Transport of pigs different with respect to the halothane gene: stress assessment.

Two transport experiments were carried out with 18 pigs each. These pigs originated from three genetic lines (homozygous halothane-positive and -negative and heterozygotes). Half the pigs were unfed for 12 h before transport. All pigs were transported twice for 2 h. Before and after transport pigs were anesthetized to take blood samples from the jugular vein and biopsies from the biceps femoris. At the same time equipment to measure body temperature and heart rate were attached or detached. Plasma cortisol and beta-endorphin concentrations were measured as well as the glycogen concentration in

Is the "K-type" caffeine-halothane responder susceptible to malignant hyperthermia?

The "K-type" designation is used to describe a patient being investigated for malignant hyperthermia (MH) when concurrent administration of caffeine and halothane induces muscle contracture (rigidity, spasm) in vitro, but when halothane and caffeine given separately produce a normal response. It is accepted in some centres that K-type individuals are susceptible to malignant hyperthermia (MHS). In this paper, the K-type is shown not to correlate with the MH susceptible (MHS) status as accepted by the European MH group.

Closed system delivery of halothane and isoflurane with a vaporizer in the anesthetic circle.

Forty-four healthy dogs undergoing elective ovariohysterectomy were anesthetized with halothane or isoflurane delivered with an in-circuit vaporizer with closed system flow rates or an out-of-circuit vaporizer with semi-closed system flow rates. When dogs were anesthetized with halothane, there were no differences in heart rate, blood pressure, body temperature, respiratory rate, or lingual venous pH, PCO2, or PO2 during induction and maintenance. Lingual venous PO2 was significantly less but still within a clinically acceptable range when isoflurane was used in an in-circuit vaporizer. Recove

Isoniazid potentiation of a guinea pig model of halothane-associated hepatotoxicity.

Isoniazid (INH) is a selective inducer of cytochrome P-450 isozymes that are involved in the biotransformation of organohalogen anesthetics. It has been used to produce a rat model of halothane-associated hepatotoxicity that was linked to enhanced oxidative biotransformation of the anesthetic. Guinea pigs were pretreated with INH in order to potentiate halothane-induced hepatic necrosis and to study the oxidative pathway as a hepatotoxic mechanism in this species. The animals received either 12.5, 25.0 or 50.0 mg kg-1 INH i.p. for 7 days. Following halothane exposure, there were dose-dependent

[Occupational exposure of the anesthesiologist to nitrous oxide and halothane. Control measures].

Anesthetic gas spillage does occur in the operating room. Anesthesiologists and other operating room personnel are likely exposed to health hazards caused by chronic exposure to anesthetic gases and vapours. The aim of the present study is to elucidate the possible relationship between the chronic exposure to anesthetic gases and their adverse effects on the health. Present data, while controversial, do not establish a cause-effect relationship. Nevertheless, until solid data refutes the existence of a health hazard for operating room personnel, the anesthesiologist must try to reduce the anes

Effect of suxamethonium, pancuronium, neostigmine and halothane on serum cholinesterase activity.

Serum cholinesterase (Ch E) estimations were done in 30 healthy Nigerians scheduled for elective surgical procedures and were studied in two groups. Half of the patients were given general anesthesia using a muscle relaxant technique and the other half had halothane as the main anesthetic. Suxamethonium reduced Ch E by 9%, pancuronium by 25% and neostigmine by 58% compared to preinduction levels. Halothane reduced Ch E by 7% and 15% at 10 minutes and 24 hours postanesthetic respectively, compared to preinduction levels. The normal control value of Ch E in 30 healthy Nigerians was on the lower

A comparison of alfentanil, halothane and enflurane as supplements for outpatient urological surgery.

Seventy-seven patients presenting for outpatient cystoscopy participated in a trial to assess postoperative recovery when either alfentanil, halothane, or enflurane were used in combination with nitrous oxide/oxygen anaesthesia. Anaesthesia was uneventful in all cases. Apnoea occurred once with alfentanil, but naloxone was not required. Vomiting occurred once with alfentanil and once with enflurane. Anti-emetics were not required. Blood pressure and pulse rate variations from preoperative levels occurred with similar frequency in all groups. Times to open eyes, show left thumb, and give correc

The Triservice anaesthetic apparatus. Trial of isoflurane and enflurane as alternatives to halothane.

Sixty male patients undergoing limb surgery were anaesthetised using a drawover technique with the Triservice apparatus. They were randomly allocated to receive trichloroethylene and one of three other volatile agents (halothane, enflurane or isoflurane) after thiopentone induction. Signs of inadequate anaesthesia were noted. The incidence of such signs was not significantly different in the three groups. Similarly, no qualitative difference could be demonstrated in the immediate recovery, but the recovery time was significantly shorter with enflurane.

