Innate and adaptive immune responses are tolerized in chimeras prepared with nonmyeloablative conditioning.

BACKGROUND: Mixed chimerism is an effective approach for tolerance induction in transplantation. Strategies to achieve mixed chimerism with relatively low toxicity have significantly expanded the clinical use of chimerism. METHODS: Allogeneic bone marrow transplants were performed between B6 (H2(b)) and BALB/c (H2(d)) mice. Recipient B6 mice were nonmyeloablatively conditioned with anti-αβ-T-cell receptor, anti-CD154, or rapamycin alone or in different combinations. A total of 15 × 10(6) BALB/c bone marrow cells were transplanted after varying doses of cGy of total body irradiation. RESULTS:

Microchimerism does not induce tolerance and sustains immunity after in utero transplantation.

BACKGROUND: To date, over 40 in utero transplants have been performed in humans; the only successes were documented in the treatment of severe combined immunodeficiency syndromes. Hemoglobinopathies and metabolic disorders are candidate diseases for this approach; however, when applied clinically, the results have been discouraging. To address the role of the fetal immune system in the outcome of in utero transplantation, we have developed a murine model of in utero transplantation in immunologically intact murine recipients and have studied chimerism and tolerance/immunity to allogeneic donor

Pancreatic islet allograft immunity and tolerance: the two-signal hypothesis revisited.

The principle assumption of this discussion is that costimulation (CoS) forms the primary stimulus that compels T cells to mount a response to their specific antigen. However, this response can be either positive or negative, depending on the developmental stage of the T cell and the microenvironment in which the antigen and CoS are received. Thus, both immunity and tolerance may represent different outcomes of a two-signal process. We would emphasize that CoS is a functional term and not a strict molecular definition. While many molecular interactions have been described as providing CoS acti

Chronic Myelomonocytic Leukemia Microenvironment in Disease Progression and Therapy: Focus on Stepwise Reconfiguring of Myeloid-Immune Cell Interactions Driving Immune Tolerance.

PURPOSE OF THE REVIEW: To summarize evidence and provide an overview of the microenvironment and myeloid-immune cell interactions within the chronic myelomonocytic leukemia (CMML) microenvironment that drive mutation-agnostic pathways of disease progression. RECENT FINDINGS: Recent work has demonstrated the importance of myeloid-immune cell interactions in CMML and related myeloid neoplasms. In particular, we focus on the bone marrow microenvironment of CMML, highlighting the role of clonal dendritic cell aggregates and their interaction with other myeloid cells such as monocytes and their pr

The Effects of Treatment with Icariin on Immune Tolerance in the Recurrent Spontaneous Abortion Mice.

Recurrent spontaneous abortion (RSA) is the most common pregnancy-related complication, affecting 1-5% of pregnancies. Currently, immune imbalance at the maternal-fetal interface is one of the main causes of recurrent abortion. Icariin (ICA) can exert immunomodulatory effects in a variety of autoimmune diseases. Nevertheless, it has not been reported for use in recurrent abortion. In this study, to clarify the effects and mechanisms of ICA for recurrent abortion, female mice CBA/J were randomly divided into Normal group, RSA group and RSA + ICA group. From 0.5 days of pregnancy to 12.5 days, t

Plac1 Is a Key Regulator of the Inflammatory Response and Immune Tolerance In Mammary Tumorigenesis.

Plac1 is an X-linked trophoblast gene expressed at high levels in the placenta, but not in adult somatic tissues other than the testis. Plac1 however is re-expressed in several solid tumors and in most human cancer cell lines. To explore the role of Plac1 in cancer progression, Plac1 was reduced by RNA interference in EO771 mammary carcinoma cells. EO771 "knockdown" (KD) resulted in 50% reduction in proliferation in vitro and impaired tumor growth in syngeneic mice; however, tumor growth in SCID mice was equivalent to tumor cells expressing a non-silencing control RNA, suggesting that Plac1 re

Duality at the gate: Skin dendritic cells as mediators of vaccine immunity and tolerance.

