Interaction between neuromuscular junction metabolic requirements in fragile X syndrome and glycogen storage disease models.

A classic human patient comorbidity of fragile X syndrome and glycogen storage disease type IX has symptoms far more severe than those for either disease alone. Causal variants result in loss of the translational regulator fragile X ribonucleoprotein 1 (FMRP) and metabolic regulator phosphorylase kinase regulatory subunit alpha 2 (PHKA2), respectively. We hypothesized FMRP-PHKA2 interaction based on unsustainably elevated metabolic demand. In Drosophila disease models, single null mutants were viable, but double knockout (DKO) animals exhibited 100% synthetic lethality, showing an essential in

A review of risk factors at the human-animal-environmental interface of garbage dumps that are driving current and emerging zoonotic diseases.

An increasing trend in zoonotic and emerging infectious diseases (EIDs) has been observed worldwide. Most EID outbreaks originate from wildlife, and these outbreaks often involve pathogen-host-environment interaction. Garbage dumps act as an interface between humans, animals, and the environment, from which EIDs could arise. Therefore, this review considers the presence of important pathogens associated with animals and vectors at garbage dumps from a One Health perspective, looking at animal, human, and environmental factors that play a role. A narrative review was performed focusing on four

Progress and obstacles in vaccine development for the ehrlichioses.

Ehrlichia are tick-borne obligately intracellular bacteria that cause significant diseases in veterinary natural hosts, including livestock and companion animals, and are now considered important zoonotic pathogens in humans. Vaccines are needed for these veterinary and zoonotic human pathogens, but many obstacles exist that have impeded their development. These obstacles include understanding genetic and antigenic variability, influence of the host on the pathogen phenotype and immunogenicity, identification of the ehrlichial antigens that stimulate protective immunity and those that elicit i

Risk Perception and Psychosocial Factors Influencing Exposure to Antimicrobial Resistance through Environmental Pathways in Malawi.

Antimicrobial-resistant (AMR) bacteria are prevalent in household and environmental settings in low-income locations. However, there are limited data on individuals' understanding of AMR bacteria exposure risks in these settings. A cross-sectional study was conducted to identify individual risk perception of AMR bacteria and its associated behavioral determinants at the household level in urban, peri-urban, and rural Malawi. We conducted interviews with 529 participants from 300 households (n = 100 households/site). The risk, attitude, norms, ability, and self-regulation model was used to asse

Barbering (fur and whisker trimming) by laboratory mice as a model of human trichotillomania and obsessive-compulsive spectrum disorders.

Animal diseases that develop spontaneously in a limited subpopulation can provide powerful models of human disease because they provide a means to investigate the interaction of a broad range of biological and environmental etiologic processes. In contrast, with experimentally induced animal models, the etiology of the model is inherently fixed, and can only speak to a limited subset of those involved in the human disease. 'Barbering' (abnormal whisker- and fur-plucking behavior) in mice resembles human trichotillomania (compulsive hair plucking) in that barbering mice pluck focused areas of h

Why it is easier to date Quaternary extinctions than human colonizations under conditions of overkill.

Faunal extinctions correlate with human colonization worldwide during the Quaternary, yet the extent to which human hunting drove these losses remains debated. Resolving these debates depends on accurately dating both human arrival and animal extinction events. Herein I present a simulated overkill scenario modeling interaction between a founding human population and a single prey species. The simulation evaluates expected age offsets, the differences between the actual and observed dates, for extinction and colonization as functions of sample size and human population growth rate. Results sho

Evolution of Bordetellae from Environmental Microbes to Human Respiratory Pathogens: Amoebae as a Missing Link.

The genus Bordetella comprises several bacterial species that colonize the respiratory tract of mammals. It includes B. pertussis, a human-restricted pathogen that is the causative agent of Whooping Cough. In contrast, the closely related species B. bronchiseptica colonizes a broad range of animals as well as immunocompromised humans. Recent metagenomic studies have identified known and novel bordetellae isolated from different environmental sources, providing a new perspective on their natural history. Using phylogenetic analysis, we have shown that human and animal pathogenic bordetellae hav

Porcine retinal cell line VIDO R1 and Chlamydia suis to modelize ocular chlamydiosis.

Human ocular Chlamydia trachomatis infections can lead to trachoma, the major cause of infectious blindness worldwide. Trachoma control strategies are very helpful but logistically challenging, and a trachoma vaccine is needed but not available. Pigs are a valuable large animal model for various immunological questions and could facilitate the study of human ocular chlamydial infections. In addition, a recent study identified the zoonotic potential of Chlamydia suis, the natural pathogen of pigs. In terms of the One Health Initiative, understanding the host-pathogen-interactions and finding a

[Oxytocin versus dopamine: The delicate balance between social bonding and addiction].

Social behaviour is fundamental to survival and adaptation, relying on complex, interacting neurobiological systems in which the neuropeptide oxytocin (OT) and the neurotransmitter dopamine (DA) play pivotal roles in regulating social bonding, motivation, and reward processing. This review synthesises recent advances in understanding the bidirectional interactions between OT and DA within key nodes of the brain's reward circuitry, including the nucleus accumbens, ventral tegmental area, amygdala, and medial prefrontal cortex. We examine how these interactions support a wide range of social beh

HIDmap: a physiology-based map of human immune system development to support animal-free research and regulatory innovation.

