Contextual and behavioral control of antipsychotic sensitization induced by haloperidol and olanzapine.

Repeated administration of haloperidol (HAL) and olanzapine (OLZ) causes a progressively enhanced disruption of the conditioned avoidance response (CAR) and a progressively enhanced inhibition of phencyclidine (PCP)-induced hyperlocomotion in rats (termed antipsychotic sensitization). Both actions are thought to reflect intrinsic antipsychotic activity. The present study examined the extent to which antipsychotic-induced sensitization in one model (e.g. CAR) can be transferred or maintained in another (e.g. PCP hyperlocomotion) as a means of investigating the contextual and behavioral controls

Aripiprazole and haloperidol: beneficial combination antipsychotic therapy for a schizophrenic patient.

OBJECTIVE: Schizophrenia is a chronic disease that is treated with dopamine antagonists. These drugs can produce intolerable side effects through their blockade of central nervous system dopamine receptors unrelated to schizophrenia. The atypical antipsychotic aripiprazole, a dopamine receptor partial agonist, has mixed agonist-antagonist effects. Its partial agonism is said to protect the patient from the side effects caused by full antagonists at the same time that its antagonism treats the schizophrenia effectively. CASE: We report a case in which a low dose of the full antagonist haloperi

Postmortem redistribution of two antipsychotic drugs, haloperidol and thioridazine, in the rat.

Antipsychotic drugs may be associated with arrhythmia, ventricular fibrillation, or torsades de pointes, which can result in sudden death. These drugs could therefore be found in postmortem toxicological analyses of autopsy specimens following unexplained sudden death. The drug concentrations in tissues and body fluids change between the death and postmortem specimens collection because of postmortem redistribution. For this reason, it is often difficult to interpret the postmortem analysis. The aim of this study was to investigate postmortem redistribution of the two cardiotoxic antipsychotic

The safety and pharmacokinetics of quetiapine when coadministered with haloperidol, risperidone, or thioridazine.

The effects of haloperidol, risperidone, and thioridazine on the pharmacokinetics and side-effect profile of quetiapine were investigated in 36 patients with schizophrenia, schizoaffective disorder, or bipolar disorder in a single-center, two-period, multiple-dose, open-label, randomized trial. Over a one-to two-week period, quetiapine doses were escalated to 300 mg twice daily (bid). Patients were then treated for at least 7 days at the target quetiapine dose and subsequently entered into the combination therapy period, receiving haloperidol (7.5 mg, bid), risperidone (3 mg, bid), or thiorida

Comparison of the effects of clozapine, chlorpromazine, and haloperidol on membrane lateral heterogeneity.

The interactions of three neuroleptic drugs, clozapine (CLZ), chlorpromazine (CPZ), and haloperidol (HPD) with phospholipids were compared using DSC and Langmuir balance. Main emphasis was on the drug-induced effects on the lateral organization of lipid mixtures of the saturated zwitterionic 1,2-dipalmitoyl-sn-glycero-3-phosphatidylcholine (DPPC) and the unsaturated acidic phosphatidylserine, brainPS. In multilamellar vesicles (MLV) phase separation was observed by DSC at X(PS)> or =0.05. All three drugs bound to these MLVs, abolishing the pretransition at X(drug)> or =0.03. The main transitio

The differential effects of chlorpromazine and haloperidol on latent inhibition in healthy volunteers.

Latent inhibition (LI) is a measure of reduced learning about a stimulus to which there has been prior exposure without any consequence. It therefore requires a comparison between a pre-exposed (PE) and a non-pre-exposed (NPE) condition. Since, in animals, LI is disrupted by amphetamines and enhanced by antipsychotics, LI disruption has been proposed as a measure of the characteristic attentional deficit in schizophrenia: the inability to ignore irrelevant stimuli. The findings in humans are, however, inconsistent. In particular, a recent investigation suggested that since haloperidol disrupte

Effect of chlorpromazine and clozapine on plasma concentrations of haloperidol in a patient with schizophrenia.

A 40-year-old patient being treated for schizophrenia developed elevated plasma levels of haloperidol (HAL) in combination with chlorpromazine (CPZ) and during overlap treatment with clozapine. Competitive inhibition of HAL by first CPZ and then clozapine is discussed as a possible mechanism.

Haloperidol-increased muscle tone in rats as a model of parkinsonian rigidity.

