Fumonisin B1 (FB1) is a possible carcinogenic molecule for humans as classified by the International Agency for Research on Cancer (IARC) in 2B group. In livestock, it is responsible for several mycotoxicoses and economic losses. Lactobacillus strains, inhabitants of a wide range of foodstuffs and the gastrointestinal tract, are generally recognized as safe (GRAS). Thus, the aim of this work was to evaluate the protective effect of Lactobacillus paracasei (LP) against FB1-induced reprotoxicities including testicular histopathology, sperm quality disturbance, and testosterone level reduction.Pu

Mycotoxins are fungal secondary metabolites that have been associated with severe toxic effects to vertebrates produced by many important phytopathogenic and food spoilage fungi including Aspergillus, Penicillium, Fusarium, and Alternaria species. The contamination of foods and animal feeds with mycotoxins is a worldwide problem. We reviewed various control strategies to prevent the growth of mycotoxigenic fungi as well as to inhibit mycotoxin biosynthesis including pre-harvest (resistance varieties, field management and the use of biological and chemical agents), harvest management, and post-

Mycotoxin contamination in Platycladi semen has become an increasing concern worldwide due to its significant threat to public health. In this study, a modified QuEChERS-based UPLC-MS/MS method was established to detect multi-mycotoxins in 190 batches of Platycladi semen samples obtained from various channels in China. Out of these samples, 188 tested positive for one or more mycotoxins, with an incidence of 98.95 %. Alternariol monomethyl ether was the most commonly detected with an incidence rate of 92.63 %, followed by fumonisin B2 at 91.58 % and sterigmatocystin at 87.89 %. Notably, the af

Due to the widely occurring co-contamination of mycotoxins in raw food materials, simultaneous monitoring of multiple mycotoxins is needed. Herein, we report the design and fabrication of an electrochemical immunosensor for simultaneous detection of two mycotoxins, fumonisin B1 (FB1) and deoxynivalenol (DON), in a single test. A dual-channel three-electrode electrochemical sensor pattern was etched on a transparent indium tin oxide (ITO)-coated glass via photolithography and was integrated with capillary-driven polydimethylsiloxane (PDMS) microfluidic channel. The two working electrodes were f

ORMDL proteins are essential negative regulators of the serine palmitoyltransferase (SPT) complex, thereby controlling the rate of de novo sphingolipid synthesis. Although mammalian ORMDLs undergo rapid turnover, the mechanisms regulating their stability remain unclear, with conflicting observations across studies. Here, we combined lipidomics, proteomics, and biochemical assays to investigate ORMDL regulation in HEK293, RPE-1, and primary mouse bone marrow-derived mast cells (BMMCs). Inhibition of SPT by myriocin or of ceramide synthases by fumonisin B1 (FB1) profoundly altered sphingolipid c

Mycotoxins have been investigated in relation to a wide range of adverse human health effects, but the evidence for all but a small number of associations is limited. Thus, the full impact on human health of the widespread exposure to mycotoxins remains to be defined. The main exception is for aflatoxins; epidemiological, experimental, and mechanistic studies have contributed to establishing aflatoxins as a cause of human liver cancer, with a particularly elevated risk in people chronically infected with hepatitis B virus. In addition, acute aflatoxicosis after exposure to high dietary toxin l

Aflatoxins and fumonisins (FB) are mycotoxins contaminating a large fraction of the world's food, including maize, cereals, groundnuts and tree nuts. The toxins frequently co-occur in maize. Where these commodities are dietary staples, for example, in parts of Africa, Asia and Latin America, the contamination translates to high-level chronic exposure. This is particularly true in subsistence farming communities where regulations to control exposure are either non-existent or practically unenforceable. Aflatoxins are hepatocarcinogenic in humans, particularly in conjunction with chronic hepatit

A screening of commercial dog and cat foods revealed considerable variation in metabolite profiles across feed categories, with standard feeds generally showing higher concentrations of Fusarium metabolites, phytoestrogens, and plant metabolites than specialized senior or intestinal diets. Concentrations of aflatoxin B1 (AFB1), deoxynivalenol (DON) DON, zearalenone (ZEN), ochratoxin A (OTA), fumonisin B1 + 2 (FB1 + FB2), and T-2/HT-2 toxins remained below existing guidance values and maximum limits. In vitro assays using peripheral blood mononuclear cells (PBMCs) from dogs and cats demonstrate

Extracellular ATP is a purinergic signal with important functions in regulating plant growth and stress-adaptive responses, including programmed cell death. While signalling events proximate to receptor activation at the plasma membrane have been characterised, downstream protein targets and the mechanism of cell death activation/regulation are unknown. We designed a proteomic screen to identify ATP-responsive proteins in Arabidopsis cell cultures exposed to mycotoxin stress via fumonisin B1 (FB1) application. Arabidopsis RIBONUCLEASE 1 (RNS1) was identified by the screen, and transgenic plant

