Epigenetic programming via lysine-specific demethylase 1 regulates responses to immune checkpoint blockade.

Immune checkpoint blockade has revolutionized cancer therapy, but durable benefit remains limited by immune-cold tumor states, poor antigen presentation, T cell exhaustion, and adaptive resistance. Recent studies position lysine-specific demethylase 1 (LSD1) as a druggable epigenetic regulator that links these barriers across tumor and immune cells. Emerging evidence shows that targeting LSD1 can restore major histocompatibility complex class I antigen presentation, activate viral mimicry and interferon programs, modulate programmed death-ligand 1 in a context-dependent manner, influence CD8+

Barrier immune memory is programmed by intestinal epithelial cell presentation of cytosol-delivered bacterial antigens.

Antigen compartmentalization is known to influence CD4 and CD8 T cell recognition but its impact on immune responses to different antigens expressed by the same pathogen is poorly understood. By tracking the clonal CD4 T cell response to a single epitope shuttled between different virulence proteins of Citrobacter rodentium, we found that response magnitude and quality depended on antigen localization. Antigens retained within bacteria elicited limited responses, whereas antigens injected into colonocyte cytosol through a type III secretion system generated robust mucosal CD4 T cell responses

GNL3 Orchestrates AR Transcriptional Programs to Drive Castration-Resistant Prostate Cancer and Immune Evasion.

Androgen receptor (AR) signaling is a primary oncogenic driver of castration-resistant prostate cancer (CRPC), yet the mechanism remains incompletely understood. Through proteomic profiling of CRPC and primary PCa cells, we identify G Protein Nucleolar 3 (GNL3) as a novel AR coregulator. GNL3 physically interacts with AR, enhances its chromatin occupancy, and directly coactivates transcriptional programs that promote cell proliferation, including NEK2 and CDC20. Concurrently, GNL3 functions as a corepressor of immune-responsive genes such as CXCL10 and TAP1 via class I histone deacetylases (HD

Transcriptional immune programs underlying inadequate response to tumor necrosis factor inhibitors in ankylosing spondylitis.

Inadequate response to tumor necrosis factor inhibitors (TNFi) in ankylosing spondylitis (AS) remains common. As the immunoregulatory and immunometabolic alterations across TNFi inadequate responder (TNFi-IR) subsets remain unexplored, we characterized molecular profiles across distinct immune subsets in TNFi-IRs and identified signatures linked to TNFi-IR. We compared subset-specific expression in 16 TNFi-IRs and 13 TNFi responders (TNFi-Rs). Gene expression demonstrated fair-good discriminative power (area under the curve: 0.70-0.87). TNFi-IRs displayed distinct transcriptomic profiles and a

Maternal undernutrition in pregnancy and lactation programs impaired immune system development and function in male offspring.

This study examined the impact of maternal undernutrition during gestation and/or lactation on neonatal immune indices. From day 10 of gestation to term, pregnant rats received either ad libitum (AdLib) feeding or 50% food restriction (FR). After birth, pups were either nursed by their own dams or cross-fostered, resulting in three groups (n = 6 per group): a control group with AdLib feeding throughout (AdLib/AdLib), a group with FR only during gestation (FR/AdLib), and a group with FR during both gestation and lactation (FR/FR). At day of life 1 and at three weeks of age, spleen and thymus we

Platelet-driven immunothrombotic remodeling programs convert ectopic endometrium into fibrotic, immune-privileged lesions.

Endometriosis is a chronic, heterogenous disorder in which cyclical hemorrhage and repetitive repair generate ectopic implants that persist, recur, and drive pain and infertility; conventional frameworks emphasizing hormonal aberration and immune failure incompletely explain lesion fibrotic maturation and spatial heterogeneity. We synthesize human, experimental, and emerging spatial-omic evidence to advance a unifying mechanistic paradigm: cyclical bleeding seeds an immunothrombotic niche in which activated platelets and fibrin scaffolds organize immune cells and stromal responders, and platel

Microbial and metabolic signatures of humanized microbiome mice associate with Tlr2 expression and Th17 immune programming.

INTRODUCTION: The gut microbiota plays a critical role in immune system development. However, whether normal variation in healthy human microbiota alone can establish distinct baseline immune states that influences responses to disease and therapy remains unclear. METHODS: To address this, we employed a humanized microbiome (HuM) mouse model where fecal matter is transplanted from healthy human donors into gnotobiotic mice. Our previous studies found that HuM mouse lines have differential responses to glioma immunotherapy, so we sought to define how microbiota composition mediates host immuni

Mapping immune responses to TAK-003 dengue vaccine: immunogenicity across the clinical development program.

Despite deployment of vector control and prevention strategies, dengue remains a substantial threat to public health. Development of an effective vaccine against dengue has been challenging due to the need for simultaneous immunity against all four dengue virus serotypes, durable protection regardless of prior dengue exposure, and the complex epidemiological patterns of serotype co-circulation. As part of the clinical development of TAK-003, a live attenuated tetravalent dengue vaccine based on a DENV-2 backbone, extensive profiling of post-vaccination immune responses has been performed in pa

Context-Dependent Epithelial and Immune Programs Shape Intestinal Resilience or Vulnerability Following Prior Colitis.

