red upon exposure to HClO or H2S, respectively, enabling both fluo

An integrated experimental and computational study on a COX-2-targeted AIE probe for precise cancer cell imaging via restriction of intramolecular rotation.

BACKGROUND: The overexpression of cyclooxygenase-2 (COX-2) and alterations in microenvironmental viscosity are recognized as pivotal hallmarks for the early diagnosis and progression of malignant tumors. Although fluorescent probes have been developed for tumor imaging, many conventional fluorophores suffer from aggregation-caused quenching (ACQ) in dense biological environments. Furthermore, existing sensors often lack the specificity to simultaneously target COX-2 and dynamically report local viscosity variations within complex cellular environments. Therefore, developing a robust molecular

Illuminating epilepsy: A deep NIR-I fluorogenic sensor reports peroxynitrite dynamics in vivo.

This work presents BDP-NIR-S-Fa, a deep NIR-I fluorogenic probe designed to report peroxynitrite (ONOO-) dynamics in epilepsy. The probe features a specific emission at 805 nm and responds rapidly and sensitively to ONOO-. It successfully visualized elevated endogenous ONOO- flux in both kainic acid-induced epileptic cells and a corresponding mouse model, demonstrating its potential as a diagnostic tool. Beyond detection, BDP-NIR-S-Fa was further applied to evaluate the therapeutic efficacy of antiepileptic drugs by monitoring the reduction of ONOO- levels in treated disease models. The probe

A dual-signal near-infrared fluorescent probe for concurrent detection of Hg²⁺ and ONOO⁻: Applications from environmental monitoring to imaging of Hg²⁺-induced oxidative stress.

As a highly toxic heavy metal, Hg²⁺ can accumulate in the human body through the food chain, causing multi-system damage, and induce the generation of peroxynitrite (ONOO⁻). The imbalance of ONOO⁻ is associated with various diseases, so it is necessary to accurately monitor both of them and the Hg²⁺-induced oxidative stress state in biological systems. Near-infrared (NIR) fluorescent probes are ideal tools, but most existing probes only detect Hg²⁺ or ONOO⁻ individually, and multifunctional probes are relatively scarce. In this study, a NIR fluorescent probe DCI-2a was developed. It uses dicya

1,2,4-Triazole-Based Excited-State Intramolecular Proton Transfer-Driven "Turn-On" Chemosensor for Selective Cyanide Detection with Test Strip Utility and Molecular Keypad Lock: An Experimental and Computational Exploration.

Cyanide (CN-) is a highly toxic anion with significant environmental and biological implications, necessitating the development of sensitive and selective detection platforms. In this work, we reported the design and synthesis of a novel excited-state intramolecular proton transfer (ESIPT)-based chemosensor (TSB) derived from 2-hydroxy-1-naphthaldehyde and 4-amino-1,2,4-triazole. Computational studies, including time-dependent density functional theory (TD-DFT), reduced density gradient scatter plots, and simulated infrared spectra, reveal the prevalence of strong intramolecular hydrogen bonds

Nitroreductase-Activated Fluorescent Probes: From Molecular Design to Disease Diagnosis.

Nitroreductases (NTRs), a class of flavin-dependent oxidoreductases, are highly upregulated in hypoxic microenvironments and are closely associated with cancer, bacterial infections, and other hypoxia-driven diseases. Precise and real-time visualization of NTR activity is therefore critical for understanding disease mechanisms and enabling accurate diagnosis and targeted therapy. Fluorescent probes have emerged as powerful and versatile chemical biology tools for NTR sensing owing to their high sensitivity, selectivity, and spatiotemporal resolution. Recent advances in two-photon fluorescence

A ratiometric ER calcium sensor for quantitative comparisons across cell types and subcellular regions.

The ER is an important regulator of Ca2+ in cells and dysregulation of ER Ca2+ homeostasis can lead to numerous pathologies. Understanding how various pharmacological and genetic perturbations of ER Ca2+ homeostasis impact cellular physiology would be facilitated by quantitative measurements of ER Ca2+ levels that allow for robust comparisons across conditions. To achieve this, we enhanced our original high dynamic range ER Ca2+ indicator, ER-GCaMP6-150, by fusing it to the HaloTag protein, which when bound to Janelia Fluor (JF) dyes creates a ratiometric ER Ca2+ probe. This probe (ER-Halo-GCa

Multicomponent Synthesis and Investigations Fluorescence Activity of Chromenone-Pyrazole Compounds.

A synthetic method is described to produce some chromenone-pyrazole derivatives through a one-pot multicomponent reaction using SrFe12O19 as a magnetic catalyst. This method provides quite a few merits, including the use of an effective and easy separable nanocatalyst, high yields of products, short reaction time, and easy work-up. Two of the products showed fluorescence properties, which have detected mercury ions without any interference with other ions. They can detect a tiny amount of mercury ions, which were comparable with other chemosensors. The detection limit is 4 × 10-7 and 3 × 10-8
