Identification and characterization of two chitin synthase genes in African malaria mosquito, Anopheles gambiae.

Chitin synthase (CHS) represents an attractive target site for combating insect pests as insect growth and development are strictly dependent on precisely tuned chitin biosynthesis and this pathway is absent in humans and other vertebrates. Current knowledge on CHS in insects, especially their structures, functions, and regulations is still very limited. We report the identification and characterization of two chitin synthase genes, AgCHS1 and AgCHS2, in African malaria mosquito, Anopheles gambiae. AgCHS1 and AgCHS2 were predicted to encode proteins of 1,578 and 1,586 amino acid residues, resp

The complete mitochondrial genome sequence of the Mormon cricket (Anabrus simplex: Tettigoniidae: Orthoptera) and an analysis of control region variability.

The Anabrus simplex is a swarming plague orthopteran found in western North America. The genome is 15 766 bp in length and genome organization follows the ancestral insect gene arrangement. atp6 lacked any readily identifiable stop codon. Examination of mRNA secondary structure for this gene suggested a stem/loop-mediated mRNA post-transcriptional processing to liberate a mature atp6 mRNA with a complete stop codon produced by polyadenylation. Comparison of similar protein with protein gene boundaries in other insect species reveal a general mechanism for mRNA excision and provide further supp

Preventive effect of insect tea against reserpine-induced gastric ulcers in mice.

The aim of the present study was to determine the preventive effect of insect tea against reserpine-induced gastric ulcers in ICR mice. A high (800 mg/kg) dose of insect tea reduced the serum levels of the proinflammatory cytokines interleukin (IL)-6, IL-12, tumor necrosis factor (TNF)-α and interferon (IFN)-γ compared with those in mice treated with a low (400 mg/kg) dose and the control mice. The serum levels of somatostatin (SS) and vasoactive intestinal peptide (VIP) in mice treated with insect tea were higher compared with those in the control mice; however, the serum levels of motilin (M

Jasmonate-Responsive MYB55 Regulates Herbivore Resistance Through the Phenylpropanoid Metabolism in Rice.

The jasmonate (JA) phytohormone signaling pathway is a key regulator of plant resistance to insect herbivores by mediating the biosynthesis of anti-herbivore metabolites and proteins. However, the JA-mediated regulatory network and its downstream defensive compounds in the monocot crop rice remain largely unknown. Here, we investigate the role of the MYB transcription factor MYB55 in JA-mediated rice defense against insect herbivores. Knockout of MYB55 enhances feeding by both brown planthopper (BPH) and leaf folder (LF), while having no effect on BPH oviposition. MYB55 directly binds to AC mo

Octopamine Signaling Regulates Phototactic Behavior via a β-Adrenergic-like Receptor in Diaphorina citri.

The Asian citrus psyllid (Diaphorina citri, ACP) is the primary vector of citrus huanglongbing and exhibits positive phototactic behavior. Octopamine (OA) is a critical neuromodulator of diverse insect behaviors, yet its regulatory mechanism in ACP phototactic behavior remains largely uncharacterized. In this study, three core genes of the OA signaling pathway in ACP were identified and characterized: the biosynthetic enzyme DcTβH and two β-adrenergic-like OA receptors (DcOctβ1R, DcOctβ2R). All three genes exhibited a U-shaped developmental expression pattern and high transcript abundance in t

miR-34 regulates cuticle pigmentation by targeting Bm-iAANAT and Bmserpin3 in Bombyx mori.

Pigmentation is an orchestrated process involved in cuticle melanization and sclerotization in insects, and plays a critical role in maintaining the structural integrity and functional completeness of the insect cuticle. Although the cascade reactions underlying pigmentation have been extensively studied, our understanding of the involvement of miRNA in this process remains limited. Here, we investigate the role of conserved microRNA-34 (miR-34) in regulating cuticular colouration in the silkworm, bombyx mori. Overexpression of miR-34 led to pronounced melanization in the larval abdomen. Mecha

Insulin receptor participates in the peripheral olfactory processes of honey bees (Apis cerana cerana).