Antiarrhythmic effect of diltiazem during halothane anesthesia in dogs and in humans.

The antiarrhythmic effects of diltiazem (DL), a slow channel inhibitor, were evaluated in the presence of epinephrine-halothane-induced arrhythmias in dogs, of premature ventricular contractions (PVCs) during anesthesia in patients (n = 10), and of tachyarrhythmias with associated atrial fibrillation (AF) during anesthesia in patients (n = 9). The arrhythmogenic dose of epinephrine (ADE) during one MAC of halothane in dogs was increased from 1.13 +/- 0.21 to 3.14 +/- 0.89 microgram X kg-1 X min-1 by the administration of 0.3 mg/kg of DL. This suggests that DL significantly increases the thresh

Cardiovascular and respiratory effects of acetylpromazine and xylazine on halothane-anesthetized horses.

Circulatory and respiratory effects of intravenously administered acetylpromazine (0.033 and 0.067 mg/kg) and xylazine (0.5 and 1.0 mg/kg) were studied in drug cross-over fashion in eight laterally recumbent horses anesthetized only with halothane (1.06%, end-tidal) in O2. Both doses of acetylpromazine caused a significant and sustained elevation in cardiac output via a rise in stroke volume. Xylazine produced an initial significant fall in cardiac output followed by a return to control levels. Halothane anesthesia did not prevent xylazine-related atrioventricular conduction block. Site of selective action of halothane on the peripheral chemoreflex pathway in humans.

Halothane in humans depresses the ventilatory response to hypoxemia in a manner that suggests a selective action on one or more components of the peripheral chemoreflex arc. To test the hypothesis that this action is at the carotid bodies themselves, the authors studied the ventilatory response to subanesthetic concentrations of halothane (0.15-0.30% inspired) in six fit volunteers maintained in a steady state of isocapnic hypoxemia (PEO2 50 mmHg). Upon exposure to halothane, hypoxemia-driven ventilation decreased promptly and progressively (from 7.5 +/- 1.2 1 X min-1 X m-2 in the control stat

Effects of induced hypotension on breathing pattern in halothane-anaesthetized man.

The effects of hypotension induced by trimetaphan on ventilatory control were assessed in sixteen normal subjects under halothane anaesthesia. The breathing pattern, mouth occlusion pressures, lung mechanics, acid-base balance, and arterial blood gases were analysed before and during trimetaphan infusion. During induced hypotension, the only significant change in the ventilatory variables was an increase in the ratio of inspiratory duration to total cycle duration from 0.39 +/- 0.05 (SD) to 0.42 +/- 0.03; P less than 0.01. The average minute ventilation remained unchanged. No modification in l

Modification of ryanodine toxicity by dantrolene and halothane in a model of malignant hyperthermia.

Ryanodine toxicity in animals has been suggested to constitute a model of malignant hyperthermia. Dantrolene is known to block the development of malignant hyperthermia triggered by halothane in susceptible swine. The authors studied the influences of dantrolene and halothane on the effects of ryanodine in vitro in isolated rat diaphragm muscle segments, and in vivo in mice, to explore the validity of this model. In the diaphragm experiments, dantrolene was found to block or delay the development of contractures produced by ryanodine and to delay the potentiation of ryanodine-induced contractu

An animal model of halothane hepatotoxicity: roles of enzyme induction and hypoxia.

Exposure of phenobarbital-pretreated male Sprague-Dawley rats to halothane, 1 per cent, for two hours under conditions of hypoxia (FIO2 0.14) resulted in extensive centrilobular necrosis within 24 hours. Accompanying the morphologic damage were an increase in serum glutamic pyruvic transminase (SGPT) and a decrease in hepatic microsomal cytochrmoe P-450. Glutathione levels in the liver were unchanged. Phenobarbital-pretreated rats anesthetized with halothane, 1 per cent, at FIO2 0.21 had only minor morphologic changes at 24 hours. Hepatic injury was not apparent in any non-phenobarbital-induce

The different responses of the hepatic arterial bed to hypovolaemia and to halothane anaesthesia.

Ten dogs were subjected to a period of hypovolaemia (bleeding volume: 2% of body weight) and to a period of halothane anaesthesia (end-tidal halothane concentration: 1%). Mean arterial blood pressure decreased to 79% of control value during hypovolaemia and to 58% of control value during halothane anaesthesia. Mean total peripheral and preportal vascular resistances increased during hypovolaemia and were unchanged during halothane. Mean hepatic arterial and portal venous blood flows decreased to 82% and 55% of control values, respectively, during hypovolaemia, and to 41% and 56% of control val
