Genetic structure of local populations of Lutzomyia longipalpis (Diptera: Psychodidae) in central Colombia.

Lutzomyia longipalpis (Lutz & Neiva), the sand fly vector of American visceral leishmaniasis in the New World tropics, has a broad but discontinuous geographical distribution from southern Mexico to Argentina. A baseline for population genetic structure and genetic variability for this species was obtained by analyzing 5 local, peridomestic populations at the approximate center of its distribution, the Magdalena River Valley of central Colombia. Three populations of L. longipalpis from El Callejón, a small rural community, were compared with 2 populations from neighboring areas 12 and 25 km di

COMPLEX COLONY STRUCTURE IN SOCIAL INSECTS: I. ECOLOGICAL DETERMINANTS AND GENETIC CONSEQUENCES.

For social insect species, intraspecific variation in colony social structure provides an opportunity to relate the evolution of social behavior to ecological factors. The species Myrmica punctiventris is a cavity-dwelling forest ant that exhibits very different colony structures in two populations in the northeastern United States. Combined data from seasonal censuses, allozyme electrophoresis, and worker hostility tests showed that a population of M. punctiventris in Vermont was strictly monogynous and seasonally polydomous. The same procedures showed that a population of M. punctiventris in

COOH-terminal propeptides of the major human procollagens. Structural, functional and genetic comparisons.

The sequences of the carboxy-terminal extensions (COOH-propeptides) of at least one chain of all of the major human procollagens have only recently been deduced, and include those of the interstitial (alpha 1(I), alpha 2(I), alpha 1(II), alpha 1(III)), basement membrane (alpha 1(IV)) and pericellular (alpha 2(V)) procollagens. Comparisons of DNA and protein sequences, corresponding to these COOH-propeptides domains, established the early divergence of the basement membrane alpha 1(IV) COOH-propeptide from the corresponding sequences of the interstitial and pericellular procollagens. The latter

Structural MRI predicts clinical progression in presymptomatic genetic frontotemporal dementia: findings from the GENetic Frontotemporal dementia Initiative cohort.

Biomarkers that can predict disease progression in individuals with genetic frontotemporal dementia are urgently needed. We aimed to identify whether baseline MRI-based grey and white matter abnormalities are associated with different clinical progression profiles in presymptomatic mutation carriers in the GENetic Frontotemporal dementia Initiative. Three hundred eighty-seven mutation carriers were included (160 GRN, 160 C9orf72, 67 MAPT), together with 240 non-carrier cognitively normal controls. Cortical and subcortical grey matter volumes were generated using automated parcellation methods

Genetic Diversity and Population Genetic Structure Analysis of Plasmodium knowlesi Thrombospondin-Related Apical Merozoite Protein (TRAMP) in Clinical Samples.

The simian malaria parasite Plasmodium knowlesi causes a high number of zoonotic infections in Malaysia. The thrombospondin-related apical merozoite protein (TRAMP) is an essential ligand for binding to the erythrocyte cell surface, whereby it facilitates the invasion. This study is the first attempt to determine the genetic diversity, phylogeography, natural selection and population structure from 97 full-length PkTRAMP gene sequences originating from Malaysia. We found low levels of nucleotide diversity (π~0.0065) for the full-length gene despite samples originating from geographically separ

Variations in genetics, biology, and phenotype of cutaneous disorders in skin of color - Part I: Genetic, biologic, and structural differences in skin of color.

Skin of color (SOC) populations include those who identify as Black/African, Hispanic/Latinx, Asian/Pacific Islander, American Indian/Native Alaskan, Indigenous Australian, Middle Eastern, biracial/multiracial, or non-White; this list is far from exhaustive and may vary between and within cultures. Recent genetic and immunological studies have suggested that cutaneous inflammatory disorders (atopic dermatitis, psoriasis, and hidradenitis suppurativa) and malignancies (melanoma, basal cell carcinoma, and cutaneous T-cell lymphoma) may have variations in their immunophenotype among SOC. Addition

Genetic diversity and population structures of Plasmodium falciparum parasites imported to China from Central and West Africa determined using a SNP barcode.

BACKGROUND: Malaria remains a highly threatening infectious disease, with Africa being the main epidemic area. China has achieved the notable feat of eliminating malaria domestically, yet the looming threat of imported malaria cases remains a concern. This study aims to apply the SNP barcode technology to investigate the population structure of P. falciparum imported to China from Central and West Africa. METHODS: We applied a 24-SNP high-resolution melting (HRM) barcode to analyze the diversity of P. falciparum populations imported to China from Central and West Africa. RESULTS: A total of

Genetic analysis of the influence of neuroantigen-complete Freund's adjuvant emulsion structures on the sexual dimorphism and susceptibility to experimental allergic encephalomyelitis.

The induction of organ-specific autoimmune diseases, such as experimental allergic encephalomyelitis (EAE) the principal animal model of multiple sclerosis (MS), relies on the use of complete Freund's adjuvant (CFA) emulsions. In this study we report that the physical structure of the particles comprising neuroantigen-CFA emulsions significantly influences the genetic control of the incidence and sexual dimorphism seen in EAE. Immunization of (B10.S/SgMcdJ x SJL/J) F(2) mice segregating the quantitative trait loci (QTL) controlling EAE in susceptible SJL/J and resistant B10.S/SgMcdJ mice with

Genetic variation and population structure in a threatened species, the Utah prairie dog Cynomys parvidens: the use of genetic data to inform conservation actions.