Insulin receptors (InR) are an integral component of the insulin/insulin-like growth factor signaling pathway, which plays a vital role in insect development, lifespan, reproduction, and olfactory sensitivity. However, whether InR participate in the peripheral olfactory system of insects remains unclear. Recently, we found that 2-heptanone (2-HT) affects AcerInR expression, the gene for an InR protein, in Apis cerana cerana. We then examined the spatiotemporal expression profile of the gene in A. cerana cerana. The mRNA of AcerInR was primarily expressed in the antennae, wings, and legs of for

Expression of the early-late gene encoding the nuclear receptor HR3 suggests its involvement in regulating the vitellogenic response to ecdysone in the adult mosquito.

The insect steroid hormone, 20-hydroxyecdysone (20E), is a key factor controlling critical developmental events of embryogenesis, larval molting, metamorphosis, and, in some insects, reproduction. We are interested in understanding the molecular basis of the steroid hormone ecdysone action in insect egg development. The yellow fever mosquito, Aedes aegypti, in addition to being an important vector of human diseases, represents an outstanding model for studying molecular mechanisms underlying egg maturation due to stringently controlled, blood meal-activated reproductive events in this insect.

Silencing Br-C impairs larval development and chitin synthesis in Lymantria dispar larvae.

In insects, 20-hydroxyecdysone (20E) mediates developmental transitions and regulates molting processes through activation of a series of transcription factors. Broad-Complex (Br-C), a vital gene in the 20E signalling pathway, plays crucial roles during insect growth processes. However, whether Br-C affects chitin synthesis in insects remains unclear. In the present study, the Br-C gene from Lymantria dispar, a notorious defoliator of forestry, was identified based on transcriptome data, and subjected to bioinformatic analysis. The regulatory functions of LdBr-C in chitin synthesis and metabol

Expression profile of genes during resistance reversal in a temephos selected strain of the dengue vector, Aedes aegypti.

BACKGROUND: The mosquito Aedes aegypti is one of the most important disease vectors because it transmits two major arboviruses, dengue and yellow fever, which cause significant global morbidity and mortality. Chemical insecticides form the cornerstone of vector control. The organophosphate temephos a larvicide recommended by WHO for controlling Ae. aegypti, however, resistance to this compound has been reported in many countries, including Brazil. METHODOLOGY/PRINCIPAL FINDINGS: The aim of this study was to identify genes implicated in metabolic resistance in an Ae. aegypti temephos resistant

Collagen type XV regulation via MMP2-dependent degradation governs ovarian development in the rice stem borer, Chilo suppressalis.

Female reproductive development is fundamental to the remarkable reproductive capacity of insects, and extracellular matrix (ECM) remodeling plays a critical role in ovarian development. However, the function of Collagen, a major ECM component, in female insect ovarian development remains poorly characterized. Using the major rice pest Chilo suppressalis as a model, we investigated the role of Col XV in insect reproduction. We cloned and characterized Cscol15, a gene encoding a 1077-amino acid protein. Real-time quantitative PCR analysis revealed peak Cscol15 expression during the egg stage an

Rickettsia symbionts favor whitefly ovary development by promoting germ cell mitosis.

BACKGROUND: Maternally inherited symbionts can impact the reproduction of their host insects in various ways. The ovary is critical for the successful reproduction of female insects. Such symbionts have strong ovary tropism. Intracellular symbionts depend on host cells for replication and transmission. However, the cellular mechanisms by which symbionts impact insect reproduction through affecting ovary development have seldom been investigated. Our previous work has shown that the facultative symbiont Rickettsia can be vertically transmitted through eggs in the whitefly Bemisia tabaci and Ric

A New Member of the Growing Family of Contact-Dependent Growth Inhibition Systems in Xenorhabdus doucetiae.

Xenorhabdus is a bacterial symbiont of entomopathogenic Steinernema nematodes and is pathogenic for insects. Its life cycle involves a stage inside the insect cadaver, in which it competes for environmental resources with microorganisms from soil and the insect gut. Xenorhabdus is, thus, a useful model for identifying new interbacterial competition systems. For the first time, in an entomopathogenic bacterium, Xenorhabdus doucetiae strain FRM16, we identified a cdi-like locus. The cdi loci encode contact-dependent inhibition (CDI) systems composed of proteins from the two-partner secretion (TP

Function of HIF-1α in Regulating Sphingolipid Metabolism and Alleviating Oxidative Stress Damage in Callosobruchus chinensis Under Hypoxia.

