Detection of hydroxyl and carbon-centred radicals by EPR spectroscopy after ischaemia and reperfusion of the rat kidney.

Recent studies suggest that oxygen-derived free radicals are involved in mediating renal reperfusion injury. EPR spectroscopy and spin trapping with the spin traps DMPO and PBN, were used to detect and quantitate the formation of hydroxyl radicals in rat kidney after ischaemia-reperfusion in vivo and in vitro in the isolated rat kidney perfused in the absence of leucocytes. EPR analysis of homogenised kidneys and of venous samples did not detect radical adducts with either spin trap. With PBN, radical adducts were not detected in vitro. When DMPO was used as the spin trap in kidneys perfused w

Defect-Mediated Charge Transfer in Y-Cd-Codoped ZnO: Enhanced Hydroxyl Radical Generation and Solar Photocatalytic Carbaryl Degradation.

In this study, yttrium-cadmium codoped ZnO (YCdZnO) photocatalysts were synthesized and investigated for the sunlight-excited photocatalytic degradation of carbaryl (CBR). The photocatalytic degradation assays showed a pseudo-first-order degradation rate constant of 12.38 × 10-3 min-1, approximately 2.08 times that of bare ZnO (5.95 × 10-3 min-1). The photocatalysts also retained over 48% degradation efficiency (DE) after four cycles. Scavenger tests indicated that hydroxyl radicals were the dominant reactive species during CBR degradation. Characterization of YCdZnO, including vibrational and

A near-infrared ratiometric optical sensor for real-time monitoring hydroxyl radical levels in ferroptosis-mediated Parkinson's disease.

Parkinson's disease (PD) is a common neurodegenerative disorder with complex pathogenesis. Ferroptosis, an iron-regulated cell death driven by lipid peroxidation, is closely linked to PD progression. Excessive hydroxyl radical (•OH) accumulation can cause severe neural damage and is associated with neurodegenerative diseases, but its role in PD remains unclear. Currently, there are no fluorescent probes to monitor •OH changes during ferroptosis and PD. Here, we developed a series of •OH-responsive ratiometric near-infrared (NIR) sensors (DCM-PD1∼8) to monitor •OH levels in ferroptosis-mediated

Synergistic induction of hydroxyl radical-induced DNA single-strand breaks by chromium(VI) compound and cigarette smoke solution.

Chromium(VI) compounds and cigarette smoke are known human carcinogens. We found that K2Cr2O7 and cigarette smoke solution synergistically induced DNA single-strand breaks (0.23+/-0.04 breaks per DNA molecule) in pUC118 plasmid DNA. K2Cr2O7 alone or cigarette smoke solution alone induced much less strand breaks (0.03+/-0.01 or 0.07+/-0.02 breaks per DNA molecule, respectively). The synergistic effect was prevented by catalase and by hydroxyl radical scavengers such as deferoxamine, dimethylsulfoxide, d-mannitol, and Tris, but not by superoxide dismutase. Ascorbic acid enhanced the synergism. G

A Low-Oxidation-State Mechanism for O─O Bond Formation in Photosystem II via Oxo-Hydroxyl Radical Coupling.

In this computational work, we identify an oxo-hydroxyl radical coupling (OHC) mechanism for O─O bond formation in PSII within the low-oxidation-state (LOS) paradigm, in which the Ca-bound W3 water and O5 act as the substrates. While several recent crystallographic and computational studies indicate that the Ox ligand is absent in the S3 state, we show that it forms upon entry into the S4 state through migration of the Ca-bound water ligand W3 into the cavity between Mn1 and Ca2 +. From this intermediate, Ox─O5 bond formation proceeds via the OHC mechanism rather than the conventional oxo-oxyl

NIR-Triggered Hydroxyl Radical Storm: A Novel Phototherapy Nanoplatform for Potent Ferroptosis‑Like Cell Death Induction in Tumors.

The development of organic photocatalysts holds considerable promise for the oxidation of NADH, thereby disrupting the NADH/NAD+ equilibrium, modulating redox homeostasis, and enhancing treatment of hypoxic tumors. However, there is still a lack of organic photocatalyst which could respond to near-infrared light. Herein, we develop an NIR-triggered organic photocatalyst (TTH) that produces hydroxyl radical and photooxidates NADH. Upon 808 nm light irradiation, TTH not only could generate hydroxyl radical in an optimized pathway of O2→O2 • -→H2O2→•OH and achieve effective oxidation of NADH and

Blockade of the renin-angiotensin system suppresses hydroxyl radical production in the rat striatum during carbon monoxide poisoning.

