Targeting the Gut Mucosal Immune System Using Nanomaterials.

The gastrointestinal (GI) tract is one the biggest mucosal surface in the body and one of the primary targets for the delivery of therapeutics, including immunotherapies. GI diseases, including, e.g., inflammatory bowel disease and intestinal infections such as cholera, pose a significant public health burden and are on the rise. Many of these diseases involve inflammatory processes that can be targeted by immune modulatory therapeutics. However, nonspecific targeting of inflammation systemically can lead to significant side effects. This can be avoided by locally targeting therapeutics to the

Mucosal immune responses following intestinal nematode infection.

In most natural environments, the large majority of mammals harbour parasitic helminths that often live as adults within the intestine for prolonged periods (1-2 years). Although these organisms have been eradicated to a large extent within westernized human populations, those living within rural areas of developing countries continue to suffer from high infection rates. Indeed, recent estimates indicate that approximately 2.5 billion people worldwide, mainly children, currently suffer from infection with intestinal helminths (also known as geohelminths and soil-transmitted helminths) . The mucosal immune system and its integration with the mammary glands.

Mucosal immunity reduces the need for elimination of penetrating exogenous antigens by proinflammatory systemic immunity. The adult gut mucosa contains some 80% of the body's activated B cells-differentiated to plasmablasts and plasma cells (PCs). Most mucosal PCs produce dimeric immunoglobulin A (IgA), which, along with pentameric immunoglobulin M (IgM), can be exported by secretory epithelia expressing the polymeric immunoglobulin receptor. Immune exclusion of antigens is performed mainly by secretory IgA in cooperation with innate defenses, but, in newborns and in IgA deficiency, secretory

Mucosal Immune Responses to Escherichia coli and Salmonella Infections.

The best-characterized mucosa-associated lymphoid tissue (MALT), and also the most relevant for this review, is the gastrointestinal-associated lymphoid tissue (GALT). The review reviews our understanding of the importance of mucosal immune responses in resisting infections caused by E. coli and Salmonella spp. It focuses on the major human E. coli infections and discusses whether antigen-specific mucosal immune responses are important for resistance against primary infection or reinfection by pathogenic E. coli. It analyzes human data on mucosal immunity against E. coli, a growing body of dat

Chemokine receptors and leukocyte trafficking in the mucosal immune system.

The CC chemokine receptors CCR6, CCR9, and CCR10 all contribute to the positioning of leukocytes at mucosal locations. Mucosal epithelial cells are major sources of the chemokine ligands for each of these receptors, although the pattern of expression of the individual ligands differs at distinct mucosal sites. CCR6 is expressed by most B cells, subsets of CD4 and CD8 memory T cells, and subsets of dendritic cells (DCs). Absence of CCR6 in mice leads to abnormal expansion of intestinal intraepithelial T cells and lamina propria T cells, smaller Peyer's patches, and defects in IgA-mediated respo

Compartmentalization of the mucosal immune responses to commensal intestinal bacteria.

Mammals coexist with a luxuriant load of bacteria in the lower intestine (up to 10(12) organisms/g of intestinal contents). Although these bacteria do not cause disease if they remain within the intestinal lumen, they contain abundant immunostimulatory molecules that trigger immunopathology if the bacteria penetrate the body in large numbers. The physical barrier consists only of a single epithelial cell layer with overlying mucus, but comparisons between animals kept in germ-free conditions and those colonized with bacteria show that bacteria induce both mucosal B cells and some T cell subset

Humans immunized with Streptococcus mutans antigens by mucosal routes.

Strategies aimed at the prevention of Streptococcus mutans infection and dental caries include mucosal immunization, which results in salivary anti-S. mutans responses. The purpose of this study was to evaluate the effectiveness of nasal vs. tonsillar immunization with S. mutans antigens in inducing salivary immune responses. Twenty-one adult subjects were immunized twice, within a seven-day interval, with a glucosyltransferase-enriched preparation (E-GTF) administered by nasal or tonsillar topical spray. Parotid saliva, nasal wash, and serum were collected prior to and at one- to two-week int

Mucosal BCG Vaccination and Immune Layering: Route-Dependent Programming of Immunity at the Respiratory Interface.