It has been suggested that hypoxia-inducible factor 1α (HIF1α) serves as a key regulatory factor in insect metabolism remodelling under hypoxia, in which lipid metabolism appears to play an important role. However, the underlying mechanism remains incompletely understood. In this paper, the function of HIF1α in regulating lipid metabolism under hypoxia in Callosobruchus chinensis (Coleoptera: Bruchidae), a typical stored-product insect, was investigated. Based on the amplification and cloning, the sequence of the CcHIF1α gene was analysed and found to be highly conserved throughout evolution.

Myriocin significantly increases the mortality of a non-mammalian model host during Candida pathogenesis.

Candida albicans is a major human pathogen whose treatment is challenging due to antifungal drug toxicity, drug resistance and paucity of antifungal agents available. Myrocin (MYR) inhibits sphingosine synthesis, a precursor of sphingolipids, an important cell membrane and signaling molecule component. MYR also has dual immune suppressive and antifungal properties, potentially modulating mammalian immunity and simultaneously reducing fungal infection risk. Wax moth (Galleria mellonella) larvae, alternatives to mice, were used to establish if MYR suppressed insect immunity and increased surviva

Generation of expression clone set for functional proteomics of human gastric and liver cancers.

Two thousand sixty-eight multi-purpose expression clones for the 326 candidate genes related to gastric or liver cancers were constructed using the Gateway system. These clones can be expressed as His, Glutathione-S-transferase (GST) or Enhanced version of the green fluorescent protein (EGFP) fusion proteins in E. coli, insect cells or mammalian cells. For the 246 E. coli expression clones, the GST fusion proteins had greater expression efficiency and solubility than the His fusion proteins. Approximately 20% of the expressed proteins had unexpected molecular weights. A detailed sequence analy

Flies and Ants from Domestic Kitchens as Sources of Clinically Important Bacteria and Antimicrobial Resistance.

Foods can be contaminated by a wide range of microorganisms, originating from multiple sources within domestic kitchens. However, the role of flies and ants as carriers of bacteria in these environments remains poorly understood, and data on the antimicrobial susceptibility profiles of insect-associated bacteria remain scarce. Therefore, this study aimed to identify Enterobacterales and Staphylococcus spp. recovered from flies and ants collected in domestic kitchens and to characterize the antimicrobial susceptibility profiles of the isolates. Additionally, selected virulence genes were invest

BdPiwi is essential for gametogenesis and post-embryonic development in the oriental fruit fly, Bactrocera dorsalis.

BACKGROUND: The PIWI-interacting RNA (piRNA) pathway is a conserved transposon silencing mechanism essential for genomic integrity and gametogenesis. While Piwi proteins are known to play essential roles in insect germline development, their functional characteristics remain largely unexplored in agricultural pests. Here, we investigated BdPiwi in Bactrocera dorsalis, a highly invasive pest whose exceptional reproductive capacity underpins its global pest status. RESULTS: Phylogenetic analysis revealed that BdPiwi is evolutionarily conserved among insects and contains the canonical PAZ and PI

Co-exposure to polystyrene microplastics and glyphosate induces gut microbiota dysbiosis and cognitive impairment in honeybees.

Microplastics, as emerging persistent environmental contaminants, can act as vectors for the absorption of other pollutants in ecosystems, such as pesticide residues. However, the combined toxicological effects of microplastics and pesticides on pollinators remain poorly understood. Here, we evaluated the toxicity of polystyrene microplastics (PS) and glyphosate (GLY), both individually and in combination, in honeybees (Apis mellifera). While no significant changes in body weight gain or food consumption were observed after 20 days of exposure, co-exposure to PS and GLY significantly increased

Microinjection Method for Anopheles gambiae Embryos.

Embryo microinjection techniques are essential for many molecular and genetic studies of insect species. They provide a means to introduce exogenous DNA fragments encoding genes of interest as well as favorable traits into the insect germline in a stable and heritable manner. The resulting transgenic strains can be studied for phenotypic changes resulting from the expression of the integrated DNA to answer basic questions or used in practical applications. Although the technology is straightforward, it requires of the investigator patience and practice to achieve a level of skill that maximize