Since Edward Jenner's discovery that intentional exposure to cowpox could provide lifelong protection from smallpox, vaccinations have been a major focus of medical research. However, while the protective benefits of many vaccines have been successfully translated into the clinic, the cellular and molecular mechanisms that differentiate effective vaccines from sub-optimal ones are not well understood. Dendritic cells (DCs) are the gatekeepers of the immune system, and are ultimately responsible for the generation of adaptive immunity and lifelong protective memory through interactions with T c

The role of immune tolerance induction in restoration of the efficacy of ERT in Pompe disease.

Pompe disease is a lysosomal storage disorder caused by deficiency in the enzyme acid α-glucosidase (GAA). Pompe disease is characterized by the accumulation of glycogen, predominantly in muscle tissue, leading to progressive muscle weakness, loss of motor, respiratory, and, in the infantile-onset form, cardiac function. Disease progression is highly variable depending on phenotype, but premature death due to respiratory complications occurs in most patients. Beginning in 2006, approved alglucosidase alfa enzyme replacement therapies [recombinant human (rh) GAA] have been available to treat Po

Staphylococcal enterotoxin B compromises the immune tolerant status in the airway mucosa.

BACKGROUND: The breakdown of immune tolerance plays a critical role in allergic disorders; the mechanism of breaching immune tolerance remains largely unknown. OBJECTIVE: The present study aimed to investigate the role of Staphylococcal enterotoxin B (SEB) in the interference of the immune tolerance in the nasal mucosa. METHODS: The immune tolerant components, tolerogenic dendritic cells (TolDC) and regulatory T cells (Treg), were assessed in the surgically removed nasal mucosa from patients with allergic rhinitis (AR) or non-AR chronic rhinitis. The contents of SEB and integrin alphavbeta6

[A comprehensive study of the neurochemical and immune mechanisms of morphine tolerance: the effects of naloxone].

To test the authors' hypothesis about the role of endopeptidase (enkephalinase A, in particular) in mechanisms of morphine tolerance and blocking action of small doses of naloxone, they studied nociception reactions, morphine antibodies titres and enkephalinase A activity after morphine, d-phenylalanine and naloxone injection in brain structures. It is shown that activity of enkephalinase A in structures of endogenous antinociceptive system increased simultaneously with morphine antibodies titres in tolerance condition. Injection of small dose naloxone inhibited enkephalinase activity in brain

Thoughts on the thymus and the T-lymphocyte repertoire. Relevance to the tolerance of the immune response.

The thymus is a central lymphoid organ, and has one important function in the immune system. It generates immunocompetent T (= thymus-dependent) cells. 'Immunocompetence' is the single most important characteristic acquired during the differentiation process of T-cells, from progenitor cell in the bone marrow to mature cell in peripheral lymphoid organs and blood. The basis of competence is recognition, mediated by the T-cell receptor on the cell surface. Using this receptor, T-cells recognize antigens only in the form of antigenic fragments complexed with molecules of the major histocompatibi

Induction of antigen-specific immunity and tolerance to Mycobacterium leprae in Lewis rats.

Intradermal (i.d.) immunization of Lewis rats with autoclaved Mycobacterium leprae resulted in antigen-specific proliferation responses and interleukin-2 release from spleen and lymph node cells that were detectable as early as 21 days, persisted for at least 9 months, and were dependent on the dose of antigen administered. Immunized animals were also completely resistant to a footpad challenge with viable M. leprae. In contrast, intravenous (i.v.) administration of at least 10(8) irradiated M. leprae isolates induced a state of nonresponsiveness characterized by the absence of proliferation a

Rescue immune tolerance induction with a recombinant factor Fc-fused VIII: prospective ReITIrate study of clinical, humoral and cellular immune responses.

BACKGROUND: Immune tolerance induction (ITI) is the gold standard for inhibitor eradication to restore the clinical efficacy of factor replacement therapy in haemophilia. However, as ITI often requires frequent administration over extended periods, it can be considered burdensome for patients and healthcare resources. Therefore, there is a need to optimise ITI treatment, particularly in patients who failed previous ITI attempts. OBJECTIVES: The ReITIrate study aimed to prospectively evaluate rescue ITI with efmoroctocog alfa, an extended half-life recombinant FVIII Fc fusion protein (herein r

Preparation of Hypoallergenic Enzymatic Hydrolysates from Crassostrea angulata with In Vitro Immune Tolerance-Inducing Potential.

