Misdiagnosis of a fungus ball in the middle ear as chronic suppurative otitis media.

BACKGROUND: Middle ear infections caused by fungi are commonly identified through laboratory tests like fungal smears, culture, and DNA detection. There are two types of ear infections caused by fungi: middle ear fungal infections and fungal otitis media. Both fungal otitis media and fungus balls in the middle ear are increasingly rare. CASE DESCRIPTION: A middle-aged immunocompetent woman suffered from long-term recurrent pus formation in the middle ear and associated hearing loss. Physical examination revealed a perforation in the pars tensa of the tympanic membrane; a pure tone audiometry

Structure, cellular distribution, antigenicity, and biological functions of Fonsecaea pedrosoi ceramide monohexosides.

Monohexosylceramides (CMHs, or cerebrosides) have been reported as membrane and cell wall constituents of both pathogenic and nonpathogenic fungi, presenting remarkable differences in their ceramide moiety compared to mammalian CMHs. Current evidence suggests that CMHs are involved in fungal differentiation and growth and contribute to host immune response. Here we describe a structural diversity between cerebrosides obtained from different forms of the human pathogen Fonsecaea pedrosoi. The major CMH species produced by conidial forms displayed the same structure previously demonstrated by ou

Inhibitors of trehalose-6-phosphate synthase activity in fungal pathogens compromise thermal tolerance pathways.

Infections caused by fungal pathogens such as Candida and Cryptococcus are associated with high mortality rates, partly due to limitations in the current antifungal arsenal. This highlights the need for antifungal drug targets with novel mechanisms of action. The trehalose biosynthesis pathway is a promising antifungal drug target because trehalose biosynthesis is essential for virulence in Cryptococcus neoformans and Candida albicans and is also a mediator of fungal stress responses, such as thermotolerance. To exploit its untapped antifungal potentials, we screened the St. Jude 3-point pharm

Structural Insights into Binding of the Antifungal Drug Fluconazole to Saccharomyces cerevisiae Lanosterol 14α-Demethylase.

Infections by fungal pathogens such as Candida albicans and Aspergillus fumigatus and their resistance to triazole drugs are major concerns. Fungal lanosterol 14α-demethylase belongs to the CYP51 class in the cytochrome P450 superfamily of enzymes. This monospanning bitopic membrane protein is involved in ergosterol biosynthesis and is the primary target of azole antifungal drugs, including fluconazole. The lack of high-resolution structural information for this drug target from fungal pathogens has been a limiting factor for the design of modified triazole drugs that will overcome resistance.

A co-evolved peptide-GPCR system senses host entry to drive fungal infection.

A successful infection requires pathogens to recognize the specific host environment in order to reprogram their physiology accordingly. One major way in which eukaryotic cells sense their surroundings is via G-Protein Coupled Receptors (GPCRs), which share a seven-transmembrane architecture and G-protein-mediated downstream signaling. While mammalian GPCRs are well-characterized and represent important drug targets, their fungal counterparts remain poorly understood. Here we uncover a GPCR-based mechanism that allows the corn pathogen Ustilago maydis to sense whether it has entered plant tiss

Fungal biofilm formation and its regulatory mechanism.

Fungal biofilm is a microbial community composed of fungal cells and extracellular polymeric substances (EPS). In recent years, fungal biofilms have played an increasingly important role in many fields. However, there are few studies on fungal biofilms and their related applications and development are still far from enough. Therefore, this review summarizes the composition and function of EPS in fungal biofilms, and improves and refines the formation process of fungal biofilms according to the latest viewpoints. Moreover, based on the study of Saccharomyces cerevisiae and Candida albicans, th

Succession of diversity, functions, and interactions of the fungal community in activated sludge under aromatic hydrocarbon stress.

Although fungi are regarded as the important degraders of aromatic hydrocarbons (AHs) in various environments, the dynamic succession and interaction of their community under aromatic hydrocarbon stress has been rarely reported. In this study, we systematically investigated the responses of the fungal community and the associations among fungal species when facing the continuous stress of two typical AHs, benzene and naphthalene. Using high-throughput sequencing technology, we demonstrated that fungal diversity displayed a significant downward trend during six weeks of continuous aromatic hydr

Therapeutic sectorial full-thickness sclero-keratoplasty for recurrent fungal keratitis.

OBJECTIVE: To report the management of a severe and recurrent fungal keratitis that required repeated penetrating keratoplasties. Despite multiple topical, intraocular and systemic antifungal treatments, superotemporal hyphal infiltration repeatedly penetrated the corneal transplant causing continuous recurrences. Cultures collected before and during surgery isolated the same organism, Fusarium spp. CONCLUSION: Corneal infection extending to the sclera and internal angle structures is the main cause of recurrence of fungal keratitis after corneal transplantation. Sectorial full-thickness scle

Synthesis and antimicrobial evaluation of some novel sulfonylamido-benzoxazoles.

A series of 2-(p-substituted phenyl)-5-[(4-substituted phenyl) sulfonylamido]-benzoxazoles were synthesized and tested for their antimicrobial activities. The structures of the new derivatives were elucidated by spectral techniques. The minimum inhibitory concentrations (MIC) of the new benzoxazoles were determined against standard bacterial and fungal strains and drug-resistant isolates and compared to those of several reference drugs.

Human-associated microbial inputs and bacterial-fungal ecological coupling shape antibiotic resistance risk in environmental dust.

Environmental dust represents a critical exposure matrix, yet the relationships between multi-kingdom dust microbiomes and antimicrobial resistance (AMR)-associated health risks remain insufficiently characterized. We applied shotgun metagenomics to dust samples from pharmaceutical factories, a dairy farm, railway stations, and schools to comprehensively characterize bacterial, fungal, and viral communities, alongside resistome structure. Microbial community composition exhibited significant differences across all three domains among the sampled environments. Specifically, dust from railway st

Compost application more strongly shapes soil fungal than bacterial communities across arable fields.

Organic amendments such as compost are widely promoted for enriching soil microbial communities and supporting key soil functions, yet their effects across diverse farming systems and pedoclimatic conditions remain poorly understood. Most evidence originates from controlled experiments that cannot account for the variability inherent in real agronomic settings. Here, using data from 56 commercially managed wheat fields under different fertilizer regimes (compost, solid digestate, or manure) across Switzerland, we show that compost application reshapes soil microbial communities under real farm

Diagnosis of fungal respiratory infections in immunocompetent children.

Fungal respiratory infections are rare in immunocompetent children, and their recognition remains challenging since clinical features are often subacute and overlapping with other causes of respiratory disease. A structured diagnostic approach is fundamental to facilitating earlier diagnosis and prompt targeted therapy, thereby improving outcomes. Fungal spores are ubiquitous in the environment and fungal colonisation of skin and mucosal membranes is common, hence differentiation of infection from colonisation is key to an accurate diagnosis and a prudent approach to management. The diagnosis

Crystal structure and transient dimerization for the FKBP12 protein from the pathogenic fungus Candida auris.

International concern over the recent emergence of Candida auris infections reflects not only its comparative ease of transmission and substantial mortality but the increasing level of resistance observed to all three major classes of antifungal drugs. Diminution in virulence has been reported for a wide range of fungal pathogens when the FK506-binding protein FKBP12 binds to that immunosuppressant drug and the binary complex then inhibits the fungal calcineurin signaling pathway. Structure-based drug design efforts have described modifications of FK506 which modestly reduce virulence for a nu