The Utah prairie dog (Cynomys parvidens), listed as threatened under the United States Endangered Species Act, was the subject of an extensive eradication program throughout its range during the 20th century. Eradication campaigns, habitat destruction/fragmentation/conversion, and epizootic outbreaks (e.g., sylvatic plague) have reduced prairie dog numbers from an estimated 95,000 individuals in the 1920s to approximately 14,000 (estimated adult spring count) today. As a result of these anthropogenic actions, the species is now found in small isolated sets of subpopulations. We characterized t

Genetic diversity, population genetic structure and demographic history of two actiniarian species (Anthopleura xanthogrammica and Diadumene lineata) along the Chinese coastline.

Anthopleura xanthogrammica and Diadumene lineata have considerable pharmaceutical potential, but their distribution and population genetics along the Chinese coastline remain poorly understood. We collected 162 samples of the two species based on external morphology from nine intertidal sites. Subsequent analyses of the mitochondrial COI segment (607 bp) confirmed 143 A. xanthogrammica individuals across all nine localities, and only 19 D. lineata individuals restricted to three localities. For A. xanthogrammica, genetic diversity, phylogenetic relationships, population structure and demograph

Genetic diversity, population structure, and benzimidazole resistance profiling of Ascaris lumbricoides in northwestern Thailand.

Ascaris lumbricoides remains one of the most prevalent soil-transmitted helminths worldwide, with ongoing transmission in endemic regions despite long-standing control programs. This study investigated the genetic diversity, population structure, and benzimidazole resistance of A. lumbricoides across three endemic regions of Thailand using mitochondrial (cox1 and nad1) and nuclear (β-tubulin) markers. Haplotype network analysis identified 13 haplotypes within the Thailand dataset, including three newly identified haplotypes, indicating measurable genetic diversity at the local scale. When anal

An Exploratory Analysis of Genetic Structure and Introgression in Salmo trutta Populations from Campania Rivers.

The Salmo trutta complex exhibits high genetic and phenotypic diversity due also to the historical climatic events and colonization processes. In Europe, several evolutionary lineages have been identified, including the Atlantic and Mediterranean. In Italy, native Mediterranean trout populations have been threatened by restocking practices involving hatchery-reared fish of Atlantic origin, thus resulting in widespread hybridization and genetic introgression. This study aimed to genetically characterize trout populations from selected river basins of the Campania region (southern Italy) and to

Genetic Structure Along Temperature Gradient in a Recent Invader Cardamine hirsuta in Eastern Japan.

Invasive species often spread rapidly, which appears discrepant because species distribution is often limited by environmental conditions. Even in widely distributed species, there are differences among genetic clusters along environmental gradients, suggesting that the environmental range of one genetic group is limited but extended distribution is possible through genetic differentiation. Cardamine hirsuta is native to Europe, invaded Japan in the 1970s, and is now found throughout Japan. To understand how C. hirsuta has rapidly spread across a climatic gradient in eastern Japan, where there

Genetic complementation reveals structure-function links in nodavirus RNA replication complex crowns.

Positive-strand RNA viruses replicate their RNA genomes in virus-induced, membrane-bounded organelles. As first found for nodaviruses, the necked cytosolic portals of these organelles bear ringed "crown" complexes of viral RNA replication proteins that drive synthesis, capping and release of new RNA genomes. Nodavirus crowns contain two 12-mer rings of viral protein A with C-proximal polymerase domains stacked. In the basal ring, protein A's N-proximal RNA capping domains form a central, toroidal floor, while in the apical ring these domains extend radially outward. A third protein A conformat

Genetic mechanisms underlying the structural elaboration and dissemination of viral internal ribosomal entry sites.

Viral internal ribosomal entry sites (IRESs) are highly structured cis-acting RNAs that mediate end-independent initiation of translation. Their origin remains obscure. The simplest IRESs (type 6) occur in the intergenic region of Dicistroviridae genomes (order Picornavirales), consist of two pseudoknots, and initiate translation by factor-independent binding to ribosomes. Larger variants contain a third pseudoknot that modifies the mechanism of IRES function by engaging with the ribosomal head and promoting binding to the ribosomal aminoacyl site. Metagenomic analyses undertaken to identify s

DNA variants affecting chromatin structure are key to the genetic architecture of obsessive compulsive disorder.

BACKGROUND: Obsessive Compulsive Disorder (OCD) affects 2 to 3% of the population and is marked by intrusive thoughts and repetitive behaviours. OCD is increasingly recognized as a polygenic disorder involving gene-environment interactions, with genetic risk largely driven by common variants and a smaller contribution from rare pathogenic variation. Despite these insights, the functional roles of implicated genes remain unclear. Recent genomic studies suggest that chromatin dysregulation contributes to the pathogenesis of OCD. RESULTS: We analysed the recently published large-scale genome-wid

Precision Medicine in patients with rare forms of genetic obesity: Necessity for coordinated and structured care.

Advances in research enable precision medicine for rare metabolic diseases. The approval of therapies as melanocortin-4 receptor (MC4R) agonists for the treatment of hyperphagia and obesity in monogenic disorders (POMC, PCSK1, and LEPR deficiencies) and Bardet-Biedl syndrome (BBS), affecting fewer than 1 in 20,000 (BBS) to fewer than 1 in 1,000,000 individuals, represents a major advance. These therapies have transformative potential but introduce new responsibilities due to limited patient numbers and lack of long-term data. Equitable access to diagnosis and treatment, expert care and structu

Genetic dissection of serum pro-neurotensin suggests potential causal impact on brain structure.

BACKGROUND: Pro-neurotensin (pro-NT) is the stable circulating precursor of neurotensin (NT), a neuropeptide expressed mainly in the central nervous system and small intestine that regulates key physiological processes like fatty acid absorption in the gut and suppresses appetite via central mechanisms. Studies in NT-deficient mice and humans implicate NT in obesity and insulin resistance, highlighting its role in metabolic regulation. METHODS: To explore the genetic determinants of circulating pro-NT and its causal relationships with obesity and brain phenotypes, we conducted a genome-wide m
