Remote ischemic conditioning provides humoural cross-species cardioprotection through glycine receptor activation.

AIMS: Remote ischaemic conditioning (RIC) releases a humoural factor able to exert cross-species cardioprotection when plasma dialysate is applied to isolated hearts. However, the exact chemical nature of this factor is currently unknown. METHODS AND RESULTS: RIC (4 × 5min femoral occlusion/5min reperfusion) was applied to 10 male pigs, and blood was taken before and after the manoeuvre. Discriminant analysis of 1H-NMR spectra (n = 10-12) obtained from plasma dialysates (12-14 kDa cut-off) allowed to demonstrate a different metabolic profile between control and postRIC samples, with lactate (

Arginine-Glycine Amidinotransferase Deficiency and Functional Characterization of Missense Variants in GATM.

Arginine-glycine amidinotransferase (GATM) deficiency is an autosomal-recessive disorder caused by pathogenic variants in GATM. Clinical features include intellectual disability, hypotonia, and myopathy. Due to normal neurodevelopment in asymptomatic individuals on creatine monotherapy, GATM deficiency is a good candidate for newborn screening. To determine the carrier frequency of GATM deficiency, we performed functional characterization of rare missense variants in GATM reported as heterozygous in the Exome Variant Server database. To assess phenotype and genotype correlation, we developed a

Visceral, inflammatory and neuropathic pain in glycine receptor alpha 3-deficient mice.

The alpha3-subunit of strychnine-sensitive glycine receptors is an important modulator of the pain-sensitizing effects of spinal prostaglandin prostaglandin E(2). Mice deficient for alpha3-subunit of strychnine-sensitive glycine receptors lack the prostaglandin E(2)-induced inhibition of glycinergic neurotransmission and recover faster from inflammation-induced hyperalgesia. It, however, remains unclear whether alpha3-subunit of strychnine-sensitive glycine receptors plays a role in other pain models involving prostaglandin synthesis, such as chemically induced pain or neuropathic pain. In thi

The glycine transporter-1 inhibitors NFPS and Org 24461: a pharmacological study.

The in vitro and in vivo pharmacology of two glycine transporter-1 (GlyT1) inhibitors, N[3-(4'-fluorophenyl)-3-(4'-phenylphenoxy)-propyl]sarcosine (NFPS) and R,S-(+/-)N-methyl-N-[(4-trifluoromethyl)phenoxy]-3-phenyl-propylglycine (Org 24461), was studied. NFPS and Org 24461 inhibited the uptake of [3H]glycine in hippocampal synaptosomal preparation with IC(50) values of 0.022 and 2.5 microM. Neither NFPS nor Org 24461 (0.1 microM) showed significant binding to alpha-1, alpha-2, and beta-adrenoceptors, D(1) and D(2) dopamine receptors, and 5-HT(1A) and 5-HT(2A) serotonin receptors in membranes

Glycine N-methyltransferase deficiency: a novel inborn error causing persistent isolated hypermethioninaemia.

This paper reports clinical and metabolic studies of two Italian siblings with a novel form of persistent isolated hypermethioninaemia, i.e. abnormally elevated plasma methionine that lasted beyond the first months of life and is not due to cystathionine beta-synthase deficiency, tyrosinaemia I or liver disease. Abnormal elevations of their plasma S-adenosylmethionine (AdoMet) concentrations proved they do not have deficient activity of methionine adenosyltransferase I/III. A variety of studies provided evidence that the elevations of methionine and AdoMet are not caused by defects in the meth

Glycine-gated chloride channels in neutrophils attenuate calcium influx and superoxide production.

Recently, it was demonstrated that liver injury and TNF-alpha production as a result of endotoxin (lipopolysaccharide, LPS) were attenuated by feeding animals a diet enriched with glycine. This phenomenon was shown to be a result of, at least in part, activation of a chloride channel in Kupffer cells by glycine, which hyperpolarizes the cell membrane and blunts increases in intracellular calcium concentrations ([Ca(2+)](i)) similar to its action in the neuron. It is well known that hepatotoxicity due to LPS has a neutrophil-mediated component and that activation of neutrophils is dependent on

Experience with isotonic 2.2% glycine as distension medium for hysteroscopic endomyometrial resection.