Oxidative stress has been suggested to play a role in brain damage during carbon monoxide (CO) poisoning. Severe poisoning induced by CO at 3000 ppm, but not 1000 ppm, enhances hydroxyl radical (˙OH) production in the rat striatum, which might be mediated by NADPH oxidase (NOX) activation associated with Ras-related C3 botulinum toxin substrate (Rac) via cAMP signaling pathway activation. CO-induced ˙OH production was suppressed by antagonists of angiotensin II (AngII) type 1 receptor (AT1R) and type 2 receptor (AT2R) but not an antagonist of the Mas receptor. Suppression by an AT1R antagonist

Endogenous 3,4-dihydroxyphenylalanine and dopaquinone modifications on protein tyrosine: links to mitochondrially derived oxidative stress via hydroxyl radical.

Oxidative modifications of protein tyrosines have been implicated in multiple human diseases. Among these modifications, elevations in levels of 3,4-dihydroxyphenylalanine (DOPA), a major product of hydroxyl radical addition to tyrosine, has been observed in a number of pathologies. Here we report the first proteome survey of endogenous site-specific modifications, i.e. DOPA and its further oxidation product dopaquinone in mouse brain and heart tissues. Results from LC-MS/MS analyses included 50 and 14 DOPA-modified tyrosine sites identified from brain and heart, respectively, whereas only a f

Electrochemiluminescence sensors for scavengers of hydroxyl radical based on its annihilation in CdSe quantum dots film/peroxide system.

This work elucidated the detailed electrochemiluminescence (ECL) process of the thioglycolic acid-capped CdSe quantum dots (QDs) film/peroxide aqueous system. The QDs were first electrochemically reduced to form electrons-injected QDs approximately -1.1 V, which then reduced hydrogen peroxide to produce OH* radical. The intermediate OH* radical was a key species for producing holes-injected QDs. The ECL emission with a peak at -1.114 V was demonstrated to come from the 1Se-1Sh transition emission. Using thiol compounds as the model molecules to annihilate the OH* radical, their quenching effec

Electron paramagnetic resonance evidence of hydroxyl radical generation and oxidative damage induced by tetrabromobisphenol A in Carassius auratus.

Tetrabromobisphenol A (TBBPA) is one of the most widely used brominated flame retardants (BFRs). To confirm its putative oxidative stress-inducing activity, freshwater fish Carassius auratus were injected intraperitoneally with TBBPA. One experiment lasted 3h to 28 days after a single injection of 100mg/kg TBBPA, and the other lasted 24h after a single injection of 0-300 mg/kg TBBPA. Reactive oxygen species (ROS) were trapped by phenyl-tert-butyl nitrone (PBN) and detected by electron paramagnetic resonance (EPR). Protein carbonyl (PCO) and lipid peroxidation product (LPO) content were also de

Allopurinol suppresses para-nonylphenol and 1-methyl-4-phenylpyridinium ion (MPP(+))-induced hydroxyl radical generation in rat striatum.

We recently demonstrated that para-nonylphenol, an environmental estrogen-like chemical, enhances hydroxyl radical (*OH) generation in the rat striatum. In the present study we have examined whether para-nonylphenol enhanced 1-methyl-4-phenylpyridinium ion (MPP(+))-induced *OH generation in the rat striatum using a microdialysis technique. Para-nonylphenol significantly enhanced MPP(+)-induced *OH generation. Further, we studied the effect of allopurinol, a xanthine oxidase inhibitor, on para-nonylphenol and MPP(+)-induced *OH generation. Allopurinol significantly suppressed para-nonylphenol a

Hydroxyl radical formation in chondrocytes and cartilage as detected by electron paramagnetic resonance spectroscopy using spin trapping reagents.

Chondrocytes have been shown to produce superoxide and hydrogen peroxide, suggesting possible formation of hydroxyl radical in these cells. In this study, we used electron spin resonance/spin trapping technique to detect hydroxyl radicals in chondrocytes. We found that hydroxyl radicals could be detected as alpha-hydroxyethyl spin trapped adduct of 4-pyridyl 1-oxide N-tert-butylnitrone (4-POBN) in chondrocytes stimulated with phorbol 12-myristate 13-acetate in the presence of ferrous ion. The formation of hydroxyl radical appears to be mediated by the transition metal-catalyzed Haber-Weiss rea

Effect of antithyroid drugs on hydroxyl radical formation and alpha-1-proteinase inhibitor inactivation by neutrophils: therapeutic implications.