Human immune system development is established through a sequence of tightly regulated processes occurring across multiple embryonic and fetal organs. Despite extensive research, our mechanistic understanding still relies largely on animal models, which often lack translational precision for human biology. This gap is particularly relevant for developmental immunotoxicity (DIT), where early-life exposure to environmental contaminants such as dioxins, PCBs, and PFAS has been associated with long-term immune dysfunction, including reduced vaccine responsiveness in children. To address this need,

Molecular detection, phylogenetic and whole genome sequence analyses of Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) Delta variant isolated from domestic cats (Felis catus) in Egypt.

Reverse zoonoses, which arise from interactions between humans and animals, are facilitated by the similarity of angiotensin-converting enzyme 2 (ACE2) cell receptors across different hosts and the high mutation rate of Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), enabling transmission across species. This study aimed to explore the transmission of SARS-CoV-2 between humans and companion animals by conducting molecular investigations on nasopharyngeal and oropharyngeal swabs from 100 domestic Cats (Felis catus) in Egypt which had contact with SARS-CoV-2 positive owners from De

Seroprevalence of Brucella Infection and Its Determinants Among Pregnant Women Attending Antenatal Care in Burao City: At the Human Livestock Interface in Northeast Somaliland, Somalia.

Background: Brucellosis typically spreads from animals to humans through contact with infected animals or their byproducts. This zoonotic disease can have serious consequences and is often caused by contact with infected livestock or their products, such as contaminated dairy, posing significant risks during pregnancy. This study aimed to evaluate the prevalence of Brucella infection among pregnant women residing in the Burao City area of northeast Somaliland, in environments where human-animal interaction is a frequent occurrence. Therefore, it is important to identify the factors that contri

Neophilia in wolves and dogs.

The study of domestication provides a unique opportunity to analyze the effects of natural selection in the attraction towards novelty, because selective pressures on domesticated animals are often greatly diminished or different from their wild counterparts. In this study, we investigated the neophilic levels of three groups of canids differing in their level of contact with human environments both from an evolutionary and ontogenetic perspective: wolves, pack-living dogs, and pet dogs. In order to study their neophilic response, we presented the animals with two objects. The first of these o

Neglected Facts on Mycobacterium Avium Subspecies Paratuberculosis and Type 1 Diabetes.

Civilization factors are responsible for the increasing of human exposure to mycobacteria from environment, water, and food during the last few decades. Urbanization, lifestyle changes and new technologies in the animal and plant industry are involved in frequent contact of people with mycobacteria. Type 1 diabetes is a multifactorial polygenic disease; its origin is conditioned by the mutual interaction of genetic and other factors. The environmental factors and certain pathogenetic pathways are shared by some immune mediated chronic inflammatory and autoimmune diseases, which are associated

The emerging role of the meningeal lymphatic and glymphatic systems in migraine pathophysiology: a systematic review.

BACKGROUND/OBJECTIVE: Migraine is a common, debilitating neurological disorder of unclear pathophysiology. Recent evidence has implicated impaired meningeal lymphatic function and its interaction with the glymphatic system in the development of neuroinflammation and pain sensitization. This systematic review aimed to summarize existing evidence on the role of the meningeal lymphatic and glymphatic systems in migraine pathophysiology. METHODS: Following the PRISMA 2020 recommendations, we searched PubMed, Web of Science, and Scopus from inception to December 15, 2025. Inclusion criteria compri

Interactions between endosulfan and dieldrin on estrogen-mediated processes in vitro and in vivo.

There is growing concern that estrogenic chemicals, both natural and human-made, may be causing a variety of reproductive disorders in wildlife and human populations. Recent in vitro data suggest that the interaction between some weakly estrogenic organochlorines, dieldrin, endosulfan, toxaphene, and chlordane, causes a synergistic increase in their estrogenic potency, an effect due to joint action on estrogen receptors (ER). As these studies were conducted using models of estrogen action derived from cells that are not physiologically controlled by estrogens, the relevance of these findings t

Neural systems underlying episodic memory: insights from animal research.

Two strategies used to uncover neural systems for episodic-like memory in animals are discussed: (i) an attribute of episodic memory (what? when? where?) is examined in order to reveal the neuronal interactions supporting that component of memory; and (ii) the connections of a structure thought to be central to episodic memory in humans are studied at a level of detail not feasible in humans. By focusing on spatial memory (where?) and the hippocampus, it has proved possible to bring the strategies together. A review of lesion, disconnection and immediate early-gene studies in animals reveals t

The Effects of Human Presence, Restraint, and Stressed Neighbors on Corticosterone Levels in Domesticated Budgerigars (Melopsittacus undulatus).

Limiting stress during interactions between captive animals and humans is important for effective husbandry. One physiological change during the vertebrate stress response is the release of glucocorticoid hormones. Here, we measured plasma corticosterone in female domesticated budgerigars (Melopsittacus undulatus) to test whether human presence, restraint, or removal and return of a newly stressed neighbor increases corticosterone. The presence of humans for 15 min at the beginning of the experiment did not significantly elevate corticosterone above baseline levels, suggesting that birds that

Should a lower treatment line be used when treating paracetamol poisoning in patients with chronic alcoholism?: a case for.

A lower threshold for treatment of paracetamol (acetaminophen) poisoning has been advocated in chronic heavy users of alcohol, based originally on animal studies indicating that chronic alcohol ingestion increased hepatotoxicity. This was attributed to increased production of the toxic metabolite, N-acetyl-p-benzoquinoneimine, by cytochrome P450 (CYP)2E1 induction. The clinical evidence for increased risk is limited to four retrospective studies with potential for referral and reporting bias and conflicting results. No study has specifically addressed the issue of the treatment threshold for a