The feasibility of performing hysteroscopic endomyometrial resection using isotonic 2.2% glycine as distension medium was studied in 181 consecutive operations. A standard continuous flow loop-resectoscope was used. Distension was achieved by means of a gravity-fed system and glycine flowed out under its own pressure. The mean (+/-SEM) glycine deficit was 160+/-20 ml and in only 7 patients was it >1 liter (maximum 1,800 ml). Only in patients with >1 liter glycine absorption was there a significant decrease in serum sodium level (mean 9 mmol/l), while serum osmolality remained normal. Apart fro

Enhancement of homomeric glycine receptor function by long-chain alcohols and anaesthetics.

1. The effects of n-alcohols (ethanol to dodecanol) and anaesthetics on strychnine-sensitive glycine receptors were studied in Xenopus oocytes expressing homomeric alpha 1 or alpha 2 glycine receptor subunits, with the two electrode voltage-clamp recording technique. 2. The glycine-induced chloride conductance of homomeric alpha glycine receptors was potentiated by all the alcohols tested when an EC2 concentration of glycine was used. Homomeric alpha 1 and alpha 2 receptors were potentiated similarly by the n-alcohols, except that low concentrations of ethanol produced greater potentiation wit

Disorder of the inhibitory glycine receptor: inherited myoclonus in Poll Hereford calves.

Inherited congenital myoclonus in Poll Hereford calves is characterized by hyperesthesia and myoclonic jerks of the skeletal musculature that occur spontaneously and in response to sensory stimuli. The symptoms of the disorder suggest a failure of spinal inhibition and are similar to those in subconvulsive strychnine poisoning. Strychnine is a high-affinity antagonist of the synaptic actions of glycine. Our recent biochemical studies revealed a specific and marked deficit in [3H]strychnine binding sites in brain stem and spinal cord membranes from myoclonic calves compared with unaffected cont

Possible defect in xenobiotic activation before glycine conjugation in protein-energy malnutrition.

1. [14C]Benzoic acid administered to rats fed a normal diet was excreted mainly (99% of 24h excretion) as hippuric acid. 2. In protein-energy malnourished rats, only about 74% of [14C]benzoic acid administered was excreted as hippuric acid. The remainder was excreted as the glucuronide conjugate. 3. The oxidative phosphorylation capacity of liver mitochondria of malnourished rats was 30% less than that of normal rat liver mitochondria. 4. The decreased rate in oxidative phosphorylation is discussed in relationship to the observed decrease in glycine conjugation.

Urinary zinc excretion following infusions of zinc sulfate, cysteine, histidine, or glycine.

Zinc sulfate (40 mg elemental zinc) or one of three amino acids (5 or 10 g cysteine, 10 g histidine, or 10 g glycine) was infused into anesthetized dogs over a 60-min period. Plasma concentrations and urine excretions of zinc and five other cations, and glomerular filtration rates were determined before, during, and after these infusions. Infusions of zinc sufficient to produce a 20-fold increase in total plasma zinc concentrations had little effect on urinary zinc excretions (threefold increase) or plasma ultrafilterable zinc concentrations. Ten grams of cysteine urinary zinc excretions more

Immunoreactive insulin (IRI) dynamics in GH-deficient patients following intravenous glycine loading.

In hypopituitary short-statured patients glycine intravenous injection (250 mg/kg of body weight) provokes a mild and inconstant stimulation of IRI release followed by a significant fall down. Though insiginificant (P 0.064), the prevalence of IRI responsive individuals to glycine in the patients group of idiopathic etiology could be of pathophysiological importance.

Interaction of benzodiazepines with central nervous glycine receptors: possible mechanism of action.