Oyster allergy has emerged as a worldwide food safety concern, and enzymatic hydrolysis is an effective strategy for preparing hypoallergenic oyster products. In this study, crude extract from Crassostrea angulata was hydrolyzed with trypsin, Neutrase, and Flavorzyme. The IgE/IgG-binding activity of the hydrolysates was compared, structures and epitopes were explored, and immune tolerance-inducing potential was evaluated using a T-cell proliferation assay. Under optimized conditions (0.5 h reaction; 1.2% trypsin, 0.8% Neutrase, and 0.4% Flavorzyme), the hydrolysates exhibited markedly reduced

IL-2-producing TFH cells sustain Treg cell homeostasis and immune tolerance in the gut.

Follicular helper T (TFH) cells are well known for their role in supporting B cell responses. Here, we identify a critical B cell-independent function for TFH cells in sustaining regulatory T (Treg) cells within gut-associated lymphoid tissues (GALT). We demonstrate that TFH cells produce interleukin-2 (IL-2) at the T cell-B cell border and within interfollicular regions and that this localized IL-2 is required to maintain GALT Treg cell homeostasis and restrain the expansion of pathogenic effector T cells. Mice selectively lacking IL-2-producing TFH cells develop colitis and fatal multiorgan

TRAIL splice variant TRAILshort disrupts T cell receptor signaling and promotes immune tolerance in vivo.

TRAIL is a TNF family ligand that trimerizes TRAIL-R1 (DR4) or TRAIL-R2 (DR5) to induce apoptosis, necroptosis, and/or NF-κB activation in receptor-bearing cells. We previously identified TRAILshort as a splice variant of TRAIL that lacks cysteine 230, cannot trimerize, and acts as a dominant-negative ligand that blocks TRAIL-mediated apoptosis. TRAILshort is expressed on cell surfaces and within extracellular vesicles, enabling it to confer TRAIL resistance to both producing and bystander cells. In this study, we showed that elevated TRAILshort levels were associated with chronic viral infect

A Flagellin-S100A9 Fusion Vaccine Breaks Immune Tolerance and Suppresses Experimental Lung Metastasis in Mice.

Lung metastasis is a major cause of cancer-related mortality, and the pre-metastatic niche (PMN) provides a window for early intervention. Here, transcriptomic profiling of pre-metastatic lungs in a subcutaneous B16-F10 melanoma model identified S100a8 and S100a9 as strongly upregulated PMN-associated genes. Exploratory TCGA analyses further linked high S100A8 or S100A9 expression with poorer overall survival in selected cancer types, although most associations were attenuated after multivariable adjustment. To evaluate these molecules as vaccine targets, we engineered recombinant fusion prote

Current understanding and advances regarding the adipose-immune-metabolic axis in disease tolerance during sepsis.

Sepsis remains a critical global health challenge characterized by high mortality and morbidity, primarily due to the limitations of current pathogen-centric therapies and a poor understanding of host-defense mechanisms. This review synthesizes the pivotal role of the adipose-immune-metabolic axis as a central regulator of disease tolerance-a host defense strategy that limits tissue damage without directly reducing pathogen load. We delineate how adipose tissue is reprogrammed from a passive energy reservoir into an active immunometabolic hub during sepsis. This functional shift is governed by

Engineering Tregs-mediated immune tolerance via foxp3a overexpression to evade allograft transplantation barriers in zebrafish.

In mammals, regulatory T cells (Tregs) are widely exploited to promote immune tolerance in organ transplantation. In zebrafish, although germline stem cell (GSC) or gonadal primordium transplantation into immunodeficient hosts can accelerate gamete production, maintaining immunocompromised lines presents substantial practical challenges. To overcome this limitation, this study generates a Tg(CMV:foxp3a) zebrafish line through systemic overexpression of Forkhead box P3a (Foxp3a), the lineage-defining transcription factor of Tregs. Transcriptomic and in situ hybridization analysis reveal downreg
