Hydroxyapatite and demineralized calf fetal growth plate effects on bone healing in rabbit model.

BACKGROUND: Synthetic hydroxyapatite (HA), beta-tricalcium phosphate (β-TCP) and their composite are promising biomaterials, specifically in the orthopedic and dental fields, as their chemical composition is similar to that of bone. Due to the need for safer bone graft applications, these bone graft substitutes are gradually gaining increased acceptability. To stimulate the process of bone healing, several methods have been used previously, including ultrasound, electrical stimulation, exposure to electromagnetic fields, bone grafts, interporous hydroxyapatite (as a bone graft substitute) and

Spatial periodicity in growth plate shear mechanical properties is disrupted by vitamin D deficiency.

The growth plate is a highly organized section of cartilage in the long bones of growing children that is susceptible to mechanical failure as well as structural and functional disruption caused by a dietary deficiency of vitamin D. The shear mechanical properties of the proximal tibial growth plate of rats raised either on normal or vitamin D and calcium deficient diets were measured. A sinusoidal oscillating shear load was applied to small excised growth plate specimens perpendicular to the direction of growth while imaging the deformation in real time with a fast confocal microscope. Weight loading young chicks inhibits bone elongation and promotes growth plate ossification and vascularization.

The mechanical stimuli resulting from weight loading play an important role in mature bone remodeling. However, the effect of weight loading on the developmental process in young bones is less well understood. In this work, chicks were loaded with bags weighing 10% of their body weight during their rapid growth phase. The increased load reduced the length and diameter of the long bones. The average width of the bag-loaded group's growth plates was 75 +/- 4% that of the controls, and the plates showed increased mineralization. Northern blot analysis, in situ hybridization, and longitudinal cell

Stress-induced Salter-Harris I growth plate injury of the proximal tibia: first report.

We describe a case of chronic Salter-Harris I injury of the proximal tibial epiphysis. To our knowledge such an injury has not been described in the English language literature. The radiological appearance can mimic chronic infection. The possibility of chronic athletic stress-related change should be considered in such scenarios to avoid a misdiagnosis.

Magnetic resonance imaging of growth plate injuries: the efficacy and indications for surgical procedures.

In 23 patients with growth plate injuries, magnetic resonance imaging (MRI) studies were performed a total of 31 times to evaluate the physis which showed plain radiographic evidence of possible damage. Fourteen patients clinically showed growth arrest, and 10 patients required a Langenskiold operation. In 3 patients who underwent this operation, subsequent premature total fusion of the physis adversely affected the postoperative results. We propose that the merging shape of the arrest line with calcification of the provisional zone of the metaphysis shown by MRI indicates poor viability of th

Distribution of hyaluronan in the epiphysial growth plate: turnover by CD44-expressing osteoprogenitor cells.

In the present study, we have examined the distribution of both hyaluronan and its receptor, CD44, during the process of endochondral ossification in the mouse tibia. Histochemical staining revealed that a large amount of hyaluronan was present in the lacunae located in the zone of hypertrophy, but it was greatly reduced or absent from the zone of erosion. In addition, hyaluronan was present in the cytoplasm of osteoprogenitor cells located in the zone of erosion. These cells also expressed CD44 on their surfaces, as revealed by double-label immunohistochemistry. These results suggested that t

Nonreducible crosslink formation in tibial dyschondroplastic growth plate cartilage from broiler chicks fed homocysteine.

In the study of tibial dyschondroplasia, scientists have for a long time thought that an altered extracellular matrix might be involved in the etiology of the disease. The results presented in this paper show that the collagen content was increased in the dyschondroplastic cartilage when compared to normal growth plate and day-old hypertrophic cartilage. Furthermore, nonreducible crosslinks were found only in dyschondroplastic cartilage, with the greatest amounts occurring in the distal region of the lesion, approximately 10-fold higher than that found in the dyschondroplastic growth plate. Calcification of cartilage matrix in chondrocyte cultures derived from rachitic rat growth plate cartilage.

Chondrocytes obtained from collagenase-digested epiphyseal growth plate cartilage of rachitic rats were grown in multilayer cultures. The cultured chondrocytes produced a metachromatic matrix and further electron microscopic examination revealed typical features of cartilage matrix collagen fibrils and matrix vesicles. The alkaline phosphatase activity in cultures was high during the entire 3-week culture period. Acid phosphatase showed a marked increase in activity during the first week of culture. The appearance of apatite crystals in the synthesized matrix was monitored by electron microsco

Growth Plate Injuries, Skeletal Maturity, and Bone Health in Young Dancers: A Bibliometric Analysis.

BACKGROUND: High demands and repetitive loads experienced by young dancers are of major concern regarding growth plate injury and issues of skeletal maturity during development. While dance medicine has become an accepted subspecialty, an evaluation of research literature regarding these specific issues among a youth population has been lacking to observe patterns and gaps. To address these challenges, bibliometric analyses are useful tools to visualize scientific productivity, leading scholars, and research clusters in this niche sports medicine topic. METHODS: A bibliometric review was cond

A comparison of tension-band plate growth modulation in mucopolysaccharidoses versus idiopathic angular deformities.