The aim of the present study was to find out whether haloperidol-induced rigidity was similar to that seen in parkinsonism. Simultaneous measurements of the muscle resistance (mechanomyogram, MMG) of the hind foot to passive flexion and extension in the ankle joint, as well as determination of the electromyographic (EMG) activity of the gastrocnemius and tibialis anterior muscles of rats were carried out. Haloperidol was injected in doses of 0.5-10 mg/kg 1 h before the start of measurements. Haloperidol increased, in a dose-dependent manner, the muscle resistance of the rat's hind leg to passi

[The effect of haloperidol at a low dose on the development of experimental parkinsonian syndrome].

Effect of galoperidole administrated in a small dose on changes in movement activity of mice in formation of hypokinetic syndrome in them using 1-methyl-4-phenyl-1,2,3,4-tetrahydropyridine (MPTP) was studied. Administration of galoperidol (0.01 mg/kg) increases the stability of structures of nigro-striate system with respect to injuring effect of MPTP; galoperidol administration in the same dose after formation of hypokinetic syndrome favors the more rapid recovering movement activity of animals. One of possible mechanisms resulting in the effect mentioned above may be an increase in a number

Haloperidol-related dyskinesias and pre- and perinatal complications in autistic children.

It has been hypothesized that an association exists between pre- and perinatal complications, central nervous system (CNS) dysfunction, and the development of tardive (TD) and withdrawal dyskinesias (WD). We assessed the relationship between pre- and perinatal complications and TD/WD in a sample of 118 children with autism who participated in an ongoing long-term prospective study of the efficacy and safety of haloperidol. The mean total Rochester Research Obstetrical Scale (ROS) score was significantly higher for children who developed TD/WD compared to those who did not (p = .007) as was the

Effects of risperidone on phencyclidine-induced behaviors: comparison with haloperidol and ritanserin.

In this study, we investigated whether risperidone, a serotonin-S2A (5-HT2A)/dopamine-D2 (D2)-receptor antagonist, inhibits phencyclidine (PCP)-induced stereotyped behaviors in comparison with haloperidol and ritanserin. Moreover, we also attempted to investigate the effects of these antipsychotics on the contents of dopamine, serotonin (5-HT) and their metabolites in rat striatum and frontal cortex. In rats, PCP (5 mg/kg, i.p.) caused hyperlocomotion and stereotyped behaviors, including sniffing, head-weaving, backpedalling and turning. Both resperidone (0.8-2.4 mg/kg, p.o.) and haloperidol (

Hyperthermic reaction to haloperidol with rigidity, associated to central core disease.

A 21-year-old female patient with psychotic symptoms developed hyperthermia, muscular rigidity and hypertension after administration of haloperidol. A muscle biopsy showed some atrophic and necrotic fibers, and a great number of fibers with central cores in the oxidative enzyme preparations. A related syndrome, Malignant Hyperthermia (MH), is sometimes associated with central core disease. The present case shows an association of a hyperthermic syndrome related to haloperidol with central core disease.

Naloxone or haloperidol but not yohimbine reverse apomorphine-induced respiratory depression.

Intravenous injection of low doses of naloxone was found to reverse the respiratory depression induced by apomorphine in chloralose-anesthesized dogs. Similar results were obtained with haloperidol, whereas yohimbine remained ineffective. These data suggest that apomorphine depresses respiratory rate through a mechanism involving both dopamine and opiate mechanisms but not alpha 2-adrenoceptors. Respiratory arrest induced by overdosages of apomorphine can be treated with naloxone.

Nicotine potentiation of haloperidol in reducing tic frequency in Tourette's disorder.

In an open, nonblind study, 10 patients with Tourette's disorder who were being treated with haloperidol were videotaped before, while, and after chewing nicotine gum. The frequency of tics was reduced significantly during the 30-minute gum-chewing period and during the 1 hour after gum chewing. Nicotine appears to potentiate haloperidol effects in patients with Tourette's disorder.

Genetics, haloperidol and pimozide: a comparative study in two mouse strains.

Haloperidol and pimozide are the only medications approved in management of Tourette's syndrome, thought due to dopaminergic overactivity with a possible genetic trait. The effect of equal dose regimens of these drugs on some brain biogenic amines and major acidic metabolites was studied in two genetically different strains of mice. These drugs exerted strain-dependent effect on regional brain levels of the compounds measured. The results suggest a higher turnover of striatal dopamine by haloperidol than by pimozide in the albino BALB/c but not in the black C57BL/6 mouse strain which may expla
