Altered DNA methylation indicates an oscillatory flow mediated epithelial-to-mesenchymal transition signature in ascending aorta of patients with bicuspid aortic valve.

Disturbed flow has been suggested to contribute to aneurysm susceptibility in bicuspid aortic valve (BAV) patients. Lately, flow has emerged as an important modulator of DNA methylation. Hear we combined global methylation analysis with in vitro studies of flow-sensitive methylation to identify biological processes associated with BAV-aortopathy and the potential contribution of flow. Biopsies from non-dilated and dilated ascending aortas were collected from BAV (n = 21) and tricuspid aortic valve (TAV) patients (n = 23). DNA methylation and gene expression was measured in aortic intima-media

Physical Parameters Influencing the Rate of Subretinal Injection: A Microfluidic Analysis.

PURPOSE: Subretinal injection during vitrectomy has become an important technique owing to the growing number of cell and gene therapy trials for retinal disease. Reducing the injection rate can mitigate trauma or shear forces on the retinal pigment epithelium, yet many surgeons alter the subretinal injection system in ways that could influence this rate. METHODS: We created a theoretical model of a standard subretinal injection system by applying the Hagen-Poiseuille equation, assuming steady, incompressible, laminar flow of a water-like Newtonian fluid. The system consisted of the vitrectom

Altered expression in liver cancer with various hepatic blood flow occlusions: A pilot RNA sequencing study.

BACKGROUND AND AIMS: Hepatocellular carcinoma is one of the most common cancers worldwide. Previous studies have reported he influence of various hepatic blood flow occlusions on tumor behavior, which is mainly mediated by liver ischemia-reperfusion. Although some genes and pathways have been determined, the whole transcriptome after various hepatic blood flow occlusions is lacking. METHODS: We systematically explored transcriptome changes after various hepatic blood flow occlusions, including sham operation (SO; n = 10), occlusion of the portal triad (OPT; n = 10), and occlusion of the porta

Diagnosis of platelet-type von Willebrand disease by flow cytometry.

Platelet-type von Willebrand disease (PT-VWD) is a rare autosomal dominant bleeding disorder which is due to a mutation in the gene encoding for platelet glycoprotein Ibalpha (GPIbalpha) resulting in enhanced affinity for von Willebrand factor (VWF). PT-VWD is often mistakenly diagnosed as type 2B VWD for the similarities between these two conditions. We characterized a new case of PT-VWD and evaluated the usefulness of a flow cytometric assay in the differential diagnosis between PT-VWD (n=1) and type 2B VWD (n=4). The flow cytometric assay was able to highlight the increased affinity of VWF

The origin and domestication of modern cultivated Chrysanthemum × morifolium.

The modern cultivated chrysanthemum (Chrysanthemum × morifolium Ramat.) is a globally important ornamental plant, ranking as the second-largest cut flower after roses. Here, to unravel its complex origin and domestication history, we achieved 6 haplotypes of the elite hexaploidy cut chrysanthemum cultivar 'Jinba' (2n = 6x = 54). By constructing a genome-wide variant map across 147 core accessions, we classified current Chrysanthemum varieties into 4 wild and 4 cultivated groups. We observed extensive, intricate gene flow among these groups, demonstrating that wild germplasms from both China an

Genomic analysis of population structure and target-site mutations of Tetranychus truncatus in China.

BACKGROUND: Tetranychus truncatus is a dominant mite pest in China, yet its population structure and pesticide resistance status remain poorly characterized. To address this, we performed whole-genome resequencing of 176 individuals, integrating them with previous data to analyze 343 individuals from 44 populations. We investigated the population genetic structure, screened for 22 target-site resistance mutations across ten genes, and reconstructed the evolutionary origins of resistance mutations in acetylcholinesterase (AChE). RESULTS: Phylogenetic analysis identified four geography-based cl

Revised taxonomy reveals sustained introgression and secondary contact in ancient lake ricefishes.