Interaction of 21 benzodiazepines with the glycine receptor in the brainstem and spinal cord of rat have been evaluated in terms of their displacement of [(3)H]strychinine binding. The rank order of potency of the 21 drugs in displacing specific [(3)H]strychinine binding correlates (p < 0.005) with their rank order of potency in a vareity of pharmacological and behavioral tests in humans and animals that predict clinical efficacy. There is a 50-fold variation in potency of the series of benzodiazepines with mean effective dose (ED(50)) values ranging from 19muM to > 1000 muM. Diazepam (Valium(

Effect of Fe on Ni-based catalysts for electrochemical glycine oxidation to ammonia.

Management of excess organic nitrogen in wastewater-derived sludge is increasingly important due to the high cost of sludge disposal. Recovering ammonia from organic nitrogen in sludge through electrolysis offers a promising approach to managing this nitrogen waste stream while generating economic value. Despite the abundance of high-molecular-weight nitrogen compounds in sludge, limited progress has been made in understanding how organic nitrogen can be directly and efficiently transformed into ammonia without microorganisms. Here, we investigate the abiotic reaction pathways for converting g

Experimental and Computational Analysis of the Roles of Polymer Templates and pH in Glycine Crystallization.

Understanding and controlling polymorph selection during amino acid crystallization is essential for applications in pharmaceuticals and bioelectronic materials. Here, we investigate the combined effects of polymer templating and pH modulation on glycine crystallization by integrating experimental results and all-atom molecular dynamics (MD) simulations. Poly(vinyl alcohol) (PVA) and a weak polyelectrolyte, poly(acrylic acid) (PAA), were employed to assess the formation of α- and γ-glycine polymorphs across a range of solution pHs. X-ray diffraction (XRD) characterization of the polymer-glycin

Functional roles of the miR1512 family in regulating soybean resistance to Heterodera glycines.

BACKGROUND: Soybean [Glycine max (L.) Merr.] is major economic crop, yet its yields are persistently eroded by soybean cyst nematode (SCN). Although several legume-specific miRNAs are known to orchestrate the soybean-SCN dialogue, the mechanisms by which they confer SCN tolerance remain largely uncharted. RESULTS: Here, we dissected the role of the miR1512 family in SCN resistance. We uncovered distinct expression patterns among its three precursors and four mature isoforms. Overexpression of the precursors of miR1512 (MIR1512) in transgenic soybean hairy roots elevated precursor transcripts

A Unique Mechanism of Glycine-Specific Inhibition of Bacterial Translation by Bottromycin A2.

The rise of antimicrobial resistance among pathogenic bacteria poses a critical challenge to modern medicine, highlighting an urgent need for novel therapeutic agents. Bottromycin A2 (BotA2) is a promising candidate for future drug development, demonstrating potent activity against clinically relevant pathogens, including methicillin-resistant Staphylococcus aureus, vancomycin-resistant Enterococcus, and Mycoplasma species, although its molecular mechanism of action has remained unclear until now. Here, we demonstrate that BotA2 inhibits bacterial translation with unique context specificity de

Cyanide Formation in Freezer Stored Foods: Freezing of a Glycine and Nitrite Mixture.

Freezing is not always the best way to keep foods safely. Some reactions are known to be accelerated in ice. Furthermore, some other reactions that are not observed in solution are also promoted in ice. We found that the formation of nitrosamines through the reaction of an amine with a nitrite is accelerated in ice. Surprisingly, cyanide is formed through the reaction of glycine with nitrite in ice but not in solution. Amines are present in many kinds of foods. Nitrite is present in vegetables and is used as a food coloring agent and to inhibit the reproduction of Clostridium botulinum. The ma

NMR study of mutations of glycine-52 of the catalytic domain of diphtheria toxin.

Cross-reacting-material 197 (CRM197) is a naturally occurring non-toxic mutant of diphtheria toxin (DT) that is one of the few carrier protein used in the manufacture of polysaccharide vaccines targeting bacterial pathogens such as Neisseria meningitidis, Streptococcus pneumaniae and Haemophilus influenzae. A detailed explanation in structural terms for the lack of toxicity has started to emerge with the report of the X-ray structure of CRM197. Here, we present an NMR spectroscopy study of the wild-type catalytic domain of diphtheria toxin and the effects of mutations at residue 52 on its conf