Microbes encounter a broad spectrum of antimicrobial compounds in their environments and often possess metabolic strategies to detoxify such xenobiotics. We have previously shown that Fusarium verticillioides, a fungal pathogen of maize known for its production of fumonisin mycotoxins, possesses two unlinked loci, FDB1 and FDB2, necessary for detoxification of antimicrobial compounds produced by maize, including the γ-lactam 2-benzoxazolinone (BOA). In support of these earlier studies, microarray analysis of F. verticillioides exposed to BOA identified the induction of multiple genes at FDB1 a

Currently there is an urgent need for multi-mycotoxin detection methods due to the co-occurrence of multiple mycotoxins in food raw materials and their augmented toxicity. Herein, a magneto-controlled aptasensor has been developed for simultaneous electrochemical detection of ochratoxin A (OTA) and fumonisin B1 (FB1), two typical mycotoxins found in food crops world-wide. This aptasensor was designed using the high specificity between the target and aptamer with heavy CdTe or PbS quantum dots (QDs) coated silica as labels and the complementary DNA functionalized magnetic beads as capture probe

Ceramide channels formed in the outer membrane of mitochondria have been proposed to be the pathways by which proapoptotic proteins are released from mitochondria during the early stages of apoptosis. We report that sphingosine also forms channels in membranes, but these differ greatly from the large oligomeric barrel-stave channels formed by ceramide. Sphingosine channels have short open lifetimes and have diameters less than 2 nm, whereas ceramide channels have long open lifetimes, enlarge in size reaching diameters in excess of 10 nm. Unlike ceramide, sphingosine forms channels in erythrocy

P-glycoprotein is a cellular efflux pump. The P-glycoprotein inhibitor PSC 833 causes apoptosis of cancer cells and induces a rise in the intracellular levels of ceramide. Our aims were to determine whether a cause and effect relationship exists between these two actions of PSC 833, and to assess whether the PSC 833-induced apoptosis is restricted to transformed cells. Apoptosis was determined by flow cytometry and radioactive quantitation of DNA fragmentation. PSC 833 induced apoptosis in the human T leukemia cell lines: Molt-4 and Jurkat. INTRODUCTION: Gut microbiota are vulnerable to foreign chemicals (xenobiotics) including pharmaceuticals, environmental pollutants, and dietary contaminants such as aflatoxin B1 (AFB1) and fumonisin B1 (FB1). Assessing the effect of these xenobiotics in the laboratory requires their dissolution in a solvent vehicle, such as dimethyl sulfoxide (DMSO). While DMSO is typically used at low concentrations under the assumption of neutrality, its independent impact on microbial dynamics is a potential experimental confounder that has not been fully explored.
METHODS: Human fecal microbiota were cult

Defined fungal starters can improve the controllability of plant-based solid-state fermentation, but candidate strains require strain-level genomic characterization and condition-specific product screening. We characterized an Aspergillus niger starter candidate used for hawk tea (Machilus rehderi) fermentation by whole-genome sequencing, comparative genomics, functional annotation, secondary-metabolite genome mining, and targeted HPLC-FLD analysis. The 35.98-Mb assembly comprised nine contigs and contained 12,409 protein-coding genes. BUSCO analysis with eurotiales_odb12.2 recovered 98.2% com

The prevalence and residue levels of mycotoxins in food have emerged as a global public health concern, exacerbated by climate change. Despite routine monitoring of mycotoxin residues in foodstuffs by health agencies, comprehensive exposure assessments remain limited due to insufficient food consumption data and complex food classification systems. This study employed the Stochastic Human Exposure and Dose Simulation High-Throughput (SHEDS-HT) model, adapted for the Taiwanese population with National Food Consumption Database (NFCD), to estimate aggregate dietary exposure to six major mycotoxi

Sixty-one samples of Portuguese cereal-based foods were analysed for the occurrence of emerging mycotoxins called enniatins (A, A1, B and B1) and beauvericin. Samples were extracted with a mixture of acetonitrile/water (85/15, v/v) using an Ultra-Turrax homogeniser, and mycotoxins were detected with liquid chromatography coupled to a mass spectrometer. This method was validated and adequate values of recovery (70-103%) and relative standard deviation (<15%) were obtained. Signal suppression/enhancement was studied and matrix-matched calibration used to minimise this effect, but no additional c

Programmed cell death via the sphingomyelin pathway has been suggested to underlie heat-shock disturbance of oocyte developmental competence. A series of experiments were performed to characterise the role of the sphingolipid ceramide in heat-shock-induced apoptosis, and to determine whether ceramide formation can be regulated. Bovine cumulus-oocyte complexes (COCs) were aspirated from ovaries collected in the cold season (November-April), in vitro-matured, fertilised and cultured for 8 days. Exposure of COCs to heat shock (41°C) during maturation reduced cleavage rate and blastocyst formation

Sphingoid bases have been implicated in various cellular processes including cell growth, apoptosis and cell differentiation. Here, we show that the regulated turnover of sphingoid bases is crucial for cell polarity development, i.e., the biogenesis of apical plasma membrane domains, in well-differentiated hepatic cells. Thus, inhibition of dihydroceramide synthase or sphinganine kinase activity with fumonisin B1 or N,N-dimethylsphingosine, respectively, dramatically perturbs cell polarity development, which is due to increased levels of sphinganine. Consistently, reduction of free sphinganine