BACKGROUND: Biotic diversification in ancient lakes is shaped by complex geological histories and genetic exchange among populations. The Malili Lake system on Sulawesi Island represents a classic natural laboratory for studying freshwater fish evolution and harbors multiple endemic Oryzias species that diversified under repeated hydrological reorganizations. Previous genomic analyses inferred that two sympatric species in Lake Towuti (O. profundicola and O. loxolepis) experienced a single ancient introgression event from a "ghost lineage" derived from O. marmoratus inhabiting another lake. Ho

Digest: Chromosomal architecture of behavioral isolation in marine isopods.

How do reproductive barriers persist when diverging species come back into contact? Ribardière et al. (2026) investigated this question in 2 marine isopods, Jaera albifrons and Jaera praehirsuta, whose males court females using divergent specialized setae. By combining population genomics and controlled crosses, the authors show that genomic regions resisting interspecific gene flow are concentrated on sex chromosomes and rearranged chromosomes-regions of low recombination that harbor the genetic bases of divergent male courtship traits.

Landscape genetics of Cochliomyia hominivorax (Diptera: Calliphoridae): predicting dispersal pathways and expansion potential after reintroduction.

The New World screwworm fly, Cochliomyia hominivorax (Coquerel, 1858) (Diptera: Calliphoridae), is an obligate parasitic blowfly that causes traumatic myiasis and represents a major threat to livestock production in the Neotropics. Although eradication programs based on the Sterile Insect Technique (SIT) successfully eliminated the species from North and Central America, the risk of reinvasion remains a persistent concern. Understanding how landscape structure influences dispersal and connectivity is therefore essential for improving surveillance and control strategies. We evaluated the influe

Label-free high-throughput live-cell sorting of genome-wide random mutagenesis libraries for metabolic traits by Raman flow cytometry.

A full spontaneous single-cell Raman spectrum captures the metabolic phenome in a label-free and noninvasive manner. However, Raman-activated cell sorting (RACS) of rare target cells from highly heterogeneous systems has remained largely conceptual. Here, we present a positive dielectrophoresis-induced deterministic lateral displacement (pDEP-DLD)-based RACS (pDEP-DLD-RACS), in which a modulated pDEP-DLD force is applied to focus, trap, and functionally sort fast-moving single cells in a wide channel. For pigment- and oil-producing yeasts, pDEP-DLD-RACS shows high sorting accuracy (>90%), high

Detecting Introgression in Shallow Phylogenies: How Minor Molecular Clock Deviations Lead to Major Inference Errors.

Recent theoretical and algorithmic advances in introgression detection, coupled with the growing availability of genome-scale data, have highlighted the widespread occurrence of interspecific gene flow across the tree of life. However, current methods largely depend on the molecular clock assumption-a questionable premise given empirical evidence of substitution rate variation across lineages. While such rate heterogeneity is known to compromise gene flow detection among divergent lineages, its impact on closely related taxa at shallow evolutionary timescales remains poorly understood, likely

Genetic architecture and evolution of color variation in American black bears.

Color variation is a frequent evolutionary substrate for camouflage in small mammals, but the underlying genetics and evolutionary forces that drive color variation in natural populations of large mammals are mostly unexplained. The American black bear, Ursus americanus (U. americanus), exhibits a range of colors including the cinnamon morph, which has a similar color to the brown bear, U. arctos, and is found at high frequency in the American southwest. Reflectance and chemical melanin measurements showed little distinction between U. arctos and cinnamon U. americanus individuals. We used a g

The population genomic structure of green turtles (Chelonia mydas) suggests a warm-water corridor for tropical marine fauna between the Atlantic and Indian oceans during the last interglacial.

The occasional westward transport of warm water of the Agulhas Current, "Agulhas leakage", around southern Africa has been suggested to facilitate tropical marine connectivity between the Atlantic and Indian oceans, but the "Agulhas leakage" hypothesis does not explain the signatures of eastward gene flow observed in many tropical marine fauna. We investigated an alternative hypothesis: the establishment of a warm-water corridor during comparatively warm interglacial periods. The "warm-water corridor" hypothesis was investigated by studying the population genomic structure of Atlantic and Sout

Pleistocene genetic legacy suggests incipient species of Sebastes mentella in the Irminger Sea.

