[Evaluation of the effectiveness of various methods of the treatment of tick-borne encephalitis in the acute period].

To reveal the dependence of the efficacy of the treatment of tick-borne encephalitis (TBE) on the patient's HLA phenotype and to elaborate recommendations for individual treatment, 98 subjects who suffered TBE and who received different types of therapy in the acute period (isolated or combined use of specific homologous immunoglobulin and RNase) were subjected to immunogenetic examinations. The clinical follow-up and immunological examination of the subjects indicated made it possible to distinguish groups with a favorable and unfavourable outcome due to the treatment carried out in the acute

[Current diagnostic methods and therapeutic principles in various forms of puerperal endometritis].

A total of 55 puerperants+ with endometritis followed by spontaneous labours (26 females) and cesarean operation (29 females) were studied. A comprehensive study involved echography, hysteroscopy, bacteriological analysis of the uterine content, and histological examination of mucosal curettage of the corpus uteri. Three types of the clinical course of endometritis were identified: 1) in the presence of placental tissue residues; 2) with necrosis of delayed decidual tissue and lochia- or hematometra; and 3) a "pure" type of endometritis without abnormal tissues in the uterus. Uterine curettage

Kaposi's sarcoma in the acquired immunodeficiency syndrome.

Kaposi's sarcoma is the most common malignancy seen in association with AIDS. Although the pathogenesis of AIDS-KS has not been clearly established, the clinical course and prognosis are closely related to the patient's immune status, prior history of opportunistic infection, and hematologic status. Treatment methods include local or regional radiation therapy, cytotoxic chemotherapy, and interferon therapy. Evaluation of the results of clinical therapeutic studies in this tumor should consider its natural history and peculiar biologic behavior and associated complications of AIDS. Effective t

Targeted Agents in the Treatment of Indolent B-Cell Non-Hodgkin Lymphomas.

Targeted therapies continue to change the landscape of lymphoma treatment, resulting in improved therapy options and patient outcomes. Numerous agents are now approved for use in the indolent lymphomas and many others under development demonstrate significant promise. In this article, we review the landscape of targeted agents that apply to the indolent lymphomas, predominantly follicular lymphoma, lymphoplasmacytic lymphoma/Waldenstrom macroglobulinaemia and marginal zone lymphoma. The review covers small molecule inhibitors, immunomodulators and targeted immunotherapies, as well as presentin

[New drugs in the treatment of multiple myeloma].

Progress in the treatment of multiple myeloma in the last decade has been able to delay, but ultimately not to prevent, the development of resistances and most patients still die of the disease or its related complications. New drugs have been developed including new alkylating agents, proteasome inhibitors and immunomodulators but also monoclonal antibodies and drugs with new mechanisms of action. Hopefully, this new generation of targeted agents will improve the results of the initial therapy, avoid relapses and development of resistances and provide better and less toxic options for the rel

Scutellaria baicalensis Georgi as a potential therapeutic drug intervention in ulcerative colitis: Mechanisms of action and clinical trials.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease characterized by clinical symptoms of diarrhea and mucopurulent bloody stools, and its incidence is increasing globally. The etiology and pathogenesis of UC remain elusive. Current therapeutic approaches, including anti-inflammatory, immunosuppressive and immunomodulating agents, are often limited in efficacy and frequently associated with adverse drug reactions. Therefore, there is an urgent need to develop safer and more effective treatment strategies to address the limitations of existing therapies. Scutellaria baicalensis Geor

Methyl-supplemented nutrition delays the development of autoimmune disease in pristane-induced murine lupus.

The aetiology and progression of systemic lupus erythematosus (SLE) resulted from a complex sequence of events generated both from genetic and epigenetic processes. In the current research, the effect of methyl-supplemented nutrition on the development of SLE was studied in the pristane-induced mouse model of the disease. The results clearly demonstrated decreased anti-dsDNA antibody and proteinuria levels, modulation of cytokines and protected renal structures in the group of treated mice. An additional increase in the DNA methylation of mouse B lymphocytes was also observed. The beneficial e

Successful management of severe neuroinvasive eastern equine encephalitis.

BACKGROUND: Eastern Equine Encephalitis (EEE) virus is an arbovirus that mostly causes asymptomatic infection in humans; however, some people can develop a neuroinvasive infection associated with a high mortality. METHODS: We present a case of a patient with severe neuroinvasive EEE. RESULTS: A 21-year-old man initially presented with headache, fever, and vomiting and was found to have a neutrophilic pleocytosis in his cerebrospinal fluid. He eventually was diagnosed with EEE, treated with high-dose methylprednisolone and intravenous immunoglobulin. His course in the NeuroIntensive Care Unit

Systemic therapy of atopic dermatitis in children and adults.

Systemic therapy is required in patients with severe atopic dermatitis (AD) refractory to adequate topical therapy. The aim of a systemic therapy is the rapid and efficient improvement of skin symptoms and pruritus in acute exacerbation and/or the long-term control of severe chronic disease. A number of immunosuppressive and immunomodulating substances are available that may efficiently reduce skin inflammation and thus lead to a relief of symptoms including pruritus. The excellent effects of cyclosporine as short-term as well as maintenance therapy have been documented in several studies in c

Optic neuritis.