BACKGROUND & AIMS: Prior intestinal inflammation can leave durable immune and epithelial alterations, yet how these changes influence responses to subsequent injury remains unclear. Infectious and sterile colitis share core features, including barrier disruption and cytokine secretion. We therefore investigated whether the nature of the initial inflammatory event shapes protection or susceptibility during later intestinal insult. METHODS: We used reciprocal mouse models of Citrobacter rodentium infection and dextran sodium sulphate-induced colitis to define how prior infectious vs sterile col

Mutation-Immune Crosstalk Contextualizes CAF-Associated ARPC1A Programs and Cisplatin Response in Gastric Cancer.

Cisplatin response in gastric cancer is shaped by malignant cell programs, stromal states, and mutation-immune crosstalk, but the CAF subtypes linked to platinum tolerance and their epithelial effectors remain incompletely defined. We integrated single-cell RNA sequencing, epithelial CNV-like inference, fibroblast reclustering, CellChat analysis, signature scoring, hdWGCNA, CAF-epithelial coupling, exploratory GSE14209 evaluation, TCGA-STAD mutation/copy-number contextualization, and DepMap CRISPR virtual knockout analysis, followed by experimental validation in AGS and HGC-27 cells. Metabolism-programming mRNA-lipid nanoparticles remodel the immune microenvironment to improve immunotherapy against MAFLD.

Metabolic dysfunction-associated fatty liver disease (MAFLD), a leading cause of hepatocellular carcinoma (HCC), poses a formidable therapeutic challenge because of the metabolic stress-induced aberrant immune microenvironment. However, no effective pharmacological therapies for the liver microenvironment remodeling in MAFLD are now available. Here, we developed a lipid nanoparticle (Def-LNP) that incorporates vitamin E-derived phosphatidylcholine (VEPC). Def-LNP effectively ameliorated the hepatic oxidative microenvironment to achieve sustained localized expression of target mRNA in hepatocyt

Impact of the indigenous rotavirus vaccine Rotavac in the Universal Immunization Program in India during 2016-2020.

In 2016, India introduced Rotavac (G9P[11]), an indigenous oral rotavirus vaccine administered at 6, 10 and 14 weeks of age through the Universal Immunization Program. Evaluating its effectiveness under routine programmatic conditions is critical, given the variable performance of rotavirus vaccines in low- and middle-income countries. Here we assessed Rotavac's real-world effectiveness and impact across 31 hospitals in 9 states between 2016 and 2020 using a test-negative case-control design. Overall, 24,624 children were enrolled in surveillance (62% male and 38% female). Of 8,372 children ag

Deficits in Long-Term Vaccine Immunity Among Childhood Cancer Survivors Despite Revaccination Programs.

BACKGROUND: Childhood cancer survivors (CCSs) often experience impaired humoral immunity because of cancer treatments that increase their susceptibility to vaccine-preventable diseases. This study aimed to assess the seroprevalence of tetanus and rubella antibodies in CCSs compared to healthy, age-matched controls. Additionally, we explored the impact of cancer treatments on vaccine-induced immunity, examined the extent of revaccination after treatment completion, and evaluated the effectiveness of revaccination on seroprevalence. METHODS: This retrospective study included 180 CCSs previously

Resumption of active recommendation of the human papillomavirus vaccine in Japan and future challenges for the National Immunization Program.

Japan's immunization program resumed proactively recommending the use of the human papillomavirus (HPV) vaccine nationwide in April 2022, after suspending this recommendation in June 2013. The promotion of catch-up vaccinations is an urgent issue to reduce the increase in cervical cancer and other cancers caused by low vaccination rates. In addition, the National Immunization Program still has issues to be considered, such as the adoption of the 9-valent vaccine, establishment of an appropriate number of vaccinations according to age, and routine immunization of males. There is a history of el

Prioritization of Vaccines for Inclusion into China's Expanded Program on Immunization: Evidence from Experts' Knowledge and Opinions.

BACKGROUND: Vaccine developers in China have made an increasing number of infectious diseases preventable through vaccination. An appropriate decision-making procedure is necessary for making wise decisions on whether to introduce new vaccines into the Expanded Program on Immunization (EPI). When there are several vaccines that could potentially be considered, a scientifically justifiable mechanism is needed for prioritizing and sequencing vaccines for consideration. METHODS: We used a modified Delphi technique (MDT) to develop and refine an indicator system to prioritize vaccines and make po

Immunologically programming the tumor microenvironment induces the pattern recognition receptor NLRC4-dependent antitumor immunity.

BACKGROUND: The efficacy of cancer immunotherapy can be limited by the poor immunogenicity of cancer and the immunosuppressive tumor microenvironment (TME). Immunologically programming the TME and creating an immune-inflamed tumor phenotype is critical for improving the immune-responsiveness of cancers. Here, we interrogate the immune modulator Flagrp170, engineered via incorporation of a pathogen-associated molecular pattern (ie, flagellin) into an immunostimulatory chaperone molecule, in transforming poorly immunogenic tumors and establishing a highly immunostimulatory milieu for immune augm