To investigate a possible speciation event within the redfish (Sebastes mentella) complex in the Irminger Sea, we examined genetics, traditional morphology, geometric morphometrics and meristics of individuals sampled throughout the Sea. Tissue samples from 1901 fish were collected in 1995 and 1996 and from 1999 to 2002, and the fish were genotyped at nine microsatellite loci, two of which were developed for this study. Individual-based genetic analyses showed that two different gene pools exist in the Irminger Sea. Although these groups overlap extensively geographically, they segregate accor

The influence of Pleistocene climatic changes and ocean currents on the phylogeography of the southern African barnacle, Tetraclita serrata (Thoracica; Cirripedia).

The evolutionary effects of glacial periods are poorly understood for Southern Hemisphere marine intertidal species, particularly obligatory sessile organisms. We examined this by assessing the phylogeographic patterns of the southern African volcano barnacle, Tetraclita serrata, a dominant species on rocky intertidal shores. Restricted gene flow in some geographical areas was hypothesized based on oceanic circulation patterns and known biogeographic regions. Barnacle population genetic structure was investigated using the mitochondrial cytochrome oxidase subunit 1 (COI) region for 410 individ

Jomon genomics reveal cold adaptation in Upper Paleolithic hunter-gatherers of eastern Eurasia.

How did Homo sapiens thrive in extreme environments during their dispersal across Eurasia? Unraveling adaptations that enabled human survival during the Last Glacial Maximum (LGM) remains critical. Here, we analyze 42 genomes from Holocene hunter-gatherers (Jomon) in the Japanese archipelago, revealing their deep ancestry from an isolated Upper Paleolithic (UP) East Eurasian lineage. We detect gene flow from northern UP Siberians into Jomon, reconciling long-standing debates between bioanthropology and archaeology regarding East Asian peopling. We find no Jomon-specific Denisovan ancestry, sug

Comparative population genetic structure of the mitochondrial 16S ribosomal RNA gene of Haemaphysalis flava (Acari: Ixodidae) between Nantong, China, and regions along the East Asia-Australasian Flyway.

Haemaphysalis flava Neumann, 1897 is recognized as a vector for Dabie bandavirus (severe fever with thrombocytopenia syndrome virus, SFTSV), which is the etiological agent of severe fever with thrombocytopenia syndrome (SFTS). The tick vector has proliferated and expanded globally in recent years, likely due to the migration of birds. In this study, we selected the mitochondrial 16S rRNA as the genetic marker to analyze the population genetics of H. flava collected from Nantong, China and other regions separated by geographical barriers, all of which are located along the East Asia-Australasia

Positive selection alone is sufficient for whole genome differentiation at the early stage of speciation process in the fall armyworm.

BACKGROUND: The process of speciation involves differentiation of whole genome sequences between a pair of diverging taxa. In the absence of a geographic barrier and in the presence of gene flow, genomic differentiation may occur when the homogenizing effect of recombination is overcome across the whole genome. The fall armyworm is observed as two sympatric strains with different host-plant preferences across the entire habitat. These two strains exhibit a very low level of genetic differentiation across the whole genome, suggesting that genomic differentiation occurred at an early stage of sp

SMAD6 transduces endothelial cell flow responses required for blood vessel homeostasis.

Fluid shear stress provided by blood flow instigates a transition from active blood vessel network expansion during development, to vascular homeostasis and quiescence that is important for mature blood vessel function. Here we show that SMAD6 is required for endothelial cell flow-mediated responses leading to maintenance of vascular homeostasis. Concomitant manipulation of the mechanosensor Notch1 pathway and SMAD6 expression levels revealed that SMAD6 functions downstream of ligand-induced Notch signaling and transcription regulation. Mechanistically, full-length SMAD6 protein was needed to

Multimodality imaging of myocardial injury and coronary microvascular dysfunction in congenital thrombotic thrombocytopenic purpura.

Congenital thrombotic thrombocytopenic purpura is an ultra-rare autosomal recessive disorder associated with mutations in the ADAMTS13 gene. The gene product is an enzyme responsible for cleavage of ultra-large von Willebrand factor multimers, and severe deficiency results in a thrombotic microangiopathy. We present the case of subclinical cardiac involvement characterized by myocardial fibrosis/scarring and impaired myocardial flow reserve in an affected patient who temporarily discontinued enzyme replacement therapy. This case highlights the complementary value of cardiac magnetic resonance