The release of proteolytic enzymes and generation of strong oxidants such as the hydroxyl radical by activated neutrophils has been proposed to play an important role in mediating toxin-induced liver injury. The antithyroid drug propylthiouracil protects against liver injury induced by many hepatotoxic agents and markedly reduces mortality in patients with alcoholic liver disease. However, the mechanism(s) by which propylthiouracil protects against liver injury is not well understood. The present studies investigate the effect of antithyroid drugs on proteolytic enzyme activity and on hydroxyl

Hydroxyl and 1-hydroxyethyl free radical detection using spin traps followed by derivatization and gas chromatography-mass spectrometry.

Detection of hydroxyl free radicals is frequently performed by electron spin resonance (ESR) following spin trapping of the radical using 5,5-dimethylpyrroline N-oxide (DMPO) to generate a stable free radical having a characteristic ESR spectrum. The necessary ESR equipment is expensive and not readily available to many laboratories. In the present study, a specific and sensitive gas chromatography-mass spectrometry (GC/MS) method for detection of hydroxyl and hydroxyethyl free radicals is described. The DMPO or N-t-butyl-alpha-phenylnitrone (PBN) radical adducts are extracted and derivatized

A high performance liquid chromatography system for quantification of hydroxyl radical formation by determination of dihydroxy benzoic acids.

The hypoxanthine/xanthine oxidase enzyme system is known to produce the superoxide ion and hydrogen peroxide during the hydroxylation of hypoxanthine via xanthine to uric acid. When chelated iron is included in this system, superoxide reduces iron (III) to iron(II) and the iron(II)-chelate further reacts with hydrogen peroxide to form the highly reactive hydroxyl radical. Because of the limitations of colourimetric and spectrophotometric techniques by which, to date, the mechanisms of hydroxyl radical formation in the hypoxanthine/xanthine oxidase system have been monitored, a high performance

Asymmetric Zn─N3O1 Single-Atom Sites Promote Hydroxyl Radical Generation for Natural-Light-Driven Inactivation of Drug-Resistant Bacteria.

Waterborne drug-resistant bacteria pose a serious global health threat, underscoring the urgent need for highly efficient disinfection strategies. The photocatalytic generation of hydroxyl radicals (•OH) represents a green and powerful route for bacterial inactivation, yet its efficiency is often limited by the sluggish kinetics of the stepwise single-electron oxygen reduction pathway. Herein, we report an atomic-level design strategy by anchoring asymmetric Zn─N3O1 sites onto ultrathin graphitic carbon nitride nanosheets (Zn1/OCN) to accelerate •OH production. The introduced Zn─N3O1 sites cre

Hydroxyl radical footprinting modification reveals an intradomain communication pathway in EFL1 disrupted by a Shwachman-Diamond syndrome-associated mutation.

Shwachman-Diamond syndrome (SDS) is a rare genetic disorder characterized by pancreatic insufficiency and neutropenia. While most cases are linked to mutations in the SBDS gene, some involve mutations in the GTPase EFL1. This protein works with SBDS to release the anti-association factor eIF6 from the 60S ribosomal subunit during ribosome biogenesis. The pathogenic EFL1 R1095Q mutant (R1086Q in yeast) exhibits altered guanine nucleotide recognition and impaired eIF6 release, prompting an investigation into its structural consequences. Using the yeast Efl1 orthologue in X-ray hydroxyl radical f

Sensitive fluorescence turn-on sensing of hydroxyl radical and glucose based on the oxidative degradation of reductive organic cage.

The accurate and sensitive quantification of hydroxyl radical (·OH) and glucose is necessary for disease diagnosis and health guidance, but still challenging owing to the low concentration of ·OH and poor water solubility of fluorescent probes. In addition, fluorescent probes may cause secondary pollution to the environment. Here an organic cage was reported as a sensitive fluorescent probe for ·OH and glucose in aqueous solution without serious secondary pollution. The prepared organic cage with good water solubility showed specific redox affinity to ·OH in acidic condition, resulting in two

Rapid Photolysis of Gaseous Organic Nitrates Formed from Hydroxyl and Nitrate Radical Oxidation of α‑Pinene and β‑Pinene.

Photolysis of gaseous organic nitrates is crucial for understanding the formation and fate of air pollutants such as nitrogen oxides (NO x ) and ozone (O3). Monoterpenes are prevalent biogenic volatile organic compounds (VOCs), greatly contributing to the formation of organic nitrates; however, there is currently a lack of experimental constraints on the photolysis chemistry of monoterpene organic nitrates. Here, we investigated the photolysis of monoterpene organic nitrates via novel dual chamber experiments, in which a large suite of organic nitrates was formed from hydroxyl and nitrate radi