This study compares the outcomes of tension-band plates (TBP) correction of angular deformities in patients with Hurler and Morquoio syndromes mucopolysaccharidoses (MPS) to those with idiopathic etiologies. A retrospective analysis of patients aged less than 18 at a single institution who received TBP between 2005 and 2017 for valgus angular deformities was conducted. Inclusion criteria were patients with complete data and MPS or idiopathic etiology. Femoral and tibial deformities were evaluated independently. Postsurgical X-rays were reported at 6-month intervals. Statistical significance wa

Radiologic study of the magnesium oxide nanoparticles effect on growth plate healing in rabbits.

BACKGROUND: The growth plate is important in new bone developing; therefore, its injury may cause closing the plate in premature period and effect on growth of the bone. This injury can lead to deformity of the skeleton, if not treated correctly and not monitored for a long time. AIM: This study was aimed to evaluate radiographically the role of autogenous cartilage grafts impregnated with Magnesium Oxide Nanoparticles (MgONPs) in growth plate regeneration and in preventing the bone bridge formation at the growth plate defect. METHODS: Ten adult rabbits were divided into two groups: a contro

Impact of All-trans Retinoic Acid on Skeletal Development: Mechanisms of Growth Plate Closure.

INTRODUCTION: All-trans retinoic acid (ATRA), a therapeutic mainstay for acute promyelocytic leukemia, is associated with off-target effects on skeletal development, including premature growth plate closure. However, the molecular mechanisms underlying ATRA-induced growth plate senescence remain poorly understood. METHODS: Using Sprague-Dawley rats, ADTC5 chondrocyte cell lines, and integrated multiomics approaches (transcriptome sequencing, weighted gene co-expression network analysis, molecular docking, and functional assays), we investigated how ATRA modulates growth plate development. Ex vivo organotypic bone slice culture reveals preferential chondrogenesis after sustained growth plate injury.

Postnatal bone growth primarily relies on chondrocyte proliferation and osteogenic differentiation within the growth plate (GP) via endochondral ossification. Despite its importance, the GP is vulnerable to injuries, affecting 15-30 % of bone fractures. These injuries may lead to growth discrepancies, influence bone length and shape, and negatively affecting the patient's quality of life. This study aimed to investigate the molecular and cellular physiological and pathophysiological regeneration following sustained growth plate injury (GPI) in an ex vivo rat femur organotypic culture (OTC) mod

The G protein-coupled receptor ADGRG6 maintains mouse growth plate homeostasis through IHH signaling.

The cartilage growth plate is essential for maintaining skeletal growth; however, the mechanisms governing postnatal growth plate homeostasis are still poorly understood. Using approaches of molecular mouse genetics and spatial transcriptomics applied to formalin-fixed, paraffin-embedded tissues, we show that ADGRG6/GPR126, a cartilage-enriched adhesion G protein-coupled receptor (GPCR), is essential for maintaining slow-cycling resting zone cells, appropriate chondrocyte proliferation and differentiation, and growth plate homeostasis in mice. Constitutive ablation of Adgrg6 in osteochondral p

Genomic Effects of Biomechanical Loading in Adolescent Human Growth Plate Cartilage: A Pilot Study.

ObjectiveThe genomic effects of biomechanical loading on human growth plate cartilage are unknown so far. To address this, we used rare human growth plate biopsies obtained from children undergoing epiphysiodesis and exposed them to precisely controlled mechanical loading using a microloading device. The biopsies were cultured 24 hours after mechanical loading, followed by RNA-sequencing analyses to decipher the genomic regulation.DesignWe conducted RNA-seq analysis of human growth plate cartilage obtained from three patients cultured ex vivo and subjected to cyclical mechanical loading with p

Microwave Ablation of the Pig Growth Plate: Proof of Concept for Minimally Invasive Epiphysiodesis.

BACKGROUND: Different surgical methods for epiphysiodesis of limb length discrepancy (LLD) have been described. Although these methods are variably effective, they are associated with morbidity (pain and limp) and potential complications. Microwave ablation is a less-invasive opportunity to halt growth by selectively destroying the growth plate via thermal energy to treat LLD in children. QUESTIONS/PURPOSES: In this proof-of-concept study using an in vivo pig model, we asked: (1) What is the durability of response 2 to 4 months after microwave ablation of the tibial growth plate as measured b

Reactivation of Vertebral Growth Plate Function in Vertebral Body Tethering in an Animal Model.

Flexible spine tethering is a relatively novel fusionless surgical technique that aims to correct scoliosis based on growth modulation due to the pressure exerted on the vertebral body epiphyseal growth plate. The correction occurs in two phases: immediate intraoperative and postoperative with growth. The aim of this study was to evaluate the reactivation of vertebral growth plate function after applying corrective forces. The rat tail model was used. Asymmetric compression and distraction of caudal growth plates were performed using a modified external fixation apparatus. Radiological and his
