Vaccine-Associated Recombination and Virulence Heterogeneity of GI-19 (QX Type) Infectious Bronchitis Virus in Eastern and Southern China, 2024-2025.

Infectious bronchitis (IB) virus (IBV) remains a major pathogen threatening the poultry industry. Its rapid mutation and recombination continuously generate variants that disseminate worldwide. Between May 2024 and February 2025, 49 field strains were isolated from chickens vaccinated with live-attenuated IBV vaccines (H120, 4/91, or QXL87) in four Chinese provinces (Jiangsu, Anhui, Shandong, and Guangdong). Based on full-length S1 gene sequencing, all isolates were classified into genotype GI, including lineages GI-13 and GI-19. Phylogenetic analysis revealed that GI-19 (QX-type) comprised as

Emerging GI-23 infectious bronchitis virus in vaccinated broiler chickens: detection and molecular characterization during a respiratory outbreak in Southern Brazil.

Infectious Bronchitis Virus (IBV) is an avian coronavirus that significantly impacts poultry production worldwide. Analysis of the complete S1 gene has classified IBV into genotypes GI to GIX and more than 40 lineages to date. This study aimed to investigate whether a new IBV variant was involved in respiratory disease outbreaks observed in 2021 in vaccinated commercial broiler flocks in Paraná State, Brazil. Day-old chicks in all affected flocks had received mass IBV vaccination at the hatchery with a combination of two live-attenuated vaccines, specifically the predominant lineages GI-1 (Mas

Regulatory T cell infiltration precedes early events associated with persistent infectious bronchitis virus (IBV) infection in the cecal tonsils of chickens.

Infectious bronchitis (IB) is a contagious respiratory disease in chickens caused by infectious bronchitis virus (IBV). While initially affecting the respiratory tract, IBV has evolved to infect other body systems. Notably, the Delmarva (DMV)/1639 genotype of IBV has been shown to persist in the cecal tonsils (CT), potentially facilitating viral transmission to naïve birds. This study aimed to uncover the early immunological mechanisms leading up to IBV persistence in the CT, compared to the spleen and to evaluate the persistence patterns among different genotypes of IBV. An animal experiment

Replication of infectious bronchitis virus (IBV) Delmarva (DMV)/1639 variant in primary and secondary lymphoid organs leads to immunosuppression in chickens.

Infectious bronchitis virus (IBV) that primarily causes respiratory infection in chickens, disseminate to multiple body systems leading to pathology, results in economic losses to poultry industry. IBV replicates in the bursa of Fabricius (BF), Harderian gland (HG), cecal tonsils (CT), and spleen. The objective of this study was to investigate the immunosuppressive effect of IBV Delmarva (DMV/1639) variant in chickens. Specific pathogen free chickens were infected with the IBV DMV/1639 variant while maintaining an age-matched uninfected control group. At predetermined time points, subsets of t

Viral subpopulation variability in different batches of Infectious bronchitis virus (IBV) vaccines based on GI-23 lineage: Implications for the field.

The control of infectious bronchitis (IB) is largely based on routine vaccine administration, often using live-attenuated vaccines. However, their capability to replicate and be transmitted among animals and farms implies significant risks. The detection of strains genetically related to vaccines complicates the diagnostic process and understanding of the viral molecular epidemiology. Moreover, reversion to virulence and associated clinical outbreaks can occur although the underlying mechanism are often unknown. In the present study, three vaccine vials, based on IBV GI-23 lineage (also known

Efficacy of infectious bronchitis virus vaccinations in the field: association between the alpha-IBV IgM response, protection and vaccine application parameters.

Infectious bronchitis virus (IBV) is, in spite of vaccination, still a major cause of respiratory problems in broilers and of poor egg production in breeders and layers in many parts of the world. A possible cause of the insufficient protection induced by vaccination is an inadequate application of the vaccine. This paper reports the results of two field studies. In the first, the results of the alpha-IBV IgM enzyme-linked immunosorbent assay (ELISA) on post-vaccination sera were compared with the efficacy of the IBV vaccination against homologous challenge of the same broilers. The results sh

Identification and fine mapping of a novel linear B-cell epitope within the S1-CTD of the QX-type infectious bronchitis virus.

Infectious bronchitis virus (IBV) causes severe economic losses to the poultry industry. The IBV S1-CTD mediates receptor binding and serves as a key target for antibody recognition. In this study, the S1-CTD protein of the QX-type IBV QXL87 strain was expressed in HEK293F cells. Four monoclonal antibodies (10H5, 11D11, 12H7, and 13H1) were generated via hybridoma technology and characterized by indirect ELISA, Western blotting, and immunofluorescence assay; variable-region sequencing showed that the four mAbs are clonally related and recognize the same antigenic region. Co-infections of chickens with avian influenza virus H9N2 and Moroccan Italy 02 infectious bronchitis virus: effect on pathogenesis and protection conferred by different vaccination programmes.

Since the emergence of low pathogenic avian influenza (LPAI) H9N2 viruses in Morocco in 2016, severe respiratory problems have been encountered in the field. Infectious bronchitis virus (IBV) is often detected together with H9N2, suggesting disease exacerbation in cases of co-infections. This hypothesis was therefore tested and confirmed in laboratory conditions using specific-pathogen-free chickens. Most common field vaccine programmes were then tested to compare their efficacies against these two co-infecting agents. IBV γCoV/chicken/Morocco/I38/2014 (Mor-IT02) and LPAI virus A/chicken/Moroc

Recombinant infectious bronchitis virus (IBV) H120 vaccine strain expressing the hemagglutinin-neuraminidase (HN) protein of Newcastle disease virus (NDV) protects chickens against IBV and NDV challenge.