Demyelinating optic neuritis is the most common cause of unilateral painful visual loss in the United States. Although patients presenting with demyelinating optic neuritis have favorable long-term visual prognosis, optic neuritis is the initial clinical manifestation of multiple sclerosis in 20% of patients. The Optic Neuritis Treatment Trial (ONTT) has helped stratify the risk of developing multiple sclerosis after the first episode of optic neuritis based on abnormal findings on brain MRI. The ONTT also demonstrated that while initial treatment of optic neuritis with intravenous corticoster

Recent Patents on development of nucleic acid-based antiviral drugs against seasonal and pandemic influenza virus infections.

Influenza viruses are etiological agents of deadly flu that continue to pose global health threats, and have caused global pandemics that killed millions of people worldwide. The global crisis involving the avian H5N1 influenza provides compelling reasons to accelerate fast track development of novel antiviral drugs against the potential pandemic virus. The availability of neuraminidase inhibitors such as oseltamivir (tamiflu) improves our ability to defend against influenza viruses, but the incidences of tamiflu-resistance are rising rapidly. Nucleic acid-based antiviral drugs are promising c

Therapeutic potential of macrolide immunosuppressants in dermatology.

Dermatologists are frequently presented with inflammatory dermatoses that are responsive to treatment with immunomodulating drugs. Corticosteroids, particularly when applied topically, have been the mainstay of treatment in the past. Their undoubted efficacy, however, has been undermined by problems with repeated use including tachyphylaxis and side effects such as skin atrophy and hypertension. Macrolide immunosuppressive drugs, originally used for prophylaxis of organ transplant rejection, have been shown to be effective in the treatment of inflammatory dermatoses. The original drugs used in

Enhancement of resistance to murine osteogenic sarcoma in vivo by an extract of Brucella abortus (Bru-Pel): association with activation of reticuloendothelial system macrophages.

The administration of an aqueous-ether extracted residue of Brucella abortus (Bru-Pel) inhibits development of transplanted osteogenic sarcomas in mice as evidenced by a decrease in mortality. At least one mechanism through which Bru-Pel modulates host resistance is activation of macrophages of the reticuloendothelial system. Peritoneal macrophages harvested from mice receiving Bru-Pel were cytotoxic for osteogenic sarcoma cells in vitro, limited the replication of vaccinia virus in cell cultures, and demonstrated enhanced emittance of chemiluminescence during phagocytosis of zymosan particles

[Pharmacological immunomodulation in sepsis].

Sepsis is defined as a dysregulated host response to infection; it is one of the leading causes of death in intensive care units worldwide. Immunological dysregulation plays a central role in the pathophysiology of sepsis and thus represents a promising therapeutic target for its treatment. However, the immune response is complex, and hyperinflammation and immune paralysis can occur simultaneously or sequentially, in different compartments, and with individually variable courses. Although a broad spectrum of immunomodulatory substances is available in principle, clinical trials have long yield

Immunomodulatory effects of medicinal and edible plants in autoimmune diseases: mechanisms and therapeutic potential.

Autoimmune diseases (ADs) and related immune-mediated inflammatory disorders are characterized by loss of immune tolerance, persistent inflammatory activation, immune-cell imbalance, oxidative stress, and tissue-specific injury. Although conventional immunomodulatory therapies remain central to disease management, their long-term use may be limited by adverse effects, variable treatment responses, and high costs. Medicinal and edible plants (MEPs) provide a diverse source of bioactive compounds with potential immunoregulatory activity. This narrative review summarizes the immunological basis o

New antimycobacterial agents.

Development of new antimycobacterial agents has gained impetus from the frequent occurrence of disseminated M. avium complex infections associated with the AIDS epidemic and the resurgence of tuberculosis. Promising new agents are being developed by modification of existing antimycobacterial agents (for example, aminoglycosides, macrolides, beta-lactams, and rifamycins) and by development of new therapeutic classes of drugs (for example, 4-quinolones). In addition, new drug delivery systems (liposome encapsulation) and immunomodulators may prove to be clinically useful in the treatment of some

Profiling Localized Immunomodulation and Drug Biodistribution within a Subcutaneous Vascularized Niche for Cell Transplantation.

Systemic immunosuppression remains essential for preventing allogeneic transplant rejection, but its chronic use causes substantial toxicity. Conceptually, local, site-specific immunomodulation offers a promising alternative, yet comparative mechanistic insight into how immunosuppressants behave when delivered directly to the graft is lacking. Here, we leveraged the Neovascularized Implantable Cell Homing and Encapsulation (NICHE) device, a subcutaneous, vascularized cell-encapsulation platform, as a spatially defined and reproducible model to study local immunomodulation in allogeneic islet t

[Muramyldipeptide - based compounds in current medicine: focus on glucosaminylmuramyl dipeptide].

The role of immune mechanisms in the pathogenesis of almost all human diseases shown in recent decades, increase in antibiotic resistance and secondary immunodeficiency, aging of the population and widespread use of immunosuppressive drugs and procedures suggest a wider use of immunomodulators in current clinical practice, but the use of most of them limits the lack of knowledge. The most promising compounds for the development as immunomodulating agents and adjuvants for a wide range of vaccines are low molecular weight fragments of peptidoglycan - muramylpeptides. The article describes the m