Mycobacterium bovis Bacille Calmette-Guérin (BCG), the only licensed vaccine against tuberculosis, provides inconsistent protection against pulmonary tuberculosis, reflecting an incomplete understanding of how vaccine-induced immunity is organized within tissues. Emerging evidence indicates that the route of vaccination is not merely a technical variable but a critical determinant of immune programming. Whereas parenteral BCG primarily elicits systemic immune responses, mucosal delivery reprograms immunity at the respiratory interface by promoting localized trained innate immunity, tissue-resi

Characterization of the immune roles of cathepsin L in turbot (Scophthalmus maximus L.) mucosal immunity.

Cathepsin L (CTSL) is one of the crucial enzymes in cathepsin family, which has been widely known for its involvement in the innate immunity. However, it still remains poorly understood how CTSL modulates the immune system of teleosts. In this study, we captured three cathepsin L genes (SmCTSL, SmCTSL.1 and SmCTSL1) from turbot (Scophthalmus maximus). The coding sequences of SmCTSL, SmCTSL.1 and SmCTSL1 are 1,026 bp, 1,005 bp and 1,017 bp in length and encode 341, 334 and 338 amino acids, respectively. In details, transcripts of CTSL genes share same domains as other CTSL genes, one signal pep

Humoral immune responses in periodontal disease may have mucosal and systemic immune features.

The humoral immune response, especially IgG and IgA, is considered to be protective in the pathogenesis of periodontal disease, but the precise mechanisms are still unknown. Immunoglobulins arriving at the periodontal lesion are from both systemic and local tissue sources. In order to understand better the local immunoglobulin production, we examined biopsy tissue from periodontitis lesions for the expression of IgM, IgG, IgA, IgE and in addition the IgG and IgA subclasses and J-chain by in situ hybridization. Tissues examined were superficial inflamed gingiva and the deeper granulation tissue

Intestinal permeability and the zonulin pathway: tight junctions, mucosal immunity, dietary modulators and clinical relevance.

The intestinal barrier separates the luminal content, including the gut microbiota, dietary antigens and toxins, from the mucosal immune system, and its integrity is essential for immune homeostasis. Loss of barrier control, commonly termed increased intestinal permeability or leaky gut, has been implicated in a broad spectrum of gastrointestinal, metabolic, autoimmune, neuropsychiatric, hepatic and cardiovascular conditions. Paracellular transport across the epithelium is governed by apical junctional complexes, of which tight junctions are the principal regulators, while the overlying mucus

Mechanisms and therapeutic prospects of DNA methylation-mucosal innate immunity crosstalk in inflammatory bowel disease.

Inflammatory bowel disease (IBD) comprises a group of chronic and relapsing intestinal inflammatory disorders whose pathogenesis and progression are closely associated with disruption of the intestinal mucosal barrier, dysregulated immune responses, and altered epigenetic regulation. The innate immune system is a central component of mucosal host defense and plays a pivotal role in pathogen recognition, inflammatory signal transduction, immune-cell functional regulation, and maintenance of barrier homeostasis. In recent years, DNA methylation has been increasingly recognized as an important me

A Thermosensitive Mucoadhesive Hydrogel for Nasal Influenza Vaccine Delivery Elicits Robust Systemic and Mucosal Immunity.

Influenza virus invades the host through the respiratory mucosa, making local immunity at the infection site critical for blocking transmission. However, the nasal mucus-cilia clearance barrier severely hinders antigen retention and limits the efficacy of mucosal vaccines. Here, we developed a thermosensitive hydrogel based on cationic chitosan (CS) for codelivery of hemagglutinin (HA) protein and CpG adjuvant (Gel-Pro-CpG). This platform remains a low viscosity liquid at 25°C for convenient nasal administration, and rapidly undergoes sol-gel transition upon contact with the nasal mucosa (∼34°

Impaired mucosal IgA response to SARS-CoV-2 in patients with inborn errors of immunity.

BACKGROUND: Patients with inborn errors of immunity (IEI) often exhibit impaired responses to vaccination and infection, yet their systemic and mucosal antibody dynamics against SARS-CoV-2 remain incompletely understood. METHODS: We investigated humoral immunity in 93 IEI patients recruited during the early phase of the pandemic, including pediatric patients with confirmed infection (n=64) and adult patients who received complete inactivated COVID-19 vaccination (n=29). Patients were classified as having primary antibody deficiency (PAD), combined immunodeficiency, or innate immune defects.