Infectious bronchitis (IB) and Newcastle disease (ND) are common viral diseases of chickens, which are caused by infectious bronchitis virus (IBV) and Newcastle disease virus (NDV), respectively. Vaccination with live attenuated strains of IBV-H120 and NDV-LaSota are important for the control of IB and ND. However, conventional live attenuated vaccines are expensive and result in the inability to differentiate between infected and vaccinated chickens. Therefore, there is an urgent need to develop new efficacious vaccines. In this study, using a previously established reverse genetics system, w

Two opposite abilities of the infectious bronchitis virus helicase Nsp13: separating the duplex and promoting the annealing of single-stranded nucleic acid.

Genome replication is a key step in the coronavirus life cycle and requires the involvement of a range of virally encoded non-structural proteins. The non-structural protein 13 (Nsp13) of coronaviruses is a highly conserved helicase and is considered an ideal drug target. However, the activity characteristics of the helicase Nsp13 of the infectious bronchitis virus (IBV) remain unclear. In this study, we expressed and biochemically characterized the purified recombinant IBV Nsp13 and found that IBV Nsp13 was able to unwind duplex substrates in a 5'-to-3' direction by using the energy from the

Significant Increase of Cinnamic Acid in Metabolites of Chicks Infected with Infectious Bronchitis Virus and Its Remarkable Antiviral Effects In Vitro and In Vivo.

Avian infectious bronchitis virus (IBV) infection has caused significant economic losses to the poultry industry. Unfortunately, there is currently no effective cure for this disease. Understanding the pathogenic mechanism is crucial for the treatment of the disease. Studying the pathogenic mechanism of IBV based on metabolomics analysis is helpful for identifying antiviral drugs. However, studies on metabolomics analysis of IBV infection have been relatively limited, particularly without metabolomics analysis in sera after IBV infection. In this study, 17-day-old SPF chicks were infected with

The modulatory effect of carvacrol on viral shedding titer and acute phase response in broiler chickens experimentally infected with infectious bronchitis virus.

Infectious bronchitis virus (IBV) is one of the major respiratory diseases of broiler causing huge economic losses. The inability to control IBV using different vaccination programs owing to the high mutation rate and recombination ability of the RNA genome generates IBV variants. This study was designed to give a specific perspective of carvacrol effect on early immune response, viral shedding titer, oxidative stress, serum biochemical parameters and clinical consequences in broilers experimentally infected by IBV. One hundred and twenty-one-day old commercial broiler chicks were equally divi

Evaluation of the Efficiency of Commercial Vaccines Against Infectious Bronchitis Virus (IBV) Belonging to the GI-16 Lineage Isolated in an Argentinean Outbreak.

In this study we evaluated the effectiveness of adding serotype 793B vaccine to an immunization program in order to control the infectious bronchitis virus (IBV) GI-16 lineage. Therefore, two different experiments were performed. First, a virus cross-neutralization test was carried out, which indicated that neither the Massachusetts (Mass) nor 793B serotypes are antigenically related to the field isolate A13 (GI-16). We also performed a challenge trial to evaluate if the Mass/793B combination is more efficient than Mass/Connecticut (Conn) to protect chickens against the Argentinian variant A13

Emerging infectious bronchitis virus (IBV) in Egypt: Evidence for an evolutionary advantage of a new S1 variant with a unique gene 3ab constellation.

Infectious bronchitis virus (IBV), a gamma-coronavirus, causes infectious bronchitis (IB), a major respiratory disease of chicken. Its high mutation rate in conjunction with recombination of the RNA genome constantly creates IBV variants that are difficult to control by currently available vaccines. In this study, we addressed the question whether small-scale holdings might harbor IBV variants that serve as a reservoir for newly emerging variants. Egyptian IBV isolate EGY/NR725/2016 (NR725/16) from a small-scale broiler farm was assigned to genotype I, clade 23 (S1:GI-23), based on partial S1

Combined adjuvant effect of ginseng stem-leaf saponins and selenium on immune responses to a live bivalent vaccine of Newcastle disease virus and infectious bronchitis virus in chickens.

Vaccination with a live bivalent vaccine of Newcastle disease virus (NDV) and infectious bronchitis virus (IBV) is a routine practice in poultry industry in China. This study was designed to evaluate ginseng stem-leaf saponins (GSLS) in combination with selenium (Se) for their adjuvant effect on the immune response to vaccination against NDV and IBV in chickens. A live bivalent vaccine of NDV and IBV was diluted in saline solution containing GSLS or Se or both and used to immunize chickens via a intraocular-and-intranasal route. Results showed that GSLS promoted significantly higher NDV- and I

Novel Taiwan-I-type infectious bronchitis virus: First evidence of testicular atrophy and efficacious protection with strain-matched oil-emulsified inactivated vaccine.

The Taiwan-I-type infectious bronchitis virus (IBV) has become one of the most dominant and threatening genotypes circulating in China poultry farms. In this study, we characterized the Taiwan-I-type IBV strain GX-NN200723 and evaluated its pathogenicity and vaccine potential. Phylogenetic and recombination analyses revealed that the strain shared the highest nucleotide similarity with the vaccine strain QXL87 and originated from a recombination event between CK/CH/LSC-99I (major parent) and TW2575/98 (I) (minor parent) within the S1 gene region. Notably, epitope mapping showed significant dif
